Garcinia kola stem bark forms part of recipes used traditionally for the treatment of malaria. In view of the prevalence of malaria in Nigeria, this study investigated the phytochemical, mineral and proximate components, as well as antiplasmodial activity and toxicological effect of Garcinia kola stem bark extract against Plasmodium berghei infected mice. The plant sample was screened for phytochemical, mineral and proximate components using standard laboratory techniques. Thirty five mice were divided into seven groups of five mice each. Malaria was induced in all the groups intraperitoneally with 0.2 mL of infected blood containing about 107 of P. berghei parasitized red blood cells, except group 6 (extract only) and group 7 (normal control). Group 1 received 100 mg/kg bodyweight of the extract orally. Group 2 received 200 mg/kg of the extract. Group 3 received 300 mg/kg of the extract. Group 4 received 5 mg/kg of chloroquine. Group 5 (induced but untreated control). The haematology, liver function enzymes and histopathology of the liver were carried out using standard protocols.  The plant was rich in alkaloids, iron and fibre. The extract treated groups (1-3) showed significant decrease (p≤0.05) in parasitemia level after seven days of treatment. There was no significant difference in AST, ALT, ALP, bilirubin and GGT activities in all the extract treated groups compared to the control. No pathological changes were evident in histopathology of all the groups treated with various concentration of the extract. The result obtained from this study confirmed the antiplasmodial activity of methanol extract of G. kola stem bark. The highest inhibition of P. berghie parasite was observed at dose 300 mg/kg comparable to chloroquine, with no hepatoxicity which confirmed the safety of G. kola. The phytochemicals and nutritional components could be responsible for the observed antiplasmodial activity of the plant.

Purpose: In this study we examined the ability of selenium and vitamin E to prevent sepsis-induced changes in lung tissue. Methods: Fifty rats were divided into five groups: Group 1: Control group; Group 2: Sepsis group. In this group only cecal ligation and perforation (CLP) was performed. Group 3: Selenium group. An intraperitoneal dose of 100 µg selenium was given for the first two days followed by a daily dose of 40 µg for the next five days. CLP was performed the following day. Group 4: Selenium and vitamin E group. In addition to selenium, vitamin E was given intramuscularly in a dose of 250 mg/kg/day for seven days. CLP was performed the following day. Group 5: Vitamin E group. Vitamin E was given intramuscularly in a dose of 250 mg/kg/day for seven days. CLP was performed the following day. Results: There were significant differences between Group 2 and all other groups in terms of blood gas values (pH, pCO2, SaO2), and leukocyte, C-reactive protein (CRP) and glutathione peroxidase levels (p < 0.005). There was no statistically significant difference between groups 3, 4 and 5 in terms of histopathological changes in lung tissue (p > 0.05), but all groups were significantly different compared with Group 2 (p < 0.05). Conclusion: Sepsis-induced lung tissue damage can be reduced or prevented by pre-treatment with of selenium and/or vitamin E in a rat model.

PURPOSE: To investigate the effect of enemas containing probiotics and budesonide on the colonic mucosa in experimental colitis. METHODS: Fifty male Wistar rats with experimental colitis induced by 10% acetic acid enema were randomized to five groups (10 rats each) according to the treatment: group 1 - saline solution, group 2 - budesonide (0.75 mg/kg/day), group 3 - probiotics (1mg/day), group 4 - probiotics plus budesonide, and group 5 - control, with not-treated rats. The following variables were studied: body weight, macroscopic and microscopic score of the colonic mucosa, and DNA content of the mucosa. RESULTS: All animals lost weight between the beginning and the end of the experiment (280+ 16 mg versus 249+21 mg, p< 0.001). There was no significant difference among the groups in relation to both the macroscopic and histological score. The budesonide + probiotic group showed higher DNA content than control group (1.24+ 0.15 versus 0.92+ 0.30 mg/100mg of tissue, p=0.01). CONCLUSION: Budesonide in addition to probiotics enhance the mucosal trophism in experimental colitis.

