This study was aimed to enhance the immune response of the broilers by vitamin E and Cod liver oil supplement with locally inactivated bivalent vaccine of Newcastle disease and Infectious bronchitis viruses. One hundred and twenty broiler chicks Ross 308 were divided to four groups at 30 chicks of each group. The 2nd group was vaccinated with 0.5ml/chick of locally inactivated ND-IB vaccine at 5 days old. The 3rd group was vaccinated with 0.5ml locally ND-IB vaccine and 0.1 ml/chick of VE. The 4th group was vaccinated with 0.5ml of locally ND-IB vaccine and 0.1ml/chick of Cod liver oil, while the first group was left without vaccination as a control group. All groups were vaccinated with attenuated ND-IB at 3 days old and attenuated IBD at 14 days old. The results of ELISA test revealed that the 3rd and 4th groups showed significantly increased (P≤0.05) in Ab titers against ND virus at aged 35 days, also, the 4th group showed significantly (P≤0.05) increased in Ab titers against IB virus at day 35 of age as compared with the other groups. The 4th group showed a significant (P≤0.05) increased in bursal index as compared with the other groups. The results of challenge test revealed that vaccinated groups resist the velogenic ND and IB viruses with relatively high productive range particularly the 4th group as compared with other groups. In conclusion, the VE and Cod liver oil supplement with local bivalent oil based vaccine of ND and IBV may enhance the immune response of the broilers particularly the 4th group which vaccinated with 0.5 ml of local bivalent oil based vaccine of ND-IB vaccine and 0.1 ml Cod liver oil.

In this study we investigated the role of the Harderian gland in the immune response following vaccination against infectious bronchitis (IB). The experiment was carried out on 100 broiler chicks which were divided into two experimental groups. Experimental group O was vaccinated on the first day of age against infectious bronchitis with vaccine Bronhivet I batches that contain a live attenuated strain H120 and the control group did not get immunized. The experiment lasted 21 days. Blood sera were examined on days 1, 7, 14 and 21 for the presence of specific antibodies against infectious bronchitis (ELISA test). Histological changes were observed in the structure of the Harderian gland in relation to the applied vaccine. This research demonstrated the role of the Harderian gland as a secondary lymphoid organ in broiler vaccination against infectious bronchitis. Maternal antibodies present in the IB had no negative impact on the vaccination of chickens on the first day of age. The vaccine virus partially damaged the Harderian gland.

Infectious bronchitis virus (IBV) primarily induces a respiratory disease in broilerscausing important economic losses in poultry industry. The aim of this study was to investigatethe immunosuppressive properties of live IB vaccine on the different programs and method ofvaccination against Newcastle disease (ND) in broilers. A total of 125 one-day-old broilerchicks were assigned into five equal groups. At day one G1 and G2groupswere vaccinated witha live (ND, IB)vaccine by dipping the head in the vaccine and intra-cloacae methodrespectively. Afterward, G3and G4 groups were vaccinated with a live ND vaccine alone by thesame method of G1 and G2. At the same time all these4 groups were vaccinated bysubcutaneous injection (S/C) with inactivated oil based vaccine ND. The G5 group was leftwithout vaccination as a control group. After that, all treated groups were vaccinated with a liveND by spray method at day 10 of the age. The immunosuppressive effects of this IB virus wasevaluated by Haemagglutination inhibition test (HI), and challenged by virulent ND virus.Bursa and Spleen indices were estimated in all groups. The results showed that the antibodytiters against ND was higher (P<0.05) in G3and G4 in compared with G1, G2 and controlgroups at 20 and 30 days of age. However, there was no significant difference of bursa indexand spleen index in all groups. Challenge test showed no mortality in the 3rd group while 10%in the 4th group, 85% in the 1st group and 90% in the 2nd group. In conclusion, it’s revealed thatthe best method and program of vaccination which can protect against ND was applied in the3rd group.

