Parents of children with special needs suffer from depression and anxiety disorder, as well as chronic insomnia and stress, this is confirmed by the presence of elevated cortisol levels and reduced immunity. In music therapy, rhythmic music and synchronization, especially music with drums, is used as a therapeutic activity capable of influencing mood improvement, stress reduction and relaxation in a therapeutic context, as it can fit into and adapt to many rhythmic complex and coherent ranges in the real world. This is unifying, creating a sense of security and belonging, allows music to be a facilitator of communicative and social interaction processes. The study uses Perceived Stress Scale (PSS), Cohen, et al., 1983), Improvisation Assessment Profile (IAP), Bruscia, 1987, Natural killer (NK) cell blood analysis, stress (cortisol) saliva analysis. The results of the study showed statistically a significant decrease in psychological and physiological stress patterns, improvements in immunity, interpersonal processes as well as intra-musical synchronization between the first and the tenth sessions.

Abstract Progenitor B cells deficient in Pax5 are developmentally multipotent, suggesting that Pax5 is necessary to maintain commitment to the B-cell lineage. Commitment may be mediated, in part, by Pax5 repression of myeloid-specific genes. To determine whether Pax5 expression in multipotential cells is sufficient to restrict development to the B-cell lineage in vivo, we enforced expression of Pax5 in hematopoietic stem cells using a retroviral vector. Peripheral blood analysis of all animals reconstituted with Pax5-expressing cells indicated that more than 90% of Pax5-expressing cells were B220+ mature B cells that were not malignant. Further analysis showed that Pax5 completely blocked T-lineage development in the thymus but did not inhibit myelopoiesis or natural killer (NK) cell development in bone marrow. These results implicate Pax5 as a critical regulator of B- versus T-cell developmental fate and suggest that Pax5 may promote commitment to the B-cell lineage by mechanisms that are independent of myeloid gene repression.

According to attachment theory, security providing attachment figures fulfill two main functions: (1) safe haven—providing safety and comfort and reducing stress—helping people regain a sense of security; and (2) secure base—providing resources and a base from which people can spring into action. According to the Dual Function of Security Priming Model, security priming can result in one of two outcomes paralleling these two functions. Which outcome is likely to present itself depends on the level of stress imposed by the context. Here we describe the Dual Function Model of Security Priming (DFSP) Model and provide evidence from a study examining the effects of attachment security priming on hypothalamic-pituitary-adrenal activity. In the study, participants were exposed to security-related cues under high or low/no-stress conditions, while their salivary cortisol concentrations were measured. Cortisol is a suitable index as it is released not only in response to stress, but also more generally when energy needs to be mobilized. We found that while security priming led to significant decreases in salivary cortisol concentrations when presented after a stressor (stress reduction), it led to a significant increase in salivary cortisol concentrations when presented before the stressor (energy mobilization).

Fractal objects are prevalent in natural scenery. Their repetition of patterns at increasingly fine magnifications creates a rich complexity. Fractals displaying mid-range complexity are the most common and include trees, clouds, and mountains. The “fractal fluency” model states that human vision has adapted to process these mid-range fractals with ease. I will first discuss fractal fluency and demonstrate how it enhances the observer’s visual capabilities by focusing on experiments that have important practical consequences for improving the built environment. These enhanced capabilities generate an aesthetic experience and physiological stress reduction. I will discuss strategies for integrating fractals into building designs to induce positive impacts on the observer. Examples include fractal solar panels, fractal window shades, and fractal floor patterns. These applications of fractal fluency represent a fundamental and potentially impactful form of salutogenesis.

The purpose of this study was to compare emotional and biological responses of men and women who are spousal caregivers of patients with Alzheimer’s disease (AD). Quality-of-life measurements, bioinstrumentation data, and immunophenotype assessments were obtained from female and male spousal caregivers of patients with AD. Spousal caregivers (women, n = 45 with average age 69.7; men, n = 16 with average age 71.4 years) completed questionnaires that assessed psychosocial variables. Blood was drawn and lymphocyte subsets (including natural killer [NK] cell number) were determined using flow cytometry. The degree of relaxation was determined measuring muscle tension (EMG) in the frontalis and trapezius muscles, skin conductance, skin temperature, and heart rate. Male spousal caregivers, as compared to female spousal caregivers, had significantly lower levels of stress, depression, caregiver burden (subjective), anxiety, anger-hostility, and somatic symptoms and higher levels of mental health, sense of coherence, NK cell number, and social and physical functioning. There were no statistically significant differences between the 2 groups in social support, coping resources, or T, T suppressor, or activated T cells. Women had more T helper cells and fewer NK cells than men. Men had fewer manifestations of a physiological stress response, as indicated by bioinstrumentation parameters. Unique sex-specific issues need to be considered when strategies are implemented to assist the increasing number of caregivers as our society ages.

