In the last years, the interest in Italian monovarietal oils has increased due to their specific organoleptic qualities. Extra virgin olive oils (EVOOs) are rich in phenolic compounds, secondary metabolites well known and studied for their nutraceutical properties. However, among EVOOs, there is great variability in phenolic composition due to the origin, the production technique, and mainly, the genotype. The aim of this work was to evaluate the different phenolic profiles and the antioxidant activities of monovarietal oils. The results confirm this variability. In fact, the overall content of oleuropein varies up to four times between the different genotypes (from 33.80 to 152.32 mg/kg oil), while the oleocanthal content is significant only in two oils. The antioxidant activity, determined with 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and oxygen radical absorbance capacity (ORAC) assays, is correlated with the content of total phenolic substances, with half maximal inhibitory concentration (IC50) values for the DPPH test ranging from 160 to 91 mg of oil, while the ORAC test shows values between 5.45 and 8.03 μmol Trolox equivalent (TE)/g oil.

Tea is among the most consumed drink worldwide, and its strong antioxidant activity is considered as the main contributor to several health benefits, such as cardiovascular protection and anticancer effect. In this study, the antioxidant activities of 30 tea infusions, which were obtained by the mimic of drinking tea of the public, from green, black, oolong, white, yellow and dark teas, were evaluated using ferric-reducing antioxidant power (FRAP) and Trolox equivalent antioxidant capacity (TEAC) assays, ranging from 504.80 ± 17.44 to 4647.47 ± 57.87 µmol Fe2+/g dry weight (DW) and 166.29 ± 24.48 to 2532.41 ± 50.18 µmol Trolox/g DW, respectively. Moreover, their total phenolic contents (TPC) were detected by Folin-Ciocalteu assay and were in the range of 24.77 ± 2.02 to 252.65 ± 4.74 mg gallic acid equivalent (GAE)/g DW. Generally, Dianqing Tea, Lushan Yunwu Tea, and Xihu Longjing Tea showed the strongest antioxidant activities among 30 teas. Furthermore, the phenolic compounds in tea infusions were identified and quantified, with catechins most commonly detected, especially in green tea infusions, which were main contributors to their antioxidant activities. Besides tea polyphenols, considerable content of caffeine also presented in 30 tea infusions.

In this study, the total phenolic and flavonoid contents and antioxidant activities of hexane and methanol extracts of Cotoneaster transcaucasicus Pojark. fruits in the Rosaceae family were investigated. Using the Folin-Ciocaltaeu method, the amount of phenolic substances was determined as 50.34 ± 2.70 mg of GAE/g in methanol extract and 38.06 ± 1.32 mg of GAE/g in hexane extract. The total amount of flavonoids was calculated as 52.01 ± 3.78 mg of QE/g in the methanol extract and 60.54 ± 6.82 mg of QE/g in the hexane extract, using the aluminum nitrate method. Antioxidant activities of C. transcaucasicus hexane and methanol extracts were evaluated by DPPH and FRAP methods. IC50 values of the extracts were calculated as 0.411 mg/mL for hexane extract and 0.237 mg/mL for methanol extract. In addition, it was determined that the reducing antioxidant power of Fe3+-Fe2+ was higher in hexane extract. As a result, it has been observed that the phenolic and flavonoid content and antioxidant activities of C. transcaucasicus fruits are high. This study is the first study in C. transcaucasicus species and it is thought that it will lead the future studies.

Xylaria species associated with termite nests or soil have been considered rare species in nature and the few which have been reported upon have been found to act as a rich source of bioactive metabolites. This study evaluated 10 ethyl acetate extracts of five new Xylaria species associated with termite nests or soil for their antioxidant activity, and cytotoxicity against different cancer and normal cell lines. DPPH and ABTS radical scavenging activities of the extracts demonstrated strong capacity with low IC50 values. The highest observed activities belonged to X. vinacea SWUF18-2.3 having IC50 values of 0.194 ± 0.031 mg/mL for DPPH assay and 0.020 ± 0.004 mg/mL for ABTS assay. Total phenolic content ranged from 0.826 ± 0.123 to 3.629 ± 0.381 g GAE/g crude extract which correlated with antioxidant activities. The high total phenolic content could contribute to the high antioxidant activities. Cytotoxicity was recorded against A549, HepG2, HeLa and PNT2 and resulted in broad spectrum to specific activity depending on the cell lines. The highest activities were observed with X. subintraflava SWUF16-11.1 which resulted in 11.15 ± 0.32 to 13.17 ± 2.37% cell viability at a concentration of 100 µg/mL. Moreover, LC-MS fingerprints indicated over 61 peaks from all isolates. There were 18 identified and 43 unidentified compounds compared to mass databases. The identified compounds were from various groups of diterpenoids, diterpenes, cytochalasin, flavones, flavonoids, polyphenols, steroids and derivatives, triterpenoids and tropones. These results indicate that Xylaria spp. has abundant secondary metabolites that could be further explored for their therapeutic properties.

