This study was carried out to determine the coliform contamination of public boreholes and pipe borne water supplies within Bosso town. Twenty (20) water samples comprising of 10 each of borehole and pipe borne samples were aseptically collected from Bosso Town and analyzed using membrane filtration technique. The results obtained revealed that most (60.0%) of the water samples from the borehole sources except the samples from Rafin-Yashi, Maikunkele, Federal University of Technology (F.U.T) Minna, Tudun Fulani, contained coliform counts within 10cfu/100ml while all (100.0%) of the pipe borne water samples had coliform counts above 10cfu/100ml. The organisms isolated included species of Escherichia, Pseudomonas, Streptococcus, Staphylococcus, Salmonella, Shigella, Clostridium, Bacillus, Yersinia and Serratia. E.coli had the highest frequency of occurrence (20%) followed by Staphylococcus aureus (11.7%), Salmonella spp (11.7%), Shigella spp (11.7%), Clostridium spp (8.3), Streptococcus faecalis (8.3%), Pseudomonas aeruginosa (6.7%), Bacillus subtilis (6.7%), Streptococcus pyrogenes (5%), Klebsiella spp (3.3%), Proteus vulgaris (3.3%), Yersinia spp (1.7%) and Serratia spp (1.7%). This study revealed that pipe borne water and borehole water samples were contaminated in Bosso, with greater contamination observed with pipe borne water. This highlights the need for continuous assessment of the quality of public water supply and intervention measures to prevent outbreak of water-borne diseases in the area.Keywords: Water; Boreholes; Pipe borne; Coliforms; Water-borne disease

A total of 50 water samples comprising; 20 well water, 15 sachet water, 10 borehole water and 5 river water samples, were obtained from Wamakko, Dange-shuni and Wurno local government areas in Sokoto Central, Sokoto West and Sokoto East, Nigeria respectively between March and May 2015. They were analyzed using membrane filtration and pour plate techniques for faecal coliforms count and total coliform count respectively. The total coliform count for all the sources ranged between 0 and 3.28 × 102 with a total mean of 1.11 × 10,2 that of faecal coliform count was found to range between 0 and 92 with a mean total count of 12.58. Among the bacteria isolated and identified, Escherichia coli had the highest total prevalence of 233 (40.31%) and occurring in all the water samples. Klebsiella spp had 99 (17.13%) occurring in all the sources, Salmonella spp 43 (7.44%) occurring in all the sources except for borehole, Pseudomonas species 88 (15.22%) occurring in all the sources, Staphylococcus aureus with 115 (19.90%) occurring in all the sources except for borehole. However, the pHs for all the samples were found to be within the range of 6.51 to 8.0 which are within WHO acceptable range of 6.0 to 8.5. From this study, it is obvious that the water sources from the areas under study are bacteriologically unfit for drinking purposes. These samples with high total coliform count are of concern and calls for urgent and stringent measures aimed at ensuring a safer drinking water for the populace.

The concern over exposure to drinking water contaminated and the resultant adverse effect on human health has prompted several studies evaluating the quality of drinking water sources. This study was carried out to determine the bacteriological and Physico-chemical qualities of commercial borehole water within Otuoke community in Bayelsa state Nigeria. Ten (10) water samples were collected from the various locations designated as (A-J). The Physico-chemical parameters were determined by using the photometric technique through the Colour Q photometer and the bacteriological analysis was determined by using aerobic plate method. The Physico-chemical parameters of the water samples analysed were within the acceptable limit of WHO standard of drinking water quality except for Bromine and cyanuric acid which ranged from 1.6±0.1 to 2.7±0.1 and 2±1 to 9±1 respectively. The Result of total bacteria count obtained from the borehole water samples ranged from 1.1 x 105cfu/ml to 6.9 x 105cfu/ml and there was no detected growth for the faecal coliform count of most of the samples except for BHW-I and BHW-J which had 1.3 x 104 to 7.9 x 104 respectively. Bacteria isolated and identified using conventional biochemical test include E. coli (17.9%), Klebsiella spp. (35.7%), Salmonella spp(25.0%), Enterobacter aerogenes (21.4%) The study therefore suggests that all the borehole water tested in Otuoke and environs are considered unsafe for consumption and therefore regular treatment before usage is recommended.

