The open rock shelter of Yabelo in Ethiopia hosts diverse Holocene paintings of great cultural importance. The paintings are characterized by the presence of different mineral coatings, whose features have not been studied yet. Our goal was to understand whether different rock samples from the Yabelo paintings collected in close proximity may reveal coatings with different minerology and biology. Thus, elemental analyses combined with microscopic and molecular investigations were performed on two coatings, one whitish (sample 1) and one reddish (sample 2). Although both samples were dominated by heterotrophic bacteria, the two coatings showed distinct mineralogical and microbiological characteristics. Sample 1 contained higher amounts of Ca and P than sample 2, which was likely related to the presence of organic matter. Sample 1 hosted bacterial genera that are potentially involved in biomineralization processes, metal redox cycles and metal resistance. In contrast, sample 2 showed mainly pathogenic and commensal bacteria that are characteristic of animal and human microbiota, and other microorganisms that are involved in nitrogen and metal biogeochemical cycles. Overall, our results indicated that the bacterial communities were particular to the coating mineralogy, suggesting a potential role of the biological components in the crust genesis.

In mammals, repression of translation during stress is associated with the assembly of stress granules in the cytoplasm, which contain a fraction of arrested mRNA and have been proposed to play a role in their storage. Because physical contacts are seen with GW bodies, which contain the mRNA degradation machinery, stress granules could also target arrested mRNA to degradation. Here we show that contacts between stress granules and GW bodies appear during stress-granule assembly and not after a movement of the two preassembled structures. Despite this close proximity, the GW body proteins, which in some conditions relocalize in stress granules, come from cytosol rather than from adjacent GW bodies. It was previously reported that several proteins actively traffic in and out of stress granules. Here we investigated the behavior of mRNAs. Their residence time in stress granules is brief, on the order of a minute, although stress granules persist over a few hours after stress relief. This short transit reflects rapid return to cytosol, rather than transfer to GW bodies for degradation. Accordingly, most arrested mRNAs are located outside stress granules. Overall, these kinetic data do not support a direct role of stress granules neither as storage site nor as intermediate location before degradation.

Abstract. It has been univocally shown that iron (Fe) is the primary limiting nutrient for phytoplankton metabolism in high-nutrient, low-chlorophyll (HNLC) waters, yet the question of how this trace metal affects heterotrophic microbial activity is far less understood. We investigated the role of Fe for bacterial heterotrophic production and growth at three contrasting sites in the naturally Fe-fertilized region east of the Kerguelen Islands and at one site in HNLC waters during the KEOPS2 (Kerguelen Ocean and Plateau Compared Study 2) cruise in spring 2011. We performed dark incubations of natural microbial communities amended either with iron (Fe, as FeCl3) or carbon (C, as trace-metal clean glucose), or a combination of both, and followed bacterial abundance and heterotrophic production for up to 7 days. Our results show that single and combined additions of Fe and C stimulated bulk and cell-specific bacterial production at the Fe-fertilized sites, while in HNLC waters only combined additions resulted in significant increases in these parameters. Bacterial abundance was enhanced in two out of the three experiments performed in Fe-fertilized waters but did not respond to Fe or C additions in HNLC waters. Our results provide evidence that both Fe and C are present at limiting concentrations for bacterial heterotrophic activity in the naturally fertilized region off the Kerguelen Islands in spring, while bacteria were co-limited by these elements in HNLC waters. These results shed new light on the role of Fe in bacterial heterotrophic metabolism in regions of the Southern Ocean that receive variable Fe inputs.

Introduction: Previous radiological studies revealed that stones lodge more frequently in the ureterovesical junction (UVJ) as well as the proximal ureter. Factors that prevent stone passage from the proximal ureter are not well studied. Aim: To explore the site of the lodged stones in the proximal ureter with direct observation during laparoscopic ureterolithotomy. Materials and methods: Between November 2014 and February 2015, we included 26 patients including 18 men and 8 women with stones larger than 10 millimeters in the proximal ureter who were candidate for laparoscopic ureterolithotomy. We prospectively recorded the site of the lodged stones in the ureter during laparoscopic ureterolithotomy in relation with the sites of ureteral stenosis as well as the gonadal vessels. Results: Among 26 patients with ureteral stone, in 19 cases stone was found close to the gonadal vein compared with seven cases that stone was in other locations of the ureter (p = 0.02). The characteristics of patients and stones were not different in cases that the stone was close to gonadal vessels compared with other locations. Conclusions: This study showed that most of the stones lodged in the proximal ureter were in close proximity with gonadal vessels. Gonadal vessels may be an extrinsic cause of ureteral narrowing.

