Abstract: Alcohol is an organic compound that is composed of the elements carbon, hydrogen and oxygen. Usually, the term of alcohol is known as liquor. One of the local products of liquor in Manado is Cap Tikus. This study aims to determine the effect of alcohol 20% on the spermatozoa quality of male Wistar (Rattus norvegicus). The research design was a completely randomized experimental design. The samples were 10 male wistar rats which consisted of 5 rats as a control group and 5 rats as the experiment group. The rats in the experiment group were given Cap Tikus for 52 days, while rats in the control group were given tap water. The results showed that spermatozoa quality from experiment group which includes the concentration of spermatozoa is 24x106 spermatozoa/ml, normal motility of spermatozoa is 23,5% and normal morphology of spermatozoa is 11%. Whereas spermatozoa quality from control group which includes the concentration of spermatozoa is59x106 spermatozoa/ml, normal motility of spermatozoa is 89% and normal morphology of spermatozoa is 97%. Conclusion: Cap Tikus decreased the spermatozoa quality which includes concentration, motility and morphology of spermatozoa. This was due to alcohol interfered the hypothalamus, the anterior pituitary gland and the testes that played an important role in the process of spermatogenesis.Keywords: cap tikus, alcohol, spermatozoa qualityAbstrak: Alkohol merupakan suatu senyawa organik yang tersusun dari unsur-unsur karbon, hidrogen dan oksigen. Dalam kehidupan sehari-hari, istilah alkohol sering dikaitkan dengan minuman keras. Salah satu jenis minuman keras produk lokal di Manado adalah Cap Tikus. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian Cap Tikus terhadap kualitas spermatozoa wistar jantan (Rattus norvegicus). Desain penelitian yang digunakan adalah eksperimental dengan rancangan acak lengkap. Sampel adalah tikus wistar jantan berjumlah 10 ekor, 5 ekor tikus digunakan sebagaikontrol dan 5 ekor tikus digunakan sebagai perlakuan. Kelompok perlakuan adalah kelompok yang diberi minuman Cap Tikus selama 52 hari sedangkan kelompok kontrol adalah kelompok yang diberi minuman air ledeng. Hasil penelitian menunjukan kualitas spermatozoa kelompok perlakuan yang meliputi konsentrasi spermatozoa adalah 24x106 spermatozoa/ml, motilitas spermatozoa normal adalah 23,5% dan morfologi spermatozoa normal adalah 11%. Kualitas spermatozoa kelompok kontrol yang meliputi konsentrasi spermatozoa adalah 59x106 spermatozoa/ml, motilitas spermatozoa normal 89% dan morfologi spermatozoa normal 97%. Simpulan: Pemberian Cap Tikus menyebabkan penurunan kualitas spermatozoa yang meliputi konsentrasi, motilitas dan morfologi spermatozoa. Hal ini dapat disebabkan karena alkohol mengganggu hipotalamus, kelenjar hipofisis anterior dan testis yang berperan dalam proses spermatogenesis.Kata kunci: cap tikus, alkohol, kualitas spermatozoa