Abstract Introduction : Acute Myeloid Leukemia (AML) is an aggressive neoplastic disorder with poor outcomes in children and adults. NK cell adoptive transfer is an anti-cancer immunotherapy that has promise for AML treatment. We aimed to improve NK cell anti-tumor efficacy with expression of a Chimeric Antigen Receptor (CAR) on the cell surface. Our CAR consists of an extracellular single-chain variable fragment targeting the AML-associated antigen CD123 (IL3Rα) and intracellular domains derived from 2B4 and TCRζ. We sought to improve the persistence and long-term functionality of our CAR-NKs by introducing transgenic interleukin-15 (IL15). Methods: CD3-depleted PBMCs were first activated with lethally irradiated feeder cells, then transduced with transiently produced replication incompetent γ-retrovirus (αCD123.2B4.ζ, αCD123.2B4.ζ-IRES-sIL15, sIL15-IRES-mOrange) on day 4 of culture. CAR expression was measured on day 8 using FACS. Secretion of IL15 was verified with ELISA. Cytotoxicity was measured using ffLuc expressing target cells and bioluminescence (BL) measurement. In serial stimulation assays, target cells were repleted daily to maintain a 1:1 effector:target ratio. Immunophenotype and cell counts were assessed by FACS. Transcriptomic analysis (RNAseq) was performed on RNA derived from NK cells purified on D10. Xenograft modeling was performed using NSG mice engrafted with MV-4-11.ffLuc or MOLM-13.ffLuc AML cell lines. Mice were treated with NK cells on D4 or D4-7-10. Untreated mice served as controls. Tumor growth was serially tracked in vivo using BL imaging. NK cell persistence and expansion were measured in peripheral blood. Results: The 2B4.ζ CAR was well expressed on the surface of transduced NK cells (median transduction efficiency 95%, range 85-97%, n=3). 2B4.ζ CAR-NK treatment prolonged survival of AML engrafted mice when compared to treatment with unmodified NKs (median survival: 63 vs 55 days; n=8 mice; p=0.014). Serial peripheral blood analysis revealed a steady decline in circulating NK cells, which were undetectable in all cohorts within 21 days. NK cells were then engineered for constitutive secretion of IL15, with and without CAR expression. 2B4.ζ/sIL15 CAR-NKs had the most potent 24h-cytotoxicity against CD123+ targets (Fig. 1). After a 10-day chronic stimulation with MV-4-11, 2B4.ζ/sIL15- and sIL15-NKs expanded (x1.2 and x5.9 respectively), while NK cells without sIL15 decreased in number. In this assay, only 2B4.ζ/sIL15 CAR-NKs exhibited sustained tumor killing. Transcriptomic analysis after 10 days of serial stimulation showed sample clustering dependent on IL15 secretion. Differential gene expression analysis (DESeq2) identified upregulation of genes associated with cell cycle progression, apoptosis regulation, chemokine signaling, and NK cell mediated cytotoxicity in NK cells secreting IL15 compared to those without. In multiparameter flow cytometric analysis, 2B4.ζ/sIL15 CAR-NKs had a higher percentage of NK cells populating clusters defined by higher surface expression of NK cell activating receptors (NKp30, NKG2D, LFA-1) compared to 2B4.ζ and unmodified NK cells. In our MV-4-11 xenograft model, NKs armed with secreted IL15 expanded in vivo and had improved persistence. A single dose (D4) of 2B4.ζ/sIL15 CAR-NKs demonstrated an initial antitumor response, equivalent to that seen following 3 doses (D4-7-10) of 2B4.ζ CAR-NKs. However, mice treated with IL15-secreting NKs had short survival (Fig. 2). Compared to control mice, peripheral blood analysis showed increasing systemic hIL15 and higher levels of hTNFα. In our more aggressive MOLM-13 xenograft model, both single dose 2B4.ζ/sIL15 CAR-NK and multiple dose 2B4.ζ CAR-NK treatment prolonged survival compared to treatment with unmodified NKs. (27 and 26 vs 20 days; n=5 mice; p<0.01; Fig. 2). Conclusion: 2B4.ζ CAR-NKs have limited antitumor efficacy and short persistence in vivo. NK cells armored with secreted IL15 have enhanced anti-AML cytotoxicity and in vitro persistence. Introduction of IL15 secretion confers a distinctly activated phenotype that is maintained during chronic antigen stimulation. Constitutive local IL15 secretion improves in vivo NK cell persistence but may cause lethal toxicity when employed against AML. These results warrant further study and should impact the development of CAR-NK clinical products for patients with AML. Figure 1 Figure 1. <p>Mindfulness training has become very popular in recent years and has proven successful for reducing anxiety and depression and enhancing coping skills (amongst other benefits). This research project explores the perceived benefits of mindfulness-based stress reduction (MBSR) training on a music therapy student working with young people with complex needs in special education. Secondary analysis of clinical records (session notes and a reflexive journal) was employed and included the use of both inductive and deductive methods of analysis. Five themes were developed (framed as themes of learning) including: Presence, Non-striving, Beginner’s Mind, Acceptance, and Patience with a final key finding being the effect of modelling to the students. Findings showed these benefits emerging through a journey from early data (before MBSR training) to later data (after MBSR training) and suggest that mindfulness training can be beneficial for both music therapy students and (indirectly) to those they work with. The themes and perceived benefits proved very interconnected with each relating to several others and ‘Being Present’ emerging as an over-arching theme. Similarities between benefits discovered and certain principles of music therapy were discussed and whether the benefits found may have occurred naturally through the course of music therapy training amongst other factors of change and natural growth.</p>

