Abstract Aim The aims of the study are to investigate the bactericidal effect of three antiseptics (chlorhexidine solution, povidone-iodine solution, and Walkhoff solution) and to determine the minimum inhibitory concentration and their effect on different microbial species. Methods and Materials The study was performed on microflora derived from root canals with simple and complicated pulp gangrene and on pure strains of Enterococcus and Candida albicans. Results Chlorhexidine and povidone-iodine proved to have antibacterial and antifungal effects if used in the treatment of pulp gangrene and in cases not responding to conventional therapy. Conclusion According to the obtained results, the spectrum of antibacterial agents used in infected canal irrigation can be enlarged to include the agents tested. Citation Shurrab MY. Abstract Background This study aimed to compare the use of photon-initiated photoacoustic streaming (PIPS) and conventional needle irrigation (CNI) in conjunction with different concentrations of sodium hypochlorite (NaOCl) to remove Enterococcus faecalis (E. faecalis) suspended bacteria and biofilms from root canal systems with different diameters or tapers. Methods Artificial root canal samples (n = 480) were randomly divided into three groups (n = 160/group). The canals were prepared to fit file sizes #10/.02, #25/.02, or #25/.06. The size #10/.02 group was incubated for seven days. The size #25/.02 or #25/.06 group was incubated for 2 days. A stable biological model of E. faecalis infection was established. The root canals were washed with distilled water or with 1%, 2%, or 5.25% NaOCl combined with CNI or PIPS. Bacterial suspensions and biofilms were assessed using an ATP assay kit and fluorescence microscopy. Image-Pro Plus was used to analyse the average fluorescence intensity to determine the most suitable root canal irrigation solution. Results In the CNI and PIPS groups, the ATP value of the 5.25% NaOCl subgroup was the lowest, followed by that of the 2% and 1% NaOCl subgroups. The ATP value of the distilled water subgroup was the highest (P < 0.05). When the root canal taper was 0.02, the ATP value of the #10/.02 + PIPS group was significantly lower than that of the #25/.02 + CNI group (P < 0.05). The average fluorescence intensity of the #10/.02 + PIPS group was lower than that of the #25/.02 + CNI group (P < 0.05). When the apical diameter was #25, the ATP value of the 0.02 taper in the PIPS group was lower than that of the 0.06 taper in the CNI group (P < 0.05), and the average fluorescence intensity of the 0.02 taper + PIPS group was lower than that of the 0.06 taper + CNI group (P < 0.05). PIPS combined with 2% and 5.25% NaOCl effectively improved the long-term antibacterial effect after irrigation and re-culture for 6 h. Conclusions Compared with CNI, PIPS has greater ability to remove bacteria in root canals with a small preparation diameter and a small taper. PIPS with 2% and 5.25% NaOCl exhibited superior antibacterial and bacteriostatic effects.

Canarium odontophyllum(CO) Miq. has been considered as one of the most sought-after plant species in Sarawak, Malaysia, due to its nutritional and pharmacological benefits. This study aimed to evaluate the pharmacodynamic interaction of crude methanol and acetone extracts from CO leaves in combination with oxacillin, vancomycin, and linezolid, respectively, against MRSA ATCC 33591 as preliminary study has reported its potential antistaphylococcal activity. The broth microdilution assay revealed that both methanol and acetone extracts were bactericidal with Minimum Inhibitory Concentration (MIC) of 312.5 μg/mL and 156.25 μg/mL and Minimum Bactericidal Concentration (MBC) of 625 μg/mL and 312.5 μg/mL, respectively. Fractional Inhibitory Concentration (FIC) indices were obtained via the chequerboard dilution assay where methanol extract-oxacillin, acetone extract-oxacillin, methanol extract-linezolid, and acetone extract-linezolid combinations exhibited synergism (FIC index ≤ 0.5). The synergistic action of the methanol extract-oxacillin combination was verified by time-kill analysis where bactericidal effect was observed at concentration of 1/8 × MIC of both compounds at 9.6 h compared to oxacillin alone. As such, these findings postulated that both extracts exert their anti-MRSA mechanism of action similar to that of vancomycin and provide evidence that the leaves ofC. odontophyllumhave the potential to be developed into antistaphylococcal agents.

The possible cochlear toxicity of topically applied povidone-iodine solution was investigated in guinea pigs by measuring transiently evoked otoacoustic emissions (TEOAEs). Seven adult pigmented guinea pigs (14 ears) with a normal acoustic pinna reflex were used. After the baseline TEOAE measurements, 0.1mL of povidone-iodine solution was applied transtympanically into the middle ear in the study group. Saline solution and gentamicin were used as negative and positive control, respectively. TEOAE measurements were repeated on the 10th day, and four weeks after the first application. Tympanometric measurements were also carried out for both ears of all animals at the end of the study. Responses in all ears, which had povidone-iodine applied to them, disappeared in a way similar to those recorded in gentamicin-treated ears. Tympanometric measurements at the end of the study showed ’type A’ tympanograms in all ears of all animals. The present study showed that povidone-iodine could have a cochleotoxic effect in guinea pigs. A study in humans with TEOAE before and after middle-ear surgery, where povidone-iodine can reach into the middle-ear cavity, may be useful for evaluating this result for clinical practice.

