Optimization of Ultrasound-Assisted Extraction of Oil from Camellia(Camellia oleifera abel) Seed

Camellia oil is the main component of the camellia seed and has many beneficial functions to human health. An investigation into ultrasound- assisted extraction was conducted for the oil extraction from camellia seed. The optimized condition of extraction parameters obtained with the response surface methodology (RSM) at a four-variable, three-level experiment Box–Behnken design (BBD), was as follows: liquid-solid ratio, 5mL/g;ultrasonic power,155W;ultrasonic treatment time,31 min; and ultrasonic treatment temperature, 55°C, under these conditions, the oil yeild was 44.55%. GC–MS analysis showed that the content of unsaturated fatty acid in camellia oil was up to 84.47% and the content of oleic acid was over 76.15%.

Ceratophyllum demersum L. (CDL) is a traditional Chinese herb to treat many diseases, but research on its anti-diabetic activity is not available. In this research, the α-glucosidase inhibitory ability and phytochemical constituents of CDL extract were firstly studied. Optimal ultrasound-assisted extraction conditions for α-glucosidase inhibitors (AGIs) were optimized by single factor experiment and response surface methodology (RSM), which was confirmed as 70% methanol, liquid-to-solid ratio of 43 (mL/g), extraction time of 54 min, ultrasonic power of 350 W, and extraction temperature of 40 °C. The lowest IC50 value for α-glucosidase inhibition was 0.15 mg dried material/mL (mg DM/mL), which was much lower than that of acarbose (IC50 value of 0.64 mg DM/mL). In total, 80 compounds including 8 organic acids, 11 phenolic acids, 25 flavonoids, 21 fatty acids, and 15 others were identified or tentatively identified from CDL extract by HPLC-QTOF-MS/MS analysis. The results suggested that CDL could be a potential source of α-glucosidase inhibitors. It can also provide useful phytochemical information for research into other bioactivities.

Ultrasound-assisted extraction (UAE) of polysaccharides from the Chinese medicinal plantRhodiola sachalinensisA. BOR (PRs) was studied. The effects of four factors (extraction time, ultrasonic power, liquid-solid ratio and extraction temperature) on the extraction yield of PRs were investigated by the single-factor test, and the optimum UAE conditions were investigated by the orthogonal test. The results demonstrated that the optimum UAE conditions for PRs were as followings: extraction time of 30 min, ultrasonic power of 85 W, liquid-solid ratio of 45 ml/g, and extraction temperature of 45°C. The present data might provide a theoretic base for full utilization ofRhodiola sachalinensisA. BOR.

The purpose of this study was to establish an extraction method for the kinsenoside compound from the whole plant Anoectochilus roxburghii. Ultrasound assisted extraction (UAE) and Ultra-high performance liquid chromatography (UPLC) method were used to extract and determine the content of kinsenoside, while response surface method (RSM) was used to optimize the extraction process. The best possible range for methanol concentration (0–100%), the liquid-solid ratio (5:1–30:1 mL/g), ultrasonic power (240–540 W), duration of ultrasound (10–50 min), ultrasonic temperature (10–60 °C), and the number of extractions (1–4) were obtained according to the single factor experiments. Then, using the Box-Behnken design (BBD) of response surface analysis, the optimum extraction conditions were obtained with 16.33% methanol concentration, the liquid-solid ratio of 10.83:1 mL/g and 35.00 °C ultrasonic temperature. Under these conditions, kinsenoside extraction yield reached 32.24% dry weight. The best conditions were applied to determine the kinsenoside content in seven different cultivation ages in Anoectochilus roxburghii.

Object. Ultrasound-assisted extraction of total flavonoids from corn silk and their antioxidant activities were studied.Methods. Response surface methodology was adopted to optimize the extraction conditions and antioxidant activities of the extracted total flavonoids were detected through ferric reducing antioxidant power (FRAP) assay.Results. Through a three-level, three-variable Box-Behnken design of response surface methodology (RSM) adopting yield as response, the optimal conditions were determined as follows: ultrasonic power 500 W, extraction time 20 min, material solvent ratio 1 : 20, and ethanol concentration 30%. Under the optimum conditions, the extraction yield of total flavonoids was 1.13%. FRAP value of total flavonoids extracted from corn silk was 467.59 μmol/L.Conclusion. The total flavonoids of corn silk could be developed as food natural antioxidant reagents.

Hibiscus sabdariffa (Roselle) is in high demand worldwide due to its beneficial health properties owing to the polyphenols content, mainly in the flower calyx. The objective of this study was to find the best conditions (time and liquid: solid ratio) to extract polyphenols from Roselle using Ultrasound-Assisted Extraction (UAE) (40 kHz, 180 W), with ethanol how solvent; as well as determine the yield of phenols, anthocyanin, flavonoids, tannins, antioxidant activity (DPPH) and antigenotoxic effect (comet assay). A traditional solid-liquid extraction was applied as a reference. Extraction times of 40 and 60 min resulted in the highest polyphenols (13.019 mg GAE/g dry weight (dw)), flavonoids (4.981 CE/g dw), anthocyanins (1.855 mg Cya3GE/g dw), and tannins (0.745 CE/g dw) recoveries and an antioxidant activity (DPPH) of 74.58%. Extracts from white calyces contained similar amounts of phenols and flavonoids, but very little condensed tannins (0.049 CE/g dw) and practically no anthocyanins. Extracts from red and white calyces, showed antigenotoxic activity and repaired capacity of damage caused by mutagens in human lymphocytes.

