The antioxidant and antifungal activities of crude hydro-ethanolic extract from Capparis spinosa L. (Capparidaceae) leaves and their fractions, obtained by liquid-liquid extraction (LLE) using solvents with increasing polarity (hexane, diethyl ether, ethyl acetate, butanol, and water), were investigated. The crude extract and the obtained fractions were characterized by colorimetric analysis, pyrolysis-gas chromatography (GC)-mass spectroscopy (MS), Fourier Transform Infrared Spectroscopy, and their antioxidant and antifungal capacity were determined. It was observed that the ethyl acetate fraction was enriched in polyphenols, the butanol fraction resulted in purified from proteins and the residual aqueous fraction contains more hydrophobic compounds. The evaluation of the antioxidant activity revealed that the ethyl acetate fraction possesses an interesting capacity 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging with a percentage of inhibition of 84.02% at a concentration of 2 mg/mL and better ferric reducing antioxidant power (FRAP) 4.275 ± 0.011 mmol/g of dry sample than the other fractions tested. Regarding the antifungal activity, the diethyl ether fraction showed the highest activity against Aspergillus niger with 58.78% of inhibition. The results obtained in this work showed the relevance of the valorization of the leaves of Capparis spinosa L., given its richness in bioactive molecules can be regarded as a natural source of antioxidant and antifungal and may be considered in the future to replace synthetic preservatives in food, pharmaceutic products and cosmetic.

The genus Minthostachys belonging to the Lamiaceae family, and is an important South American mint genus used commonly in folk medicine as an aroma in cooking. The phytochemical-rich samples of the aerial parts of Minthostachys diffusa Epling. were tested for pharmacological and health-promoting bioactivities using in vitro chemical and enzymatic assays. A range of radical scavenging activities of the samples against biological radicals such as nitric oxide and superoxide anion and against synthetic 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radicals, the ferric reducing antioxidant power and the lipid peroxidation inhibition were determined and ranked using the ‘relative antioxidant capacity index’ (RACI). The ethyl acetate fraction showed the highest RACI of +1.12. Analysis of the various fractions’ inhibitory ability against enzymes involved in diabetes (α-amylase and α-glucosidase), and against enzymes associated with Parkinson’s or Alzheimer’s diseases (acetylcholinesterase and butyrylcholinesterase) also suggested that the ethyl acetate fraction was the most active. Liquid chromatography–tandem mass spectrometry analysis of the ethyl acetate fraction showed more than 30 polyphenolic compounds, including triterpenes. The inhibitory cholinesterase effects of the triterpenes identified from M. diffusa were further analysed by in silico docking of these compounds into 3D-structures of acetylcholinesterase and butyrylcholinesterase. This is the first study on pharmacological activities and phytochemical profiling of the aerial parts of M. diffusa, showing that this plant, normally used as food in South America, is also rich in health-promoting phytochemicals.

Background: The prevalence of diabetes mellitus is increasing on a global trend. The aim of the present study is to identify the most effective antioxidants and hypoglycemic fraction of Azanza garckeana. Methods: The fractions (nhexane or ethyl-acetate or aqueous) of A. garckeana were administered to the alloxan-induced diabetic rats at doses of 100, 200, and 400 mg/kg for 15 days. Antioxidants activities were evaluated at concentrations of 62.5, 125, 250, and 500 µg/mL using the DPPH scavenging assay. Results: Results revealed that both the hexane, ethyl-acetate, and aqueous fractions exhibited hypoglycemic and antioxidants activities in a dose-dependent manner. The n-hexane fraction demonstrated highest percentage DPPH scavenging effect of 26.34±3.43, 38.44±4.35, 59.34±3.45, and 74.83±5.35 at 62.5, 125, 250, and 500 µg/mL respectively. The ethyl-acetate fraction demonstrated 19.33±2.98, 28.94±3.24, 47.34±2.90, and 57.82±4.54 respectively while the aqueous fraction exhibited the least activities of 12.45±23.45, 18.64±2.94, 27.94±3.89, and 39.43±3.89 at concentrations of 62.5, 125, 250, and 500 µg/mL respectively. In addition, the n-hexane fraction demonstrated the most significant hypoglycemic effect with the highest glucose reduction of 58.97 ±3.45 %, 63.86±5.35 %, and 66.51±4.35 %, ethyl acetate fraction demonstrated glucose reduction of 7.55±0.54%, 21.77±2.35 %, and 29.56±3.46 % while the aqueous fraction demonstrated the least hypoglycemic effect of 9.89±2.67 %, 18.09±3.45 %, and 18.87±3.24 at 100, 200 and 400 mg/kg bw respectively. Conclusion: The n-hexane fraction of Azanza garckeana extract could serve as a reservoir of bioactive agents that could be useful for the development of a new anti-diabetic agent