Because damaged cells may account for a substantial proportion of the bacterial population in processed foods, the food microbiologist must choose the most appropriate methods for detecting damaged as well as noninjured cells. Any method intended to recover damaged organisms should include a resuscitative, or repair, process that will restore the injured cells to a sound physiological condition before subjecting them to the severity of selective enrichment media. It should also provide a reliable indication of the microbiological safety and quality of any particular food. This paper reviews various factors that affect the recovery of Salmonella spp., which include: (a) sample rehydration, (b) period of preenrichment, (c) incubation in both aerobic and anaerobic environments, (d) media composition and (e) the relative merits of preenrichment and direct selective enrichment. Because resuscitation of injured Salmonella cells does not occur during the selective enrichment step and beyond, the effect and interaction of these factors are considered primarily for the preenrichment step of the isolation procedure for Salmonella. This paper also reviews five methods recently developed for recovery of coliforms, which include: (a) hydrophobic grid membrane filtration, (b) radiometry, (c) electrical impedance, (d) fluorogenic assay and (e) the Petrifilm system. Each of these methods may incorporate a step for resuscitation of injured organisms.

A study of imported unprepared whole lettuces sampled from supermarkets, greengrocers, shops, and market stalls found that all were of acceptable microbiological quality. Twenty-seven out of 151 (18%) imported lettuce samples had Enterobacteriaceae levels of 104 CFU/g or more. However, these bacteria that constitute part of the natural microflora of unprepared vegetables may also be derived from the soil and/or by poor handling. The pathogens, Salmonella spp., Shigella spp., Campylobacter spp., Escherichia coli O157:H7, Vibrio cholerae, Listeria monocytogenes, and also Escherichia coli, an indicator of fecal contamination, were not detected in any imported lettuces, indicating that hygiene, harvesting, and production practices were good. Imported lettuces with Enterobacteriaceae levels of 104 CFU/g or more varied with type of retail premises and the temperature at which the lettuces were displayed. Samples from greengrocers, shops, and market stalls were more likely to contain Enterobacteriaceae at levels in excess of 104 CFU/g than those from supermarkets.

Water from boreholes and packaged commercial sachet water from different areas in a community in southern Nigeria was analyzed with membrane filtration for a snapshot of heterotrophic count and coliforms. Two boreholes out of the 20 analyzed had counts of over 500 Cfu/mL and 7 boreholes indicated the presence of coliforms. Sixteen samples out of 20 sachet water brands analyzed showed a regulatory product registration code, whereas 4 samples had no number or code indicating that they were not registered. The heterotrophic count of all sachet water was well within the limit for all samples analyzed, and coliform was detected in only two samples. The overall quality of borehole water in the community studied was rated D (65%), whereas the sachet water was rated C (90%) according to the World Health Organization (WHO) surveillance guidelines. Improvements in water quality structure in the community studied are required to help achieve WHO sustainable development goals on water sanitation. The etiology, virulence properties, epidemiology, and pathogenicity of bacteria associated with borehole and sachet water are also discussed.