Abstract: It is known that Tribulus terrestris can improve sperm quality. One of the Indonesian traditional medicine, jamu, contains this Tribulus terrestris. This study aimed to investigate the effect of jamu containing Tribulus terrestris on sperm quality which consisted of concentration, motility, and morphology of spermatozoa of male wistar rats (Rattus norvegicus). This was an experimental study. Nine male wistar rats were randomly divided into 3 groups of 3 rats each: Group P0 (control), group P1 (given 5 mg of Tribulus terrestris), and group P2 (given 10 mg of Tribulus terrestris). This study was conducted for 52 days. The results showed that the spermatozoa concentration of P1 was 49x106 sperm/ml suspension and of P2 53x106 sperm/ml suspension; both were lower than of P0 (59x106 sperm/ml suspension). The sperm motility category (A+B) of P1 was 33% and of P2 (37%); both were lower than of P0 (45%). The motility category A was not found in P1 and P2. The percentage of spermatozoa with normal morphology of P1 was 45% and the abnormal ones was 55%, meanwhile in P2 there was 49% of normal sperm morphology and 51% of abnormal morphology. Conclusion: Jamu containing Tribulus terrestris had no effect in improvement of the sperm quality.Keywords: jamu, Tribulus terrestris, sperm qualityAbstrak: Tribulus terrestris dikenal sebagai bahan yang dapat memperbaiki kualitas sperma. Penelitian dilakukan selama 52 hari. Hasil penelitian menunjukkan bahwa konsentrasi spermatozoa P1 49x106 sperma/ml suspensi dan P2 53x106 sperma/ml suspensi lebih rendah dibandingkan P0 59x106 sperma/ml suspensi. Motilitas spermatozoa kategori A+B pada P1 (33%) dan P2 (37%) lebih rendah dibandingkan pada P0 (45%). Tidak ditemukan motilitas kategori A pada P1 dan P2. Morfologi spermatozoa normal pada P1 sebesar 45% dan abnormal 55%. P2 memiliki morfologi spermatozoa normal sebesar 49% dan abnormal 51%. Simpulan: Pemberian jamu dengan Tribulus terrestris tidak meningkatkan kualitas sperma yang mencakup konsentrasi, motilitas, dan morfologi spermatozoa.Kata kunci: jamu, Tribulus terrestris, kualitas sperma

The purpose of this study was to prove that the administration of Jati (Guazuma ulmifolia Lamk) leaves extract improves the lipid profile of dyslipidemic male Wistar rats. Subjects were 20 rats (Rattus norvegicus), male, Wistar strain, dyslipidemia (total cholesterol ≥ 200 mg dl-1), aged 2 months old, weighing 180-200 grams. The control group (10 rats) were given a placebo of 3 ml aquadest (P0) and the treatment group was given extracts of the Jati (Guazuma ulmifolia Lamk) leaves extract of 25 mg kg-1 BW (P1). Before and after treatment for 14 days, total cholesterol, triglyceride, LDL, and HDL levels were examined. The results showed that in the P0 group there were no changes in total cholesterol, triglyceride, LDL, and HDL levels (p>0.05), whereas the P1 group experienced a decrease in total cholesterol, triglyceride and LDL levels (p<0.05) and an increase in HDL levels (p<0.05). The results of this study indicated that the Jati leaves extract was effective to improve the lipid profile of dyslipidemic rats. It was necessary to compare the effectiveness of Jati leaves extract with synthetic dyslipidemia drugs used in the community such as statin.

Abstract Background We investigated the effects of a polyherbal formulation prepared from the extracts of Mondia whitei (Periplocaceae), Dracaena arborea (Dracaenaceae), and Bridelia ferruginea (Euphorbiaceae) (MDB) on the sexual behavior of normal rats (NR) and prediabetic rats (PR). Methods Male Wistar rats were administered with drinking fructose solution (21%) or tap water for 16 weeks. After induction of prediabetic status, NR (n = 30) and PR (n = 30) were randomly distributed into 10 groups of six animals each and orally treated with distilled water (10 mL/kg), sildenafil citrate (5 mg/kg), or MDB (50, 100, or 500 mg/kg) for 21 days. Sexual behavior parameters per series (S) of ejaculation were evaluated on days 1, 7, 14, and 21. Results Drinking fructose solution (21%) induced prediabetic status in rats, characterized by a significant (p < 0.01) increase in glycemia by 43.41% compared with the control group. MDB improved sexual performances of NR and PR by increasing the mount frequency (MF) and the intromission frequency (IF) as well as the number of rats capable of ejaculating. For instance, the MF and the IF were significantly increased in animals administered with MDB for 7 (50 mg/kg, S1 and S2), 14 (100 mg/kg, S3), or 21 days (100 or 500 mg/kg, S2). This increase was more pronounced on days 7 and 14 in NR and PR treated with sildenafil citrate or MDB extracts, respectively. Conclusions The mixture of MDB improved sexual activity in NR and PR. This result may further justify the traditional use of these plants as sexual performance enhancers.

