Introduction: Nicotine, a major component of cigarette smoke, alters ovarian function and Ajwa (Phoenix dactylifera) being an antioxidant prevents its toxicity. Aims & Objectives: This experimental study was designed to evaluate protective effect of Ajwa against ovarian toxicity induced by nicotine in adult albino rats. Place and duration of study: The study was conducted in the Department of Anatomy, Federal Postgraduate Medical Institute, Shaikh Zayed Medical Complex, Lahore for duration of 3 months (January 2017-March 2017). Material & Methods: 40 adult female albino rats were divided into four equal groups. 1ml distilled water was given to the rats of control (A) group by gastric intubation, experimental group B was given nicotine injection intraperitoneally (0.1 mg/kg body weight) for 28 days. The rats of experimental group C were given Ajwa fruit extract 1gm/kg body weight daily by gastric intubation, while rats of experimental group D were given 0.1mg/kg body weight nicotine intraperitoneally plus 1gm/kg body weight Ajwa fruit extract by gastric intubation for 28 days. Results: In group B, secondary and Graafian follicles were mostly atrophic, degeneration of granulosa cells, pyknotic nuclei and stromal vacuolization were present. Zona pellucida was torn and irregular. Mean diameters of secondary and Graafian follicles were significantly lower in nicotine group B (p-value<0-001*). Experimental group C revealed normal histological architecture and diameter of ovarian follicles. Near normal restoration of follicular structure was seen in group D. Conclusion: Ajwa fruit extract is counterbalancing for nicotine induced oxidative stress in ovarian follicles of rat, so it may be useful to improve infertility or sub-fertility induced by smoking in females.

Objective: To determine the anti-hyperglycemic effects of interleukin-1 inhibitor (diacerein) in alloxan induced diabetic albino wistar rats. This experimental study was performed at the Department of Animal Husbandry and Veterinary Sciences, Sindh Agriculture University, Tando Jam within 6 months from April 2016 to September 2016. Total of 160 adult Albino Wistar Rats having an average of 200 to 300 grams body weights were selected. Animals were categorized into 4 groups as; Group A (n=15): Control rats – receive 0.9% normal saline as placebo Experimental Groups Group B (n=15): Experimental Control (Diabetic rats) - Alloxan50 mg/kg body weight intraperitoneal. Group C (n=15): Diabetic rats + Diacerein (30 mg/kg/day) orally daily. Group D (n=15): Diabetic rats + Diacerein (50 mg/kg/day) orally daily. Animals were kept and treated as per the NIH Guideline for Use and Care of Laboratory Animals. Diabetes mellitus was induced via a single intraperitoneal injection of 50 milligram/kg alloxan monohydrated dissolved in aseptic 0.9% saline. After 72 hours, blood specimens were taken from the caudal vein of the rats and glucose level>200 mg/dL was taken as diabetes. Experimental rats were given diacerein approximately 30 and 50 mg orally for 6 weeks. At the completion of experiment the body weight was measured of each animal by electronic measuring balance and blood sample was taken from each animal of all groups to assess the blood glucose level and HbA1c level. Data were recorded via self-made proforma and analysis was done by using SPSS version 20. Results: Average body weight of Diabetic control (Group B) was 193.33±22.50 grams, which was lower in contrast to Diacerein treated group C 202.47±25.70 grams and significantly lower as compared to Diacerein treated group D as  212.6±23.43 grams. A significant increase in blood glucose levels 182.07±10.63 mg/dl was noted in the Diabetic control (Group B) compared to Diacerein treated group C (110.13± 8.54 mg/dl) and group D (85.87±8.41 mg/dl) (P=0.001). HbA1c was markedly raised in the Group B- diabetic controls, while diacerein treated diabetic rats (groups C and D) showed a significant decrease in HbA1c (P=0.001). Conclusion: It was concluded that Diacerein achieves the Euglycemic state by reducing the levels of blood glucose and glycated hemoglobin (HbA1c) in Alloxan-Induced diabetes mellitus in Wistar Albino Rats.

