The soil-borne fungi Fusarium pseudograminearum and Rhizoctonia cerealis are the major pathogens for the economically important diseases Fusarium crown rot (FCR) and sharp eyespot of common wheat (Triticum aestivum), respectively. However, there has been no report on the broad resistance of wheat genes against both F. pseudograminearum and R. cerealis. In the current study, we identified TaWAK-6D, a wall-associated kinase (WAK) which is an encoding gene located on chromosome 6D, and demonstrated its broad resistance role in the wheat responses to both F. pseudograminearum and R. cerealis infection. TaWAK-6D transcript induction by F. pseudograminearum and R. cerealis was related to the resistance degree of wheat and the gene expression was significantly induced by exogenous pectin treatment. Silencing of TaWAK-6D compromised wheat resistance to F. pseudograminearum and R. cerealis, and repressed the expression of a serial of wheat defense-related genes. Ectopic expression of TaWAK-6D in Nicotiana benthamiana positively modulated the expression of several defense-related genes. TaWAK-6D protein was determined to localize to the plasma membrane in wheat and N. benthamiana. Collectively, the TaWAK-6D at the plasma membrane mediated the broad resistance responses to both F. pseudograminearum and R. cerealis in wheat at the seedling stage. This study, therefore, concludes that TaWAK-6D is a promising gene for improving wheat broad resistance to FCR and sharp eyespot.

In two experiments on a farm practicing conservation agriculture, the grain yield of a range of wheat cultivars was significantly (p < 0.001) negatively related to the post-harvest population densities of Pratylenchus thornei in the soil profile to 45 cm depth. In a third and fourth experiment with different rotations, methyl bromide fumigation significantly (p < 0.05) decreased (a) a low initial population density of P. thornei in the soil profile to 90 cm depth and (b) a high initial population of P. thornei to 45 cm depth, and a medium level of the crown rot fungus, Fusarium pseudograminearum, at 0–15 cm depth to a low level. For a range of wheat and durum cultivars, grain yield and response to fumigation were highly significantly (p < 0.001) related to (a) the P. thornei tolerance index of the cultivars in the third experiment, and (b) to both the P. thornei tolerance index and the crown rot resistance index in the fourth experiment. In the latter, grain yield was significantly (p < 0.001) positively related to biomass at anthesis and negatively related to percentage whiteheads at grain fill growth stage. One barley cultivar was more tolerant to both diseases than the wheat and durum cultivars. Crop rotation, utilizing crop cultivars resistant and tolerant to both P. thornei and F. pseudograminearum, is key to success for conservation farming in this region.

AbstractUtilization of biocontrol agents is a sustainable approach to reduce plant diseases caused by fungal pathogens. In the present study, we tested the effect of the candidate biocontrol fungus Aureobasidium pullulans (De Bary) G. Armaud on strawberry under in vitro and in vivo conditions to control crown rot, root rot and grey mould caused by Phytophthora cactorum (Lebert and Cohn) and Botrytis cinerea Pers, respectively. A dual plate confrontation assay showed that mycelial growth of P. cactorum and B. cinerea was reduced by 33–48% when challenged by A. pullulans as compared with control treatments. Likewise, detached leaf and fruit assays showed that A. pullulans significantly reduced necrotic lesion size on leaves and disease severity on fruits caused by P. cactorum and B. cinerea. In addition, greenhouse experiments with whole plants revealed enhanced biocontrol efficacy against root rot and grey mould when treated with A. pullulans either in combination with the pathogen or pre-treated with A. pullulans followed by inoculation of the pathogens. Our results demonstrate that A. pullulans is an effective biocontrol agent to control strawberry diseases caused by fungal pathogens and can be an effective alternative to chemical-based fungicides.

