Benign prostatic hyperplasia (BPH) is the most common benign proliferative disease among men during aging. The herbal extract of P. africanus has been used for the treatment of BPH since time immemorial. However microstructural changes of this extract to the prostate of animal or human models are still elusive. Therefore, histological and histomorphometric changes in rats with testosterone-induced BPH due to P. africanus herbal extracts were investigated. Twenty-eight male Wistar rats (200 ± 50 g) were divided into four groups each with 7 rats. Group 1 (Negative control) was given 2 ml/day of olive oil subcutaneously and 2 ml/day of normal saline intragastrically for 28 days. Three groups were induced with BPH by subcutaneous injection of testosterone propionate 3 mg/kg body weight/day for 28 days. Thereafter Group 2 (BPH) was sacrificed, while Group 3 (Positive control) and Group 4 (Treatment group) were administered with finasteride 5 mg/kg/day and P. africanus extract 400 mg/kg/day intragastrically for 28 days, respectively. BPH group revealed thickening and hyperplasia of tubular epithelium with involutions with the stroma showing large spaces and dilated blood vessels. These features were restored with P. africanus extract administration. High epithelial height, large stromal area and lower luminal area observed in the BPH were greatly reversed with P. africanus extract comparable to negative controls. Generally, P. africanus extract restored and ameliorated histological and histomorphometrical changes of the BPH-induced rat’s prostates. Anredera cordifolia or binahong is one of the Indonesian medicinal plants that is used to treat peptic ulcer. The purpose of this study was to evaluate the effect of the addition of sodium carboxymethylcellulose (CMC) and sorbitol on anti-peptic ulcer activity of A. cordifolia leaves extracts in male Wistar rats. The plants were extracted using decoction method and freeze dried. Three liquid formulas were used i.e., i) a combination of sodium CMC and sorbitol; ii) only sorbitol; iii) extract only. The rats were divided into 6 groups, i.e., positive control (sucralfate 35 mg/kg body weight); negative control (80% ethanol); normal control; and 3 formulas. After the administration of the liquid formula, all groups, except normal control, were given 80% ethanol (l5 ml/kg body weight) to induce peptic ulcer. Antipeptic ulcer activity was evaluated using direct observation on rats gastric mucosa, and histopathology assessment. The result showed that the strongest anti-peptic ulcer  was shown by sorbitol only (96.95% inhibition),  followed by the combination of sodium CMC and sorbitol (92.68% inhibition). The formula which only contained extract showed only  31.70% inhibition.  Statistical analysis showed significant differences between formula 1 and 2 with negative controls. In conclusion, A. cordifolia leaves extract with the addition of sorbitol showed the strongest anti-peptic ulcer activity. Keyword: Anredera cordifolia, peptic ulcer, suspense, Wistar rat.

The present study was conducted to evaluate the mutagenic and antimutagenic potential of Cousinia thomsonii (CT) extract in bone marrow cells of male wistar rats using some important parameters like micronucleated polychromatic erythrocyte (MnPCE), mitotic index (MI), chromosomal aberrations (CA) and polychromatic erythrocyte to normochromatic erythrocyte ratio (PCE/NCE). 30 male rats of wistar strain were divided into 6 groups with 5 rats each group. Group 1 rats were taken as negative control having free access to distilled water and rat feed. Group 2 rats were taken as positive control treated with mutagen cyclophosphamide (CP) at dose of 60 mg/kg b wt. for 2 days. Group 3 and 4 were treated with CT extract at dose of 100 and 200 mg/kg b wt. for 20 days. Group 5 and 6 were treated with 100 and 200 mg/kg b wt of CT extract for first 18 days and for last 2 days with CP at dose concentration of 60 mg/kg. It was found that rats treated with CT extract alone did not produce any significant changes in MnPCE, PCE/NCE ratio, CA and MI when compared with control treated rats (group 1). However in group 5 and 6 rats treated with CT extract in combination with CP a protective effect was observed against the cyclophosphamide induced cellular mutagenicity. In concluding remark Cousinia thomsonii was found to show antigenotoxic potential and also produce protective antimutagenic effects against CP induced chromosomal damage.

