For the first time, organogenesis and physiological characteristics of Fragaria ananassa microclones (cvs. ‘Alpha’ and ‘Solnechnaya polyanka’) under the influence of mechanocomposite (MC) based on rice husks amorphous silica and flavonoids of green tea during the multiplication stage in in vitro conditions were studied. The addition of the MC (0.0, 2.5, 5.0 and 10.0 mg·L-1) to the Gamborg- Eveleg’s basal salt medium supplemented with 0.75 mg·L-1 6-benzylaminopurine has shown beneficial action on processes of organogenesis followed by enzymatic, photosynthetic, and hormonal activities of in vitro cultured strawberry plantlets. In both cultivars, the high frequency of proliferation (100 %) and maximum number of axillary shoots increased by 1.8–2.0 times on medium supplemented with 5.0 mg·L-1 MC. The concentrations of 2.5 and 5.0 mg·L-1 MC were optimal for obtaining plantlets with high physiological state in in vitro conditions. The results may be used for the development of production systems for a healthy planting material using biotechnological approaches and recommended for commercial strawberry micropropagation.

Culture medium requirements for micropropagation of Isoplexis chalcantha was achieved for the first time after high degree of contamination and phenolic exudates were detected and solved. Cultures were established from axillary shoots using juvenile branches collected from this medicinal plant. Most satisfying results were obtained using a solidified and a modified MS medium (NO3- : NH4+ ratios) enriched with ascorbic acid or soluble PVP plus GA3, BAP and NAA. Explants (nodal segments) were used for in vitro shoots multiplication and best results were achieved with modified MS plus BAP and auxins. AbstractIn this study, the effects of compounds providing Fe in chelated (NaFeEDTA and Fe(III)AC) and non-chelated (FeSO4·7H2O) forms as components of culture media, onin vitroshoot multiplication and rooting ofMagnolia soulangeana‘Alexandrina’,Magnolia grandifloraandPrunus cerasifera‘Nigra’ were comparatively evaluated. Each of the tested chemicals was used as a single Fe source in the basal salt medium. In the stages of shoot multiplication and rooting plant response was scored by biometrical indices (number of shoots, leaves and roots, shoot and root length, percent of rooted plants and root hairs). The occurrence of physiological disorders was estimated by visual observations. In presence of FeSO4, symptoms of chlorosis, hyperhy-dricity, early senescence and specific morphology of roots, suggesting Fe deficiency, were observed. These deteriorations were entirely prevented at the application of Fe chelates of which, in this experimental systems, Fe(III)AC was tested for the first time. The addition of Fe(III)AC positively affected the plant quality to extent comparable to that of NaFeEDTA. The obtained data suggest that both applied Fe chelates are more appropriate than non-chelated Fe form and can be alternatively used in the optimization of nutrient media for micropropagation ofMagnoliaandPrunus cerasiferagenotypes.

AbstractThe coconut palm or “tree of life” is one of nature’s most useful plants and the demand for its fruit is increasing. However, coconut production is threatened by ageing plantations, pests and diseases. Currently, the palm is exclusively propagated via seeds, limiting the amount of planting material. A novel micropropagation method is presented, based on axillary shoot formation. Apical meristems of in vitro coconut seedlings are cultured onto Y3 medium containing 1 µM TDZ. This induces the apical meristem to proliferate through axillary shoots in ~ 27% of the initiated explants. These axillary shoots are seen as white clumps of proliferating tissue and can be multiplied at a large scale or regenerated into rooted in vitro plantlets. This innovative micropropagation method will enable the production of disease-free, high quality in vitro plantlets, which will solve the worldwide scarcity of coconut planting material.

This long term study demonstrates for the first time that it is possible to propagate embryogenic Vigna trilobata and to subsequently initiate the differentiation of embryos into complete plantlets. Initiation of callus was possible on 2,4-D. Somatic embryos differentiated on modified MS basal nutrient medium with 1.0 mg/l  of 2,4-D and 0.5 mg/l  of Kn. Sustained cell division resulted in globular and heart shape stages of somatic embryos. Transfer of embryos on to a fresh modified MS basal medium with 0.5 mg/l of Kn and 0.5 mg/l of GA3 helped them to attain maturation and germination. However, the propagation of cells, as well as the differentiation of embryos, were inhibited by a continuous application of these growth regulators. For this reason, a long period on medium lacking these growth regulators was necessary before the differentiation of embryos occurred again. ABSTRACT Of the information available on steroid hormone metabolism in responsive tissues, only that relating hormone metabolism to physiological activity is reviewed, i. e. metabolite activity in isolated in vitro systems, binding of metabolites to target tissue receptors, specific steroid hormone metabolizing enzymes and relationship of hormone metabolism to target organ physiological state. Further, evidence is presented in the androgen field, demonstrating 5α-reduced metabolites, formed inside the target cells, as active compounds. This has led to a consideration of testosterone as a »prehormone«. The possibility that similar events take place in tissues responding to progesterone is discussed. Finally, the role of hormone metabolism in the regulation of hormone availability and/or renewal in target cells is discussed. In this context, reference is made to the potential role of plasma binding proteins and cytosol receptors.

