This study aims to determine the dose of Trichoderma koningii which can induce soursop germination; know the best soaking time that can affect the induction of soursop germination; to know the interaction between soaking time and Trichoderma koningii dose on soursop germination. This research was conducted in April - June 2019 at the Green House of Plant Sciences, Faculty of Agriculture, Sam Ratulangi University, Manado. The material used in this study was soursop seed obtained from Lota Village, Pineleng Tomohon Subdistrict, Trichoderma koningii with 106 solids obtained from the North Sulawesi Province Plant Protection Center Collection, Aquadest, 70% Alcohol, 80% Acetone, sandalready sifter and sterilized. This research was arranged in factorial with a Completely Randomized Design (CRD) consisting of two factors, namely 5 treatments of Trichoderma koningii dose and 4 immersion treatments repeated three times (3x) so that a total of 60 treatment units. Each treatment unituses 10 seeds. The results of this study indicated that the dose of Trichoderma koninggi 200 g without soaking can increase the germination power by 86.67% and the vigor index by 32.96%; Immersion with Koninggi Trichoderma suspension has not been able to induce soursop seed germination rate; Theinteraction between Trichoderma koningii dose and soaking time did not affect the speed of soursop seed germination.*eprm*

Setting storage space temperature and time saving becomes an important application in supporting the development of plantations to provide seeds as needed and sustainable. The purpose of this research is to delay the rapid of seed germination in Magnolia champaca by assessing the effect of various suspend temperatures and times saving on seed germination of Magnolia camphaca . The research was conducted in Plant Physiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University in February until June 2016. The method was used completely randomized design pattern Factorial by 3 factors with 12 treatments and 3 replications with the temperature: temperature ± 31°C, temperature ± 25 °C, temperature ± 18 °C, temperature ± 4 °C and the retention of time 0 week, 2 weeks , 4 weeks and 6 weeks. The parameters include the observation of germination rate, germination, vigor index and sprout morphology. The results of the research the treatment of interaction of temperature and time saving significantly affected on germination rate, germination and seed vigor index of Magnolia champaca. Germination speed, germination and vigor index were both produced by the treatment of storage at a temperature of ± 18 °C and a temperature of ± 25 °C at all times to keep (2 weeks, 4 weeks, 6 weeks) compared to control treatment. A normal sprout produced in temperature treatment ± 18 °C and ± 25 °C temperature ranges between 90-93%. An abnormal sprout produced in control treatment (0 week) by 25%. Measurement of moisture content and physiological maturity level of Magnolia champaca seed needs to be done before it is stored and germinated to test the viability of seeds of sprouts.

The experiment was conducted in the Agronomy Laboratory of Agrotechnology Discipline, Khulna University, Khulna from 29th August to 17th September, 2009. The experiment was laid out in a Completely Randomized Design (CRD) with two factors and three replications. The aim of the experiment was to study the efficacy of three osmopriming agents viz ., NaCl, KC1 and CaCb with control on seed germination and seedling growth of three T. aman rice varities viz., BRRI dhan40, BRRI dhan41 and BINA dhan7. Among the chemicals CaCb showed best as osmopriming agents and it enhanced all germination parameters. BRRI dhan41 showed the best performance. Better root length and shoot length were observed in control. Seeds of BRRI dhan41 when treated with NaCl showed the highest germination percentage (98.67%), germination energy (88%), germination speed (96.83%), but vigor index (10.63) was found highest in BRRI dhan40 when it was treated with KC1. BRRI dhan41 produced the largest root (8.56 cm) when seeds were treated with NaCl. BRRI dhan40 (10.51 cm) and BINA dhan7 (10.53 cm) produced the highest shoot length at controlled condition and CaCl2 treated seed, respectively.   DOI: http://dx.doi.org/10.3329/jbau.v10i1.12013   J. In developing productive forests, seeds play a crucial role. The seeds of forests tree have physical condition of hard seed coat, especially Leguminoceae family. Silvicultural techniques that can overcome seed coat dormancy are dyeing and submersion of seeds. The knowledge and experience in breaking seed dormancy of Sengon (Paracerianthes falcataria L.) is rare, therefore it is necessary to study how to overcome seed dormancy. The objective of this study was to determine the effect of dyeing and submersion treatments to overcome seed dormancy of Sengon seed to improve its viability. This research used Completely Randomized Design (CRD) with two factors: namely Seed (Factor A), dipped in hot water at 60 º C for (A0 = Control, A1 = 2 minutes, A2 = 4 minutes, A3 = 6 minutes), Factor B, seeds soaked in cold water: (B0 = control, B1 = 6 hours of soaking, B2 = 12 hours, B3 = 18 hours, B4 = 24 hours). The parameters observed were germination percentage, germination rate and vigor index. The results showed that among all parameters observed the interaction of factors A1B2, A2B2 and A3B2 were the best treatment for germination percentage which was equal to 100 percent, the best germination rate of 3.897 days and the best vigor index of 27.02