AIM: This research aims to examine the effects of soy milk on mounting latency (ML), mounting frequency (MF), estrogen levels, androgen-binding protein (ABP) expression, and spermatogenesis in male rats (Rattus norvegicus). METHODS: Twenty-four male wistar rats (Rattus norvegicus) aged 4 weeks were divided into four groups. Control group (given a normal diet), P1; P2; P3 (given the normal diet and soy milk powder at doses of 7.1; 14.2; 21.3 g/KgBW/day, respectively) for 6 weeks. Observation of ML and MF were performed at 9 weeks 5 days of age, and rat surgery was performed at 10 weeks of age. Analysis of estrogen hormone levels was conducted by enzyme-linked immunosorbent assay (ELISA), ABP staining was using immunohistochemistry method, testicular spermatogenesis was observed using histopathological methods, and observation of spermatozoa was performed under the microscope.  RESULTS: The results showed no significant reduction of ML and MF, estrogen levels, and ABP expression (p ≤ 0.256; 0.865; 0.959, respectively) in male rat, but there was a significant decrease in the number, morphology, motility of spermatozoa, and testicular histophatology, (p ≤ 0.000, 0.003, 0.008, 0.000, respectively). CONCLUSION: The administrassion of soy milk in various doses (7.1;14.2;21.3 g/KgBW/day) in male Wistar rats (Rattus norvegicus) had showed significantly difference on histopathological evaluation using Johnson’s scoring system, sperm quantity and quality, while on mounting latency and frequency, estrogen levels, and ABP expressions did not show significantly difference between groups. That describe of isoflavone in soy milk can affect several aspects related to male endocrine and reproductive development.

Objective: Smoking are known to have detrimental effect on sperm. The effects are suspiciously come from nicotine as its main pharmacologic component. This study is carried out to analyze the effect of nicotine exposure per inhalation to the difference of motile sperm percentage and normal morphology sperm percentage of male wistar rats. Material & Methods: Thirty male rats were used for this research. They were randomly divided into 5 groups and treated with nicotine inhalation 0.5 mg/kgBW,  1 mg/kgBW, 2 mg/kgBW and 4 mg/kgBW for 30 days while the control rats were not given any treatment. At the end of experiment, sperm analysis was carried out to measure the percentage of motile sperm and percentage of sperm with normal morphology. Results: Percentage of motile sperm significantly decreased (p<0.05) in all treatment groups compared to control group. Comparison between treatment groups revealed that the percentage of motile sperm significantly decreased in 0.5 mg/kgBW vs 2 mg/kgBW, 0.5 mg/kgBW vs 4 mg/kgBW, and 1 mg/kgBW vs 4 mg/kgBW. Percentage of normal morphology sperm decreased significantly (p<0.05) in 1 mg/kgBW, 2 mg/kgBW, and 4 mg/kgBW compared to control groups. Comparison between treatment groups revealed that there were no significant decreased between each treatment groups. Conclusion: This research conclude that nicotine exposure per inhalation can deteriorate the percentage of motile sperm and the percentage of normal morphology sperm of male wistar rats.

Introduction: Diabetes mellitus is a complex and progressive disease which often lead to several debilitating complications that partly caused by by free radicals which can be overcome with antioxidants. Bay leaves contain essential oils, tannins, flavonoids and terpenoids which have considerable antioxidant properties. Therefore, this study aims to determine the effect combination treatment of vildagliptin and bay leaf (Syzygium polyanthum) extract toward pancreatic beta cells density and glycated albumin levels in diabetic male Wistar rats (Rattus norvegicus). Methods: An experimental posttest only control group study was conducted using 36 albino male rats, aged 2-3 months, weighing 180-200 grams. The rats were divided into 2 groups (n= 18) with control group treated with 1.8mg/200g body weight vildagliptin and 2cc placebo (aquabidest) while the treatment group received 1.8mg/200g BW vildagliptin and 250 mg/200g BW bay leaves extract. All treatment lasted for 21 days. Results: The results showed that pancreatic beta cell counts in the treatment group  was significantly higher than the control group  (109.07 ± 20.47 cells/field of view vs 90.87 ± 13.91 cells/field of view; p<0.01). However, the levels of Glycated albumin between two groups were not significantly different (treatment vs control: 17.33 ± 4.51 vs 20.18 ± 4.57; p=0.068). Conclusion: This study suggested that combination treatment of vildagliptin and bay leaf extract increased pancreatic beta cells but did not reduce glycated albumin levels in diabetic male Wistar rats.