Fusarium crown rot is a significant disease of durum wheat (Triticum turgidum L. var. durum), which exhibits high levels of disease susceptibility. The most extreme symptom of crown rot is a prematurely senescing culm that typically fails to set grain. Individual crown rot-affected durum wheat plants displaying both nonsenescent and prematurely senescent culms were harvested to compare visual discoloration, Fusarium pseudograminearum biomass, and vascular colonization in culm sections sampled at three different heights above the crown. Field samples of EGA Bellaroi were collected at Wellcamp, QLD, in 2011, 2012, 2013, and 2014, and of Hyperno at Narrabri, NSW, in 2014. Prematurely senescent culms exhibited greater visual discoloration, F. pseudograminearum biomass, and vascular colonization than nonsenescent culms in each year they were examined. The extent of these differences varied between environments and timing of collection in each year. Vascular colonization initially occurred in xylem vessels and spread into phloem tissues as disease severity increased. The increased presence of hyphae in vascular bundles of prematurely senescing culms provides strong evidence for the hypothesis that restriction of water and nutrient movement in a diseased culm is a key factor in crown rot severity.

Abstract A cDNA encoding gallerimycin, a novel antifungal peptide from the greater wax moth Galleria mellonella, was isolated from a cDNA library of genes expressed during innate immune response in the caterpillars. Upon ectopic expression of gallerimycin in tobacco, using Agrobacterium tumefaciens as a vector, gallerimycin conferred resistance to the fungal pathogens Erysiphe cichoracearum and Sclerotinia minor. Quantification of gallerimycin mRNA in transgenic tobacco by real-time PCR confirmed transgenic expression under control of the inducible mannopine synthase promoter. Leaf sap and intercellular washing fluid from transgenic tobacco inhibited in vitro germination and growth of the fungal pathogens, demonstrating that gallerimycin is secreted into intercellular spaces. The feasibility of the use of gallerimycin to counteract fungal diseases in crop plants is discussed.

Durum wheat (Triticum turgidum ssp. durum) is susceptible to Fusarium pseudograminearum and sensitive to zinc (Zn) deficiency in Australian soils. However, little is known about the interaction between these two potentially yield-limiting factors, especially for Australian durum varieties. The critical Zn concentration (concentration of Zn in the plant when there is a 10% reduction in yield) and degree of susceptibility to F. pseudograminearum was therefore determined for five Australian durum varieties (Yawa, Hyperno, Tjilkuri, WID802, UAD1153303). Critical Zn concentration averaged 24.6 mg kg–1 for all durum varieties but differed for the individual varieties (mg kg–1: Yawa, 21.7; Hyperno, 22.7; Tjilkuri, 24.1; WID802, 24.8; UAD1153303, 28.7). Zinc efficiency also varied amongst genotypes (39–52%). However, Zn utilisation was similar amongst genotypes under Zn-deficient or Zn-sufficient conditions (0.51–0.59 and 0.017–0.022 g DM μg–1 Zn, respectively). All varieties were susceptible to F. pseudograminearum but the development of symptoms and detrimental effect on shoot biomass and grain yield were significantly greater in Tjilkuri. Even though crown rot symptoms may still be present, the supply of adequate Zn in the soil helped to maintain biomass and grain yield in all durum varieties. However, the extent to which durum varieties were protected from plant growth penalties due to crown rot by Zn treatment was genotype-dependent.