Seed priming is an effective, eco-friendly method to promote seed germination and seedling vigor of okra to overcome the reduced and delayed germination in fresh or stored okra seeds caused by seed hardness. An experiment was carried to evaluate the effects of different priming on okra seeds germination and seedling vigor using Arka Anamika variety at Horticulture lab of Agriculture and Forestry University, Rampur, Chitwan, Nepal. Investigation was carried out with 6 treatments (T1: seed priming with tap water, T2: seed priming with 200ppm NAA solution, T3: seed priming with 10% PEG-200 solution, T4: seed priming with 200ppm GA3 solution, T5: seed priming with 5% Trichoderma solution and T6 no priming) with 4 replications in Complete Randomized Design (CRD). Seeds primed with T1 to T5 were soaked for 24 hours and shade dried for 6 hours before sowing. Priming with T4 was found to be best in terms of maximum seed germination (60.12%), seed vigor index (5772.68 cm), mean germination rate (7.53 seeds per day). The highest shoot length (81.40 mm) was observed at T1 whereas enhancement of root length occurred with the priming with T3. All treatments had a significant positive effect on all the germination parameters in comparison to control. The study concluded that GA3 priming enhanced germination as well as seed vigor in okra and hydro priming and tricho-priming can be used as an alternative to GA3 priming among farmers in Nepal.

Peanut (Arachis hypogaea L.) seeds were germinated to investigate the effect of the fermentation period of oak tree sawdust on germination viability and seedling characteristics. Its germination rate, seedling weight, length, and total vigor index were assessed. The seeds were sown in oak tree sawdust fermented for 0, 30, 45, and 60 days. The germination rates of the seeds in fermented sawdust were significantly different. The seeds in the 45-day fermented sawdust produced the heaviest biomass weight (4.6 g) with the longest true leaf (1.7 cm) and hypocotyl (3.4 cm) resulting in the highest total vigor index (925.8). In contrast, seeds in 0-day fermented sawdust had the lowest total vigor index (18.3). Microbiome analysis showed that the microbial community in the sawdust changed as the fermentation progressed, indicating that the microbial community seems to affect seed germination physiology. Taken together, 45-day fermented sawdust is recommended for optimal peanut seed germination and seedling growth.

SUMMARYA laboratory experiment was conducted to determine the effect of ultrasound (US) treatment on seed germination of chickpea, wheat, pepper and watermelon. All tests were carried out at 40 kHz in a water bath ultrasonic device varying two factors, treatment duration (5, 10, 15, 30, 45 or 60 min) and germination temperature (15 or 20 °C). Parallel tests were run in which seeds were soaked in water without sonication in order to eliminate the effect of water from US test results. The effects of US on seed germination varied between crops and were more obvious on germination speed, expressed as germination rate index (GRI), rather than on germination percentage (GP). In particular, US treatment significantly increased the GRI of chickpeas, wheat and watermelon, resulting in a maximum increase of 133% (at 45 min), 95% (30 min) and 45% (5 min), respectively, above control seeds. The beneficial effects of US on the GRI of these crops were observed at both 15 and 20 °C, suggesting that US treatment offers a practical priming method to overcome the slow germination that may occur at low temperatures. Water-soaking treatment improved the GP of both chickpea and pepper seeds by 59 and 24%, respectively, compared to the control but neither water nor US had any positive effect on pepper GRI. Post-treatment measurement of moisture content of these seeds produced variable results depending on crop species and US treatment duration. Results of this research indicated that US treatment effectively enhanced speed of germination of chickpea, wheat and watermelon seeds. This increase in speed of germination may improve early field establishment of these crops in the semiarid Mediterranean region and thus needs further investigation. The US technique may also be very useful for plant propagators in nurseries to achieve fast seedling establishment of watermelon.