Introduction: Monosodium glutamate (MSG) is one of the most widely employed food enhancers. Although the umami compound, controversy persists regarding the effects of MSG intake on body weight. Chronic MSG intake may result in excessive body weight gain and obesity. Consumption of MSG result in organ damage, cardiovascular disease, oxidative stress, and also risk factors for obesity. This study aims to determine the effect of oral MSG on obesity in adult male Wistar rats (Rattus norvegicus).Methods: This true experimental study used the post-test control group design. Twenty-four adult male Wistar rats were randomly divided into four groups: control (received distilled water), Group 1 (MSG 0.378 mg/gr BW), Group 2 (0.756 mg/gr BW) and Group 3 (1.512 mg/gr BW). The obesity parameter was obtained by the Lee index. Kruskal-Wallis test follows by Mann-Whitney test were used to compare the Lee index between groups.Results: Lee’s index mean for each group was 358.4%, 314.1%, 287.8%, and 320.9%, respectively. The Kruskal Wallis test showed a significant difference in the Lee index between groups (p = 0.043). A follow-up test using Mann-Whitney found a significant difference between group 2 and the control group (p = 0.043, p <0.05). The mean of Lee index of group 2 was 70.51% lower than the control group.Conclusion: This study concluded that Lee index was not increased in MSG-treated rats than in the control group after oral MSG intervention for 21 days.

Background: Physical exercise aims to improve or maintain physical fitness. However, excessive physical exercise may cause increase of oxidative stress which leads to cellular injury, including in the proximal tubules of kidney. This research aims to find out the effect of single- and repeated- excessive swimming exercise to the kidney histopathology of male wistar rats (Rattus norvegicus). Methods: This research was an experimental study with complete random and post test only control group design. Twenty seven male wistar rats were divided into three treatment groups: control group, single- excessive swimming exercise group (45 minutes for one day), and repeated- excessive swimming exercise group (45 minutes/day for seven days). In the end of the treatment, the kidney of the rats were taken to be analyzed by using hematoxylin-eosin stain. The mean of proximal tubules injury of kidneys were counted by two observers using blinded method. Data were analyzed by using one way anova test. Result: The analysis showed there was a significant difference in the mean percentage of proximal tubules injury between control group and single swimming group as well as the repeated swimming group (Anova p=0,000), also there was significant difference between the treatment groups (p=0,020). The highest average of proximal tubules injury was in the single swimming group. Conclusion: Single- and repeated- excessive swimming exercise causes the proximal tubules injury of kidney.