The methanolic extract of Artemisia incisa Pamp. was dissolved in distilled water and successively partitioned with n-hexane, chloroform, ethyl acetate and n-butanol. The antioxidant potential of all these fractions and remaining aqueous fraction was evaluated by four methods, i.e., the scavenging activity of the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), the total antioxidant activity, the ferric reducing antioxidant power (FRAP) assay and the ferric thiocyanate assay. In addition, the total phenolics was determined. The obtained results revealed that among the studied fractions the ethyl acetate soluble fraction showed the most potent DPPH-radical scavenging activity with an IC50 value of 5.3 ? 0.71 ?g mL-1, which is even more effective than the standard antioxidant butylated hydroxytoluene (BHT) (IC50 value 0f 12.1? 0.92 ?g mL-1). The ethyl acetate fraction also showed the highest FRAP value (3677.13 ? 27.1 ?g TE mL-1), inhibition of lipid peroxidation (60.93 ? 0.84 % at 500 ?g mL-1) and total phenolic content (95.5 ? 0.05 ?g GAE g-1) as compared to other fractions. However, the remaining aqueous fraction was found to posses the highest antioxidant activity of all the fractions.

Afzelia bella Harms (Fabaceae), a plant widely distributed in Africa, is used in traditional medicine for varied disease conditions including the treatment of topical skin infections. The present study investigated the antimicrobial and antioxidant activities of the methanol extract and various solvent fractions of the leaves of the plant. The methanol leaf extract was partitioned to yield petroleum ether, chloroform, ethyl acetate and residual aqueous fractions. Total phenol and flavonoid contents, radical scavenging activity and ferric reduction antioxidant power (FRAP) were determined by spectrophotometry, while antimicrobial activity and minimum inhibitory concentration (MIC) of the extract and fractions were determined using agar-well diffusion and agar dilution methods, respectively against clinical bacterial isolates of Bacillus subtilis, Escherishia coli, Klebsiella pneumoniae, Staphylococcus aureus, and fungi, Aspergillus flavus, Candida parapsilosis, Microsporium audiounii. Bacillus subtilis was the most susceptible among the bacterial strains tested, while Microsporium audiounii was the most susceptible fungus. The alcoholic extract and all solvent fractions demonstrated a concentration dependent antimicrobial activity with inhibition zone diameter range of 7.5 to 35.0 mm. MIC ranged from 0.1 - 8 mg/ml and activity was highest in ethyl acetate fraction with MIC of 0.1 mg/ml. FRAP ranged from 0.161 - 0.319 mmol Fe2+/g extract and was highest in the ethyl acetate fraction. These results give an indication that A. bella leaf has high antioxidant and antimicrobial activities and support the folkloric claim of the therapeutic potential of the plant. Methanolic extract of Java Plum (Syzygium cumini L. (Skeel) seed (MEJS) is potential source of natural antioxidant. As indicated by several in vitro measurements, the extract had strong DPPH (1,1 diphenyl, 2–picryl hydrazyl) and ABTS (2,2-azinobis, 3-ethylbenzothiazoline-6-sulphonate) radical scavenging activity, strong Ferric Reducing Antioxidant Power (FRAP) and moderate inhibition activity of linoleic acid oxidation. This study aimed to determine antioxidant activity and phenolic compound of Java Plum seed (Syzygium cumini L. (Skeel) methanolic extract fractions. Phenolics compound identification using Thin Layer Chromatography (TLC) showed that all fractions (polar, semi polar and hydrolyzed semi polar fraction) contained Gallic acid, Tannic acid and flavonol’s Rutin. HPLC-DAD analysis showed that its polar fraction contained 25 ppm flavonol’s Quercetine and 55181 ppm flavonol’s (+)- Catechin, ethyl acetate fraction contained 54 ppm flavonol’s Rutin and 528 ppm (+)- Catechine, while hydrolyzed ethyl acetate fraction contained 404 ppm Rutin and 28692 ppm (+)- Catechine.

The ethanol-water (7 : 3, v/v) extract of Cotoneaster multiflorus sarcocarp was sequentially fractionated by liquid-liquid partition using n-hexane, diethyl ether, methylene dichloride, and ethyl acetate. The contents of total polyphenols, total flavones, and oligomeric proanthocyanidins in the five parts (including the ethanol-water extract) were determined. In addition, 2,2-diphenyl-1-picrylhydrazyl free radical-scavenging, 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical cation decolorization, reducing power, ferric reducing antioxidant power, and lipid peroxidation inhibition assays were conducted to test the antioxidant activities of Sample 1 (the ethanol-water fraction) and Sample 2 (the ethyl acetate fraction) in vitro. In the above five assays, Sample 2 showed greater antioxidant capacities than Sample 1. Furthermore, Sample 2 was better able to protect low-density lipoproteins from oxidation in a dose-dependent manner. The test results show that C. multiflorus sarcocarp, especially the ethyl acetate-soluble fraction, may be a potential source of natural antioxidants.

