Assisted reproductive technologies play a major role in the cattle industry. An increase in the use of in vitro-derived embryos is currently being seen around the globe. But the efficiency and quality of the in vitro-derived embryos are substandard when compared to the in vivo production. Different protocols have been designed to overcome this issue, one of those being the use of reproductive fluids as supplementation to embryo culture media. In this study, in vitro-derived calves produced with reproductive fluids added to their embryo production protocol were followed for the first year of life pairwise with their in vivo control, produced by artificial insemination (AI), and their in vitro control, produced with standard supplementation in embryo production. The objective was to assess if any differences could be found in terms of growth and development as well as hematological and biochemical analytes between the different systems. All the analysed variables (physical, hematological, and biochemical) were within physiological range and very similar between calves throughout the entire experiment. However, differences were more evident between calves derived from standard in vitro production and AI. We concluded that the use of reproductive fluids as a supplementation to the embryo culture media results in calves with closer growth and development patterns to those born by AI than the use of bovine serum albumin as supplementation.

AbstractThe present study examined differences in adults’ spatial-scaling abilities across three perceptual conditions: (1) visual, (2) haptic, and (3) visual and haptic. Participants were instructed to encode the position of a convex target presented in a simple map without a time limit. Immediately after encoding the map, participants were presented with a referent space and asked to place a disc at the same location from memory. All spaces were designed as tactile graphics. Positions of targets varied along the horizontal dimension. The referent space was constant in size while sizes of maps were systematically varied, resulting in three scaling factor conditions: 1:4, 1:2, 1:1. Sixty adults participated in the study (M = 21.18; SD = 1.05). One-third of them was blindfolded throughout the entire experiment (haptic condition). The second group of participants was allowed to see the graphics (visual condition); the third group were instructed to see and touch the graphics (bimodal condition). An analysis of participants’ absolute errors showed that participants produced larger errors in the haptic condition as opposed to the visual and bimodal conditions. There was also a significant interaction effect between scaling factor and perceptual condition. In the visual and bimodal conditions, results showed a linear increase in errors with higher scaling factors (which may suggest that adults adopted mental transformation strategies during the spatial scaling process), whereas, in the haptic condition, this relation was quadratic. Findings imply that adults’ spatial-scaling performance decreases when visual information is not available.

Hyperspectral imaging offers enormous potential for measuring grape composition with a high degree of representativity, allowing all exposed grapes from the cluster to be examined non-destructively. On-the-go hyperspectral images were acquired using a push broom hyperspectral camera (400–100 nm) that was mounted in the front part of a motorized platform moving at 5 km/h in a commercial Tempranillo vineyard in La Rioja, Spain. Measurements were collected on three dates during grape ripening in 2018 on the east side of the canopy, which was defoliated in the basal fruiting zone. A total of 144 grape clusters were measured for Total soluble solids (TSS), Titratable acidity (TA), pH, Tartaric and Malic acid, Anthocyanins and Total polyphenols, using standard wet chemistry reference methods, throughout the entire experiment. Partial Least Squares (PLS) regression was used to build calibration, cross validation and prediction models for the grape composition parameters. The best performances returned determination coefficients values of external validation (R2p) of 0.82 for TSS, 0.81 for Titratable acidity, 0.61 for pH, 0.62 for Tartaric acid, 0.84 for Malic acid, 0.88 for Anthocyanins and 0.55 for Total polyphenols. The promising results exposed in this work disclosed a notable methodology on-the-go for the non-destructive, in-field assessment of grape quality composition parameters along the ripening period.