Background: Despite best medical and surgical practice, some cases of chronic rhinosinusitis (CRS) can remain recalcitrant. Bacterial biofilms have been associated with the recalcitrance of sinonasal inflammation. Biofilms are highly resistant to commonly prescribed antibiotics. Accordingly, more effective antimicrobial treatment options are needed to treat refractory CRS. The aim of this study was to determine the in vitro efficacy of neutral electrolysed water (NEW) and povidone-iodine (PVI) against CRS-associated Staphylococcus aureus biofilms. Methods: Mature S. aureus biofilms were grown in a Centre for Disease Control (CDC) biofilm reactor. The antimicrobial activity of NEW, PVI and doxycycline was determined for both planktonic and biofilm cultures of a clinical S. aureus isolate using minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and minimum biofilm eradication concentration (MBEC) assays. Results: MICs and MBCs were determined for all antimicrobials. MBC values were similar to MICs for both antiseptics, but doxycycline MBCs were significantly higher than the associated MICs. Biofilms were highly resistant to NEW and doxycycline. The MBEC for doxycycline was between 500 and 1000 µg/mL. NEW was ineffective against biofilms and no MBEC could be determined. In contrast, a concentration of 10% of the commercial PVI solution (10 mg/mL PVI) led to effective eradication of mature biofilms. Conclusion: In this study, only PVI showed promising antibiofilm activity at physiological concentrations. The in vivo efficacy of PVI warrants further investigation of its potential as a treatment for recalcitrant CRS.

In this work, six complexes (2–7) of Cr(III) and Co(II) transition metals with triazole ligands were synthesized and characterized. In addition, a new ligand, 3,5-bis(1,2,4-triazol-1-ylmethyl)toluene (1), was synthesized and full characterized. The complexes were obtained as air-stable solids and characterized by melting point, electrical conductivity, thermogravimetric analysis, and Raman, infrared and ultraviolet/visible spectroscopy. The analyses and spectral data showed that complexes 3–7 had 1:1 (M:L) stoichiometries and octahedral geometries, while 2 had a 1:2 (M:L) ratio, which was supported by DFT calculations. The complexes and their respective ligands were evaluated against bacterial and fungal strains with clinical relevance. All the complexes showed higher antibacterial and antifungal activities than the free ligands. The complexes were more active against fungi than against bacteria. The activities of the chromium complexes against Candida tropicalis are of great interest, as they showed minimum inhibitory concentration 50 (MIC50) values between 7.8 and 15.6 μg mL−1. Complexes 5 and 6 showed little effect on Vero cells, indicating that they are not cytotoxic. These results can provide an important platform for the design of new compounds with antibacterial and antifungal activities.

We present a case of a 15-month-old boy with Cornelia de Lange Syndrome (NIPBL gene mutation). On a PSG, central sleep apnea (central apnea-hypopnea index of 19/hour) and nocturnal hypoventilation (transcutaneous CO2> 50 mmHg for 53% of the night) were found. A positive pressure initiation study was aborted because the patient developed a serious adverse reaction. The differential diagnosis included a skin fragility condition versus an allergic contact dermatitis to the interface; this could be from the povidone-iodine solution used to clean the NiPPV interface or from the plastic of the interface itself. A skin biopsy was performed which was normal. The reaction was likely secondary to an allergic contact dermatitis from the povidone-iodine solution used to clean the NiPPV interface. The patient is currently tolerating NiPPV.

Abstrak Candida albicansb (C. albicans) adalah salah satu mikroorganisme penyebab masalah kesehatan reproduksi wanita, yaitu keputihan (fluor albus). Penggunaan larutan povidone iodine dan bahan alam seperti ekstrak daun sirih menjadi pilihan masyarakat sebagai pembersih alat kewanitaan. Tujuan penelitian ini adalah untuk membandingkandaya hambat larutan antiseptik povidone iodine dan ekstrak daun sirih terhadap jamur C. albicans secara in vitro. Penelitian dilakukan terhadap lima isolat jamur C. albicans dengan larutan kontrol akuades.Perlakuan terdiri dari povidone iodine, ekstrak daun sirih dengan konsentrasi 5%, 10%, dan 20%.Hasil penelitian menunjukkan bahwa povidone iodine memiliki daya hambat terhadap C. albicans. Ekstrak daun sirih dengan konsentrasi 5% dan 10% tidak memiliki daya hambat terhadap C. albicans, namun ekstrak daun sirih konsentrasi 20% memiliki daya hambat terhadap C. albicans. Analisis statistik dengan uji ANOVA yang dilanjutkan dengan uji Post-hoc menunjukkan perbedaan bermakna antara daya hambat larutan povidone iodine dan ekstrak daun sirih 20% terhadap kontrol(p < 0.05).Larutan povidone iodine memiliki daya hambat dua kali lebih besar terhadap pertumbuhan C. albicans dibandingkan ekstrak daun sirih 20%. Dari penelitian ini dapat disimpulkan bahwa larutan povidone iodine dan ekstrak daun sirih 20% dapat menghambat pertumbuhan jamur C. albicans secara in vitro. The antiseptic solution, povidone iodine, is still an option to overcome vaginal discharge. The use of natural materials such as betel (Piper betle L.) leaves extract also become a popular choice as adouche for women. The objective of this study was to compare the inhibitory activity of povidone iodine solution and betel leaf extract against the growth of C. albicans in vitro. We used five different isolates of C. albicans with distilled water as control. Each isolate was treated with povidone iodine solution, betel leaf extract at concentration of 5%, 10%,and 20%. The results showed that povidone iodine had inhibitory effect on C. albicans. Betel leaf extract at concentration of 5% and 10% did not have inhibitory effectwhile betel leaf extract at concentration of 20% hadinhibitory effect on C. albicans. Analysis by ANOVA followed by Post-hoc tests showed a significant difference in inhibitory activity of povidone iodine and betel leaf extract at 20% concentration compared to control (p < 0,05). Povidone iodine solution showed twice as much as inhibitory effect on C. albicans compared to betel leaf extract (20% concentration). It is concluded that povidone iodine solution and betel leaf extract at 20% concentration can inhibit the growth of C. albicans in vitro.Keywords: povidone iodine, betel leaf extract, Candida albicans