     This study aimed to investigate the effects of injection hatching eggs with different doses of vitamin E on productive traits, some physiological and immune response to Newcastle disease vaccine. Two hundred fertile eggs of broiler breeder Ross (308) strain were incubated in automatic incubator machine after divided into four treatments (50 eggs per treatment) with two replicates. First treatment was injected 0.1ml/egg of phosphate buffer sterile into amniotic fluid at day 18th of incubation which it was considered as control group, second treatment was injected with 0.1ml/egg of inactivated Newcastle disease vaccine, third treatment was injected with 0.1ml/egg of inactivated Newcastle disease vaccine and 0.1 ml/egg of vitamin E, fourth treatment was injected with 0.1ml/egg of inactivated Newcastle disease vaccine and 0.15 ml/egg of vitamin E. All injected eggs were carried back into incubator for complete hatching process. Hatched chicks were transferred to the hall in farm of the Veterinary Medicine College/University of Baghdad; therefore, the chicks were distributed into four treatments with two replicates depending on the previously treated groups until fifth week of age. The results revealed that the treated groups with vitamin E were significantly (P≤0.05) increased in hatchability percentage, body weight, weight gain, as well as significant reduce in feed intake and improvement in feed conversion ratio as compared with second group and control, the results of third group showed significantly increased (P≤0.05) in antibody titers against Newcastle disease virus at aged (21 and 35) day as compared with the other treated groups and control.

Four hundred fertile eggs of broiler breeder Ross strain were incubated in commercial Hatchery. Eggs were injected into amniotic fluid on 18th day of incubation after divided into four treatment groups (100 eggs per treatment) with two replicates. First treatment was injected 100 µl of PBS, second treatment was injected with100 µl of inactivated ND vaccine, the third treatment was injected with 100 µl of inactivated ND vaccine and 50 µl vitamin E (oily form) and finally the fourth treatment was injected with 100 µl of inactivated ND vaccine and 50 µl Cod liver oil. All injected eggs were carried back into hatchery for complete hatching process. Hatched chicks were transported to the farm of the Veterinary Medicine College/University of Baghdad for 42 days from 6/10/2011 to 16/11/2011. The hatched chicks from the previously treated groups were distributed into four treatments with two replicates for up to the experimental end. Results of treated groups showed significant (P ≤ 0.05) increase in body weight, weight gain, as well as significant reduce in feed intake and improvement in feed conversion ratio, also, antibody titers against Newcastle disease virus of treated groups showed significant (P ≤ 0.05) increase progressively with age until the end of the experiment compared with broiler chicks of control.

This study was conducted to investigate the safety of the most commonly used Newcastle disease vaccines in Al-Sulaimaniyah province (LaSota and Clone30). A total of 225 one-day-old broiler chicks of Ross 308 breed were investigated for their maternal-derived antibody (MDA) titers by ELISA test. Subsequently, these chicks were divided randomly into 3 equal groups; ( 2 treatment groups, a T1 group which was vaccinated by LaSota vaccine and T2 group which was vaccinated by the Clone30 vaccine). and control non-vaccinated group. ELISA test was used to investigate the antibody titers against NDV in all groups on day 10 post second vaccination at 34 days of chicks age and tissue biopsies were obtained for histopathological examination from the trachea, spleen, Bursa of Fabricius and thymus to explore the tissue changes that may be induced by the vaccine. Significant variation was observed in the means of antibody titers against NDV between the control and treatment groups, whereas, no significant variation was observed between the treatment groups themselves. The histopathological examination results showed that a reactive lymphocytic response was observed in both treatment groups compared to the control group. In addition, focal epithelial sloughing and mucopurulent exudates were observed in the trachea of T1 group chicks only. The result of this study showed that the NDV vaccine of clone30 is approximate of the same efficiency and more secure than LaSota vaccine.

The research was conducted to study the effect of feeding broiler chickens on diets containing isomaltooligosaccharides on the growth performance, carcass traits and immune response. 90-one day old broiler chicks were used according to completely randomized two treatment groups and one control, 30 birds each. Birds fed ad-libitum on basal starter and grower-finisher diets for 35 day. Diets of treatment`s groups contained 0.5 g/Kg and 1 g/Kg of Isomaltooligosaccharides, while the control group fed on the basal diets without Isomaltooligosaccharides supplementation. Dietary supplementation of broiler chickens with Isomaltooligosaccharides improved body weight, feed conversion, carcass traits, two lymphoid organs weight and log antibody titer against avian flu vaccine. Most of the highest values were for birds fed low levels of Isomaltooligosaccharides. Feed intake decreases as Isomaltooligosaccharides level increases. Dietary supplementation with Isomaltooligosaccharides did not affect the lipids profile (triglycerides, total cholesterol, LDL and HDL), however the blood VLDL levels decreased with increased levels of Malondialdehyde and Glutathione reductase. Collectively, Dietary supplementation of broiler chickens with 0.5 g/Kg diet of Isomaltooligosaccharides improved growth performance, carcass traits and immune status.

