This study aimed at providing a route towards the production of a novel exopolysaccharide (EPS) from fermented bamboo shoot-isolated Lactobacillus fermentum. A lactic acid bacteria strain, with high EPS production ability, was isolated from fermented bamboo shoots. This strain, R-49757, was identified in the BCCM/LMG Bacteria Collection, Ghent University, Belgium by the phenylalanyl-tRNA synthetase gene sequencing method, and it was named Lb. fermentum MC3. The molecular mass of the EPS measured via gel permeation chromatography was found to be 9.85 × 104 Da. Moreover, the monosaccharide composition in the EPS was analyzed by gas chromatography–mass spectrometry. Consequently, the EPS was discovered to be a heteropolysaccharide with the appearance of two main sugars—D-glucose and D-mannose—in the backbone. The results of one-dimensional (1D) and two-dimensional (2D) nuclear magnetic resonance spectroscopy analyses prove the repeating unit of this polysaccharide to be [→6)-β-D-Glcp-(1→3)-β-D-Manp-(1→6)-β-D-Glcp-(1→]n, which appears to be a new EPS. The obtained results open up an avenue for the production of novel EPSs for biomedical applications.

Polyhydroxyalkanoates (PHAs) are microbially synthesized biopolyesters which have attracted great attentions as a new biological material, potential alternative to traditional fossil fuel-based plastic due to their biodegradability and biocompatibility. Poly-3-hydroxybutyrate (PHB) and poly-(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) are the most common members of PHAs. In this study, the nonsulfur and facultatively phototrophic bacterium Rhodospirillum rubrum was cultivated to accumulate PHA by a two-step culture strategy. Gas chromatography (GC), gas chromatography-mass spectrometry (GC-MS), and nuclear magnetic resonance spectroscopy (NMR) analyses showed that PHAs synthesized from fructose was PHBV, in which the 3HV content was 46.5 mol%, which means the better mechanical property. The molecular weight, distribution, and thermal features were characterized by gel permeation chromatography (GPC), differential scanning calorimeter (DSC), and thermo gravimetric analysis (TGA), respectively. The low PDI of 1.08 revealed the narrow and evenly molar mass distribution which shows the stable features. The high melting temperature and their other physical properties implied their potential applications. The traditional process of producing PHBV involves related carbon sources such as valeric acid. However, our study clearly described a new medium formula with fructose and a complete fermentation method to produce PHBV with a high 3HV faction and low molecular distribution.

Bamboo shoots are considered as one of the useful health foods because of their rich contents of proteins, carbohydrates, vitamins, fibres, and minerals and very low fat. Though bamboo shoots provide lots of health benefits, their consumption is confined mostly to Southeast Asian and East Asian countries. The acceptability of bamboo shoots as popular vegetable crop is very less due to their high pungent smell and bitter acidic taste. The use of bamboo as food in India is mainly restricted to Northeastern part of the country where they form an indispensable part of several traditional speciality dishes. The different ethnic communities take fresh or fermented bamboo shoot as one of most preferred traditional food items. Some of the important bamboo based traditional foods are ushoi, soibum, rep, mesu, eup, ekhung, hirring, and so forth. Bamboo shoots should be properly processed before they are consumed as freshly harvested shoots have high content of toxic cyanogenic glycosides which may pose serious health problems. The prospect of bamboo shoot industry in Northeast India is bright due to its rich genetic resources of bamboos. However, habitat destruction and extensive use of bamboos for food, handicraft, and construction purposes have resulted in severe depletion of natural bamboo resources. This review stresses upon the high nutritive values and health benefits of bamboo shoots and their usage as important traditional foods in Northeast India. The bamboo market potential of the region and use of in vitro plant micropropagation methods as effective means of bamboo conservation are also emphasized in this paper.

In Streptococcus thermophilus, EpsC is a polysaccharide co-polymerase which is involved in determining the chain length of EPS synthesized by the Wzx/Wzy-dependent pathway. Our previous study found that there was a positive correlation between transcription level of epsC and molecular weight of EPS in S. thermophilus 05-34. To further investigate the effects of EpsC on EPS biosynthesis, this gene was over-expressed in S. thermophilus 05-34 in this study. Reverse transcription qPCR and Western blotting confirmed the successful transcription and translation of epsC in 05-34, respectively. The yield of EPS was not affected by the over-expression of EpsC. Gas chromatography-mass spectrometry (GC-MS) showed that the monosaccharide composition was still composed of galactose and glucose in a molar ratio of 1.0:0.8, whereas high performance gel permeation chromatography (HPGPC) indicated that the molecular weight of EPS was increased from 4.62 × 105 Da to 9.17 × 105 Da by the over-expression of EpsC. In addition, S. thermophilus 05epsC which could produce higher molecular weight EPS improved the viscoelasticity and water-holding capacity of yogurt, but significantly reduced the level of syneresis in yogurt. In summary, these results indicated that homologous over-expression of EpsC in S. thermophilus could increase the molecular weight of EPS and improve the microrheological or physical properties of yogurt.