The analysis of phenolic compounds in extra virgin olive oils was carried out by high-performance liquid chromatography utilizing photodiode array and mass spectrometry detectors. The chromatographic profile of thirty samples from four Italian Regions highlighted the presence of secoiridoids, phenolic alcohols, flavonoids, and phenolic acid classes. A similar qualitative profile was observed with some differences in peak area and fifteen compounds were tentatively identified. Quantitative analysis was performed by UV detection considering eight standard phenolic compounds. The chromatographic method, after optimization, was validated studying some parameters, e.g., intra-day and inter-day retention time precision, limit of detection, limit of quantification, and linearity. Recovery of the method was performed achieving good results (10 and 50 g·g−1 with recovery of 72.9–92.1% (w/w) and 79.1–102.8% (w/w), respectively). In all samples secoiridoids were the main compounds ranging from 85 to more than 99% (w/w) of the total concentration of detected phenolic compounds while phenolic acids accounted for the lowest percentage (0.1–0.6%, w/w). Finally, total concentration of phenolic compounds and antioxidant activity were determined with different chemical assays. A good and significant correlation among total phenolic compound concentration and antioxidant activity was observed. A significant different phenolic compound concentration and antioxidant activity was determined between samples from Puglia and Sicily. This was studied performing statistical analysis by one-way analysis of variance (ANOVA) followed by Bonferroni post-hoc test.

This study was conducted to determine the pH, protein concentration, mineral, phenolic, flavonoid, and organic acid contents, as well as the antioxidant activity of three batches of fruit jiaosu (F1B3, F1B4, and F1B5). The jiaosu was prepared by mixing ten types of fresh fruits, raw honey, and water (3:1:10, w/w/w), and fermented at room temperature for six months. All fruit jiaosu had low pH (2.96–3.69), and the protein concentration was ranged from 8.45 to 16.2 µg/mL. Potassium (605.0–657.2 mg/L) was found to be the most abundant mineral in all the samples, followed by magnesium, calcium, and sodium. The total phenolic and flavonoid contents were 183.1–210.5 µg gallic acid equivalent/mL and 45.2–53.8 µg quercetin equivalent/mL, respectively. Acetic acid was the major organic acid in the F1B4 and F1B5 samples (6.58 g/L and 20.7 g/L, respectively) while the F1B3 sample contained the highest amount of lactic acid (12.1 g/L). Ascorbic and citric acids were present in small amounts in all samples. All the samples showed stronger scavenging activity against 2,2-diphenyl-1-picrylhydrazyl radicals (half-maximum inhibitory concentration, IC50: 13.6–16.8% v/v) than against hydrogen peroxide (IC50: 47.2–50.6% v/ v). The ferric reducing antioxidant power and oxygen radical absorbance capacity values for the samples were 14.9–20.9 µg ascorbic acid equivalent/mL and 23.9–25.4 mg Trolox equivalent/mL, respectively. The results indicated that the fruit jiaosu is a complex acidic solution containing proteins, minerals, phenolic compounds, and organic acids with antioxidant activities. Natural fermentation may cause variations in the organic acid content of the fruit jiaosu.