Trees with stem bark lesions are frequently observed in Eucalyptus globulus Labill. plantations, particularly in the central west region of Uruguay. These lesions constitute a problem for trunk decortications at harvest and they also facilitate the access of fungi that could cause wood decay. Seven, three and oneyear-old plantations, located at three sites in close proximity were selected. Four types of trunk lesions were present in trees regardless the age of plantation and more than one type was found in each plantation. The aim of this study was to investigate the fungal composition associated with these lesions and compare them to healthy tissues and try to find out the origin of these symptoms. Another purpose was to elucidate the real role of the fungi considered pathogens by means of experimental inoculations. Segments from lesions and healthy tissues yielded 897 fungal isolates belonging to 32 taxa, 681 isolates from bark lesions and 216 from healthy tissues. Both healthy and symptomatic tissues showed similar fungal species composition, but with differences in frequencies of colonization. Cytospora eucalypticola Van der Westhuizen, Botryosphaeria spp., Pestalotiopsis guepinii (Desm.) Stey. and Penicillium spp. were the dominant species isolated. As symptoms were not reproduced after experimental inoculation with Botryosphaeria ribis Grossenb. & Duggar and B. eucalyptorum Crous, & M.J. Wingf, it could be suggested that these lesions were originated by unfavorable environmental conditions. The frost that occurred for several days out of season and flooding may have been involved in the development of bark lesion.

<p><strong><em>ABSTRAKSI: </em></strong><em>Penelitian ini bertujuan untuk menjelaskan peranan seorang seniman bernama Ateng Japar dalam mengembangkan seni pertunjukan Longser di Kabupaten Bandung, Jawa Barat, Indonesia, dari tahun 1975-2002. Masalah utama yang dibahas adalah bagaimana dinamika perkembangan seni pertunjukan Longser pada masa Ateng Japar, 1975-2002. Metode yang digunakan dalam penelitian ini yaitu metode historis. Hasil penelitian menunjukkan bahwa Longser merupakan seni teater rakyat yang hidup di daerah Priangan, khususnya di Kabupaten Bandung. Longser seringkali disebut sebagai seni sandiwara rakyat yang banyak menampilkan tarian-tarian berpasangan dan memberikan kesempatan kepada para penonton untuk menari bersama dengan penarinya. Pada tahun 1939, Ateng Japar mendirikan kelompok Longser yang diberi nama “Pancawarna”. Awalnya, kelompok ini melakukan pertunjukan dengan cara mengamen, namun memasuki tahun 1970-an terjadi peralihan tempat pertunjukan, yakni kedalam gedung kesenian. Pada tahun 2002, sang seniman Ateng Japar diberitakan wafat dan hal ini berdampak pada perkembangan Longser yang semakin meredup. Selain itu, masuknya pengaruh globalisasi yang diikuti oleh perubahan zaman yang semakin maju dan modern, pada akhirnya membuat seni pertunjukan Longser yang masih bertahan hingga saat ini kurang mendapat tempat di hati masyarakat luas. Eksistensi Longser semakin diperparah ketika sebagian masyarakat, terutama dari kalangan generasi muda, masih banyak yang belum mengetahui tentang seni pertunjukan Longser. </em></p><p><strong><em>KATA KUNCI:</em></strong><em> Longser, teater rakyat, Ateng Japar, perubahan zaman, dan pengaruh globalisasi.</em><em> </em></p><p><strong><em>ABSTRACT: </em></strong><em>“Ateng Japar: The Legend of Longser Performing Arts and His Roles in Bandung Regency, 1975 - 2002”.<strong> </strong>This study aims to explain the role of an artist named Ateng Japar in developing the performing arts of Longser in Bandung regency, West Java, Indonesia, in the period of 1975-2002. The main issue discussed is the development of Longser performing arts organized by Ateng Japar from 1975-2002. The method used in this research is the historical method. The results showed that Longser is the art of theater folks who live in the Priangan region, especially in Bandung regency. Longser, often referred to as a theatrical art, displays many folk dances in pairs and provide an opportunity for the audience to dance along with the dancers. In 1939, Ateng Japar established a Longser group named “Pancawarna” (five colours). Initially, this group performed by singing, but by the 1970s, there was a shift in the venue into an art gallery. In 2002, the artist Ateng Japar passed away, and his death negatively affected the development of Longser. Besides, the influence of globalization followed by a change of more advanced and modern age, eventually, makes the performing arts of Longser, although still survives today, less appreciated by the wider community. Methyltransferase like-3 (METTL3) and METTL14 complex transfers a methyl group from S-adenosyl-L-methionine to N6 amino group of adenosine bases in RNA (m6A) and DNA (m6dA). Emerging evidence highlights a role of METTL3-METTL14 in the chromatin context, especially in processes where DNA and RNA are held in close proximity. However, a mechanistic framework about specificity for substrate RNA/DNA and their interrelationship remain unclear. By systematically studying methylation activity and binding affinity to a number of DNA and RNA oligos with different propensities to form inter- or intra-molecular duplexes or single-stranded molecules in vitro, we uncover an inverse relationship for substrate binding and methylation and show that METTL3-METTL14 preferentially catalyzes the formation of m6dA in single-stranded DNA (ssDNA), despite weaker binding affinity to DNA. In contrast, it binds structured RNAs with high affinity, but methylates the target adenosine in RNA (m6A) much less efficiently than it does in ssDNA. We also show that METTL3-METTL14-mediated methylation of DNA is largely restricted by structured RNA elements prevalent in long noncoding and other cellular RNAs.