ABSTRACT Objective: The aim of this study was to compare the incidence of root cracks after root canal instrumentation with thermomechanically processed nickel-titanium (Ni-Ti) files with different instrumentation kinematics. Materials and Methods: A total of 150 extracted mandibular premolars with mature apices and straight root canals were divided into five groups and used in this study. In Group 1, 30 teeth were prepared using hand K-files and assigned to control group, Group 2 was instrumented using K3XF Rotary files (SybronEndo, Glendora, CA, USA) with continuous rotary motion. The teeth in Group 3 were instrumented by ProTaper Next (Dentsply Maillefer, Ballaigues, Switzerland) rotary files which make asymmetric rotary motion, In Group 4, teeth were instrumented by RECIPROC (VDW, Munich, Germany) with reciprocation motion and in Group 5, teeth were instrumented by Twisted File (TF) Adaptive (SybronEndo, Orange, CA, USA) files that use combination of continuous rotation and reciprocation motion (n = 30/per group). All the roots were horizontally sectioned 3, 6, and 9 mm from the apex with a low speed saw under water cooling. Then, the slices were examined through a stereomicroscope to determine the presence of dentinal microcracks.Results: For the apical (3-mm) and coronal (9-mm) sections, the ProTaper Next and TF Adaptive produced significantly more cracks than the hand files, RECIPROC, and K3XF (P < 0.05). There was no significant difference between the experimental groups and control group at the 6-mm level (P > 0.05). Conclusions: Within the limitations of this in vitro study, all thermal-treated Ni-Ti instruments and hand files caused microcracks in root canal dentin.

Introduction: Khaya senegalensis is a genus of seven species of trees in the mahogany family Meliaceae, native to tropical Africa and Madagascar. Mahogany  in  English, Aganwo  in  Yoruba,  Madachi  in  Hausa  and  Ono  in  Igbo. All species become big trees 30–35m tall, rarely 45m, with a trunk over 1 m trunk diameter, often buttressed at the base. The leaves are pinnate, with 4-6 pairs of leaflets, the terminal leaflet absent; each leaflet is 10–15 cm long abruptly rounded toward the apex but often with an acuminate tip. Aim: The  aim of  the study  was  to determine  the Toxic  effect of prolonged oral administration  of  the  aqueous stem bark of  Khaya senegalensis extract  on the histology of  Kidneys and its biochemical parameters in wistar rats. Methods: This work is an experimental research. A total of 20 wistar rats were randomly divided in to 5 groups each of which consist of 4 rats. Group 1 received distilled water to serve as control while group 2, 3, 4, and 5 received 500 mg/kg bw, 1000 mg/kg bw 2000 mg/kg bw  and 4000 mg/kg bw of the aqueous extract respectively for 60 days after which they were sacrificed, processed in Automatic Tissue Processor machine, Sectioned and stained with H &E. Results: There was statistical significant increase in urea and potassium in all the test groups but is not dose dependent. The creatinine was significantly increased in groups 2, 4 and 5. While other parameters such as sodium, chloride and bicarbonate no significant difference when compared to the control group. The kidney sections showed normal structure in group 1 when compared with the test groups. However, there was significant infiltration of inflammatory cell across all the groups which were suggestive of kidney damage or injury. Similarly phenomenon was noticed in group 5 with additional congestion in the glomerulus and more polymorphs seen. Conclusion: The LD50 was found to be greater than 5000 mg/kg bw, therefore, 400 mg/kgbw was used as higher dose in the experimental wistar rats. There were statistical significant increases in some parameters groups while some groups not significant. The kidney section showed significant infiltration of polymorphs across all the groups more marked in group 5 with distension and damaging of the glomerulus indicating renal injury.

Abstract Objective:The aim of this study is to evaluate the effect of iohexol as a contrast agent on the anticoagulant activity of oral factor Xa inhibitors. Methods:The study included 65 people who underwent contrast computerized tomography (CT). Patients in group 1 were using rivaroxaban (20 patients), patients in group 2 were using apixaban (20 patients), patients in group 3 were using edoxaban (20 patients), and group 4 was the control group (5 volunteers). Iohexol (60ml) was used as a contrast agent. Two tubes were used to collect 2 ml of blood from the patients at 4 hours after the drug dose (rivaroxaban, apixaban, or edoxaban) and 1 hour after the contrast CT (CT was performed 3 hours after the drug was taken). In the control group, at any time and 1 hour after contrast CT, 2 tubes of 2 ml of blood were collected. The anticoagulant properties of rivaroxaban, apixaban, and edoxaban were evaluated using anti-factor Xa levels. Results:The anti-factor Xa level was increased after using the contrast agent in the rivaroxaban group (0.66±0.32U/ml vs. 0.67±0.32U/ml; p=0.01) and the edoxaban group (0.74±0.35 U/ml vs. 0.76±0.36 U/ml; p=0.006). However, there was no significant difference in the apixaban group (0.66±0.33U/ml vs. 0.66±0.32U/ml; p=0.21) and control group (0.02±0.01U/ml vs. 0.03±0.01U/ml; p=0.33).Conclusion:The anticoagulant properties of rivaroxaban and edoxaban tended to increase significantly, but there was no statistically significant difference in the anticoagulant properties of apixaban with contrast agent. The increasing is too small so that these laboratory results need to validate with larger clinical trials(NCT04611386).