Epimedium elatum (Morren & Decne) of family Berberidaceace is a rare perennial medicinal plant, endemic to high altitude forests of Northwestern Himalayas in India. Ethnobotanically, it has been used as an ingredient for treatment of bone-joint disorders, impotence and kidney disorders in Kashmir Himalayas. Phytochemically, it is rich in Epimedin ABC and Icariin; all of these have been demonstrated to possess remarkable biological activities like PDE-5 inhibition (treatment of erectile dysfunction), anticancer, antiosteoporosis antioxidant and antiviral properties. The present investigation reports its traditional usage, comprehensive distribution and conservation status from twenty ecogeographical regions in Kashmir Himalayas, India. The species was reported from Gurez valley for the first time. Numerous threats like excessive grazing, deforestration, habitat fragmentation, tourism encroachment, landslides and excessive exploitation have decreased its natural populations in most of the surveyed habitats. Consequently, its existence may become threatened in near future if timely conservation steps are not taken immediately by concerned stakeholders involved in medicinal plant research. Moreover, use of plant tissue culture techniques is recommended for development of its in vitro propagation protocols. Therefore, introduction of this medicinal plant in botanical gardens, protected sites and development of monitoring programmes are needed for its immediate conservation in Northwestern Himalayas, India.

The only drug for treatment of Schistosomiasis is Praziquantel, and the possible emergence of resistance makes research on novel therapeutic agents necessary. Targeting of Schistosoma mansoni epigenetic enzymes, which regulate the parasitic life cycle, emerged as promising approach. Due to the strong effects of human Sirtuin inhibitors on parasite survival and reproduction, Schistosoma sirtuins were postulated as therapeutic targets. In vitro testing of synthetic substrates of S. mansoni Sirtuin 2 (SmSirt2) and kinetic experiments on a myristoylated peptide demonstrated lysine long chain deacylation as an intrinsic SmSirt2 activity for the first time. Focused in vitro screening of the GSK Kinetobox library and structure-activity relationships (SAR) of identified hits, led to the first SmSirt2 inhibitors with activity in the low micromolar range. Several SmSirt2 inhibitors showed potency against both larval schistosomes (viability) and adult worms (pairing, egg laying) in culture without general toxicity to human cancer cells.<br>

Green tea (Camellia sinensis) is one of traditional plants that have the potential as an anticancer. The sample used in this research commercial green tea extract. The purpose of this study was to test the antiproliferation activity of green tea extract on breast cancer cell MCM-B2 in vitro. Green tea extract fractionated using three solvents, ie water, ethanol 70%, and n-hexane. Extract and fraction of green tea water have value Lethality Concentration 50 (LC50) more than 1000 ppm. The fraction of ethanol 70% and n-hexane had an LC50 value of 883.48 ppm and 600.56 ppm, respectively. The results of the phytochemical screening of green tea extract are flavonoids, tannins, and saponins, while the phytochemical screening results of n-hexane fraction are flavonoids and tannins. Antiproliferation activity was tested on breast cancer cells MCM-B2 and normal cells Vero by trypan blue staining method. The highest MCM-B2 cell inhibitory activity was achieved at a concentration of 13000 ppm green tea extract and 1000 ppm of n-hexane fraction, 59% and 59%, respectively. The extract and n-hexane fraction of green tea are not toxic to normal Vero cells characterized by not inhibiting normal cell proliferation. The different integrated production systems can directly interfere with its bacterial community. The present study aimed to assess density, bacterial diversity and the influence of dry and rainy season in different integrated and an exclusive production system. The fallow and a native forest area was assessed to. Samples were collected in 2012 March and September. The isolation were carried out into Petri dishes containing DYGS medium. The number of colony forming units (CFU) was counted after 48 hours and. The bacterial density ranged between 106 and 107 CFU g-1 soil. The crop system affected the dynamics of the bacterial community only in the rainy season. The rainy season showed greater density of total bacteria when compared to the dry period regardless of the cropping system. The dendrograms with 80 % similarity showed thirteen and fourteen groups in the rainy and dry seasons. Isolates with the capacity to solubilize phosphate in vitro were obtained from all areas in the two seasons, but this feature has been prevalent in bacteria isolated during the rainy season