Background: Liver injuries induced by different toxic substances have been recognized as one of the great toxicological problem for years. However, there are a lot of herbal medicines existing to compensate these disorders.Alcohols are one of the most important organic compounds in different areas of our daily lives. The health benefits of Ajwa Date Fruit Extract are well documented in literature. Objective: To determine the effect of alcohol on weight of liver and possible protective role of Ajwa date fruit extract (ADFE) in adult male albino rats. Study Design:Experimental Study Place and Duration of Study: Department of Anatomy, Postgraduate Medical Institute, Lahore from 1st October 2018 to 31st March 2019. Methodology: Twenty four adult male albino rats were used in this research and divided into the 4 groups A, B, C and D having 6 rats in each group. Group A was designated as control group;group B received ethanol 3g/kg/day of 40% v/v prepared in distilled water; group C received ethanol and ADFE1g/kg/day and group D received ethanol and ADFE 2g/kg/day by oral gavage once daily for 28 days.The 100% pure alcohol was dissolved in distilled water for preparation of 40% alcohol. Dates palm (Phoenix dactylifera L.) fruits washed with tap water and removed the seeds. The extract of the date fruits made by adding distilled water to date fruit (3:1) and leaving for 48 hours in refrigerator (4°C). Results: The increase in mean weight of liver in group B on 29th day of experiment were statistically significant (p=0.001) in comparison with the control. The mean weight of liver was decreased significantly after 28 days of experiment in experimental groups C and D. Conclusions: Alcohol administration in the male albino rats significantly increase their weight of liver which was improved by the use of ADFE. Key words: Alcohol, Ajwa date fruit extract, Rats

Objective: to reveal the effects of sea buckthorn seed oil on acetaminophen induced variations (Diameter of hepatocytes and vacuolization) in liver of rats. Methods: it was experimental study, conducted at Post Graduate Medical Institute. This study was performed on 24 adult rats, which were then randomly separated into 4 groups A,B,C and D. the experiment lasted for 13 days. Group A was control received normal saline and distilled water. Group B, received acetaminophen750mg/kg as single dose on day 10. Group C and D received acetaminophen at dose of 750mg/kg along with sea buckthorn seed oil extract 2.6mg/kg and 5.2mg/kg. Liver was removed and fixed in 10% formalin. To observe the effect of acetaminophen and sea buckthorn seed oil , slides were prepared for histological examination. The diameter of hepatocytes and vacuolization was observed. The evaluation of results was done by using SPSS 21. Results: In Group B, vacuolization was seen in all animals and significant increase in mean diameter (11.9± 1.5 μm) of hepatocytes was seen. However in Group C, vacuolization was absent in all animals and mean hepatocyte diameter was (6 ± 0.6 μm). In Group D, no vacuolization was seen in all animals and mean hepatocyte diameter decreased to (5.2± 1.5 μm) after administration of SBT seed oil. Conclusion: Administration of SBT( 2.6mg/kg and 5.2mg/kg) exerts a protective effect against damaging effects of ACM ( 750 mg/kg) on hepatocytes in albino rats. This hepatoprotective role becomes greater with increasing dose of SBT.

Objective: To observe the nephroprotective role of berberis vulgar is on renal parenchyma against microscopic and morphometric changes induced by Gentamicin in albino rats. Study Design: Lab based experimental study. Place and Duration of Study: It was conducted in Baqai Medical University in collaboration with department of Anatomy Liaquat National Hospital and Medical College, from Jan to Jul 2017. Methodology: A total of 40 male adult albino rats were used in the study. Four groups were made. Each group contained 10 rats. Group A was a control group, group B received only berberis vulgaris fruit extract orally per day for 21 days, group C received Gentamicin 100 mg/kg/day intraperitonially daily for 21 days. Group D received Gentamicin 100 mg/kg/day intraperitonially along with berberis vulgaris fruit extract 100 mg/kg/day orally. Both kidneys were removed. H&E and PAS stains were used for observing histological alterations and protective role of berberis vulgaris fruit extract. Results: Glomerulus and proximal convoluted tubules were observed histologically in all 4 groups. Microscopy of group B showed parameters nearly similar to control group. Microscopy of group C showed significant derangement in all parameters when compared with control group. Group C showed decrease glomerular, proximal convoluted tubular count was noted. Glomerular diameter increases and there was glomerular hypertrophy and tubular necrosis. Microscopy of group D showed significant improvement due to berberis vulgaris which restored normal renal architecture. Conclusion: Berberis vulgaris has a nephroprotective effect and it can be used as a new medicine against nephrotoxic drugs like Gentamicin.