Diabetes is growing public health. The research investigated the modulatory roles of the aqueous stem-bark extract of Moringa oleifera on glucose utilization. A modified Oral Glucose Tolerance Test (OGTT) was used in studying the effect of the extract on glucose absorption, on four groups of six rats and standard methods were used to test the effect of the extract on enzyme activities (hexokinase and glucose 6 phosphatase) on three groups of six rats. In OGTT, rats in group-1(diabetic control) and group-4 (normal control) were administered with the vehicle only. The other groups were administered different concentrations of the extract in the vehicle (group-2 and 3 were 200mg/kg body weight and 300mg/kg respectively). In the enzymes activities, 200mg/kg body weight of the extract was administered to diabetic treated group whereas normal untreated and diabetic untreated received 5ml of water only. Glucose concentrations of OGTT showed increased concentration in the first 30 minutes after administration of the extract and steady time-dependent decreased concentration through 120 minutes. Group-3, showed a significant difference in each 30 minutes interval, compared to the 120 minutes (p<0.05). Each interval is significantly different from the preceded 30 minutes interval (p<0.005). Group-2 was significantly different in the first and second 30-minutes intervals, compared to the preceded interval (p<0.005). The first 30 minute interval was significantly different from the baseline and 120 minutes (p<0.05). In enzyme activities, the diabetic treated and normal untreated were significantly different from the diabetic untreated (p<0.05). The extract improved glucose utilization.

Background: Malaria is a parasitic infection that is common in the tropical and sub-tropical regions of the world. It is estimated to kill one million individuals annually. Detarium microcarpum (Fabaceae family) is used traditionally for the treatment of malaria, diabetes, hypertension, convulsions, pneumonia, leprosy, and meningitis. The purpose of this research was to evaluate the antiplasmodial potential and safety profile of the methanol extract of D. microcarpum stem bark. Methods: The extract was subjected to phytochemical screening and oral median lethal dose (LD50) estimation. Using curative, suppressive, and prophylactic experimental animal models, antiplasmodial activity was assessed in mice infected with chloroquine-sensitive Plasmodium berghei berghei. Biochemical and hematological tests were conducted in rats orally administered with the extract for 28 days. One-way analysis of variance (ANOVA) was used to analyze the data, followed by Dunnett's post hoc test. Results: Alkaloids, flavonoids, saponins, tannins, triterpenes, and glycosides were found in the extract through phytochemical analysis. When compared to the negative control group, the extract at the tested doses (200, 400, and 800 mg/kg) showed a significant (P < 0.001) curative, suppressive, and prophylactic effect and significantly (P < 0.001) prolonged the survival time of mice in the treated groups. The extract had an insignificant (P > 0.05) effect on the biochemical and hematological parameters tested. Conclusions: The results suggest that the stem bark extract of D. microcarpum at the doses tested possesses antiplasmodial activity and is relatively safe after short-term use.