Abstract: Excessive physical activity increases the consumption of oxygen which can result in increased levels of reactive oxygen species (ROS) causing oxidative stress characterized by the elevation of F2-isoprostane as a byproduct of lipid peroxidation. Willow (Justicia gendarussa Burm. f.) has an antioxidant activity that is able to inhibit and break the chain reaction of free radicals and prevent lipid peroxidation. This study was aimed to prove that willow leaf extract could inhibit the elevation of F2-isoprostane levels in male Wistar rats (Rattus norvegicus) urin induced by excessive physical training. This was a true experimental study using 16 male Wistar rats (Rattus norvegicus), aged 2-3 months, weighing 180-200 g as subjects. The control group (P0) was treated with a placebo and excessive physical training, and the treatment group (P1) was treated with the willow leaf extract of 200mg/rat/day and excessive physical training. Before and after 14 days of treatment, the urine samples were collected and their levels of F2-isoprostane 8-isoPGF2α were examined by using enzyme immunoassay kit (EIA). The results showed that the mean level of F2-isoprostane in the control group (P0) before treatment (pretest) was 5.20±0.483 ng / mL while in the treatment group (P1) was 5.46±0.655 ng / mL (P > 0.05). On the other hand, after treatment for 7 days (post-test), the mean level of F2-isoprostane in the control group (P0) was 5.34±0.476 ng/mL and in the treatment group (P1) was 3.61±0.389 ng/mL (P < 0.01). Conclusion: The extract of willow (Justicia gendarussa Burm. f.) leaf could inhibit the elevation of F2-isoprostane levels in male Wistar rats (Rattus norvegicus) urin induced by excessive physical training.Keywords: willow leaf, F2-isoprostane, excessive physical trainingAbstrak: Aktivitas fisik berlebih akan meningkatkan komsumsi oksigen yang dapat mengakibatkan peningkatan kadar reactive oxygen species (ROS) sehingga menyebabkan stres oksidatif yang ditandai dengan meningkatnya F2-isoprostan sebagai salah satu produk peroksidasi lipid. Tanaman gandarusa (Justicia gendarussa Burm. f.) mengandung antioksidan yang dapat mencegah reaksi peroksidasi lipid. Penelitian ini bertujuan untuk membuktikan bahwa pemberian ekstrak daun gandarusa dapat menurunkan kadar F2-isoprostan urin tikus Wistar (Rattus norvegicus) jantan yang diinduksi latihan fisik berlebih. Jenis penelitian ialah eksperimental dengan subjek 16 ekor tikus (Rattus norvegicus), galur Wistar, sehat, berumur 6 bulan, berat badan 180-200 gr, dengan kadar F2-isoprostan >2ng/mL. Kelompok kontrol (P0) diberikan pelatihan fisik berlebih dan plasebo aquadest 2ml sedangkan kelompok perlakuan (P1) diberikan pelatihan fisik berlebih dan ekstrak daun gandarusa dosis 200mg/ekor/hari. Sebelum dan setelah 14 hari perlakuan, sampel urin ditampung dan diperiksa kadar F2-isoprostan menggunakan 8-isoPGF2α enzyme immunoassay kit (EIA) dari assay design untuk data pretest dan post-test. Hasil penelitian menunjukkan rerata kadar F2-isoprostan pada kelompok P0 sebelum perlakuan (pretest) ialah 5,20±0,483 ng/mL, sedangkan pada kelompokP1 ialah 5,46±0,655 ng/mL (P > 0,05). Setelah perlakuan selama 14 hari (post-test), rerata kadar F2-isoprostan pada kelompok P0 ialah 5,34±0,476 ng/mL dan pada kelompok P1 ialah 3,61±0,389 ng/mL (P < 0,01). Simpulan: Ekstrak daun gandarusa (Justicia gendarussa burm. f.) dapat menurunkan kadar F2-isoprostan urin tikus Wistar (Rattus norvegicus) jantan yang diinduksi latihan fisik berlebih.Kata kunci: daun gandarusa, F2-isoprostan, pelatihan fisik berlebih

Background: Artesunate (AS) is an artemisinin antimalarial drug used as a single drug or in combination with other antimalarials. Objective: This study was to find its effect on some brain biomolecules and behavioural activities in Wistar rats. Methods: Forty adult male Wistar rats weighing between 150- 180g were divided into four groups of A, B, C and D with 10 animals each. Group A served as the control that received tap water, while groups B, C and D served as the experimental groups that received 2.85mg/kg (therapeutic dose-TD) and 5.71mg/kg (high pharmacologic dose-HPD) of AS per day for 3 days, and 2.85mg/kg (long duration therapeutic dose -LDTD) of AS per day for six days respectively. Half of the dose was administered twelve hourly (twice a day), and twelve hours after the last treatments, behaviour test using the ‘open field maze' was carried out. Immediately after, the animals were sacrificed with chloroform anaesthesia and the whole brain removed and weighed. Whole brain homogenates were used to determine brain total protein (TP), triacylglycerol (TAG) and cholesterol (CH).Data were analyzed statistically by ANOVA and Tukey-Kramer Multiple Comparative Test as applicable. Results: There were no difference (p<0.05) between the experimental groups and the control group in the anthropometric parameters and behavioural activities. In the brain biomolecules concentration, TP was lower in concentration in the HPD group, TAG was lower in concentration in the LDTD group, while the HPD and LDTD groups had lower CH concentration compared to the control. In all the parameters studied no difference was found between the TD group and the control. Conclusion: AS at recommended dose may not affect some behaviour and brain biomolecule concentration, unlike when taken in excess of dose and or time. Even at these doses/time there may have been no behavioural manifestation.