Aim: The present study was undertaken to establish a suitable drying process for Garcinia cowa and Garcinia pedunculata fruits for maximum retention of their nutritional qualities. Methodology: Slices of three different thickness with 0.3, 0.6, 0.9 cm of G. cowa and G. pedunculata fruits were dried by sun drying, solar drying and by cabinet drying at three different temperatures (50°C, 70°C and 90°C). Results: The mean crude fat and ash content were higher in G. cowa than G. pedunculata while later was found rich in crude protein and Fe contents. Total phenol content ranged between 881.31-888.65 and 719.00-736.74 mg gallic acid equivalent 100 g-1, total flavonoid content between 89.21-90.06 and 51.00-52.54 mg quercetin equivalent 100 g-1 and mean HCA contents ranged between 3.13-3.92% and 1.84-1.99% for G. cowa and G. pedunculata, respectively. The methanolic extracts exhibited significant antioxidant activity. With lower IC50 values, G. cowa had higher antioxidant activity than G. pedunculata. Interpretation: Oven drying at 50°C and 0.9 cm slice thickness for drying resulted in better retention of total phenolic and flavonoid contents with better antioxidant activities.

Eleven Italian monovarietal extra virgin olive oils (MEVOOs) (Carboncella, Coratina, Frantoio, Leccino, Marzio, Maurino, Moraiolo, Piantone di Falerone, Pendolino, Rosciola, Sargano di Fermo) from olives grown in the same experimental olive orchard, under the same conditions (fertilization, irrigation), and processed with the same technology (three-way continuous plant) were investigated. As a result, the impact of the olive cultivar on fatty acid and triacylglycerols composition, oxidative stability, polar phenolic profile and sensory properties (panel test) of the oil was assessed. Pendolino, Maurino and Marzio oils presented the highest levels (p < 0.01) of palmitic, linoleic and linolenic acids % and the lowest oleic:linoleic ratio. Within triacylglycerols, triolein (OOO) strongly varied among the oils, with Coratina and Leccino having the highest content. Frantoio showed the lowest 1-Stearoyl-2-palmitoyl-3-oleylglycerol and 1,3-Distearoyl-2-oleylglycerol amounts. Rosciola showed the highest level (p < 0.01) for two of the most abundant secoiridoid derivatives (the dialdehydic forms of decarboxymethyl elenolic acid linked to hydroxytyrosol and tyrosol). A good correlation was found between total phenolic content and oxidative stability, indicating Marzio and Leccino respectively as the richest and poorest genotypes. Sensory variability among varieties was mainly linked to perceived bitterness, pungency and fruitiness, while no effects were found on secondary flavors.

Sweet potato is the sixth most important crop widely cultivated around the world with abundant varieties. Different varieties gain different phenolic profiles which has drawn researchers’ attention for its unique health benefits. Our study evaluated the phenolic profiles, total and cellular antioxidant activities, antiproliferative activities, and cytotoxicity in 10 cultivated varieties of sweet potato in different colours. Among fourteen metabolites detected in our study, hyperoside, ferulic acid and caffeic acid were considered as prominent in SPSRs. According to the principle component analysis, phytochemical composition of HX22, YS15 and YS7 was quite similar. The results also evidenced that purple-fleshed varieties, such as YS43, YZ7 and YY153, have higher total phenolics content and corresponding stronger total antioxidant capacities as well as cellular antiproliferative activities against human liver cancer HepG2 cells than other varieties. The extremely significant correlation between phenolics and total antioxidant activity was also revealed by Pearson correlation analysis (p < 0.05). However, no significant relevance was found between intracellular antioxidant activity and total phenolic content or flesh colour of sweet potatoes.

The present study aims to investigate the antioxidant activity and cytotoxicity of white mugwort (Artemisia lactiflora) leaf extract in the human liver cancer cell line (HepG2 cell). Dried leaves of white mugwort were macerated in 80 % (v/v) aqueous ethanol to get white mugwort leaf extract (WMLE).  Spectrophotometrically methods were conducted to quantify the total phenolic content and the antioxidant capacity of the extract, and were found to be 213.33±17.80 mg gallic acid equivalent and 27.20±1.06 mg trolox equivalent per gram of dry extract, respectively. Cytotoxic effects of white WMLE on cell viability and cellular antioxidant defense against reactive oxygen species (ROS) were also examined. The WMLE showed weak ability to inhibit cell growth, with a 50 % inhibitory concentration (IC50) value of 148 µg/mL. Furthermore, the extract exhibited antioxidant activity in HepG2 cells via the ability to lower cellular ROS in a dose-dependent fashion. The preliminary results of this investigation supported that WMLE exhibits strong antioxidant activities while remain considerably non-cytotoxic effect. However, more intensive studies are required to clarify the use of WMLE as a novel alternative medicine for the treatment of liver cancer.
