Afzelia bella Harms (Fabaceae), a plant widely distributed in Africa, is used in traditional medicine for varied disease conditions including the treatment of topical skin infections. The present study investigated the antimicrobial and antioxidant activities of the methanol extract and various solvent fractions of the leaves of the plant. The methanol leaf extract was partitioned to yield petroleum ether, chloroform, ethyl acetate and residual aqueous fractions. Total phenol and flavonoid contents, radical scavenging activity and ferric reduction antioxidant power (FRAP) were determined by spectrophotometry, while antimicrobial activity and minimum inhibitory concentration (MIC) of the extract and fractions were determined using agar-well diffusion and agar dilution methods, respectively against clinical bacterial isolates of Bacillus subtilis, Escherishia coli, Klebsiella pneumoniae, Staphylococcus aureus, and fungi, Aspergillus flavus, Candida parapsilosis, Microsporium audiounii. Bacillus subtilis was the most susceptible among the bacterial strains tested, while Microsporium audiounii was the most susceptible fungus. The alcoholic extract and all solvent fractions demonstrated a concentration dependent antimicrobial activity with inhibition zone diameter range of 7.5 to 35.0 mm. MIC ranged from 0.1 - 8 mg/ml and activity was highest in ethyl acetate fraction with MIC of 0.1 mg/ml. FRAP ranged from 0.161 - 0.319 mmol Fe2+/g extract and was highest in the ethyl acetate fraction. These results give an indication that A. bella leaf has high antioxidant and antimicrobial activities and support the folkloric claim of the therapeutic potential of the plant. The radish (Raphanus sativus L.) is a vegetable of the Brassicaceae family cultivated worldwide and has several medicinal properties. Its biological activities are related to various secondary metabolites present in the species, especially phenolics. Thus, the objectives of this study were the chemical analysis and evaluation of the antioxidant and antimicrobial activities of the dry extract and fractions of the fodder turnip leaves (R. sativus var. oleiferus Metzg.). Samples were analyzed by mass spectrometry and the antioxidant activity was evaluated using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical method and the reducing power method. Antimicrobial activity was determined by the agar diffusion and microdilution methods. The total phenols were concentrated in the butanol fraction (121.27 mg GAE/g) and the flavonoids were concentrated in the ethyl acetate fraction (98.02 mg EQ/g). The ethyl acetate fraction showed the best antioxidants results, with 83.45% of free radical scavenging and 11.34% of ferric ions reduction. The analysis of antimicrobial activity showed that the dry extract had the highest average zone of inhibition against Bacillus subtilis (18.67 mm). Smaller values of the minimum inhibitory concentration for Micrococcus luteus were, and the ethyl acetate fraction showed a lower minimum inhibitory concentration (0.1 mg/ml) for that microorganism. There was a strong correlation between the antioxidant activity and the content of phenols and flavonoids. The results showed the potential antioxidant and antimicrobial activities of this extract with the ethyl acetate fraction being most promising for further studies.

The genus Minthostachys belonging to the Lamiaceae family, and is an important South American mint genus used commonly in folk medicine as an aroma in cooking. The phytochemical-rich samples of the aerial parts of Minthostachys diffusa Epling. were tested for pharmacological and health-promoting bioactivities using in vitro chemical and enzymatic assays. A range of radical scavenging activities of the samples against biological radicals such as nitric oxide and superoxide anion and against synthetic 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radicals, the ferric reducing antioxidant power and the lipid peroxidation inhibition were determined and ranked using the ‘relative antioxidant capacity index’ (RACI). The ethyl acetate fraction showed the highest RACI of +1.12. Analysis of the various fractions’ inhibitory ability against enzymes involved in diabetes (α-amylase and α-glucosidase), and against enzymes associated with Parkinson’s or Alzheimer’s diseases (acetylcholinesterase and butyrylcholinesterase) also suggested that the ethyl acetate fraction was the most active. Liquid chromatography–tandem mass spectrometry analysis of the ethyl acetate fraction showed more than 30 polyphenolic compounds, including triterpenes. The inhibitory cholinesterase effects of the triterpenes identified from M. diffusa were further analysed by in silico docking of these compounds into 3D-structures of acetylcholinesterase and butyrylcholinesterase. This is the first study on pharmacological activities and phytochemical profiling of the aerial parts of M. diffusa, showing that this plant, normally used as food in South America, is also rich in health-promoting phytochemicals.