The seed coat of red sword bean (Canavalia gladiata (Jacq.) DC.) is rich in antioxidant polyphenols. It is often discarded as a byproduct with the consumption of red sword bean, since it is very thick and not consumed by people. The aim of this study was to develop an ultrasound-assisted extraction method to extract natural antioxidants from the seed coats. The extraction process was optimized by using response surface methodology. After the single-factor experiments, three key factors, including ethanol concentration, liquid/solid ratio, and extraction time, were selected and their interactions were studied using a central composite design. The optimal extraction condition was 60.2% hydroethanol, a liquid/solid ratio of 29.3 mL/g, an extraction time of 18.4 min, an extraction temperature of 50 °C, and ultrasound power of 400 W. Under the optimal conditions, antioxidant activity of the extract was 755.98 ± 10.23 μmol Trolox/g dry weight (DW), much higher than that from maceration (558.77 ± 14.42 μmol Trolox/g DW) or Soxhlet extraction (479.81 ± 12.75 μmol Trolox/g DW). In addition, the main antioxidant compounds in the extract were identified and quantified by high-performance liquid chromatography–diode array detection–tandem mass spectrometry (HPLC–DAD–MS/MS). The concentrations of digalloyl hexoside, methyl gallate, gallic acid, trigalloyl hexoside, and digallic acid were 15.30 ± 0.98, 8.85 ± 0.51, 8.76 ± 0.36, 4.27 ± 0.21, and 2.89 ± 0.13 mg/g DW. This study provides an efficient and green extraction method for the extraction of natural antioxidants from the bean coat of red sword bean. The extract of antioxidants might be added into functional foods or nutraceuticals with potential beneficial functions.

Enzyme-assisted extraction (EAE) and ultrasound-assisted extraction (UAE) were popular methods used to extract bioactive compounds from citrus peels, by-products of fruit processing industry. In this study, the total phenolic content (TPC), total flavonoid content (TFC), naringin and hesperidin contents, and antioxidant and antimicrobial activities of the extracts from pomelo peels using the combined enzyme and ultrasound-assisted extraction (E-UAE) or ultrasound and enzyme-assisted extraction (U-EAE) technique were investigated and compared with those extracted using the EAE and UAE. The optimal EAE conditions were as follows: enzyme concentration of 2%, water-solid ratio of 40 ml/g, incubation temperature of 50°C, and extraction time of 60 min, whereas the optimal UAE conditions were ultrasonic energy of 40 kHz, water-solid ratio of 40 ml/g at room temperature, and extraction time of 60 min. The results indicate that the total phenolics, total flavonoids, naringin, and hesperidin contents of the extracts significantly increased in the following order of the extraction techniques: UAE < EAE < U-EAE < E-UAE (p<0.05). The combined E-UAE technique was the most effective technique for bioactive compound extraction with the highest antioxidant and antimicrobial activities. The results also indicate that chemical composition and antioxidant and antimicrobial activities of the extracts were different depending on the pomelo species.

The feasibility of improving the extraction rate of common bean antioxidants by ultrasonic treatment was investigated. Scanning electron microscopy (SEM) and spectrum Fourier transform infrared spectrophotometer (FT-IR) analysis revealed that ultrasonic treatment substantially altered the cellular structure of common bean seed, resulting in increased surface area, eroded cell walls, and greater exposure of cellulose and hemicellulose. The highest antioxidant activity was obtained at optimal extraction conditions (68 min, 55% acetone, 36:1 liquid to solid ratio, 30 ℃, and 480 W) which were optimized by response surface methodology. In terms of the extraction rate of common bean antioxidants, ultrasound-assisted extraction (UAE) exhibits about seven-fold higher extraction efficiency than conventional solvent extraction (CSE). In addition, 10 phenolic compounds in the common bean extracts were detected and quantified by high performance liquid chromatography (HPLC), including protocatechuic acid, catechin, chlorogenic acid, epicatechin, ferulic acid, coumarin, rutin, myricetin, cinnamic acid, and genistein. In summary, ultrasonic treatment is an ideal candidate methodology for improving the extraction rate of common bean antioxidants.

The purpose of this study was to select the optimum ultrasound-assisted extraction (UAE) conditions for the extraction of polysaccharides from Pleurotus eryngii (DC. ex Fr.) Que (P. eryngii ). The main factors that affect the extraction yield of polysaccharides such as solid:liquid ratio, ultrasonic power, extraction time and extraction temperature were studied individually. An orthogonal experiment was designed to optimize the extraction parameters. It was found that UAE method was a reliable, simple and effective method for fast extraction of polysaccharides from P. eryngii. The optimum UAE conditions were as followings: Solid:liquid ratio of 1:35, Ultrasonic power of 55 W, Extraction time of 30 min and extraction temperature of 45°C.

Vietnamese Cordyceps militaris (C. militaris) has long been recognized as one of the most valuable traditional Chinese medicines. In this study, adenosine was extracted from Vietnamese C. militaris by ultrasound-assisted enzymatic extraction method (UAEE) using water as a solvent. Then, the effects of five single factors on adenosine content including pH, enzyme-to-material ratio, ultrasonic power, ultrasonic time, and ultrasonic temperature were determined. After that, three factors consisting of ultrasonic power, ultrasonic time, and ultrasonic temperature were chosen based on their effects on adenosine content. The simultaneous influence of these factors on the adenosine content was investigated by response surface method using central composite design. The adenosine content was evaluated by high-performance liquid chromatography method. Under the optimal conditions, the extract was evaluated for antioxidant and anticancer bioactivities. In addition, different extraction methods including aqueous extraction (AE), ultrasound-assisted extraction (UAE), and enzyme-assisted extraction (EAE) methods were carried out to compare with UAEE. As a result, it can be concluded that UAEE is a promising method for adenosine extraction and further studies regarding isolation and purification need to be conducted.
