Urea-N, NH3-N, creatinine-N and amino-acid-N accounted for 77% of the total urinary nitrogen excreted by young pigs. The N from glycine, alanine and taurine made up 75% of the total urinary amino-acid-N. The N from indispensable amino acids, however, accounted for less than 10% of the total amino-acid-N. In a cascade hydroponic system, the used nutrient solution drained from a primary crop is directed to a secondary crop, enhancing resource-use efficiency while minimizing waste. Nevertheless, the inevitably increased EC of the drainage solution requires salinity-tolerant crops. The present study explored the salinity-tolerance thresholds of basil to evaluate its potential use as a secondary crop in a cascade system. Two distinct but complemented approaches were used; the first experiment examined basil response to increased levels of salinity (5, 10 and 15 dS m−1, compared with 2 dS m−1 of control) to identify the limits, and the second experiment employed a cascade system with cucumber as a primary crop to monitor basil responses to the drainage solution of 3.2 dS m−1. Growth, ascorbate content, nutrient concentration, and total amino acid concentration and profile were determined in both experiments. Various aspects of basil growth and biochemical performance collectively indicated the 5 dS m−1 salinity level as the upper limit/threshold of tolerance to stress. Higher salinity levels considerably suppressed fresh weight production, though the total concentration of amino acids showed a sevenfold increase under 15 dS m−1 and 4.5-fold under 5 and 10 dS m−1 compared to the control. The performance of basil in the cascade system was subject to a compromise between a reduction of fresh produce and an increase of total amino acids and ascorbate content. This outcome indicated that basil performed well under the conditions and the system employed in the present study, and might be a good candidate for use as a secondary crop in cascade-hydroponics systems.

Studies were carried out to determine the effect of micronization on energy, starch and amino acid digestibilities in hulless barley. Six pigs (Canabrid × Camborough) were weaned at 21 d of age and fitted with a simple T-cannula at the distal ileum on days 23 or 24. The pigs were fed one of three diets with major constituents of hulless barley and soybean meal (HB + SBM), micronized hulless barley and soybean meal (MHB + SBM) and corn starch and soybean meal (C + SBM) according to a repeated Latin square design. The pigs were fed three times daily, equal amounts at 8-h intervals. The diets were supplied at a rate of 5% (wt/wt) of body weight. The average body weight of the pigs was 9.3 kg at start and 15.9 kg at the conclusion of the experiment at 58 d of age. Faeces were collected for 48 h on days 6 and 7 and ileal digesta for 24 h on days 8 and 9. Chromic oxide was used as digestibility marker. The apparent ileal amino acid digestibilities in HB and MHB were determined by the difference method. The apparent ileal digestibilities of the indispensable amino acids were higher in MHB than in HB and ranged from 5.3 to 10.0 percentage units. Of the indispensable amino acids, the differences were significant (P < 0.05) for arginine, histidine, isoleucine, phenylalanine and valine. Micronization of HB improved (P < 0.05) the ileal digestibility of starch from 79.0 to 97.3%. Micronization resulted in an increase in the digestion of energy in the small intestine and a decrease in microbial fermentation of energy in the large intestine. This shift in the disappearance of energy from the large to the small intestine should also result in an improvement in the efficiency of energy utilization. These studies show a positive effect of micronization on the digestibilities of energy and amino acids in young pigs fed HB. Key words: Micronization, barley, digestibility, pigs

Objective: In this paper amino acid profile of Lentinus sajor-caju (Fr.) Fr., a basidiomycetous mushroom has been investigated.Methods: During the evaluation 15 amino acids (lysine, aspartic acid, serine, threonine, glutamic acid, cysteine, glycine, alanine, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine and histidine) were determined from the dried sample of Lentinus sajor-caju by following the standard technique of biochemistry using ion-exchange chromatography.Results: The total amino acid content has been evaluated at 18.82 g/100g. Amongst the evaluated amino acids, exogenous amino acid lysine (6.66 g/100g) is preponderantly present in comparison to all other amino acids. The essential amino acid (EAA) index (44.64%) and biological value (BV=36.93%) has also been determined for the examined sample.Conclusion: Lentinus sajor-caju (Fr.) Fr. is a potential source of quality protein with a substantial proportion of exogenous and endogenous amino acids.

Ideal protein (amino acids) concept focused on broiler ABSTRACT. Accurate estimation of the amino acid requirement is very importance for diet formulation in growing animals such as broiler. Inadequate formulation of dietary amino acids will impair protein utilization and increase the total nitrogen output to the environment. Amino acids requirement of animals changes due to dietary, sex, and genetic and environmental factors. To address all potential combinations of factors with dose-response experiments is almost impossible. Thus the ideal protein concept provides a simple and effective approach to solve this problem. Based on this concept, the requirement of indispensible amino acids can be determined. For this concept, lysine has been used as the reference amino acid to calculate other indispensable amino acids due to several considerations. This paper will review more detailed about protein concept based on amino acid profile by using broiler as sampel review.

Background. Urolithiasis is the process of stone formation in the urinary tract. Its etiology is only partly known, and efficient therapeutic approaches are currently lacking. Metabolomics is increasingly used in biomarkers discovery for its ability to identify mediators of relevant (patho)physiological processes. Amino acids may be involved in kidney stone formation. The aim of the present study was to investigate the presence of an amino acid signature in stone former urine through a targeted metabolomic approach. Methods. A panel of 35 amino acids and derivatives was assessed in urines from 15 stone former patients and 12 healthy subjects by UPLC-MS. Partial Least Squares Discriminant Analysis (PLS-DA) was used to define amino acid profiles of cases and controls. Results and Discussion. Our approach led to the definition of a specific amino acid fingerprint in people with kidney stones. A urinary amino acid profile of stone formers was characterized by lower levels of α-aminobutyric acid, asparagine, ethanolamine, isoleucine, methionine, phenylalanine, serine, tryptophan, and valine. Metabolomic analysis may lend insights into the pathophysiology of urolithiasis and allow tracking this prevalent condition over time.