The purpose of the current study was to determine the phenolic composition, antioxidant, and antimicrobial activities in grape cane extracts from typical cultivars of Southern Italy. Aqueous extracts at different pHs (1–13) were prepared from “Aglianico”, “Fiano”, and “Greco” grape canes. The results demonstrated that an alkaline pH (13.00) produced the best polyphenol-rich extracts, as the total phenolic content was more than double when compared to the respective extracts prepared at pH 1.00. “Greco” grape canes gave the highest quantity of phenolic compounds at each pH, ranging from 42.7 ± 0.4 to 104.3 ± 3.0 mg Gallic Acid Equivalents (GAE)/g Dry Extract (DE) from pH 1.00 to 13.00. The Radical Scavenging Activity (RSA) and the Ferric Reducing Antioxidant Power (FRAP) were measured. The highest antioxidant activity was showed by “Greco” extract at pH 7.00. Seventy-five compounds were identified in the extracts by HPLC-MS with six of them described for the first time in grape canes. Procyanidins were highly abundant in extracts at pH 7.00, whereas stilbenoids were the most represented compounds at pH 13.00. Very strong antiviral activity against herpes simplex viruses was recorded for the extracts at pH 7.00 and 13.00 that were active in the early stages of infection by acting directly against the viral particles. The overall results suggest that grape canes, currently underutilized, can be usefully valorised by providing active extracts to use as antioxidant and antiviral agents.

AbstractThe phytochemical and antimicrobial activity of the petroleum ether and crude methanol extracts, chloroform and ethyl acetate fractions of the leaves of Lannea kerstingii were investigated. Phytochemical screening revealed the presence of steroids and triterpenes in the petroleum ether extract, steroid, triterpene, flavonoids and tannins in both crude methanol extract and chloroform fraction while the ethyl acetate fraction contained only flavonoids and tannins. The extracts exhibited antimicrobial activities with zones of inhibition ranging from 17.00 to 21.03, 20.10 to 25.24, 25.32 to 34.02 and 22.28 to 27.20 mm for petroleum ether extract, methanol extract, chloroform and ethyl acetate fractions respectively. The minimum inhibitory concentration was between 5 and 10mg/ml, 5mg/ml for the petroleum ether and methanol extract respectively, and between 2.5 and 5 mg/ml, 5mg/ml for the acetate fractions. The minimum bactericidal concentration for all the extracts was 40mg/ml respectively except for chloroform fraction which ranged from 20 to 40mg/ml. The minimum fungicidal concentration for all the extracts was found to be 40mg/ml respectively. This result indicates the broad spectrum antimicrobial potential of L. Kerstingii and justifies the use of this plant in traditional medicine.

Mallotus oppositifolius (Geiseler) Mull. Arg.  (Euphorbiaceae) is a predominant edible shrub in Nigeria. It is locally used for treatment of common infections and wounds. This study evaluated the toxicity and antioxidant potential of the crude extract and fractions of M. oppositifolius using invitro 1,1-diphenyl-2-picrylhydrazine (DPPH) radical scavenging assay. The result showed abundance of flavonoids (28.21), tannins (17.72), alkaloids (7.79) and Saponins (0.23) in the crude extract of M. oppositifolius leaf extract. Also, the result showed that the LD50 is above 5000 mg/kg, therefore the crude extract is safe for consumption. The result further showed that the ethyl-acetate and butanol had significant (p<0.05) antioxidant activity when compared with the control at 125, 62.5 and 31.125 µg/ml respectively while at 250 and 500 µg/ml there was no significant different in the level of reduction of oxidation between the butanol and ethyl acetate when compared with the control. The result further showed that the butanol and ethyl-acetate fraction of M. oppositifolius had the best antioxidant potential among the fractions studied. The result of GCMS studies of ethyl-acetate fraction showed the presence of 7,10,13-eicosatrienoic acid methyl ester, korormicin, agosterol, phyllanthin, ricinoic acid, 2,4,6-decatrienoic acid 1a,2,5,5a,6,9,10,10a-octahydro-5,5a-dihydroxy-4-(hydroxymethyl)-1,1,7,9, limonin, com-pounds that have shown to possess strong antioxidant activities. Also, the GCMS result of butanol fraction showed the presence of seven compounds which include: ferulylhistamine, hippeastrine, 10,13-eicosadienoic acid methyl ester, ethyl 5,8,11,14-eicosatetraenoate, 6-hydroxy-powelline-N-nitroso-7-demethoxy-aldehyde, 6,7-epoxypregn-4-ene-9,11,18-triol-3,20-dione 11,18-diacetate and 5Alpha-androstane-3,17-dione 17-monooxime,  compounds which may serve as a new lead for the treatment of diseases associated with the oxidative stress.

Dryopteris filix mas (D filix-mas) is wildly used in ethnomedicine for the management of rheumatoid arthritis, wounds and other diseases. We investigated the anti-oxidant activities of its leaf extract, and chromatographic fractions. The ethanol leaf extract was partitioned into four fractions; n-hexane, ethyl acetate, n-butanol and water. Ferric reducing anti-oxidant power (FRAP), 1, 1-diphenyl-2-picrylhydrazil (DPPH) and nitric oxide (NO) scavenging in vitro assays were carried out on the extract and fractions at 6.25, 12.5, 25, 50, 100, 200, 400 and 800 µg/mL. The most active fraction (ethyl acetate fraction) was further purified using chromatographic techniques to isolate its major compound whose structure was elucidated using ID nuclear magnetic resonance (NMR) and mass spectrometry. The ethyl acetate fraction produced the highest free radical scavenging activity among the other fractions. The fraction (VLC-E7) from which the bioactive compound, quercetin-3-O-αL-rhamnopyranoside, was isolated had the best FRAP and DPPH scavenging activities with EC50 and IC50 values of 88.81 ± 3.41 and 26.87 ± 0.24 respectively more than the ethyl acetate fraction. This study revealed that the polyphenol flavonoid, quercetin-3-O-αL-rhamnopyranoside could be responsible for antioxidant activity of ethno-medicinal property of D filix-mas leaf.