Commercial drinking water may serve as potential threat to public health if these items are contaminated with a number of pathogenic microorganisms due to faulty manufacturing process. Present study attempted to isolate and quantify the microorganisms from various jar and bottle water samples collected from various areas of Dhaka city. Antibiotic susceptibility pattern of suspected bacterial isolates were also determined in this study. Out of the eighteen samples studied, ten were jar water samples and eight were bottled water samples. The range of total viable bacterial count (TVBC) in these samples ranged from 102 to 105 cfu/ml. Specific pathogens such as, Salmonella spp., Shigella spp., Vibrio spp. and fecal coliforms could not be found in these samples. However coliforms could be detected in 10 samples. The antibiogram study showed that all Escherichia coli and Klebsiella spp. isolates found from these samples were sensitive against gentamicin (10 μg) and azithromycin (30 μg). Variable antibiotic resistance among these bacterial isolates was detected against cefotaxime (30 μg), streptomycin (10 μg) and erythromycin (15 μg). Due to delicious taste and readily availability, desserts are one of the most popular foods in Dhaka city. High amount of carbohydrate and protein in dessert items make them more susceptible to proliferation of microbial growth. Present study depicted a complete microbiological profile of some popular desserts such as, sweet, pastry, ice cream, pudding, falooda, yogurt and custard available in different food shop at Dhaka city, Bangladesh. All the samples were found to be contaminated with heterotrophic bacteria as well as fungi within the range of 103 to 106 cfu/g. In case of specific microflora, the growth of Staphylococcus spp., Klebsiella spp. and Pseudomonas spp. were observed in most of the samples indicates the poor quality of these products. Bioburdens of E. coli in sweet, pudding and yogurt were found in the range of 1.2×102 to 2.7×103 cfu/g. Salmonella spp., Shigella spp. and Vibrio spp. could not be isolated from any of the samples. Current study indicates that hygienic conditions should be maintained during preparation, packaging and retailing of dessert items in order to reduce the load of contamination in ready to eat foods which will ensure good health of consumers. The Sustainable Development Goal on sanitation aims to achieve universal access to good health, affordable drinking water, sanitation and an end to open defeacation by 2030. The recent ranking of Nigeria as first globally for open defecation is of public and environmental health concern. This study assessed the sanitary condition and the microbiological quality of well and surface waters of Panhauya community and Ahmadu Bello University farm, Zaria, and the antibiogram of the bacterial isolates.. Based on the WHO criteria, the sanitary inspection showed that 16.7%, 54.2%, 25% and 4.2% of the water sampling points had a very high, high, intermediate and low risk of contamination respectively. Occurrence of Escherichia coli, Giardia lamblia, Entamoeba histolytica, Pseudomonas aeruginosa, Salmonella spp and Vibrio cholerae in water samples from Panhuaya community was 87.5%, 75%, 68.8%, 50%, 25% and 12.5% respectively. In ABU farm Shika, the occurrence was; E. coli (75%), E. histolytica (63%), G. lamblia and Salmonella spp. All E. coli isolates exhibited high multidrug resistance to antibiotics screened with a MAR index of 0.3-0.8. The drinking water sources in Panhuaya and ABU farm were unsafe and the presence of these pathogens in the water samples may be attributed to a number of factors including poor sanitation, manure application and open defecation practice. This indicates a public health risk to the residents and emphasises the need for safe water supplies sanitation and antibiotic stewardship. Bermuda residents collect rainwater from rooftops to fulfil their freshwater needs. The objective of this study was to assess the microbiological quality of drinking water in household tanks throughout Bermuda. The tanks surveyed were selected randomly from the electoral register. Governmental officers visited the selected household (n = 102) to collect water samples and administer a short questionnaire about the tank characteristics, the residents’ habits in terms of water use, and general information on the water collecting system and its maintenance. At the same time, water samples were collected for analysis and total coliforms and Escherichia coli were determined by 2 methods (membrane filtration and culture on chromogenic media, Colilert kit). Results from the 2 methods were highly correlated and showed that approximately 90% of the samples analysed were contaminated with total coliforms in concentrations exceeding 10 CFU/100 mL, and approximately 66% of samples showed contamination with E. coli. Tank cleaning in the year prior to sampling seems to protect against water contamination. If rainwater collection from roofs is the most efficient mean for providing freshwater to Bermudians, it must not be considered a source of high quality drinking water because of the high levels of microbial contamination.

Eighty-eight samples of ground meat were randomly collected from retail stores in the metropolitan area of Monterrey, Mexico, and were analyzed for microbial contamination. Methods were those recommended by the Mexican regulation and/or the U.S. Food and Drug Administration. Over 75% of the samples contained 105 total mesophilic microorganisms per g, and over 40% had 106 total coliforms per g. Fecal coliforms were present in most samples. Staphylococcus aureus was detected in 2.3% of the samples, Salmonella spp. in 11.4%, Listeria spp. in 62%, and L. monocytogenes in 16%. Escherichia coli was detected in 76% of samples, but none was serotype O157:H7. Shigella spp. was not found in any sample. Fusarium spp. and Mucor spp. were detected in 3.4% of the samples, and low levels of yeast in 93%. The microbiological quality of the ground meat analyzed was unsatisfactory, and the product could be an important cause of food poisoning.