Abstract Background MYB transcription factors are involved in many biological processes, including metabolism, development and responses to biotic and abiotic stresses. In our previous work, a new MYB transcription factor gene, ZmMYB59 was induced by deep sowing and down-regulated during maize seed germination via Real-Time PCR. However, there are few reports on seed germination regulated by MYB proteins and the functions of ZmMYB59 remain unknown. Results In this study, to examine its functions, Agrobacterium -mediated transformation was exploited to generate ZmMYB59 transgenic tobacco and rice. In T 2 generation transgenic tobacco, germination rate, germination index, vigor index and hypocotyl length were significantly decreased by 25.0~50.9%, 34.5~54.4%, 57.5~88.3% and 21.9~31.2% compared to wild-type (WT) lines. In T 2 generation transgenic rice, germination rate, germination index, vigor index and mesocotyl length were notably reduced by 39.1~53.8%, 51.4~71.4%, 52.5~74.0% and 28.3~41.5%, respectively. On this basis, relative physiological indicators were determined. The activities of catalase, peroxidase, superoxide dismutase, ascorbate peroxidase and proline content of transgenic lines were significantly lower than those of WT, suggesting that ZmMYB59 reduced their antioxidant capacity. As well, ZmMYB59 expression extremely inhibited the synthesis of gibberellin A1 (GA 1 ) and cytokinin (CTK), and promoted the synthesis of abscisic acid (ABA) concurrently, which implied that seed germination was repressed by ZmMYB59 in hormone levels. Furthermore, cell length and cell number of hypocotyl/mesocotyl in transgenic plants were notably decreased. Conclusions Taken together, it proposed that ZmMYB59 plays a negative regulation during seed germination in tobacco and rice, which also contributes to illuminate the molecular mechanisms regulated by MYB transcription factors.

It was estimated that North Sulawesi needs 9,870,093.33 coastal vegetation seeds to rehabilitate the damage of coastal ecosystems which reach 14,805.14 ha, where the largest area 13.884 ha located in other land use. This study aims to provide information of seed germination techniques in Permanent Nursery Kima Atas, Manado. Research was arranged in complete randomized design as factorial with three replications. Ketapang (Terminalia cattapa) will obtain high viability in fluctuation of temperature and humidity, i.e used sand media, without wildlings and placed under 25% shade and rain. Keben (Baringtonia asiatica) through the incision and decay that placed in the open location will obtain viability until 90% within 2 months. Nyamplung (Calophyllum inophyllum) requires shell splitting in order to improve the viability and it can be done by cracking and stripping.