Introduction: Mechanisms leading to diminished platelet counts in immune thrombocytopaenia (ITP) appear to be multifactorial: autoantibodies, autoreactive CD8+ cytotoxic T cells, enhanced apoptosis and loss of sialic acid which mediates platelet clearance through the Ashwell-Morell receptors present in hepatocytes. Differential involvement of each of them might condition the ability of patients with ITP to respond to treatments. We aimed to examine platelet features and the immunological state of patients with ITP who do not respond to any treatment to detect the unique characteristics of this group. Methods: This was an observational, prospective and transversal study. Patients with chronic primary ITP were included: 28 ITP patients without treatment for at least 6 months (UT-ITP); 36 responders to agonists of thrombopoietin receptors (TPO-RA); and 14 ITP patients who did not respond to first- and second-line treatments (NR-ITP). A healthy control group (n=104) was also included in the study. Active caspase-3, -7, -8 or -9 were determined by flow cytometry using CaspaTag kits (Millipore, Madrid, Spain) in PRP diluted with HEPES-buffer containing 2 mM Ca2+ and 2 mM Gly-Pro-Arg-Pro (Sigma-Aldrich, Madrid, Spain) to prevent fibrin formation . Platelet surface glycan exposure was analysed by determining the binding of lectins by flow cytometry. To do so, washed platelets were incubated with 1 μg/ml Alexa fluor 488-conjugated wheat germ agglutinin lectin (WGA, Invitrogen, Spain) or with 1 μg/ml FITC-conjugated Ricinus communis agglutinin (RCA, Vector Labs, UK). WGA binds to sialic acid and N-acetylglucosaminyl residues, and RCA is a galactose-specific legume lectin which binding serves as an indirect measurement of the loss of sialic acid. Peripheral blood mononuclear cells (PBMCs) subsets were analysed by flow cytometry using specific antibodies. Experimental data was analysed using SPSS 9.0 software (SPSS Inc., Chicago, IL). Results: Platelets from TPO-RA treated and from NR-ITP patients had increased caspase-3, -7, -8 and -9 activities (Figure 1A). Platelets from NR-ITP patients exposed less sialic acid and more N-acetylglucosaminyl residues than the other groups (Figure 1B). Binding of WGA and RCA correlated with caspase activities (Table 1). Distribution of lymphocytes, monocytes and natural killer cells is shown in Table 1. NR-ITP patients had an increased proportion of B lymphocyte (LB), maybe due to a significant rise in the fraction of naive LB cells, and a diminution in LTreg subset. Whereas classical monocytes was increased, nonclassical monocyte fraction was decreased in the UT-ITP and NR-ITP groups. NR-ITP patients also presented an increased CD16+CD56bright cells fraction and a diminished NK CD16+CD56dim subset. TPO-RA-treated patients seemed to recover an immune homeostasis similar to healthy controls (monocyte and NK cells subset distribution and LTreg count similar to control group). It is of interest to note the relationship between loss of sialic acid from platelet surface glycans and Tregs count: the most reduced surface exposure of sialic acid, the less Treg count (Figure 2). Conclusions: Platelets from NR-ITP patients had more signs of apoptosis and a different composition of surface glycans, accompanied by a diminished LTreg population, a higher LB naïve percentage, and an increased CD16+CD56bright cells fraction in circulation, indicating a severe deregulation of the immune system. Since an inverse correlation was observed between loss of sialic acid and LTreg count, a potential relationship between glycan composition on the platelet surface and immune response is suggested, positing terminal sugar moieties of the glycan chains as aetiopathogenic agents in ITP. On the other hand, TPO-RA appears to have a beneficial effect on immune response. Nevertheless, one of the limitations of our study was that patients were recruited once the response to TPO-RA was achieved; therefore, a longitudinal study would provide more information regarding TPO-RA effects.