AbstractIn the field study period from 2006 to 2010, the incidence and severity of sharp eyespot caused byRhizoctoniawere recorded on 36 cultivars of triticale at the milk ripe growth stage. Four localities in north-central Poland were included: Chrząstowo, Dębina, Kończewice and Minikowo. The susceptibility of the seedlings of 30 cultivars of triticale toR. cerealis(AG-D subgroup I) andR. solani(AG-5) was studied in the laboratory. There was much variation in incidence and severity of sharp eyespot between years and locations. The disease was most intense at Chrząstowo. At this location, the mean percentage of diseased stems on 28 cultivars was 2.6-35.7 (-55.0), and the mean disease index was 0.7-15.6 (-24.5), with the lowest and highest values in 2008 and 2009, respectively. At Minikowo, the disease was least intense. At this location, the mean percentage of diseased stems on 23 cultivars was 1.0-4.6 (-18.0), and the mean disease index was 0.3-1.4 (-6.3), with the lowest and highest values in 2006 and 2007, respectively. The cultivars with least intense disease were Tulus and Atletico (Chrząstowo), Grenado and Zorro (Dębina), Krakowiak and Tornado (Kończewice), and Woltario and Constans (Minikowo). The cultivars with most intense disease were Alekto (Chrząstowo), Baltiko (Dębina), Pawo (Kończewice) and Borwo (Minikowo). MostlyR. cerealiswas isolated from the diseased stems;R. solaniwas isolated only sporadically. There was a wide variation in the susceptibility of triticale cultivars toRhizoctonia. Most triticale seedlings inoculated withR. cerealisproduced symptoms typical of sharp eyespot. Seedlings inoculated withR. solaniformed extended lesions with no defined borders. Most symptoms developed on coleoptiles, with less symptoms on the leaves and the least on the roots. There was much variation in susceptibility of triticale cultivars to bothRhizoctoniaspecies. Cultivars were grouped into six categories according to the intensity of seedling infection. Categories 1, 2 and 3, representing low, moderate and high susceptibility toR. cerealis, included 17, 10 and 3 cultivars, respectively.Categories 4, 5 and 6, representing low, moderate and high susceptibility toR. solani, included 3, 12 and 15 cultivars, respectively.Cultivars Baltiko and Zorro had low, and cv. Cultivo had high susceptibility to bothRhizoctoniaspecies. No cultivar was resistant toRhizoctonia. There was a positive correlation between infection byR. cerealisandR. solani.Infection of coleoptiles byR. cerealisorR. solaniwas significantly correlated with infection of leaves. No correlation between intensity of sharp eyespot on triticale plants in the field and on seedlings in controlled conditions was found.

It has long been known that the size and shape of sampling units used to assess crop yield may have significant effects upon the precision of the estimates (Smith, 1938; Hudson, 1939) but little is known about the effects when estimating incidence of disease. Gilligan (1980) showed that estimates of the incidence (i.e. presence or absence of disease) of stem canker (Phomx, lingam, perfect state Lepto sphaeria maculans)of oil-seed rape were more precise when large square sampling units rather than long rectangular units of similar area or small square sampling units were used. Moreover, estimates derived from the frequently used method of sampling by removal of 25 stems, supposedly at random from each plot, were shown to be biased.

AbstractHexaploid spring wheat (Triticum aestivum) may exhibit significant crown rot disease responses to infection by Fusarium pseudograminearum, with a range of susceptibility levels available in commercial cultivars. Dry conditions during grain-fill may lead to the expression of prematurely senescing culms, which typically fail to set grain. Assessment of hexaploid spring wheat plants exhibiting both non-senescent and prematurely senescent culms was performed using visual discolouration, Fusarium pseudograminearum biomass, vascular colonisation and quantification of wheat DNA in culm sections sampled at three different heights above the crown and at the peduncle. A comparison of these parameters at four time points from milk development, when senescent culms are first observed, to maturity was conducted. Samples from six commercial cultivars were collected in 2014 from Narrabri and Tamworth, New South Wales and Wellcamp, Queensland. Prematurely senescent culms exhibited greater visual discolouration, Fusarium pseudograminearum biomass and vascular colonisation than non-senescent culms in each cultivar. Colonisation of xylem and phloem tissue was extensive in the basal portions of prematurely senescent culms (36 to 99%), and suggests significant impacts on water and nutrient movement during crown rot disease. Maturation coincided with significant changes in Fusarium pseudograminearum biomass and vascular colonisation. Wheat DNA content varied among cultivars, culm conditions, culm sections and sampling times. The variation in the severity of disease states between culms of the same plant suggests that the timing of initiation of infection in individual culms may vary.