Sulfur nutrition is a critical part of proper crop growth and development. In our study, biomass yields (BY) and S uptake were investigated on long-term maize monoculture on haplic luvisol soil during the 23 years of this trial, as well as changes in water extractable (Sw), adsorbed (Sads), mineral (Sav), and pseudo-total S (St) fractions. Treatments used in this study are: (1) Control (Cont); (2) ammonium sulfate (AS); (3) urea and ammonium nitrate (UAN); (4) UAN + phosphorus and potassium (UAN + PK); (5) UAN + phosphorus, magnesium, sulfur (UAN + PMgS); and (6) Fallow. Recently, the Mehlich 3 method started to be used in the Czech Republic to determine content of plant available S. Using this method, it was found that the content of S extracted by Mehlich 3 (SM3) closely correlates to Sav in both topsoil and subsoil (r = 0.958 in 1997 and 0.990 in 2019, both at p < 0.001). We also found that, on average, during the entire experiment, all treatments had increased yields over Cont (135–147%) and increased S uptake (291, 192, 180, and 246% of Cont for AS, UAN, UAN + PK, and UAN + PMgS, respectively). Examining the changes from 1997 to 2019 in topsoil (0–30 cm depth), we discovered a decrease of S content in Sw, Sads, Sav, and St fractions on all treatments to an average of 34.6%, 65.8%, 42.2%, and 78.6% of their initial values. The exception was AS treatment, which doubled its initial content in mineral fractions and maintained the same levels of St, and which we attribute to the very high dose of S on this treatment (142 kg ha−1 year−1). Using the simple balance method, AS and UAN + PMgS treatments lost 142.2 and 95.3 kg S ha−1 year−1 to other sinks, except plant uptake, from the entire soil profile (0–60 cm) during 23 years of experiment. Other treatments also show significant losses with the exception of Fallow. Given these results, it is clear that content of sulfur in soil is generally decreasing and attention should be paid mainly towards minimizing of its losses.

The objective of this study was to determine the influence of phytogenic product-supplemented, organic acid-supplemented, and prebiotic-supplemented diets on the production results, antioxidative status, and selected welfare indices in broiler chickens. A total of 1155 one-day old male Ross 308 broilers were randomly assigned to one of three treatment groups: Group C, no additives; Group A, supplemented with phytogenic supplement (50% hop); and Group P, supplemented with 65% organic acids and their salts, and 30% prebiotic complex. Health condition and production results were monitored during the entire experiment. After 42 days, 10 birds from each dietary treatment group were selected for blood sampling and slaughter analysis. The results obtained revealed that over the whole feeding period, none of the investigated additives significantly affected broiler performance indices. However, feeding the birds treatment-A increased the relative abundance of Bifidobacterium in caecal digesta compared to the other treatments, whereas feeding treatment-P increased the relative abundance of Lactobacillus compared to the control treatment. Overall, treatment-A was more effective at increasing relative abundance of Clostridia in birds at 42 days of age than treatment-P. Finally, there were no changes in blood levels of antioxidant indices or liver function indicators.

The purpose of this study was to explore the feasibility of a urination management system by developing a smart band-based algorithm that recognizes the urination interval of women. We designed a device that recognizes the time and interval of urination based on the patient’s specific posture and posture changes. The technology used for recognition applied the Radial Basis Function kernel-based Support Vector Machine, a teaching and learning method that facilitates multidimensional analysis by simultaneously judging the characteristics of complex learning data. In order to evaluate the performance of the proposed recognition technique, we compared actual urination and device-sensed urination. An experiment was performed to evaluate the performance of the recognition technology proposed in this study. The efficacy of smart band monitoring urination was evaluated in 10 female patients without urination problems. The entire experiment was performed over a total of 3 days. The average age of the participants was 28.73 years (26–34 years), and there were no signs of dysuria. The final accuracy of the algorithm was calculated based on clinical guidelines for urologists. The experiment showed a high average accuracy of 91.0%, proving the robustness of the proposed algorithm. This urination behavior recognition technique shows high accuracy and can be applied in clinical settings to characterize urination patterns in female patients. As wearable devices develop and become more common, algorithms that detect specific sequential body movement patterns that reflect specific physiological behaviors could become a new methodology to study human physiological behavior.