Objectives: To observe the effect of lagenaria siceraria on inflammation and fibrosis brought about by arsenic in liver of Sprague Dawley rat. Study Design: Laboratory based randomized control trial. Place and Duration of Study: This experiment was performed at Department of Anatomy, Army Medical College Rawalpindi in co-operation with National Institute of Health (NIH) Islamabad for eight weeks (1st March 2017 to 25th April 2017). Material and Methods: Fifty Sprague Dawley rats (both male and females housed separately) were carefully chosen and distributed randomly into five groups, each consisting of 10 animals. A and B were the control groups whereas C, D and E served as experimental groups. During the first four weeks, experiment groups C, D and E were given a dosage of 5milligram/kilogram body weight of sodium arsenite. At the end of four weeks, animals from control group A and experimental group C were dissected and liver samples were processed for microscopic studies. In the next 4 weeks, group D animals were set aside without any further intervention. At that time, sodium arsenite at a dose of 5 milligram/kilogram body weight and lagenaria siceraria at a dose of 100 milligram/kilogram bodyweight were administered to group E animals. Group B animals served as control for experimental groups D and E. At the end of these 4 weeks animals of groups B, D and E were dissected. Liver was processed, fixed and stained for microscopic study. Area of portal triad as well as liver lobules were studied for inflammation and fibrosis and results were analysed. Statistical tool used to analyse the data was SPSS v 22. Results were considered to be significant when p-value is ≤ 0.05. Results: Experimental group C developed moderate grade fibrosis and inflammation (grade 2 to 3) as compared to group A. Degree of inflammation and fibrosis was mild to moderate (grade 1 to 2) in group D. There was no inflammation and fibrosis (Grade 0 to 1) in group E. Group B served as a control for group D and E. Conclusion: Inflammation and fibrosis developed in the liver of adult rats when they were subjected to sodium arsenite even for a brief calculated period. Simultaneous administration of lagenaria siceraria can shield and diminish the toxic effects of arsenic. Oxidative potential and immunomodulatory properties and presence of flavonoid like substances renders lagenaria siceraria to act as ameliorative against this fibrosis and inflammation in liver lobules and surrounding area of portal triads.

Background: Kidney is an important excretory organ. Its damage can be occurred due to prolonged use and higher doses of drugs, exposure to some chemicals, toxins, or infectious agents. Herbal plants as Ashwagandha (Withania somnifera) may have free radical scavenging activity thereby can be used for the prevention and treatment of kidney damage. Objective: To observe the nephroprotective effect of Ashwagandha (Withania somnifera) root against gentamicin induced nephrotoxicity in Wistar albino rats. Methods: This experimental study was carried out in the Department of Physiology, Sir Salimullah Medical College (SSMC), Dhaka from 1st July 2010 to 30th June 2011. A total number of 35 Wistar albino rats, age ranged from 90 to 120 days, weighing between 150 to 200 grams were included in this study. After acclimatization for 14 days, they were divided into control group (Group A) and experimental group (Group B). Control group was again subdivided into group A1 (baseline control, consisted of 10 rats) and group A2 (gentamicin treated control group, consisted of 10 rats). Again, experimental group (Group B- Ashwagandha pretreated and gentamicin treated group) consisted of 15 rats. All groups of animals received basal diet for 22 consecutive days. In addition to this, group A2 also received gentamicin subcutaneously (100mg /kg body weight/day) for the last eight (15th to 22nd day) consecutive days. Again, group B received ashwagandha root extract (500mg/kg body weight/ day; orally) for 22 consecutive days and gentamicin subcutaneously (100mg/kg body weight /day) for last eight (15th to 22nd day) days. All the animals were sacrificed on 23rd day. Then blood and kidney sample were collected. Estimation of serum urea, creatinine levels were done by using standard Laboratory kits. The statistical analysis was done by one way ANOVA and Bonferroni test as applicable. Results: The mean serum urea, creatinine levels were significantly (p<0.001) higher in gentamicin treated control group in comparison to those of baseline control. Again, these levels were significantly (p<0.01) lower in ashwagandha pretreated and gentamicin treated group (experimental group) when compared to those of gentamicin treated group (control). Conclusion: Ashwagandha (Withania somnifera) root may have some nephroprotective effect against gentamicin induced nephrotoxicity. This study was designed to investigate the allethrin induced toxico-pathological changes in adult male albino rats. A total of 60 adult male albino rats were divided randomly into 4 equal groups. Group A was treated as control while group B was given Allethrin @ 0.5g/kg, group C @ 1.0g/kg and group D @1.5g/kg body weight respectively, orally once at day 14 of experiment. Dullness, erected hairs, alopecia, less response towards feed and water, watery droppings, tremors, convulsions and coma were more prominent signs in treated groups. Feed intake and body weight was decreased in all treated groups. Acute mortality was seen in group C and D. The total erythrocyte and leukocyte count, hemoglobin concentration and packed cell volume (P≤0.05) was deviated from normal in all the groups treated with allethrin. Histopathologically, kidneys exhibited condensed nuclei, necrotic tubules and congested renal parenchyma. In liver, vacuolar degeneration in nucleus and cytoplasm was observed. Micro nucleated lymphocytes were also evident in group D treated with highest dose of allethrin. In conclusion, allethrin induced dose and time-dependent toxico-pathological effects in adult male albino rats were observed and presented.

