The aim of this work was to contribute to the knowledge on the chemical composition and bioactive properties of two species of the Ocimum genus, namely O. basilicum cultivar ’Cinammon’ and O. × citriodorum. For this purpose, samples of these plants grown in Portugal were evaluated for their composition in phenolic and volatile compounds, and the infusion and hydroethanolic extracts were assessed for their in vitro antioxidant, antimicrobial, cytotoxic, and anti-inflammatory activities. In total, the two basil samples showed the presence of seven caffeic acid and derivatives (dimers, trimers, and tetramers) and five flavonoids, mainly glycoside derivatives of quercetin. Despite some qualitative and quantitative differences, in both samples rosmarinic acid was the major phenolic compound, and linalool the predominant volatile compound. In general, the tested extracts provided relevant bioactive properties since both basil species showed higher antioxidant activity in Thiobarbituric Acid Reactive Substances (TBARs) and Oxidative Hemolysis Inhibition (OxHLIA) assays when compared with the positive control Trolox. Despite O. × citriodorum extracts showing slightly better activity against some strains, both types of extracts evidenced similar antimicrobial activity, being more active against Gram-positive bacteria. The extracts also revealed interesting cytotoxicity, particularly the O. × citriodorum hydroethanolic extract which was also the only one exhibiting anti-inflammatory activity.

The present work aimed to characterize leaves, stems, and flowers of Carissa macrocarpa (Eckl.) A.DC., by performing an analysis of the phenolic compounds by HPLC-DAD/ESI-MS, correlating them with bioactive properties, such as antioxidant, anti-inflammatory, cytotoxic, and antimicrobial activities. Thirty polyphenols were identified in the hydroethanolic extract, including phenolic acids, flavan-3-ols, and flavonol glycosides derivatives (which presented the highest number of identified compounds). However, flavan-3-ols showed the highest concentration in stems (mainly owing to the presence of dimers, trimmers, and tetramers of type B (epi)catechin). Leaves were distinguished by their high antioxidant and anti-inflammatory activities, as well as their bactericidal effect against E. coli, while stems presented a higher cytotoxic activity and bactericidal effect against Gram-positive bacteria. Moreover, a high correlation between the studied bioactivities and the presence of phenolic compounds was also verified. The obtained results bring added value to the studied plant species

Metabolic diseases have increased considerably such as diabetes mellitus (DM). Since diabetes is a systemic disease, it implies high cardiovascular risks. It has been widely established that cardiac injury is related to mitochondrial dysfunction through increment of reactive oxygen species (ROS). Synthetic antioxidants can have important side effects; therefore natural sources may represent a better option. Traditional Mexican medicine has been using Eryngium carlinae (EC) for medical treatment. Also our group showed that hexanic extract possesses in vitro antioxidant capacity. Experimental diabetes in Wistar rats was generated by streptozotocin (STZ) and hexanic extract of EC was supplied for 7 weeks (30 mg/kg). Cholesterol, triacylglycerides, glucose, and thiobarbituric acid reactive substances (TBARS) levels were determined in serum. Mitochondria from left ventricle were used in the quantification of TBARS, reduced glutathione, nitric oxide (NO) levels and activity of superoxide dismutase (SOD) enzyme was performed.  Biochemical parameters of glucose and triacylglycerides, as well as TBARS levels in serum show a significant reduction in diabetic group supplied with EC hexanic extract. Thus, we can conclude that the EC hexanic extract possesses antioxidant activity in vitro, and in vivo, by reducing glucose and triacylglycerides levels during hyperglycemia, which may eventually reduce the risk of developing diabetic cardiomyopathy.

