This study aimed to evaluate the changes in aromatic components and other chemical properties of Tainong mango during fruit development, ripening, and storage. As the volatiles of Tainong mango and their related molecular mechanisms remain unclear, volatile profile, metabonomics, and transcriptome analyses were applied to investigate the molecular determinants of the synthesis of aroma components in mango during fruit development and storage. Total acids, total sugar, total carotenoids, enzyme activities of the mango pulp samples were also determined. Volatile components of the mango pulp samples were identified using a gas chromatography-mass spectrometric method. Ribonucleic acid (RNA) sequences of the samples were analyzed by real-time polymerase chain reaction. The results showed that 181 volatiles were isolated and identified in the fruit at seven stages. Compared to the other stages, mango collected on day 8 and day 12 had higher concentrations of 17 volatile components, especially (E,Z)-2,6-nonadienal, 53384 transcripts were also detected through RNA sequencing. The differentially expressed genes analyses included catalytic activity, transferase activity, adenosine diphosphate binding, transcription factor activity, and oxidoreductase activity. α-Pinene content and expression of the differentially expressed genes involved in terpenoid metabolism and enzyme activities in the terpenoid metabolic pathways gradually increased during the maturity of the fruit, and had maximum values at day 8 of storage. Moreover, the integrative analyses revealed potential molecular insights of mango development and aroma formation in the fruit.

Strawberry (Fragaria × ananassa) is an ideal plant for fruit development and ripening research due to the rapid substantial changes in fruit color, aroma, taste, and softening. To gain deeper insights into the genes that play a central regulatory role in strawberry fruit development and ripening characteristics, transcriptome profiling was performed for the large green fruit, white fruit, turning fruit, and red fruit stages of strawberry. A total of 6,608 differentially expressed genes (DEGs) with 2,643 up-regulated and 3,965 down-regulated genes were identified in the fruit development and ripening process. The DEGs related to fruit flavonoid biosynthesis, starch and sucrose biosynthesis, the citrate cycle, and cell-wall modification enzymes played important roles in the fruit development and ripening process. Particularly, some candidate genes related to the ubiquitin mediated proteolysis pathway and MADS-box were confirmed to be involved in fruit development and ripening according to their possible regulatory functions. A total of fiveubiquitin-conjugating enzymesand 10MADS-box transcription factorswere differentially expressed between the four fruit ripening stages. The expression levels of DEGs relating to color, aroma, taste, and softening of fruit were confirmed by quantitative real-time polymerase chain reaction. Our study provides important insights into the complicated regulatory mechanism underlying the fruit ripening characteristics inFragaria × ananassa.

Red and blue light have great effects on physiological processes and growth of plants. In this experiment, we investigated the physiological and growth response of pepper (Capsicum annuum L.) to supplementary red:blue (4:1) light for 1 h (T1), 3 h (T2), and 5 h (T3), and the full-spectrum light-emitting diodes, LEDs, as control (CK). Thirty-day-old seedlings were grown under these treatments for 20 days in a climate-controlled room before data measurement. The results showed that the light treatments significantly (p < 0.05) affected the photosynthesis and growth indexes as well as gene expression in the pepper seedlings. Plants under T2 generally had better performance in terms of seedling growth. A total of 124, 1283, and 1091 differentially expressed genes were found in CK vs. T1, CK vs. T2, and CK vs. T3, respectively. Among the treatments, T2 in comparison with CK had 705 upregulated and 578 downregulated differentially expressed genes (DEGs). We also found that CPRF2, Paggis, HLIPS, GIGANTEA, LSH1, and FTSH genes were expressed differently under the various light treatments. Based on GeneOntology (GO) enrichment analysis, DEGs were significantly enriched on 15 GO terms of which xyloglucan:xyloglucosyl transferase activity and apoplastic, cellular polysaccharide metabolic, and cellular carbohydrate metabolic processes were closely related to light responses. A total of 96 genes that are related to plant–pathogen interaction, zeatin biosynthesis, plant hormone signal transduction, and wax/cutin/suberine biosynthesis which are involved in the pathway of light reaction in plants were significantly enriched in T2 plants compared with plants under CK. The application of red:blue light at 4:1 for 3 h improved the growth of pepper seedlings better than the other treatments and this can be tested under the Chinese Solar Greenhouse condition.

