Thirty clinically healthy Holstein-Friesian cows underwent twice daily machine milking and ultrasonographic examinations of the udder just prior to and after milking. Digital ultrasonographic images of each udder quarter were subjected to computer-assisted echotextural analyses to obtain mean numerical pixel values (NPVs) and pixel heterogeneity (PSD) of the mammary gland parenchyma. The average milk yield and pH were higher (p < 0.05) in the morning, whereas crude fat, total solids, solids non-fat and citric acid content were higher (p < 0.05) during the evening milking period. Mean NPVs and PSDs of the mammary gland parenchyma were greater (p < 0.05) after than before milking. There were significant correlations among echotextural characteristics of the udder and protein percentage, lactose content and freezing point depression determined in the milk samples collected in the morning and crude protein, casein, lactose and solids non-fat in the evening. Our results can be interpreted to suggest that computerized analysis of the mammary gland ultrasonograms has the makings of a technique for estimating non-fat milk constituents in cows. However, future validating studies are necessary before this method can be employed in commercial settings and research. Moreover, significant inter-quarter differences in udder echogenicity may necessitate further echotextural studies of separate quarters.

Values of milk indicators (MIs) can be influenced by sampling errors and milk manipulation. This paper estimated the freezing point depression (FPD) and other MIs drifts which can cause fat movement. That is important for: – preparation of reference milk samples (MSs) for proficiency testing and instrument calibrations; – estimation of the impact of milk treatment as centrifugation in dairy plants on FPD. Five MSs (A = original milk; milk with modified fat (F) content; B = less F, C = low F, D = more F, E = high F) were created (gravitation F separation at 4 °C for 12 hours) with the same milk matrix 12× per year. F averages increased by 4.80% (122.1%) from 1.68 to 6.48% due to manipulation. It increased variability of MIs especially for SNF (solids non fat), L (lactose) and CP (crude protein). SCC (somatic cell count) averages increased by 803 (196.8%) from 9 to 812 thousand.ml−1. Correlation (r) F × SCC was 0.85 (P < 0.001). SNF, L and CP averages decreased by 0.47% (5.3%), 0.31% (6.3%) and 0.17% (5.0%). Correlations were −0.78, −0.75 and −0.64 (P < 0.001). Urea decreased along with F increase by 1.05 mg.100ml−1 (2.9%) but with r −0.13 (P > 0.05). Acetone increased by 1.37 mg.l−1 (47.6%) with r 0.21 (P > 0.05). Electrical conductivity decreased by 0.23 mS.cm−1 (6.0%) with r −0.15 (P > 0.05). Alcohol stability was reduced by 0.14 ml (23.3%) with r −0.15 (P > 0.05). FPD, titration and actual acidity were not influenced.

Abstract The aim of this study was to determine the fatty acid (FA) profile of Simmental cow milk in relation to lipid intake in feeds and their composition, and to compare it with FA composition of milk from cows of Polish Holstein-Friesian (PHF) breed. To this end, 104 milk samples were collected from Simmental cows and 52 milk samples derived from PHF breed were taken over a 2-year period. The season significantly influenced both contents of lipid, dry matter and crude protein as well as FA content in lipids extracted from forages. Winter and autumn seasons were characterized by a significantly higher intake of lipids with forages resulting from the intake of fresh silages, compared with the other seasons when the animals were fed stored silages. This was reflected in the highest content of conjugated linoleic acid (CLA) c-9 t-11 C18:2 in milk from the autumn season. Compared with PHF, milk from Simmentals had a higher content of short-chain saturated FAs (SCSFAs) and was characterized by great differences in FA contents determined in the group of odd- and branched-chain FAs (OBCFAs). Milk from Simmental cows had the butyric acid C4:0 and iso C15:0 content of 4.99 and 1.20 g/100 g FA on average, respectively, which have been associated with benefits for human health.

