The knowledge of the effects of fire on soil properties is of particular concern in Mediterranean areas, where the effects of vegetation type are still scarce also. This research aimed: to assess the properties of burnt soils under different vegetation types; to highlight the soil abiotic properties driving the soil microbial biomass and activity under each vegetation type; to compare the biological response in unburnt and burnt soils under the same vegetation type, and between unburnt and burnt soils under different vegetation types. The soils were collected at a Mediterranean area where a large wildfire caused a 50% loss of the previous vegetation types (holm oak: HO, pine: P, black locust: BL, and herbs: H), and were characterized by abiotic (pH, water, and organic matter contents; N concentrations; and C/N ratios) and biotic (microbial and fungal biomasses, microbial respiration, soil metabolic quotient, and hydrolase and dehydrogenase activities) properties. The biological response was evaluated by the Integrative Biological Responses (IBR) index. Before the fire, organic matter and N contents were significantly higher in P than H soils. After the fire, significant increases of pH, organic matter, C/N ratio, microbial biomass and respiration, and hydrolase and dehydrogenase activities were observed in all the soils, especially under HO. In conclusion, the post-fire soil conditions were less favorable for microorganisms, as the IBR index decreased when compared to the pre-fire conditions.

From March 30th to April 9th, 2021, 814 million liters of legacy phosphate mining wastewater and marine dredge water from the Piney Point facility were released into lower Tampa Bay (Florida, USA). This resulted in an estimated addition of 186 metric tons of total nitrogen, exceeding typical annual external nitrogen load estimates to lower Tampa Bay in a matter of days. Elevated levels of phytoplankton (non-harmful diatoms) were first observed in April in the lower Bay. Filamentous cyanobacteria blooms (Dapis spp.) peaked in June, followed by a bloom of the red tide organism Karenia brevis. Reported fish kills tracked K. brevis concentrations, prompting cleanup of over 1600 metric tons of dead fish. Seagrasses had minimal changes over the study period. By comparing these results to baseline environmental monitoring data, we conclude that many of the biological responses observed after the release from Piney Point are abnormal relative to historic conditions.

Ionizing radiation (IR)-induced bystander effects contribute to biological responses to radiation, and extracellular vesicles (EVs) play important roles in mediating these effects. In this study we investigated the role of bone marrow (BM)-derived EVs in the bystander transfer of radiation damage. Mice were irradiated with 0.1Gy, 0.25Gy and 2Gy, EVs were extracted from the BM supernatant 24 h or 3 months after irradiation and injected into bystander mice. Acute effects on directly irradiated or EV-treated mice were investigated after 4 and 24 h, while late effects were investigated 3 months after treatment. The acute effects of EVs on the hematopoietic stem and progenitor cell pools were similar to direct irradiation effects and persisted for up to 3 months, with the hematopoietic stem cells showing the strongest bystander responses. EVs isolated 3 months after irradiation elicited no bystander responses. The level of seven microRNAs (miR-33a-3p, miR-140-3p, miR-152-3p, miR-199a-5p, miR-200c-5p, miR-375-3p and miR-669o-5p) was altered in the EVs isolated 24 hour but not 3 months after irradiation. They regulated pathways highly relevant for the cellular response to IR, indicating their role in EV-mediated bystander responses. In conclusion, we showed that only EVs from an early stage of radiation damage could transmit IR-induced bystander effects.

AbstractThe exposome, which is defined as the cumulative effect of environmental exposures and corresponding biological responses, aims to provide a comprehensive measure for evaluating non-genetic causes of disease. Operationalization of the exposome for environmental health and precision medicine has been limited by the lack of a universal approach for characterizing complex exposures, particularly as they vary temporally and geographically. To overcome these challenges, passive sampling devices (PSDs) provide a key measurement strategy for deep exposome phenotyping, which aims to provide comprehensive chemical assessment using untargeted high-resolution mass spectrometry for exposome-wide association studies. To highlight the advantages of silicone PSDs, we review their use in population studies and evaluate the broad range of applications and chemical classes characterized using these samplers. We assess key aspects of incorporating PSDs within observational studies, including the need to preclean samplers prior to use to remove impurities that interfere with compound detection, analytical considerations, and cost. We close with strategies on how to incorporate measures of the external exposome using PSDs, and their advantages for reducing variability in exposure measures and providing a more thorough accounting of the exposome. Continued development and application of silicone PSDs will facilitate greater understanding of how environmental exposures drive disease risk, while providing a feasible strategy for incorporating untargeted, high-resolution characterization of the external exposome in human studies.

