Cascabela thevetia (Thevetia peruviana) giant milkweed, is known for its pharmacological importance for centuries. The chemical constituents of Thevetia peruviana leaves are amyrin, amyrin acetate, ?-sitosterol, urosolic acid, cardenolides, calotropin. The anti-arthritic activity of Thevetia peruviana leaves in comparison with Diclofenac was evaluated through in-vitro studies. In-vitro studies revealed anti-arthritic effects (p< 0.05) of crude extracts of aqueous, methanolic, ethanolic and chloroform of Thevetia peruviana leaves on bovine serum albumin as evident for % inhibition protein denaturation. Different extracts of Thevetia peruviana leaves were found to possess in-vitro antiarthritic activity against BSA, using diclofenac as the reference standard which are Dose-dependent (100µg/ml, 250µg/ml, 500µg/ml). Chloroform extract has shown better activity comparing the other extracts.

SummaryThe 3-morpholinosydnonimine (SIN-1) generates both nitric oxide (NO) and superoxide anion (O2−). It elicits dose-dependent vasodilation in vivo, in spite of the opposite effects of its breakdown products on vascular tone and platelet aggregation.This study was designed to investigate the influence of intravenous SIN-1 injection on platelet Ca2+ handling in patients undergoing coronary angiography. SIN-1 administration reduced cytosolic [Ca2+] in unstimulated platelets by decreasing Ca2+ influx. It attenuated Ca2+ mobilization from internal stores evoked by thrombin or thapsigargin. In vitro studies were used as an approach to investigate how simultaneous productions of NO and O2− from SIN-1 modify thrombin- or thapsigargin-induced platelet Ca2+ mobilization. Superoxide dismutase, the O2− scavenger, enhanced the capacity of SIN-1 to inhibit Ca2+ mobilization but catalase had no effect.This suggests that the effects of SIN-1 on platelet Ca2+ handling resemble those of NO, but are modulated by simultaneous O2− release, independently of H2O2 formation.

SummaryThis study examined the effects of meiosis inhibition during bovine oocyte transportation on developmental competence and quality of produced embryos. The transportation medium was supplemented with: 100 μM butyrolactone I (BL), 500 μM IBMX + 100 μM forskolin (mSPOM), 100 μM milrinone (MR) or follicular fluid (bFF), and was carried out in a portable incubator for 6 h. Next, oocytes were in vitro matured (IVM) for 18 h, without the meiotic inhibitors, with the exception of mSPOM group, in which was added 20 μM cilostamide. The three control groups were IVM with 10% fetal calf serum (FCS) (Control Lab FCS) or 0.6% bovine serum albumin (BSA) (Control Lab BSA) in a CO2 in air incubator or in the portable incubator with 0.6% BSA (Control Transp BSA). Higher cleavage rates (P < 0.05) were obtained in the Control Lab FCS group (84.5 ± 5.3%) compared with the other groups (59.6 ± 3.4% to 70.9 ± 2.3%). Embryonic development was higher (P < 0.05) in the Control Lab FCS group (39.8 ± 4.7%) than in the Control Transp BSA (22.7 ± 3.4%) and MR (21.6 ± 2.3%) groups. However, they were similar (P > 0.05) to the other groups (23.6 ± 3.3% to 28.8 ± 2.7%). The total number of blastomeres was higher (P < 0.05) in the Control Lab FCS group (85.2 ± 5.6) than in Control Lab BSA (53.6 ± 2.9), Control Transp BSA (55.5 ± 4.4), BL (58.2 ± 3.0), mSPOM (57.9 ± 4.9) and MR (59.2 ± 3.9), but all these treatments did not differ (P > 0.05) from bFF (67.7 ± 4.2). No differences (P > 0.05) were found in apoptosis by the activity of caspases (139.0 ± 3.2 to 152.4 ± 6.5, expressed in fluorescence intensity) as well as the percentage of TUNEL-positive cells (12.3 ± 2.0% to 15.7 ± 1.7%). In conclusion, the transportation of oocytes over 6 h with BL, mSPOM or bFF enabled the acquisition of developmental competence at similar rates to the Control Lab FCS group.

Two proteins, haemoglobin and bovine serum albumin, have been studied with respect to their rates of alkylation by chlorambucil in vitro at 37 °C and pH 8.4. The proteins are of nearly the same molecular weight and free carboxylic acid content, but the alkylation reaction is 30 times faster with haemoglobin. On the other hand, the adsorption of chlorambucil by albumin is 20 times greater than that exhibited by haemoglobin. This inverse relationship between extent of adsorption and reaction rate suggests that adsorption protects the chlorambucil from activation in the solvent.