Aim: The aim is to evaluate the antioxidant potentials of Eucalyptus camaldulensis methanol stem bark extract (ECMSBE) on Wister albino rats. Methodology: The phytochemical screening of ECMSBE was conducted using standard methods. A total of 36 albino rats were used for the antioxidant studies. The rats were divided into six (6) groups of six rats. Control group received distilled water orally at 2ml/kg. Oxidative stress was induced in groups 2 to group 6 CCl4 (1ml/kg, s.c) at every 72hrs for 10 days. Group 2 was untreated while groups 3 – 6 received doses of 50, 100, 150 and 200mg/kg of ECMSBE respectively. On the 11th day, the rats were sacrificed and the liver was removed and homogenised and oxidative stress parameters were determined. Results: Phytochemical analysis of ECMSBE revealed the presence of saponins, flavonoids, tannins, phenols, glycosides, steroids, terpenoids and resins. There was no significant difference (P<.05) between the CCl4 induced group and the group treated with ECMSBE (50mg/kg). However, their concentrations were significantly different from the group treated with ECMSBE (100mg/kg – 200mg/kg) when compared to the group treated with CCl4 Only. The CCl4-induced group had its vitamin A, vitamin C and vitamin E concentrations significant different (P<.05) from the groups treated with ECMSBE (100, 150 and 200mg/kg body weight). There was no significant difference (P>.05) in the levels of SOD, CAT and GPx between the group induced with CCl4 only and the group treated with ECMSBE (50mg/kg), however, it these concentrations were significantly higher (P<.05). The enzymatic antioxidants concentration in the normal control group was not significantly different (P>.05) when compared the group that was treated with ECMSBE (200mg/kg) Conclusion: The result suggest that the extract of E. camaldulensis possessed antioxidant properties which can be used as effective protecting agents against oxidative stress and other diseases.

Aim: This study aimed to investigate the antiplasmodial activity and effect of stem bark of Terminalia avicennioides made as dietary feed fed to mice infected with Plasmodium berghei, on some serum biochemistry. Methodology: Twenty (20) mice were divided into four groups. Group 1 was not infected with Plasmodium berghei (normal control), Group 2 was infected with P. berghei but not treated (negative control). Group 3 was infected and treated with 5.0 mg/kg of Arthemeter-Lumefantrine (positive control). Groups 4 was infected and fed with treated feed (T. avicennioides). Treatments were carried out for five days. Blood was taken daily from the tail of the mice before treatment for the assessment of parasitaemia. The animals were sacrificed on the fifth day and the whole blood was collected into EDTA bottle. Serum obtained was used to assay for biochemical parameters. Results: Parasitaemia count was significantly lower (p<0.05) in all the treated groups when compared with the negative control group. The high-density lipoprotein was significantly higher (P<0.05) in the normal control (123.14±3.19) when compared with the positive control (99.18±2.76), negative control (85.29±0.85) and the group treated with T avicennioides (86.14±3.21). The serum alanine aminotransferase, alkaline phosphatase and aspartate aminotransferase level in the group treated with T. avicennioides (167.90±4.13, 15.87±1.32 and 17.50±1.95) respectively were significantly reduced (p<0.05) when compared with the negative control (197.25±5.44, 20.01±1.32 and 26.71±0.45) respectively. The mean bilirubin and albumin level in the negative control showed no significant difference when compared with the group fed with T. avicennioides. Conclusion: The study concluded that T. avicennioides have antiplasmodial activity with a mild adverse effect on liver function.

An interesting feature of ionizing radiation, especially Gamma and X-rays as a DNA damaging factor is the range of lesions it induces. γ-H2AX foci are documented to represent DNA double-strand breaks (DSBs) as a biomarker for radiation-induced damage. Study design 42 adult male mice Albino BALB/c, had been divided randomly into 6 groups of seven mice each. Group 1 received a standard saline solution untreated also, do not expose to radiation. Group 2 mice received vitamin C (VC) (200 mg/kg.day) intra-peritoneal (i.p.) injected for 8 days without radiation. Group 3 control was exposed to γ-radiation. Group 4 control was exposed to X-ray radiation. Group 5 mice had been administrated with vitamin C in the identical dose of group 2 for 8 days, then exposed to (4 Gy) of γ-ray. Group 6 was administrated with vitamin dose in the same above and the same period, then exposed to (4 Gy) of X-ray. All groups had been sacrificed by cervical dislocation at (1, 3, and 24 h). Post radiation testis mice tissues were collected. A significant difference (P 0.05) between the group of vitamin C and with a control group exposed to both (γ, X-rays) in foci forming, but there is no significant difference (P 0.05) between γ and X- rays for the control and vitamin C groups. The results demonstrate that vitamin C is a good radioprotective agent for testis mice tissues; the effect of (γ and X-rays) had almost the same results on the mice testicle tissues with the same dose.