Ekosistem lamun merupakan salah satu ekosistem yang memiliki kompleksitas dan keanekaragaman hayati yang tinggi. Padang lamun merupakan hamparan vegetasi lamun yang menutupi suatu kawasan pesisir. Selain memiliki fungsi ekonomi, lamun juga memiliki fungsi ekologis yakni berperan penting sebagai pendaur zat hara oleh mikroorganime yaitu bakteri. Penelitian ini bertujuan untuk mengetahui kerapatan lamun, kelimpahan bakteri heterotrof yang berasosiasi dengan lamun serta pengaruh kerapatan lamun dengan kelimpahan bakteri heterotrof di Pantai Prawean, Jepara. Metode yang digunakan yakni deskriptif eksplanatif dengan pengambilan sampel secara purposive dan dianalisis dengan IBM SPSS Statistic 22. Jenis lamun yang ditemukan di Pantai Prawean ada 5 (lima): Enhalus acoroides, Thalassia hemprichii, Cymodocea rotundata, Halodule uninervis dan Halodule pinifolia. Kerapatan tertinggi didapat dari jenis Thalassia hemprichii sebesar 78 Ind/m2 dan terendah adalah Enhalus acoroides 10 Ind/m2 dan kelimpahan bakteri heterotrof tertinggi diperoleh dari tingkat kerapatan rapat di stasiun 3 yakni 29,4x108 Upk/ml dan kelimpahan terendah diperoleh dari tingkat kerapatan jarang di stasiun 2 yakni 3,3x108 Upk/ml. Korelasi antara kerapatan lamun dengan kelimpahan bakteri heterotrof tinggi atau kuat yakni 0,896 dan korelasi ini dinyatakan sangat signifikan terbukti nilai sig. 0,001 dengan tingkat kesalahan 0,1%. Artinya bertambahnya kerapatan lamun dapat meningkatkan pula kelimpahan bakteri heterotrof. Seagrass ecosystem is one ecosytems that has high complexity and biodiversity. Seagrass beds are a stretch of seagrass vegetation that covers a coastal area. Beside its economic function, seagrass also have ecological function that play an important role of nutrient cycle for microorganism its bacteria. This study aims to determine the density of seagrass, the abundance of heterothropic bacteria and influence of seagrass density with abundance of heterotrophic bacteria at Prawean beach, Jepara. The method used in this study is descriptive explanative with purposive sampling and the data analyzed by IBM SPSS Statistic 22. There are 5 (five) species of seagrass that can be found in Prawean beach: Enhalus acoroides, Thalassia hemprichii, Cymodocea rotundata, Halodule uninervis and Halodule pinifolia. The highest density obtained from Thalassia hemprichii species is 78 sprouts of seagrass/m2 and the lowest density obtained from Enhalus acoroides is 10 obtained from seagrass density at station 3 its value 29,4x108Cfu/ml and the lowest abundance of heterotrophic bacteria was obtained from rare seagrass at station 2 its value 3,3x108Cfu/ml.  The corelation between seagrass density with abundance heterotrophic bacteria is high or strong that has value 0,846 and this correlation is very significantly proven has sig value 0,001 with error rate 0,1%, it can be conclude that increase of seagrass density can also increase the abundance of heterotrophic bacteria.
