Caries are a pathological process of extracorporeal nature, characterized by demineralization of inorganic substances as well as proteolysis triggered by acids produced by bacteria present in dental plaque, as a result of metabolism of sugars of both external and internal origin. Periodontal disease, on the other hand, is a multifactorial degenerative disease associated with inflammation, involving a group of tissues that surround the dental cervix and root of the tooth. It is believed that one of the mechanisms in the etiopathogenesis of caries and periodontitis are disorders of local and/or general oxidative stress (OS) parameters. Numerous clinical studies have confirmed the relationship between oxidative stress markers and oral diseases. In most analyzed studies, technical and biological variability was so high that none of the markers so far has proven suitable for routine clinical use. The aim of systematic reviews of the literature is to present the existing studies on OS parameters, mainly concerning the activity of antioxidant enzymes in saliva of patients with caries and periodontitis.

(1) Background: Current evidence suggests that salivary markers of oxidative stress are indicative of clinical disease indices such as the papillary bleeding index (PBI) and the caries index (CI). The aim of this study was to assess the relation of oxidative stress markers with oral dental caries and periodontal problems in a pediatric population. (2) Methods: In this case-control study, unstimulated whole saliva was collected from individuals aged 3–18 years (n = 177); 14 individuals were excluded. Study subjects were divided into those with caries (CI = 2, n = 78) and those who were caries-free (n = 85). These groups were then divided into another subset consisting of children (mean age 7.3 years, n = 121) and adolescents (mean age 16.1 years, n = 42). The PBI was determined in all groups. We then assessed salivary levels of oxidative stress markers. (3) Results: Total antioxidant capacity (TAC) level increased in patients with more gingival bleeding (p < 0.05) in the study group aged 3–18 years. In addition, TAC showed a significant decrease in samples with caries when compared to the caries-free group in adolescents (p = 0.008). (4) Conclusions: TAC levels may be a marker of both gingival bleeding and dental caries in young adult populations. We hope that in the near future, prophylaxis, control, follow up and even possible therapeutic use of oxidative stress markers in a chairside way will become possible as antioxidants have been shown to be effective against oral diseases.

Objective: Children with nephrotic syndrome (NS) have a stressful condition, and oxidative damage may impair their treatment response. This study aims to gain a better understanding of the relationship between oxidative stress and NS to lay the basis for further research into improved diagnostic options, treatment, and prevention of the disease. Methods: We took a blood sample from 100 Indian patients aged 2-14 y. Each patient was tested for oxidative stress. The buege method was used to assess MDA levels in patients. The pyrogallol method was used to measure SOD activity in blood serum, and the jollow method was used to measure glutathione levels. Results: The levels of oxidative stress markers (MDA, SOD, and GSH) were compared between NS patients and the control. SOD and GSH concentrations were significantly decreased in the NS group when compared to the control. In contrast, MDA level was significantly higher in the NS group than in the control. In the correlation analysis, we found that the serum SOD activity was significantly positively correlated with serum albumin and creatinine level in patients with NS. Thus, oxidative stress in children with NS is indicated by reduced antioxidant potential because of low albumin. Therefore, it is thought that oxidative stress is implicated in the development of NS in Indian children. Conclusion: We concluded that oxidative stress was intensified in children with NS due to decreased antioxidant levels caused by hypoalbuminemia.

Sirt6 is a member of the sirtuin family involved in physiological and pathological processes including aging, cancer, obesity, diabetes, and energy metabolism. This study is aimed at evaluating the relationship between liver SIRT6 gene expression and the oxidative stress network depending on adiposity levels in Zucker rats, an animal model of metabolic syndrome. We observed that liver-specific SIRT6 expression is reduced in an in vivo model of spontaneous obesity and metabolic syndrome. We also observed that SIRT6 expression in the liver is positively associated with SIRT1 and GST-M2 expressions, two proteins involved in antioxidant protection pathways and inversely related to body weight and plasmatic oxidative status. Interestingly, the SIRT6 expression is upregulated after energy restriction-induced weight loss concomitantly with an improvement in oxidative stress markers. These results suggest that SIRT6 may be a potential therapeutic target for the treatment of obesity and associated metabolic disorders, such as liver disease.