SummaryUltra-low-molecular-weight heparins (ULMWHs) with better efficacy and safety ratios are under development; however, there are few structural data available. The main structural features and molecular weight of ULMWHs were studied and compared to enoxaparin. Their monosaccharide composition and average molecular weights were determined and preparations studied by nuclear magnetic resonance spectroscopy, scanning ultraviolet spectroscopy, circular dichroism and gel permeation chromatography. In general, ULMWHs presented higher 3-O-sulphated glucosamine and unsaturated uronic acid residues, the latter being comparable with their higher degree of depolymerisation. The analysis showed that ULMWHs are structurally related to LMWHs; however, their monosaccharide/oligosaccharide compositions and average molecular weights differed considerably explaining their different anticoagulant activities. The results relate structural features to activity, assisting the development of new and improved therapeutic agents, based on depolymerised heparin, for the prophylaxis and treatment of thrombotic disorders.

One of the solutions to fulfill  the food sustainability is diversification of local food. One of the local food that potential to be used and processed is bamboo shoots. In South Kalimantan, the potential of bamboo as a producer of bamboo shoot plants with an estimated total area of 2158 hectares with a potential of 6 million stems. To increase the value and health of bamboo shoots can be made with fermentation. Fermentation is done by two methods, enzimatic fermentation and spontaneous fermentation. The results of the highest crude fiber obtained in spontaneous fermentation of bamboo shoots Haur 44.46% while the highest starch content present in fermented bamboo shoots Paring stater of 13.91%. Metal content, everything is still fulfill the quality standard. Flour bamboo shoots can be used as food supplements or raw materials of fiber flour.Keywords: bamboo shoots,  fermentation, fiber flour

Abstract The effects of thermal compression on the organic-soluble material of a uniform wood substrate, hybrid poplar (clone OP-367), has been studied. Poplar veneers were preconditioned to 0% or 8% moisture content and subsequently hot pressed at 150°C, 200°C, and 250°C. The dichloromethane (DCM) extracts were characterized by various hyphenated analyses [gas chromatography-mass spectrometry (GC-MS), analytical pyrolysis (Py-GC-MS), electrospray ionization-mass spectrometry (ESI-MS), and gel permeation chromatography (GPC)]. The yields and composition of extractable compounds were markedly influenced by treatment at 250°C compared to those obtained at lower temperatures. Most importantly, the organic extracts contained a significant portion of lignin-derived compounds after treatment at 250°C.

Dialdehyde starches with different aldehyde content from native corn starch were prepared by sodium periodate oxidation to be used as a tanning agent in leather making. For this purpose, native corn starch was oxidized with sodium metaperiodate in different molar ratios. After oxidation processes, the yields, solubility in water and aldehyde contents of the obtained dialdehyde starches were determined as well as structure characterizations by Proton Nuclear Magnetic Resonance Spectroscopy, Fourier Transform Infrared Spectroscopy and Gel Permeation Chromatography. Evaluating the gel permeation chromatography data, the dialdehyde starch samples which were thought to be in appropriate molecular weight/size to penetrate into skin fibers were selected to be used in the tanning process. Their tanning abilities were evaluated by investigating hydrothermal stabilities, filling and fiber isolation characteristics and physical properties determined by mechanical tests and organoleptically. From the evaluation of the results, it was revealed that sodium metaperiodate oxidized starches which have appropriate molecular weight and adequate aldehyde content has a remarkable tanning effect and can be utilized as a tanning agent with the advantages of not necessitating pickling process which means saving time and simplifying the production but more importantly offering an important advantage from an environmental point of view.

A novel flame retardant polymethylsilsesquioxane (PMSQ) was successfully obtained via combination of non-hydrolytic and hydrolytic sol-gel routes. Chemical structure of the resultant PMSQ was determined by nuclear magnetic resonance spectroscopy, Fourier-transform infrared spectrometry and powder X-ray diffraction, respectively. All the measurements demonstrated that the product possessed regular structure with chain-to-chain width of 0.87nm and chain thickness of 0.40nm. Weight average molecular weight of PMSQ was measured to be 3.5×105 using gel permeation chromatography. Numerical simulations of the molecular structure suggested that PMSQ should exhibit cis-isotactic configuration and double helical conformation at undisturbed condition.