Abstract The objective of this study was to estimate the influence of lactation number, month of milk sampling, lactation stage and herd size on the freezing point of milk of Polish Holstein‑Friesian cows. Data comprised 4,719,787 milk samples from the first seven lactations of 752,770 Polish Holstein- Friesian cows. Milk freezing point (MFP), milk yield, and fat and protein content were analyzed. The mean MFP of milk samples (-0.5326°C) as well as more than 92% of all milk samples did not exceed the quality limit for the freezing point of cows′ raw milk, which, following Polish standards, was taken to be -0.52°C. The freezing point of milk samples, milk yield, fat and protein content were highly significantly affected by all examined factors: month of sampling, successive lactations, stage of lactation, and herd size. MFP was lowest in milk samples taken from January to March, and highest in samples from November to December. MFP increased with lactation number. Mean MFP decreased with time within lactation, except the first stage (5-35 days in milk). Generally, MFP was highest in small herds (up to 9 cows) and lowest in large herds (more than 150 cows).

Although milk output is known to be affected by dietary manipulation, studies have so far failed to provide sufficient information to successfully predict dietary effects on both milk protein content and yield (Thomas 1983). In a further effort to determine the metabolic control of milk protein synthesis three diets containing different levels of crude protein were fed to four Holstein-Friesian cows in early- to mid-lactation, and metabolite flow across the mammary gland was measured in relation to milk protein production.

SummarySimilar, marked variations in the freezing-point depressions of jugularvenous blood and of milk throughout a day were observed in cows when drinkingwater was offered for a single, short period each day. Values for milk were found to agree more closely, however, with those for mammary-venous blood than with those for jugular-venous blood. It appears that milk is in osmotic equilibrium with the blood flowing through the udder continuously throughout the period the milk remains within the udder and not only during its formation, and that milk secretion causes a slight alteration in the osmotic pressure of fluids within the immediate locality of the mammary gland. Changes in the milk composition that occurred in association with the observed changes in freezing-point depression were consistent with a movement of water into or out of the udder in response to any change in the osmotic pressure of blood.

An experiment was conducted with 36 Holstein-Friesian cows to determine the effect on milk yield and milk composition of feeding maize grain with or without meat-and-bone meal (MBM) at high levels of pasture on offer. Cows were offered ground maize with and without MBM to give supplements of 3.0, 5.5 and 8.0 kg DM/day at 16% or 10% crude protein. Animals grazed nitrogen-fertilised tropical grass and grass-legume pastures. Milk production was recorded over a 300-day lactation. Milk yields over 300 days at 3.0, 5.5 and 8.0 kg DM/day averaged 5435, 5605 and 5882 kg/cow, respectively. For milk yields over both 100 and 300 days, a linear response to grain supplement occurred. This represented 0.22 kg milk/kg DM (P<0.05) for the first 100 days, and 0.30 kg milk/kg DM (P=0.087) over the 300-day lactation. Cows receiving MBM tended to lose less (P=0.068) liveweight in the period 1-100 days and to gain more (P=0.054) between 100 and 300 days than cows without MBM. Milk yields across treatments for cows fed with and without MBM averaged 2143 and 2061 kg/cow (P>0.05) for days 1-100, and 5668 and 5614 kg/cow (P>0.05) for days 1-300 of lactation.

AbstractThe objective of this study was to determine the relationship between milk β-hydroxybutyrate (BHB), acetone (ACE) as well as parity and lactation stage and milk freezing point (MFP) in Polish Holstein-Friesian cows in early lactation. Additionally, we studied the relationship between milk ketone bodies and daily milk yield (DMY), fat (MF) and protein (MP) content in milk. The data obtained from the Polish Federation of Cattle Breeders and Dairy Farmers, comprised 749,894 test day milk samples, collected between 6 and 60 days in milk (DIM) from 521,049 lactations of 514,066 cows. Milk BHB and ACE were determined using the Fourier transform infrared (FTIR) technology. Four classes of parities were created: first, second, third, and fourth to seventh and two classes of lactation stage: 5–21 and 22–60 DIM. BHB was grouped into five classes: ≤0.05, 0.06–0.10, 0.11–0.20, 0.21–0.50 and >0.50 mmol/L, and ACE was also classified into five classes: ≤0.05, 0.06–0.10, 0.11–0.15, 0.16–0.30 and >0.30 mmol/L. Data on MFP, DMY, and MF and MP content were analyzed using the MIXED procedure of SAS and a linear model in which effects of parity, lactation stage, BHB and ACE classes were included, together with interactions between lactation stage and BHB classes, parity and BHB classes, lactation stage and ACE classes, and parity and ACE classes. The differences among parity, lactation stages, BHB and ACE classes in MFP, DMY, MF and MP were highly significant. There was a clear tendency for decreasing of MFP with increasing of BHB. Such a trend did not occur in case of ACE. DMY and MP decreased and MF increased with increasing BHB or ACE. In conclusion, since MFP can be measured relatively easily and is well related to milk BHB content, it may be used in the prediction of diagnostic models of ketosis based on milk composition.