The present study was carried out to investigate the nephroprotective effect of the ethanolic extract of Allium saralicum R.M. Fritsch (ASRMF) in mice. Thirty-five male mice were divided into five groups (n=7). Group 1 (positive control) received 1 mL/kg olive oil intraperitoneally (i.p.) and 0.5 mL distilled water orally; Group 2 (negative control) received CCl4 (50% in olive oil, 1 mg/kg; i.p.); Groups 3, 4 and 5 received CCl4 and 200, 800 and 1600 ?g/kg of ASRMF extract, respectively. The renal volume and cortex in Groups 1 and 2 were increased by 55% and 62% (p?0.001) following CCl4 administration, respectively, and were improved after ASRMF administration. The volume of proximal convoluted tubules (PCTs), glomeruli, vessels and interstitial tissue increased 80%, 150%, 83% and 64% (p?0.05), respectively, in CCl4-treated mice, and decreased significantly with 800 and 1600 ?g/kg of ASRMF. The length of PCTs and vessels increased 51% and 45% and decreased (p?0.05) with 200, 800 and 1600 ?g/kg of ASRMF, respectively. CCl4-treated mice lost 22.5% of glomeruli; the loss was inhibited significantly (p?0.05) by ASRMF. Urea and creatinine concentrations were increased (p?0.05) in CCl4-induced nephrotoxicity as compared to the controls, whereas different doses of ASRMF restored the levels of these biomarkers compared to the negative controls. In conclusion, ASRMF has a potent nephroprotective property and can improve renal structural and serum biomarkers in CCl4-induced nephrotoxicity in mice.

AbstractAlpha-1 blockers, often used to treat benign prostate hyperplasia (BPH), have been hypothesized to prevent COVID-19 complications by minimising cytokine storms release. We conducted a prevalent-user active-comparator cohort study to assess association between alpha-1 blocker use and risks of three COVID-19 outcomes: diagnosis, hospitalization, and hospitalization requiring intensive services. Our study included 2.6 and 0.46 million users of alpha-1 blockers and of alternative BPH therapy during the period between November 2019 and January 2020, found in electronic health records from Spain (SIDIAP) and the United States (Department of Veterans Affairs, Columbia University Irving Medical Center, IQVIA OpenClaims, Optum DOD, Optum EHR). We estimated hazard ratios using state-of-the-art techniques to minimize potential confounding, including large-scale propensity score matching/stratification and negative control calibration. We found no differential risk for any of COVID-19 outcome, pointing to the need for further research on potential COVID-19 therapies.

Objective: Our study aimed to the efficacy of silodosin in patients with LUTS associated with BPH who were not-responder to previous ARs blocker therapy. Methods: Patients who did not benefit from alpha blockers treatment, but did not want surgical treatment are included in this study. At enrollment, 75 patients and 75 patients were assigned to group 1 and group 2, respectively. Group 1 received silodosin 8 mg, group 2 received their previous α blocker. Results: Although, in group 1 mean IPSS score at baseline was 20,81±0,97, and it significantly decreased to 17,12±1,25 at third months, in group 2 no significant changes were observed. On the other hand in group 1 a decrease was also observed for both IPSS sub score was significantly reduced at third months, when it is compared with baseline. At the end of the third month, a significant improvement in this parameter was observed after switching to silodosin, as compared with first value (p< 0,05). As to the residual urine, significant improvement was observed in the silodosin group but no significant improvement was observed in group 2. Conclusion: This study was conducted to investigate the effectiveness and safety of silodosin in patients with BPH who had not achieved satisfactory symptom control with other α blockers in routine clinical practice before surgical treatment. Thus, at least patients who have different comorbidities will be protected from morbidities of surgery.
