Comparison of three halophilic archaea on growth and Vibrio anguillarum challenge resistance of Artemia under gnotobiotic conditions

Oxytetracycline (OTC) has been commonly used as an effective antibiotic against various fish bacterial diseases, including vibriosis. In this study, the absorption-enhancing effect of citric acid on oral OTC pharmacokinetics and treatment of artificial Vibrio anguillarum infection was evaluated in juvenile yellowtail Seriola quinqueradiata followed by serum OTC concentration analysis. When 25 mg kg-1 body weight (BW) OTC was administered in combination with 1250 mg kg-1 BW citric acid, the serum OTC concentration reached almost the same concentration as that of the group treated with 50 mg kg-1 BW OTC. This coadministration successfully suppressed mortality due to vibriosis similar to the group treated with 50 mg kg-1 BW OTC. Conversely, poor efficacy was observed when only 25 mg kg-1 BW OTC was administered. These results suggest that coadministration of citric acid can be beneficial in reducing the dose of OTC needed for effective treatment, and thus contributes to the goal of reduced use of this antibiotic in aquaculture.

Fructans are fructose-based (poly)saccharides with inulin and levan being the best-known ones. Thanks to their health-related benefits, inulin-type fructans have been under the focus of scientific and industrial communities, though mostly represented by plant-based inulins, and rarely by microbial ones. Recently, it was discovered that some extremely halophilic Archaea are also able to synthesize fructans. Here, we describe the first in-depth functional and molecular characterization of an Archaeal inulosucrase from Halomicrobium sp. IBSBa (HmcIsc). The HmcIsc enzyme was recombinantly expressed and purified in Escherichia coli and shown to synthesize inulin as proven by nuclear magnetic resonance (NMR) analysis. In accordance with the halophilic lifestyle of its native host, the enzyme showed maximum activity at very high NaCl concentrations (3.5 M), with specific adaptations for that purpose. Phylogenetic analyses suggested that Archaeal inulosucrases have been acquired from halophilic bacilli through horizontal gene transfer, with a HX(H/F)T motif evolving further into a HXHT motif, together with a unique D residue creating the onset of a specific alternative acceptor binding groove. This work uncovers a novel area in fructan research, highlighting unexplored aspects of life in hypersaline habitats, and raising questions about the general physiological relevance of inulosucrases and their products in nature.

Filtration (25 nm) and ultraviolet irradiation dosages of 13,100–29,400 microwatt seconds per square centimetre (μW∙s∙cm−2) effected a 99.98–100% reduction of five gram-negative fish pathogens — Aeromonas salmonicida, A. hydrophila, Vibrio anguillarum, Pseudomonas fluorescens, and the enteric redmouth organism in 12.5 °C clear spring water or spring water containing particulate matter. Filtration and a dosage of 4500 μW∙s∙cm−2 killed 99.83–100% of test strains in spring water and 4000–4750 μW∙s∙cm−2 killed 99.33–99.99% in water with particulate matter. Irradiation of unfiltered water containing particulate matter was less effective, especially at dosages of 5000 μW∙s∙cm−2 or less, which killed 97–99.94% of strains. Filtration and 13,100 μW∙s∙cm−2 irradiation of water containing A. salmonicida prevented transmission of furunculosis. AbstractAs a critical evolutionary pivot between invertebrates and vertebrates, lampreys provide rich genetic information. Lamprey immune protein (LIP) is a key immune regulator. MicroRNAs, well-conserved in the response to immunological stress, remain understudied in lamprey immunity. We generated a lamprey microRNA expression atlas, using deep sequencing, upon Vibrio anguillarum infection. Using comparative methods, we found that miR-4561 potentially regulates innate immunity via interaction with lip. We found a sequence in the 3′-UTR region of LIP mRNA complementary to the miR-4561 seed region; miR-4561 expression was negatively correlated with LIP. During V. anguillarum infection, miR-4561 inhibited LIP expression and bacterial clearance. Notably, LIP expression in supraneural body cells was necessary for the Gram-negative immune response. Additionally, we observed that overexpression of miR-4561 induced apoptosis in embryonic cells, suggesting a role in embryonic development. Collectively, we show lamprey microRNAs may significantly affect gene regulation and provide new insights on LIP-mediated immune regulation.