BackgroundThe oriental melon (Cucumis meloL. var.makuwa) is one of the most important cultivated cucurbits grown widely in Korea, Japan, and northern China. It is cultivated because its fruit has a sweet aromatic flavor and is rich in soluble sugars, organic acids, minerals, and vitamins. In order to elucidate the genetic and molecular basis of the developmental changes that determine size, color, and sugar contents of the fruit, we performedde novotranscriptome sequencing to analyze the genes expressed during fruit development.ResultsWe identified a total of 47,666 of representative loci from 100,875 transcripts and functionally annotated 33,963 of the loci based on orthologs inArabidopsis thaliana. Among those loci, we identified 5,173 differentially expressed genes, which were classified into 14 clusters base on the modulation of their expression patterns. The expression patterns suggested that the differentially expressed genes were related to fruit development and maturation through diverse metabolic pathways. Analyses based on gene set enrichment and the pathways described in the Kyoto Encyclopedia of Genes and Genomes suggested that the expression of genes involved in starch and sucrose metabolism and carotenoid biosynthesis were regulated dynamically during fruit development and subsequent maturation.ConclusionOur results provide the gene expression patterns related to different stages of fruit development and maturation in the oriental melon. The expression patterns give clues about important regulatory mechanisms, especially those involving starch, sugar, and carotenoid biosynthesis, in the development of the oriental melon fruit.

Peroxidase (POD), polyphenoloxidase (PPO), and superoxide dismutase (SOD) activities were measured during several phases of bud development, ripening, and storage of `Oregon Spur Red Delicious' (Malus domestica Borkh) apple fruit. POD, PPO, and SOD activities were significantly altered as a result of bud development. At the early stages of fruit development, the activities of these enzymes were significantly higher, then declined as the fruit increased in size. The activities of these enzymes declined as the fruit reached full maturity. Fruits harvested 3 weeks before commercial maturity and stored for up to 6 months have higher enzyme activities than fruits harvested at or beyond commercial maturity. The activities of these enzymes in relation to fruit firmness, soluble solids, and ethylene will be determined and related to overall fruit quality.

Abstract Background: Fruit flesh color in watermelon (Citrullus lanatus) is a great index for evaluation of the appearance quality and a key contributor influencing consumers' preferences, but the molecular mechanism of this intricate trait remain largely unknown. Here, the carotenoids and transcriptome dynamics during the fruit development of cultivated watermelon with five different flesh colors were analyzed.Results: A total of 13 carotenoids and 16781 differentially expressed genes (DEGs) including 1295 transcription factors (TFs) were detected in five watermelon genotypes during the fruit development. The comprehensive accumulation patterns of carotenoids were closely related to flesh color. A number of potential structural genes and transcription factors were found to be associated with the carotenoid biosynthesis pathway using comparative transcriptome analysis. The differentially expressed genes were divided into six subclusters and distributed in different GO terms and metabolic pathways. Furthermore, we performed weighted gene co-expression network analysis and predicted hub genes in six main modules determining carotenoid contents. Cla018406 (a chaperone protein dnaJ-like protein) may be a candidate gene for β-carotene accumulation and highly expressed in orange flesh-colored fruit. Cla007686 (a zinc finger CCCH domain-containing protein) was highly expressed in the red flesh-colored watermelon, maybe a key regulator of lycopene accumulation. Cla003760 (membrane protein) and Cla021635 (photosystem I reaction center subunit II) were predicted to be hub genes and may play an essential role in yellow flesh formation.Conclusions: The composition and contents of carotenoid in five watermelon genotypes vary greatly. A series of candidate genes were revealed through combined analysis of metabolites and transcriptome. These results provide an important data resource for dissecting the candidate genes and molecular basis governing flesh color formation in watermelon fruit.

This study was performed to determine the possibility of using mango fruit (Mangifera indica) in brewing technology. The aim of using the SPME-HS-GC-MS technique was to assess what changes occurred in the volatile composition of mango beers brewed in this study. Mango fruit was added to the beer in five different forms to ascertain what kind of preparation should be used to improve beer aroma. Analysis of the volatile components in mango beer showed that beer without mango addition was characterized by the lowest content of volatile compounds (1787.84 µg/100 mL). The addition of mango fruit increased the concentration of compounds, such as α-pinene, β-myrcene, terpinolene, α-terpineol, cis-β-ocimene, caryophyllene, and humulene, in beer. Beer prepared with mango pulp addition was characterized by the highest concentration of volatile components from mango beers (2112.15 µg/100 mL). Furthermore, beers with mango addition were characterized by a higher polyphenol content (up to 44% higher than control beer) and antioxidant activity than control beer and were evaluated by a trained panel as having a better taste and aroma than beer without fruit addition.