Background: For a long time, Diabetes mellitus has been treated with medicines derived from plants. Objective: To evaluate the effect of Ginger aqueous extract on Glomerular mesangial matrix in Alloxan induced diabetic nephropathy of albino rats. Materials and Methods: In this study we induced diabetes mellitus with Alloxan intraperitoneally (150 mg/kg body weight) in Experimental groups B & C. Then the rats of Experimental group C received 200mg/kg body weight of ginger aqueous extract by gavage daily for five weeks starting from 8th day after Alloxan injection. Results: We observed that on histopathological examination, Experimental group B kidneys revealed highly increased mesangial matrix while the animals of experimental group C treated with ginger aqueous extract showed less increase in mesangial matrix as compared to experimental group B but it was more than control group A. Three groups had significant difference among them having p-values <0.001. Conclusion: The results of the present study indicated that the co-treatment of Ginger aqueous extract prevented alloxan induced diabetic nephropathy in albino rats. The aqueous extract of Ginger showed amazing results regarding renal histopathology of diabetic rats. The overall nephroprotective effect of Ginger is probably due to a counteraction of free radicals by its antioxidant components

Objective: The main purpose of this study is to evaluate the hepatoprotective effects of propolis in hepatocytes injury caused by ATT due to isoniazid and rifampicin. Methods: Healthy albino rats of with average weight of 200-250g were under this study. These rats dividing into main four groups, A group is taken a control group and then further into the group B, group C, and group D as group for experiments. The control group had 15 rats with measured weight, they were given distilled water. Group B had 15 rats, they were given with standard dose of rifampicin and isoniazid. Group c had 15 rats, they were also given with standard dose of rifampicin and isoniazid. Group D had 15 rats, they were given with standard dose of rifampicin and isoniazid and also extract of the propolis we prepared. Results: Serum ALT in the experimental group B with group C, group D were also found to be of statistically significant with p-value < 0.001. ALT serum level observed high in group B. Multiple comparison between groups revealed that group B with a significantly increase in the serum enzyme AST level in comparison to group A, group C and group D with p-value <0.001. Conclusion: This study showed that ethanolic extract of propolis prevents isoniazid and rifampicin induced hepatotoxicity in the albino rats. Background: The extract of Corchorus olitorius has a reasonable content of vitamins A and C, calcium, iron and fibre, and therefore enjoys a universal application in the treatment of some disease conditions, even as the whole leaf is a very important component of food in so many cultures. Purpose: The study was designed to investigate the effect of Corchorus olitorius extracts on the testis of adult Wistar rats. Method: A total number of thirty two adult Wistar rats weighing between 180 and 200 grammes separated into four groups labeled A,B,C and D respectively were used for the study. Animals in group A which served as the control group were fed with the normal rat chow and water only. The group B rats were administered 100mg/kg body weight of aqueous extract of Corchorus olitorus; while those in group C were administered 500mg/kg body weight of the extract. The group D rats received 1000mg/kg body weight of the extract. For a period of four weeks, the different experimental animal groups received the respective aforementioned treatments once daily, around nine- o’clock in the morning through oral intubation. At the end of the 28 day treatment, the animals were sacrificed and the testes harvested for histological, investigation, and through cardiac puncture, blood samples for some hormonal studies was also collected and investigated using standard laboratory standards. Results: Observations made showed that the extract produced no histological distortions, degenerative or defective effects on the testicular tissues.