Obesity and diabetes are major metabolic disorders which are prevalent worldwide. Algae has played an important role in managing these disorders. In this study, Gracilaria edulis, a marine red algae, was investigated for antioxidant and hypoglycemic potential using in vitro models. De-polysaccharide methanol extract of G. edulis was sequentially partitioned with hexane, chloroform, ethyl acetate, and antioxidants, and hypoglycemic potentials were evaluated using multiple methods. High antioxidant potential was observed in the ethyl acetate fraction in terms of ferric reducing antioxidant power, iron chelating, and DPPH and ABTS radical scavenging activities, while the crude methanol extract exhibited potent oxygen radical-absorbance capacity. Potent α-amylase inhibitory activity was observed in the ethyl acetate fraction, while the ethyl acetate fraction was effective against α-glucosidase inhibition. Glucose diffusion was inhibited by the ethyl acetate fraction at 180 min, and the highest antiglycation activity was observed in both chloroform and ethyl acetate fractions. Additionally, gas chromatography-mass spectrometry analysis of the ethyl acetate fraction revealed the presence of several potent anti-diabetic compounds. In conclusion, G. edulis exhibited promising antidiabetic potential via multiple mechanisms. The ethyl acetate fraction exhibited the strongest hypoglycemic and antiglycation potential among the four fractions, and hence the isolation of active compounds is required to develop leads for new drugs to treat diabetes.

Abstract Background Diabetes mellitus is a metabolic disorder of glucose metabolism and management of blood glucose level is the hallmark in the treatment of this disease. The present study investigated chemical composition, in vitro antioxidant and antidiabetic activity of different fractions of 80% methanol Piper guineense leaves extract. Materials and methods The crude methanolic extract of P. guineense was obtained following 80% methanol cold extraction and was successively partitioned with dichloromethane (DCM), ethyl acetate (EtOAc), n-butanol (nBuOH) and aqueous solvents to give four fractions. The chemical composition of the fractions from P. guineense was determined using gas chromatography coupled with mass spectrometry (GC-MS) and their potentials as antioxidant and anti-diabetes were evaluated. Results The percentage yields were 3.16, 2.22, 0.68 and 0.66% (w/w) in n-butanol, DCM, aqueous and ethyl acetate fractions of P. guineense methanolic extract, respectively. The GC-MS analyses identified a total of 71 and 34 phytochemicals in n-butanol and ethyl acetate fractions respectively. Tributyl acetylcitrate (10.95%) and phytol (9.11%) were the major components in the n-butanol fraction while ethyl piperonyl cyanoacetate (27.35%) and phytol (15.17%) were the major constituents in the ethyl acetate fraction. Ethyl acetate fraction had the highest ferric reducing antioxidant power with a value of 53.96 ± 0.40 mgAAE/g while n-butanol fraction possessed highest total antioxidant power (9.98 ± 0.15) followed by aqueous fraction (9.72 ± 0.02). The ethyl acetate and n-butanol fractions with IC50 value of 0.24 ± 0.07 and 0.83 ± 0.15 μg/mL respectively elicited significant inhibitory activities against α-glucosidase while only n-butanol fraction (IC50 = 0.33 ± 0.09 μg/ml) exhibited appreciable inhibition against α-amylase activity. However, none of the four fractions showed significant inhibitory activity towards dipeptidyl-peptidase-IV. Conclusion n-butanol and ethyl acetate fractions of 80% methanol P. guineense leaves extract can be a potential source of bioactive compounds of pharmacological importance in the management of diabetes.

Objective: In an attempt to explore herbal drug which may become useful in the prevention of diabetes and antioxidant potential by the ethanol extracts of Maytenus heyneana (MH) root belonging to the family Celastraceae and their different fractions were studied.Methods: Different fractionation was done using chloroform, ethyl acetate, and methanol on ethanolic extract of MH and preliminary phytochemical analysis was done by standard methods to identify the presence of important compounds. In vitro antioxidants activities were carried by 2,2-diphenyl- 2-picrylhydrazyl (DPPH) and 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation radical scavenging assays. For antidiabetic potential, α-amylase and α-glucosidase enzyme inhibitory studies were carried on different fractions.Results: Phytochemical studies show the presence of alkaloids, flavonoids, phenols, cardiac glycosides, and terpenoids in all fractionations; however, tannins and quinones were present in ethyl acetate fraction and saponins in methanolic fraction. For antioxidant activity, ethyl acetate fraction shows concentration of the sample causing 50% inhibition (IC50) values in 22.31 μg/ml and methanolic fraction shows in 12.82 μg/ml concentrations for DPPH and ABTS radical scavenging assay, respectively. In case of antidiabetic activity, methanolic fraction offered significant result in inhibitory action of α-glucosidase and also for α-amylase assay IC50 (5.28 and 3.14 μg/ml) than other fractions.Conclusion: From the results of our studies, it can be concluded that MH shows antidiabetic and antioxidant values and methanolic fraction of MH could be possessed potential constituents in the prevention of diabetes and antioxidant than other fractions. However, further studies are required to validate.
