Purpose: To investigate Heliotropium strigosum whole plant extract for its potential to reduce the blood glucose level of alloxan-induced diabetic mice. Methods: Preliminary phytochemical analysis was carried out using standard procedures. Diabetes was induced in Balb/c mice by injecting alloxan (200 mg/kg i.p.). The crude methanol extract of Heliotropium strigosum (Hs.Cr, 30, 100 and 300 mg/kg doses) was administered daily to alloxan-induced diabetic mice for 15 days and its effect on fasting blood glucose levels, body weight and oral glucose tolerancewas evaluated. Two control groups (non-diabetic control and diabetic control) received normal saline (0.2 ml). Metformin (500 mg/kg) was used as reference standard. Results: Heliotropium strigosum showed positive for the presence of alkaloids, tannins and flavonoids. The extract (30, 100 and 300 mg/kg) caused significant reduction in the fasting blood glucose level of alloxan-induced diabetic mice on days 5, 10 and 15 compared to diabetic control (p < 0.001). In this regard, the anti-hyperglycemic effect compared to the reference (metformin). The extract also timedependently decreased the body weight of the treated animals as well as improved tolerance of the oral glucose overload. Conclusion: These results indicate that Heliotropium strigosum possesses anti-hyperglycemic effect, reduces body weight and enhances the tolerance of glucose overload in mice. Further studies are therefore required to determine its feasiilty as an alternate herbal medicine in the management of diabetes in humans. Keywords: Heliotropium strigosum, Anti-hyperglycemic, Alloxan-induced diabetic mice, Blood glucose, Oral glucose tolerance, Body weight

Objective: To evaluate the anti-diabetic and anti-diarrheal activity of methanol extract of Flemingia stricta Roxb. (Fabaceae) leaf. Methods: In anti-diabetic study, the extract was administered to alloxan-induce diabetic mice at two concentrations (200mg/kg and 400mg/kg body weight) for acute (12 hours) and prolong treatments (15 days) and blood glucose levels (Blood glucose level) of diabetic mice were monitored at intervals of hours and days throughout the duration of treatment. Antidiarrheal test was conducted by castor oil induced diarrhea and enteropooling as well as intestinal motility in mice at three different concentration (100mg/kg, 200mg/kg and 400mg/kg body weight). Results: Treatment of alloxan induce diabetic mice with the extract caused a significant reduction in fasting blood glucose level of the diabetic mice both in acute (12 hours) and prolong treatment (15 days) and it was determined that the extract at both concentration (200mg/kg and 400mg/kg) showed the significant (P<0.05) hypoglycemic effect in comparison to the standard drug Metformin (10mg/kg). In the case of castor oil induced diarrheal test, enteropooling test and gastrointestinal motility test, the extract of F. stricta at 100mg/kg, 200mg/kg and 400mg/kg has given significant effect (P<0.05) in comparing to standard drug Loperamide (5mg/kg). Conclusion: These result suggested that the methanol extract of F. stricta Roxb. possess promising anti-diabetic effect on alloxan-induced mice and significant antidiarrheal effect on castor oil induced diarrheal mice.

The purpose of this study was to show the antidiabetic effect of ethanolic extract of Aloe vera gel in alloxan induced hyperglycaemic mice. Ethanolic extract of Aloe vera gel was orally administered for four weeks at a dose of 300mg/kg body weight. Weekly estimates of fasting blood glucose level in normal non-diabetic and alloxan induced hyperglycaemic mice were assessed. Ethanolic Aloe vera gel extract revealed no blood glucose lowering action upon non-diabetic mice. But, a significant reduction in blood glucose level (p<0.001) was seen in diabetic mice, when compared with diabetic control. Similar results were found when compared with a standard antidiabetic drug, gliclazide. To conclude, as administration of ethanolic extract of Aloe vera gel significantly reduces blood glucose level in hyperglycaemic mice, it can optimistically emerge as a new hope in the management of diabetes mellitus. Some lactic acid bacteria (LAB) are observed to be potential probiotics with functional properties such as lowering fasting blood glucose (FBG), as a promising hyperglycemia management. This study investigated the ability and mechanism of Lactobacillus rhamnosus BSL and Lactobacillus rhamnosus R23 on lowering FBG in diabetic rats induced by streptozotocin (STZ). The rats were orally administered with L. rhamnosus BSL and L. rhamnosus R23 by giving 1 mL cell suspension (109 CFU/mL) daily for 30 days. The body weight (BW) was recorded once in three days, and FBG was recorded once in six days. An oral glucose tolerance test (OGTT) was measured 1 week after injection with STZ and before sacrifice. Fecal samples were collected on days 0, 15, and 30 for LAB population and identification, performed by PCR detecting 16S rRNA. Oral administration of L. rhamnosus BSL and L. rhamnosus R23 decreased FBG and improved glucose tolerance via downregulation of glucose-6-phosphatase (G6pc) expression by 0.57- and 0.60-fold change, respectively (P<0.05). The lipid profiles, BUN, creatinine, SGOT, and SGPT were significantly (P<0.05) different between normal and diabetic rats, but they were not significantly (P>0.05) different among diabetic rats. Both strains were effective in increasing fecal LAB population. Molecular identification of the isolated LAB from fecal sample indicated that they were able to survive and pass through the digestive tract. These results suggested that both strains have the ability to manage blood glucose level and become a promising agent to manage hyperglycemia and diabetes.