This paper studies polyphenols and flavonoids level evolution in Juglans regia L. leaves and mesocarp during the growing season, as well as compounds evolution in the walnut kernel for fully mature picked nuts, stored for eight months at room temperature. Both compounds, polyphenols and flavonoids were significantly influenced by cultivar, harvest moment, and, respectively, by the storage time. The highest content of polyphenols was found in ′Jupânești′ cultivar’s leaves, ′Vina′ cultivar’s kernel, and ′Franquette′ cv. mesocarp. Also, nut mesocarp and leaves samples harvested in July and nut kernel stored till next March contained maximum polyphenols levels. The highest flavonoid content was determined for ′Vina′ cultivar’s leaves, ′Franquette′ cultivar’s fruit mesocarp, ′Vina′’s kernel, leaves and mesocarp sampled in July and nut kernel stored until March. For the entire experiment, maximum polyphenols level was found in ′Jupânești′ cultivar’s leaves harvested in July. Flavonoids reached their highest level in case of ′Franquette′ kernel stored until March. The three studied cultivars: ′Jupânești′, ′Vina′ and ′Franquette′ can be valuable sources of phenolic compounds.

This paper proposed a liquid level measurement and classification system based on a fiber Bragg grating (FBG) temperature sensor array. For the oil classification, the fluids were dichotomized into oil and nonoil, i.e., water and emulsion. Due to the low variability of the classes, the random forest (RF) algorithm was chosen for the classification. Three different fluids, namely water, mineral oil, and silicone oil (Kryo 51), were identified by three FBGs located at 21.5 cm, 10.5 cm, and 3 cm from the bottom. The fluids were heated by a Peltier device placed at the bottom of the beaker and maintained at a temperature of 318.15 K during the entire experiment. The fluid identification by the RF algorithm achieved an accuracy of 100%. An average root mean squared error (RMSE) of 0.2603 cm, with a maximum RMSE lower than 0.4 cm, was obtained in the fluid level measurement also using the RF algorithm. Thus, the proposed method is a feasible tool for fluid identification and level estimation under temperature variation conditions and provides important benefits in practical applications due to its easy assembly and straightforward operation.

Researchers have conducted in-depth research on DNA methylation mechanism, which is related to various diseases such as deficiency of imprinted gene and occurrence of tumors. This study provides a novel rapid quantitative detection assay and real-time fluorescence recombinase-aided amplification assay (RAA) for DNA methylation. Firstly, specific sequence of methylation genes was chosen and primers and fluorogenic probe for RAA experiment were designed and synthesized. Lastly, these amplification products were proven by sequencing and analysis. Results showed that the amplification efficiency and template concentration of RAA had linear dependent (R2 > 95%) when the concentration range was 4.64×108 copies/μL˜4.64×104 copies/μL. The test assay can also detect positive samples when the template concentration is below 4.64×104 copies/μL. Remarkably, the entire experiment process only takes 15–20 minutes, so it is beneficial for rapid bedside simple screening of some special DNA methylation sites, such as detection of resistance genes. In a word, this method has very great potential for diseases with DNA methylation in clinical settings, especially if methylation analysis needs to be done quickly and easily.

This study demonstrated that a microbial community dominated by fungi can be selected and maintained in the long-term under non-sterile conditions, in a pilot-scale packed-bed reactor fed with tannery wastewater. During the start-up phase, the reactor, filled with 0.6 m3 of polyurethane foam cubes, was inoculated with a pure culture of Aspergillus tubingensis and Quebracho tannin, a recalcitrant compound widely used by tannery industry, was used as sole carbon source in the feeding. During the start-up, fungi grew attached as biofilm in carriers that filled the packed-bed reactor. Subsequently, the reactor was tested for the removal of chemical oxygen demand (COD) from an exhaust tanning bath collected from tanneries. The entire experiment lasted 121 days and average removals of 29% and 23% of COD and dissolved organic carbon (DOC) from the tannins bath were achieved, respectively. The evolution of the microbial consortium (bacteria and fungi) was described through biomolecular analyses along the experiment and also developed as a function of the size of the support media.