The carbon ion-beam has emerged as a novel physical mutagen for creating genetic variability and crop improvement. In this study, seeds of a high-yielding pyramided rice line MR219-PL-5 were exposed to carbon ion beam irradiation at 10, 20, 40, 60, 80, and 100 Gy. The radiosensitivity test was conducted to determine the optimum dose of carbon ion beam irradiation based on the lethal dose 50% (LD50) using Sandwich Blotter Technique. The biological responses of carbon-ion beam irradiation were also observed in other characteristics such as germination rate (GeR), survival rate (SR), growth rate (GRoR), shoot length (SL), root length (RL), seedling height (SH), days to flowering (DTF), fertility rate (FR) and thousand-grains weight (TGW). Based on the polynomial curve of SR graph, the lethal dose 50% (LD50) value was 86.12 Gy. However, the optimum dose range of carbon ion-beam irradiation was between 40 and 60 Gy as these two doses recorded the highest SR, 63 and 67%, respectively. Furthermore, the shoulder dose in this study was 60 Gy since SR decreased significantly at higher doses. M1 individuals irradiated at 40 and 60 Gy had the best biological responses where significant differences were found for SR, SL, RL, GRoR, SH, DTF and FR at these two doses compared to the other doses. Further studies on M2 and M3 populations could help to identify potential individuals as well as to understand the inheritance of each trait of interest from one generation to the next.

Adiponectin is a multifunctional adipocytokine produced predominantly by adipocytes, with potent antiinflammatory, insulin-sensitizing, and cytoprotective properties. Autophagy is a lysosome-dependent self-degradative process that mediates the degradation of damaged organelles, invading pathogens and protein aggregates, thus maintaining cellular homeostasis. Adiponectin performs different biological functions by regulating autophagy. This review attempts to elucidate the biological responses and potential mechanisms underlying adiponectin-induced autophagy, with an aim to guide the identification of new therapeutic targets of related diseases. The cyclic nucleotides, cAMP and cGMP, are ubiquitous second messengers responsible for translating extracellular signals to intracellular biological responses in both normal and tumor cells. When these signals are aberrant or missing, cells may undergo neoplastic transformation or become resistant to chemotherapy. cGMP-hydrolyzing phosphodiesterases (PDEs) are attracting tremendous interest as drug targets for many diseases, including cancer, where they regulate cell growth, apoptosis and sensitization to radio- and chemotherapy. In breast cancer, PDE5 inhibition is associated with increased intracellular cGMP levels, which is responsible for the phosphorylation of PKG and other downstream molecules involved in cell proliferation or apoptosis. In this review, we provide an overview of the most relevant studies regarding the controversial role of PDE inhibitors as off-label adjuvants in cancer therapy.

Nootropic drugs are the class of drugs or supplements that are claimed to enhance cognitive functions, specifically executive functions, memory and creativity in healthy individual. They are sometime referred as cognitive enhancers or smart drugs as they are associated with memory improvement functioning. Some of them are well known drugs and clinically approved by the Food and Drug Administration (FDA). All metabolic reactions are purely dependent on enzymatic actions as they play a very important role in regulating and maintaining most of the biological responses and various processes. An enzyme Acetylcholinesterase (AChE) seems to play an essential role in the conduction of cholinergic brain synapses and neuromuscular junctions. There have been different nootropic drugs identified and approved for curing neurodegenerative disorders such as Alzheimer, Parkinson and Huntington's disease. Their binding efficiency and energy have been well studied an established by using the in-silico docking tools. There are different docking tools available today for analysis of molecules such as PyRx, Auto dock and schrodinger suite. The advent of these tools is being widely used by the pharmaceutical industries for the virtual screening of the formulated drugs against the desired target molecule. It has made the drug formulation process more time efficient and cost effective. Thus, an in-silico approach has been widely accepted for drug discovery and its design.