In modern society, stress reduction in the workplace is a pressing issue. Although many studies have been done on the psychological and physiological effects of indoor plants, the majority of them have been conducted in laboratory or quasi-office settings. The objective of this study was to verify the stress reduction effects of the presence of small indoor plants on employees in a real office setting. We investigated the changes in psychological and physiological stress before and after placing a plant on a worker’s desk. Sixty-three office workers at an electric company in Japan were the participants of this study. The participants were directed to take a 3-minute rest while sitting at their desk when they felt fatigue. There were two phases of the study: a control period without plants and an intervention period when the participants were able to see and care for a small plant. We measured psychological stress in the participants using the State-Trait Anxiety Inventory (STAI). As an index of physiological stress, the participants measured their own pulse rate throughout the study. STAI scores decreased significantly after the intervention period (P < 0.05). The ratio of the participants whose pulse rate lowered significantly after a 3-minute rest increased significantly during the intervention period (P < 0.05). Our study indicates that having opportunities to gaze intentionally at nearby plants on a daily basis in the work environment can reduce the psychological and physiological stress of office workers.

Research suggests that young sexual minority men (YSMM), particularly YSMM of color, are more likely to have dysregulated physiological stress responses as compared to their heterosexual counterparts. In addition, the quality of social relationships has been demonstrated to impact physiological stress patterns among young adults. One key indicator of social relationships is that of adult attachment, which refers to the types of socioemotional bonds that individuals form with peers, family, and romantic partners. However, the association between adult attachment and physiological stress functioning among diverse samples of YSMM has been underexplored. Thus, the current study sought to bridge this gap in the literature by exploring how race/ethnicity moderates the association between adult attachment and physiological stress patterns among Black versus White YSMM. N = 63 YSMM participated in a 5-day daily diary study in which they completed a baseline survey and provided saliva samples over the 5-day period in order to measure diurnal cortisol. Three-level hierarchical linear modeling was used in order to examine the association between race/ethnicity, adult attachment, and diurnal cortisol over the 5-day period. Results suggest that adult attachment avoidance was associated the cortisol awakening response (CAR) among YSMM. In addition, race/ethnicity moderated the association between adult attachment avoidance and the CAR such that YSMM who identified as Black had evidence of a more dysregulated physiological stress response as compared to YSMM who identified as White. Future research should focus on further unpacking the drivers of physiological stress patterns among White and Black YSMM.