ABSTRAK Buah pisang Cavendish merupakan salah satu produk hortikultura yang rentan mengalami kemunduran fisiologis dan sangat rentan terhadap kerusakan fisik setelah panen. Penggunaan berbagai konsentrasi asap cair bambu tabah dengan lama perendaman yang berbeda dapat digunakan sebagai pengawet alami. Buah pisang yang digunakan adalah pisang Cavendish yang berasal dari Kabupaten Buleleng, dalam satu sisir berisi paling sedikit10 buah dengan panjang rata-rata 18.89 cm dan berat rata-rata 125  gram (g). Tujuan penelitian ini adalah menemukan kombinasi perlakuan terbaik dari perlakuan konsentrasi asap cair dan lama perendaman untuk menjaga kesegaran dan masa simpan pada buah pisang Cavendish selama penyimpanan. Rancangan percobaan yang digunakan dalam penelitian ini adalah Rancangan Acak Lengkap (RAL) faktorial dengan dua faktor, yaitu faktor pertama adalah konsentrasi asap cair bambu tabah dan faktor kedua adalah lama perendaman. Faktor pertama terdiri dari empat taraf konsentrasi yaitu 1%, 3%, 5%, dan 7%. Faktor kedua terdiri dari tiga taraf lama perendaman yaitu 10 menit, 20 menit dan 30 menit dengan suhu penyimpanan menggunakan suhu ruang (28±2oC).  Parameter yang diamati dalam penelitian ini adalah laju respirasi, susut bobot, kekerasan, TPT, tingkat kerusakan, dan organoleptik. Hasil interaksi terbaik ditunjukkan pada perlakuan 7% konsentrasi asap cair bambu tabah dan perendaman 10 menit, dengan nilai rata-rata parameter laju respirasi 51.18 ml CO2/kg.jam, nilai rata-rata susut bobot dengan nilai 17.61%, nilai rata-rata kekerasan 3.82 kg, nilai rata-rata TPT 15.65 %Brix, nilai tingkat kerusakan 2.40, nilai rata-rata organoleptik rasa sebesar 4.67, aroma sebesar 4.54 dan warna sebesar 3.87. Disimpulkan bahwa kombinasi perlakuan A4B1 (konsentrasi asap cair bambu tabah 7% dengan perendaman selama 10 menit) adalah kombinasi terbaik untuk menjaga kesegaran dan memperlambat penurunan kualitas buah pisang Cavendish hingga akhir periode penyimpanan hari ke-15. ABSTRACT Cavendish banana one of the horticultural products that are prone to physiological setbacks and very vulnerable to physical damages after harvest. Various concentrations of tabah bamboo liquid smoke with different soaking times can use as natural preservatives. The bananas used in this study were Cavendish bananas from Buleleng, every bunch consists of at least ten banana with a length of 18.59 cm and a weight of 125 ± 5 g. This study aims to find the best treatment combination of the treatmentoof liquid smoke concentration and soaking time to maintain freshness and shelf life of Cavendish bananas during storage. The experimental design used in this study was a Completely Randomized Design (CRD) with two factors, the first factor was the concentration of tabah bamboo liquid smoke, and the second factor was the soaking time. The first factor consists of four levels of concentration namely 1%, 3%, 5%, and 7%. The second factor consists of three levels of soaking time, namely 10 minutes, 20 minutes and 30 minutes with storage temperature using room temperature (28 ± 2oC).  The parameters observed in this study are: repiration rate, weight loss, hardness, level of damages, TPT and organoleptics. The best interaction results are shown in the treatment of 7% concentration of tabah bamboo liquid smoke with 10 minutes of soaking with an average value of respiration rate parameters of 51.18 ml CO2 / kg.hour, an average value of weight loss with a value of 17.61%, an average value of hardness of 3.82 kg, the average value of TPT was 15.65% Brix, the value of the level of damage was 2.40, the average organoleptic value of taste was 4.67, aroma was 4.54 and color was 3.87. The conclus of this study indicates that the combination of A4B1 (7% tabah bamboo liquid smoke concentration with soaking for 10 minutes) is the best combination to keep freshness and the quality of green bananas until the storage period of the 15th day.

However, the germination growth of red palm is slower than other kinds of palm, so the best way to germinate the red palm seed is needed. The aim of this research is to determine the effect of apliccation some young coconut water concentration and kascing fertilizer to red palm seed germination. The research used factorial completely randomized design with 5 replication, the first factor is young coconut water concentration (0%, 25%, 50%, 75%, and 100%) and the second factor is germination media (steril sand and kascing fertilizer). Variables measured were live germination percentage, speed germination rate, radicula length, and plumula height. Results of this study showed that the best germination media is steril sand and 0% concentration of young coconut water.