Methanolic extract of Java Plum (Syzygium cumini L. (Skeel) seed (MEJS) is potential source of natural antioxidant. As indicated by several in vitro measurements, the extract had strong DPPH (1,1 diphenyl, 2–picryl hydrazyl) and ABTS (2,2-azinobis, 3-ethylbenzothiazoline-6-sulphonate) radical scavenging activity, strong Ferric Reducing Antioxidant Power (FRAP) and moderate inhibition activity of linoleic acid oxidation. This study aimed to determine antioxidant activity and phenolic compound of Java Plum seed (Syzygium cumini L. (Skeel) methanolic extract fractions. Phenolics compound identification using Thin Layer Chromatography (TLC) showed that all fractions (polar, semi polar and hydrolyzed semi polar fraction) contained Gallic acid, Tannic acid and flavonol’s Rutin. HPLC-DAD analysis showed that its polar fraction contained 25 ppm flavonol’s Quercetine and 55181 ppm flavonol’s (+)- Catechin, ethyl acetate fraction contained 54 ppm flavonol’s Rutin and 528 ppm (+)- Catechine, while hydrolyzed ethyl acetate fraction contained 404 ppm Rutin and 28692 ppm (+)- Catechine.

The ethanol-water (7 : 3, v/v) extract of Cotoneaster multiflorus sarcocarp was sequentially fractionated by liquid-liquid partition using n-hexane, diethyl ether, methylene dichloride, and ethyl acetate. The contents of total polyphenols, total flavones, and oligomeric proanthocyanidins in the five parts (including the ethanol-water extract) were determined. In addition, 2,2-diphenyl-1-picrylhydrazyl free radical-scavenging, 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical cation decolorization, reducing power, ferric reducing antioxidant power, and lipid peroxidation inhibition assays were conducted to test the antioxidant activities of Sample 1 (the ethanol-water fraction) and Sample 2 (the ethyl acetate fraction) in vitro. In the above five assays, Sample 2 showed greater antioxidant capacities than Sample 1. Furthermore, Sample 2 was better able to protect low-density lipoproteins from oxidation in a dose-dependent manner. The test results show that C. multiflorus sarcocarp, especially the ethyl acetate-soluble fraction, may be a potential source of natural antioxidants.

Obesity and diabetes are major metabolic disorders which are prevalent worldwide. Algae has played an important role in managing these disorders. In this study, Gracilaria edulis, a marine red algae, was investigated for antioxidant and hypoglycemic potential using in vitro models. De-polysaccharide methanol extract of G. edulis was sequentially partitioned with hexane, chloroform, ethyl acetate, and antioxidants, and hypoglycemic potentials were evaluated using multiple methods. High antioxidant potential was observed in the ethyl acetate fraction in terms of ferric reducing antioxidant power, iron chelating, and DPPH and ABTS radical scavenging activities, while the crude methanol extract exhibited potent oxygen radical-absorbance capacity. Potent α-amylase inhibitory activity was observed in the ethyl acetate fraction, while the ethyl acetate fraction was effective against α-glucosidase inhibition. Glucose diffusion was inhibited by the ethyl acetate fraction at 180 min, and the highest antiglycation activity was observed in both chloroform and ethyl acetate fractions. Additionally, gas chromatography-mass spectrometry analysis of the ethyl acetate fraction revealed the presence of several potent anti-diabetic compounds. In conclusion, G. edulis exhibited promising antidiabetic potential via multiple mechanisms. The ethyl acetate fraction exhibited the strongest hypoglycemic and antiglycation potential among the four fractions, and hence the isolation of active compounds is required to develop leads for new drugs to treat diabetes.

In today’s world, search for natural medicines is increasing as a result of drug resistance of pathogens and also due to negative consequences of antibiotic. Presence of phytochemicals, antioxidant potential and antimicrobial activity of Artocarpus heterophyllus was carried out in this study. Leaf of this plant was subjected to warm extraction with three different solvents namely methanol, aqueous methanol and ethyl acetate. Leaf extract showed the presence of coumarin, alkaloid, terpenoid in methanol solvent; tannin, coumarin, saponin in aqueous methanol extract and coumarin, terpenoids in ethyl acetate solvent. Further, antimicrobial activity was assessed through disc diffusion method with six pathological bacteria and two fungi strains in four different concentrations of plant extract. Largest ZOI of 16mm was obtained against B. subtilis in 200mg/ml concentration for ethyl acetate extract. Antioxidant potential was measured by DPPH (Diphenyl-2-picrylhydrazyl) assay. DPPH free radical Scavenging Activity was expressed in % inhibition with L Ascorbic acid as standard and leaf extract in methanol showed the best activity.