Gynura divaricata (L.) DC and G. bicolor DC are used as secret recipes to treat diabetes mellitus in some parts of China. Pharmacological tests were performed to prove the anti-hyperglycemic effect of these two plants of genus Gynura Cass. in this study. Both water and 95% ethanol extracts of fresh G. divaricata had significant effects on lowering blood glucose level in normal mice, in which the dose of 0.4 g (crude drug)/kg of 95% ethanol extract was more effective than 50 mg/kg glyburide. The ethyl acetate and n-butanol extracts of dried G. divaricata had significant effects on lowering blood glucose level in normal and alloxan diabetic mice too. Both ethyl acetate and n-butanol extracts of dried G. bicolor showed very significant effect on lowering blood glucose level to normal and alloxan-diabetic mice, and the dose 4.0 g (crude drug)/kg had a more hypoglycemic effect than 50 mg/kg glyburide in normal mice.

Combination therapy is considered as a new and effective therapeutic strategy for control of hyperglycemia. The present study aimed to determine the hypoglycemic effect of Afghan Hibiscus sabdariffa L. and Carum Carvi L. hydro-alcoholic extracts either alone or in combination on diabetic rats. Streptozotocin-induced diabetic rats were administered intraperitoneally with hydro-alcoholic extracts of H. sabdariffa (75, 150 and 300 mg/kg), C. carvi (150, 300 and 500 mg/kg) and their combinations for 21 days. The Fasting blood glucose and body weight of rats were determined on 0, 7, 14 and 21st days. The results showed a significant (P<0.0001) hypoglycemic effect of H. sabdariffa extract (75, 150 and 300 mg/kg) on 7, 14 and 21th days, C. carvi extract (500 mg/kg), and their effective doses combination (75:25), especially on 7th day. Only 75 mg/kg of H. sabdariffa had positive effect on body weight loss of diabetic rats (P<0.05). One can conclude that Afghan H. sabdariffa and C. carvi hydro-alcoholic extracts and their combination have a hypoglycemic effect. However, their combinations do not have any synergic effects, so the hypoglycemic effect of H. sabdariffa was much greater and even normalized the blood glucose level.

Background: Diabetes mellitus is a genetic and endocrine system disease that creates injurious health effects not in Pakistan but throughout the world. Several functional compounds are present in buckwheat which has distinct biological properties of cholesterollowering, anti-hypertension, improving constipation and obesity. The recent research was directed to study the nutritional and hypoglycemic effect of buckwheat in Type 2 diabetic rats induced by alloxan. Methods: Two varieties of buckwheat were examined for their nutritional composition and mineral content according to the methods of AACC (2000) while antioxidant activity was measured by using DPPH and a spectrophotometer. Alloxan induced diabetic mice were applied to investigate the blood-glucose-lowering effect of common buckwheat extract (CBE) and Tartary buckwheat extract (TBE). After type 2 diabetic induced rats were separated randomly into three groups. Results: The results revealed that Common buckwheat flour (CBF) contains12. 93% crude protein, 2.39% crude fat, 9.42% crude fiber and 2.90% ash content, while Tartary buckwheat flour (TBF) contains14.88% crude protein, 3.47% crude fat, 12.03% crude fiber, and 2.86% ash content. TBF contains significantly higher minerals as compared to CBF and also exhibited higher antioxidant activity. The results for diabetes-induced mice showed a significant difference for fasting blood glucose level, glucose tolerance test and body weight among control diabetic mice (group I) and diabetic mice given the CBE (group II) and TBE (group III). The fasting blood glucose level of group I increased while it decreased in group II and group III over time. Group II and group III had more glucose tolerance as compared to group I. Conclusion: The present study demonstrated that TBF contains higher nutritional contact as compared to CBF. Furthermore, it was indicated that the oral administration of CBE and TBE could lower the blood glucose level and can increase glucose tolerance effectively.