AbstractBasil plant (Ocimum americanum) is efficacious as an anti-inflammatory and analgesic activity. According to research by Sarma and Babu, 2011, Verma and Kothiyal, 2012 showed basil activity as an antioxidant, antimicrobial, anti-diabetic, anthelmintic, antifungal, insecticide, anti-inflammatory, analgesic, and lowering total cholesterol and LDL-C levels. The purpose of this study was to determine the stabilization activity of red blood cell membranes on methanol partitioning, n-hexane partitioning and ethanol extract of basil leaves in vitro. This study used the erythrocyte membrane stabilization method from the induction of a hypotonic solution with samples of methanol partitioning, n-hexane partitioning and ethanol extract to be compared with a positive control, namely Na diclofenac. By analyzing the data using UV-Vis spectrophotometry test. These results were supported by the ANOVA statistical test which stated that there was a difference in each treatment and continued with the Tukey test which stated that there was no difference between 100 ppm diclofenac sodium and 400 ppm ethanol extract.Keywords: Extract, Basil (Ocimum americanum) Leaf, In Vitro. AbstrakTumbuhan Kemangi (Ocimum americanum) berkhasiat sebagai aktivitas sebagai anti-inflamasi dan analgesik. Tujuan dari penelitian ini yaitu untuk mengetahui aktivitas stabilisasi membran sel darah merah pada partisi metanol, partisi n-heksan dan ekstrak etanol daun kemangi secara in vitro. Penelitian ini menggunakan metode stabilisasi membran eritrosit dari induksi larutan hipotonik dengan sampel partisi metanol, partisi n-heksan dan ekstrak etanol yang akan dibandingkan dengan kontrol positif yaitu Na diklofenak. Dengan analisis data menggunakan uji spektrofotometri UV-Vis. Hasil ini didukung dengan uji statistik ANOVA yang menyatakan terdapat perbedaan pada setiap perlakuan dan dilanjutkan uji tukey yang menyatakan tidak ada perbedaan pada natrium diklofenak 100 ppm dengan ekstrak etanol konsentrasi 400 ppm.Kata Kunci : Ekstrak, Daun Kemangi (Ocimum americanum), In Vitro.

The purpose of the present study was to determine whether it is possible to alter the development of fatigue and ablate free radical-mediated lipid peroxidation of the diaphragm during loaded breathing by administering oxypurinol, a xanthine oxidase inhibitor. We studied 1) room-air-breathing decerebrate, unanesthetized rats given either saline or oxypurinol (50 mg/kg) and loaded with a large inspiratory resistance until airway pressure had fallen by 50% and 2) unloaded saline- and oxypurinol-treated room-air-breathing control animals. Additional sets of studies were performed with animals breathing 100% oxygen. Animals were killed at the conclusion of loading, and diaphragmatic samples were obtained for determination of thiobarbituric acid-reactive substances and assessment of in vitro force generation. We found that loading of saline-treated animals resulted in significant diaphragmatic fatigue and thiobarbituric acid-reactive substances formation ( P < 0.01). Oxypurinol administration, however, failed to increase load trial time, reduce fatigue development, or prevent lipid peroxidation in either room-air-breathing or oxygen-breathing animals. These data suggest that xanthine oxidase-dependent pathways do not generate physiologically significant levels of free radicals during the type of inspiratory resistive loading examined in this study.

The hydroethanolic extract of Anthocephalus cadamba displayed remarkable antioxidative potential in the 1,1-diphenyl-2-picrylhydrazyl (DPPH), the hydrogen peroxide, the nitric oxide scavenging, the reducing power, the total antioxidant capacity, the lipid peroxidation inhibition (thiobarbituric acid-reactive substances production), and the RBC membrane stabilization assays. While in the DPPH assay the IC50 value of the extract was 146.5 μg/mL, it was 24.8 μg/mL in the nitric oxide scavenging assay.

 Objective: The aim of this study was to investigate in vitro anti-inflammatory activity of tinocrisposide using lipopolysaccharides (LPS)-stimulated RAW 264.7 macrophage cells. Tinocrisposide is a furano diterpene glycoside that was isolated in our previous study from Tinospora crispa.Methods: Anti-inflammatory effect was quantified spectrometrically using Griess method by measuring nitric oxide (NO) production after the addition of Griess reagent.Results: The sample concentrations of 1, 5, 25, 50, and 100 μM and 100 μM of dexamethasone (positive control) have been tested against the LPS-stimulated RAW 264.7 cells, and the results showed NO level production of 39.23, 34.00, 28.9, 20.25, 16.3, and 13.68 μM, respectively, and the inhibition level of 22.67, 33.00, 43.03, 60.10, 68.00, and 73%, respectively.Conclusions: From the study, it could be concluded that tinocrisposide was able to inhibit the formation of NO in the LPS-stimulated RAW 264.7 cells in concentration activity-dependent manner, with half-maximal inhibition concentration 46.92 μM. It can be developed as anti-inflammatory candidate drug because NO is a reactive nitrogen species which is produced by NO synthase. The production of NO has been established as a mediator in inflammatory diseases.