The present study was designed to investigate in vitro anthelmintic and cytotoxic activities of crude methanolic extract of two plants(Polygonum viscosum and Aphanamixis polystachya) grown in Bangladesh. Evaluation of cytotoxic activity was done using the brine shrimp lethality bioassay. The crude methanolic extract of Polygonum viscosum showed significant cytotoxic potential (LC50 value of 6.34 ?g/ml) among all the fractions comparing with that of standard vincristine sulphate (0.825 ?g/ml). Besides, the LC50 values of crude methanolic extract, pet ether and chloroform extracts of Aphanamixis polystachya showed good cytotoxic activities 11, 10.36, and 16.45 µg/ml, respectively. The other study was undertaken to evaluate anthelmintic activity (using Pheretima posthuma model) where piperazine was used as reference standard. The crude methanolic extract of Polygonum viscosum leaves produced a significant anthelmintic activity in dose dependent manner and the activity of crude extract was comparable with that of standard drugs. Besides, the Aphanamixis polystachya extract revealed moderate anthelmintic activity. Here, the anova testing was done with the P < 0.05. Medicinal plants possess therapeutic potential and are used to treat various diseases around the world. Eclipta prostrate (L.) is a medicinal herb that has extensive application in the native medicinal system. In any therapeutic activity chemical constituents play an important role. Eclipta prostrata has been investigated in this study for its antioxidant, antimicrobial and antidiabetic activity in vitro. The well-known research protocol available in the literature established qualitative analysis of the different phytochemical constituents and quantitative analysis of total phenol and flavonoids. The hydroalcoholic extracts of the leaves and seeds of Eclipta prostrata exhibited significant and dose-dependent antioxidant activity including ability to donate electron. To analyze the antimicrobial activity, Leaves hydroalcoholic extracts and Eclipta prostrate seeds were tested against two selected strains using a well-diffusion method and showing significant inhibitory action against all the strain tested. In addition, the dose-dependent α-amylase inhibitory activity with an IC50 value of acarbose, leaves, and seed extract was found to be 364.89μg/ml and 438.43μg/ml, respectively, indicating that Eclipta prostrate is a promising source as an herbal medicine. Background: Indian medicinal plants have great potential towards curing many diseases. Medicinal plant Coleus aromaticus is known for its wide medical applications. The main objectives of the study undertaken were to analyze the phytochemicals and compare the concentration present in the dialyzed Coleus aromaticus protein extract with various solvents.Methods: Various extracts of the medicinal plant Coleus aromaticus leaves such as dialyzed Coleus aromaticus protein, hydro alcoholic extract, ethanol extract and chloroform extract were prepared and analyzed for various phytochemical concentrations such as Total phenols (Folin-Ciocalteu method), Flavonoids (Aluminum chloride method), total sugars (Dubois method) and Proteins (Bradford’s method). Standard graph for each component was plotted.Results: The protein concentration in dialyzed Coleus aromaticus protein extract is considerably higher (5.8μg/10μl) than hydro alcoholic extract (1.6μg/10μl), ethanol extract (5.2μg/10μl) and chloroform extract (2.8μg/10μl). The other phytochemicals like total phenol, flavonoids and total sugars were low in concentration in the dialyzed Coleus aromaticus protein extract compared to hydro alcoholic extract, ethanol extract and chloroform extract.Conclusions: In this study, the protein concentration in dialyzed Coleus aromaticus protein extract is considerably higher than the same in other solvent extracts. This is the basis for further studies to unfold the antioxidant activity of Coleus aromaticus protein in vitro.

In the present work antifilarial active fraction was isolated from the leaves Chloroform extract of Aegle marmelos Corr. evaluated in vitro for antifilarial activity and studied the possible oxidative role against Setaria cervi parasite. Antifilarial study was carried out with isolated fractions by worm motility and MTT assays. Complete parasite motility inhibition was observed at 0.002 to 0.08 mg/mL in motility assay and in MTT assay plant fraction gave > 50% reduction 58.9, 74.6 and 97.2% at concentrations 0.02, 0.04 and 0.08 mg/mL at 10, 6 and 2 hours incubation period respectively (p< 0.05). Inhibitory concentration (IC50) was found to be 0.015 mg/mL. Oxidative parameters levels for MDA, Carbonyl content and Nitric oxide were identified as antifilarial activity achieved. The level of oxidative parameters was calculated in dose dependent manners as compared to the control level. The antifilarial activity of isolated fraction is associated with the oxidative mechanism in this study.

A new phenyl glycoside, equisetumoside D (1), was isolated from the aerial parts of Equisetum debile, along with equisetumoside B (2), dehydrovomifoliol (3), corchoionoside C (4), (-)-isolariciresinol-3a-O-β-D-glucopyranoside (5), and kaempferol 3-O-sophoroside-7-O-β-D-glucopyranoside (6). Their structures were elucidated by NMR spectroscopic and MS experiments. Compound 1 was found to be cytotoxic against both tested human cancer cell lines, hepatocellular carcinoma (Hep-G2, IC50: 1.12 μg/mL) and rhabdosarcoma (RD, IC50: 0.25 μg/mL), while the other compounds showed no activity against these cell lines by in vitro assay.

We studied ten patients on hemodialysis (HD) treated in sequence with cuprophan (CU), ethylenevinylalcohol (EVAL), polyacrylonitrile (A-69) and polysulphone (PSP) membrane dialyzers. ß2-microglobulin (ß2m) was measured by radioimmunoassay in plasma and dialysate samples. Plasma concentrations were corrected for changes in extracellular volume (ECV). We also studied adsorption in vitro by incubating the above membranes with I-125-labelled ß2m. There were no changes in ß2m plasma concentration after HD with CU dialyzers, but a significant decrease was observed with the other membranes tested. Filtration of ß2m across the dialyzer was absent with CU and minimal with EVAL. However, large amounts were recovered from dialysate with the high-permeability dialyzers, AN-69 and PSP. In vitro studies showed that maximal adsorption capacity was obtained with AN-69 (73%) compared to 9% with CU, 4% with EVAL and 7% with PSP. In summary, ß2m clearance with PSP is achieved through greater removal of this protein by mass transport across the membrane. The mechanism by which ß2m is removed from blood during AN-69 dialysis seems to include both adsorption to and filtration by the membrane itself.