AbstractControlled breeding schemes for oestrous detection constitutes a proactive technical response that balances the infrastructural requirement for a profitable dairy operation and the demands for optimal animal performance. The present study compared (a) the reproductive response of a treatment based on a short vs longer-acting PGF2α analogue (tiaprost vs luprostiol), and (b) the reproductive response after a treatment of GnRH-PGF2α vs PGF2α alone for synchronizing dairy cows. Holstein-Friesian cows averaging 9000 kg milk/lactation and fed according to their requirements were used in the study. Cows were cyclic, at least 60 days post partum and were clinically sound before being considered for the experiments. In Experiment 1, animals were synchronised using an i.m. injection of either 15 mg of luprostiol or 0.75 mg of tiaprost, based on ultrasonic diagnosis of a corpus luteum. Animals were inseminated at observed oestrus. In Experiment 2, cows were synchronised, at random, by either an injection of 10pg ofbuserelin (day 0) followed by 0.75 mg of tiaprost at day 7 (GnRH-PGF2α) orjust 0.75 mg of tiaprost (PGF2α). For both treatments only cows with an ultrasonically detected corpus luteum were treated. Animals were inseminated at oestrus. At the time of treatment and again 3 days later, milk samples were collected and assayed for progesterone by RIA. Cows with progesterone concentrations >1 ng/ml were considered to have corpus luteum. Luteolysis was considered to have occurred when concentrations of progesterone were > 1 ng/ml at day 0 and <0.8 ng/ml at day 3. In Experiment 1, both analogues gave similar results in terms of induced luteolysis [luprostiol: 36/39 (92.3%) vs tiaprost: 36/41 (87.8%)], oestrous detection efficiency [luprostiol: 26/36 (72.2%) vs tiaprost: 30/36 (83.3%], oestrous distribution [day 2, 3 and 4, respectively: luprostiol: 26.9%, 50.0%, 19.2% vs tiaprost: 36.7%, 50.0%, 13.3%], and conception rates [luprostiol: 12/25 (48.0%) vs tiaprost: 14/28 (50.0%); P>0.05]. In Experiment 2, oestrous detection efficiency, interval to oestrus and conception rate were similar between treatments [97/149 (65.1%), 71.1 h, 43/95 (45.3%) for PGF2α vs 130/188 (69.1%), 68.2h, 65/126 (51.6%) for GnRH-PGF2α, respectively]. However the oestrous distribution was more concentrated in GnRH-PGF2α treated animals (P<0.01).

Milk output has been manipulated by dietary methods for many years, however the underlying mechanisms for changes in milk composition are as yet unclear. In an attempt to further understand these mechanisms we have compared the mammary uptake of metabolites on two isoenergetic diets with different protein levels expected to provide different amounts of amino acid to the mammary gland.Early- to mid-lactation Friesian cows were used to examine the effect of increased dietary protein supply on the supply of metabolites to and uptake by the mammary gland in relation to milk protein synthesis. Two barley based concentrates were formulated to contain 20.8 (Cl) or 29.1 (C2) g N/kg DM, using fishmeal (Provimi 66, high UDP) as the supplementary protein and fed with grass silage (28.3 g N/kg DM) at a total dry matter intake of 17 kg/day. In Experiment 1 these diets were fed at 50:50 (concentrate:silage) to four lactating cows fitted with rumen and duodenal cannulae in a simple crossover design.