AbstractXanthone derivatives of acetic, propionic and 2-methylpropionic acids were synthesized and assayed for their anti-inflammatory, analgesic and ulcerogenic activities. Compound 8 causes a dose-dependent diminution of paw edema (up to 61%) in the carrageenan model and at the highest tested dose reduces mechanical hyperalgesia in the Randall-Selitto test more effectively than the reference compound (~75% and ~32%, respectively). It shows high in vitro metabolic stability (Clint=12.5 μL/mg/min, t1/2=138.6 min) in the rat liver microsomes. None of the studied xanthone derivatives are ulcerogenic. The results of the present study suggest that compound 8 can be of interest in the future for the search for antinociceptive and antiedematous agents devoid of ulcerogenic effect.

Epidendrum coryophorum belongs to the Orchidaceae family. Traditional uses of some species for this genus include infusions of the leaves used for kidney problems, treat influenza, conjunctivitis, liver pain, relieve kidney symptoms, and hypoglycemic effect. This work's objective was to determine the phytochemical profile of the ethanolic extract of Epidendrum coryophorum leaves and to evaluate the potential anti-inflammatory activity in vitro of the extract employing the erythrocyte membrane stabilization method. The phytochemical screening carried out in this work suggested phenols, coumarins, flavonoids, tannins, steroids, and sterols in the ethanolic extract of Epidendrum coryophorum leaves. Cardiotonic glycosides and carbohydrates were also found. The ethanolic extract's UV-Vis spectrum showed absorption maxima at 268 nm and 332 nm, which could correspond to flavonoids of the flavonoid classes, 3-OH substituted flavonols, or isoflavones. The quantitative determination of total phenols of the ethanolic extract was carried out using the Folin-Ciocalteu method. The total phenolic content expressed as mg Gallic acid equivalent (G.A.E.) per gram of extract was found to be 19,96 mgGAE/g of Epidendrum coryophorum. The ethanolic extract of Epidendrum coryophorum leaves showed hemolysis inhibition values ​​of 18,19% at 1,0 mg/mL, 38,98% at 1,5 mg/mL and 40,94% at 2,5 mg/mL compared with aspirin (positive control) giving values ​​of 65,33% at 1,0 mg/mL, 72,26% at 1,5 mg/mL and 73,75% at 2,5 mg/mL. The values ​​obtained for inhibition of hemolysis with ethanolic extract, compared with the values ​​obtained with a pure anti-inflammatory, are significant and demonstrate anti-inflammatory activity in Epidendrum coryophorum. Abstract A simple, reliable, and reproducible fluorometric method for measuring thiobarbituric acid-reactive substances (TBARS) in serum is proposed, based on the reaction between malondialdehyde (MDA) and thiobarbituric acid. Formation of TBARS was complete at pH 2.4-2.6, but extraction with n-butanol proved complete only at lower pH, i.e., 1.6-1.7. Analytical recoveries of MDA added to serum were 94%-101%; within- and between-run CVs were 2.4-3.6% and 4.6-5.5%; and the detection limit for TBARS in serum was 0.10 mumol/L. Optimized conditions included: (a) collection of either serum or heparinized plasma, (b) preservation from in vitro autoxidation by glutathione and EDTA, and (c) storage at -20 degrees C up to 35 days. The mean (+/- SD) TBARS concentration in 47 healthy adults was 1.01 (0.21) mumol/L; no sex-related difference was observed. Higher concentrations were measured in patients with renal insufficiency undergoing hemodialysis and in patients with insulin-dependent diabetes, chronic pancreatitis, or liver cirrhosis.