Aim and Objective:: To determine the mechanisms present in the etiopathogenesis of nasal polyposis. It is not clear whether amino acids contribute in a causal way to the development of the disease. Therefore, the aim of this study was to determine the plasma-free amino acid profile in patients with nasal polyposis and to compare the results with a healthy control group. Materials and Methods:: This was a prospective controlled study that took place in the Otolaryngology Department at the Harran University Faculty of Medicine between April 2017 and April 2018. Plasmafree amino acid profile levels were studied in serum samples taken from a patient group and a healthy control group. Patients who were diagnosed with bilateral diffuse nasal polyposis and were scheduled for surgical interventions were included in this study. Individuals whose age, gender, and body mass index values were compatible with that of the patient group and who did not have any health problems were included in the control group. All the participants whose levels of plasma-free amino acid were thought to be affected by one or more of the following factors were excluded from the study: smoking and alcohol use, allergic rhinitis presence, the presence of acute or chronic sinusitis, a history of endoscopic sinus surgery, unilateral nasal masses, a history of chronic drug use, systemic or topical steroid use in the last three months for any reason, and liver, kidney, hematological, cardiovascular, metabolic, neurological, or psychiatric disorders or malignancies. However, the gamma-aminobutyric acid (gaba: ng = 0.16 (0.10 – 0.24); cg = 0.21 (0.19 – 0.29); p = 0.010) plasma concentration was significantly lower in the nasal polyposis group than in the healthy control group. Conclusion: In this study, plasma levels of 15 free amino acids were significantly higher in the nasal polyposis group than in the healthy control group. A plasma level of 1 free amino acid was found to be significantly lower in the nasal polyposis group compared to the healthy control group. Therefore, it is important to determine the possibility of using the information obtained to prevent the recurrence of the condition and to develop effective treatment strategies. This study may be a milestone for studies of this subject. However, this study needs to be confirmed by further studies conducted in a larger series.

Aim. Amino acid metabolism in cancer patients differs from that in healthy people. In the study, we performed urine-free amino acid profile of gastric cancer at different stages and health subjects to explore potential biomarkers for diagnosing or screening gastric cancer.Methods. Forty three urine samples were collected from inpatients and healthy adults who were divided into 4 groups. Healthy adults were in group A (n=15), early gastric cancer inpatients in group B (n=7), and advanced gastric cancer inpatients in group C (n=16); in addition, two healthy adults and three advanced gastric cancer inpatients were in group D (n=5) to test models. We performed urine amino acids profile of each group by applying ion chromatography (IC) technique and analyzed urine amino acids according to chromatogram of amino acids standard solution. The data we obtained were processed with statistical analysis. A diagnostic model was constructed to discriminate gastric cancer from healthy individuals and another diagnostic model for clinical staging by principal component analysis. Differentiation performance was validated by the area under the curve (AUC) of receiver-operating characteristic (ROC) curves.Results. The urine-free amino acid profile of gastric cancer patients changed to a certain degree compared with that of healthy adults. Compared with healthy adult group, the levels of valine, isoleucine, and leucine increased (P<0.05), but the levels of histidine and methionine decreased (P<0.05), and aspartate decreased significantly (P<0.01). The urine amino acid profile was also different between early and advanced gastric cancer groups. Compared with early gastric cancer, the levels of isoleucine and valine decreased in advanced gastric cancer (P<0.05). A diagnosis model constructed for gastric cancer with AUC value of 0.936 tested by group D showed that 4 samples could coincide with it. Another diagnosis model for clinical staging with an AUC value of 0.902 tested by 3 advanced gastric cancer inpatients of group D showed that all could coincide with the model.Conclusions. The noticeable differences of urine-free amino acid profiles between gastric cancer patients and healthy adults indicate that such amino acids as valine, isoleucine, leucine, methionine, histidine and aspartate are important metabolites in cell multiplication and gene expression during tumor growth and metastatic process. The study suggests that urine-free amino acid profiling is of potential value for screening or diagnosing gastric cancer.

Abstract The objective was to determine the influence of amino acid (AA) supplementation during the adaptation period on the ileal digestibility of crude protein and AA in corn and soybean meal (SBM). Six barrows with an initial body weight of 30.9 ± 2.6 kg fitted with a T-cannula in the distal ileum were assigned to a 6 × 6 Latin square design with 6 dietary treatments and 6 periods. Two experimental diets contained corn or SBM as the sole source of AA and an N-free diet was additionally prepared. For AA supplementation groups, an AA mixture consisted of Gly, Lys, Met, Thr, Trp, Ile, Val, His, and Phe was added to the corn diet and the N-free diet at the expense of cornstarch, and an AA mixture of Lys, Met, and Thr was added to the SBM diet. All diets contained 0.5% of chromic oxide. The 6 experimental diets were fed to the pigs for 4 and half days, and the 3 diets containing AA mixture were switched to the respective diets without AA mixture during the following 2 and half days. Ileal digesta were collected during the last 2 days. The addition of AA mixture during the adaptation period caused increased apparent ileal digestibility of Arg and Trp in corn (P &lt; 0.05), but did not affect that in SBM. The addition of AA mixture during the adaptation period caused increased apparent ileal digestibility of Pro and Gly regardless of feed ingredient (P &lt; 0.05), but did not affect that of other AA. All AA except Pro in corn and SBM were unaffected by the addition of AA mixture during the adaptation period. In conclusion, the addition of amino acid during the adaptation period does not affect the standardized ileal digestibility of indispensable amino acids in feed ingredients.
