In order to use lactic acid bacteria isolated from Mulkimchi as a raw material for cosmetics, strain screening was carried out. And we measured the antioxidant activity of the lactic acid bacteria heat-killed cells obtained by heating lactic acid bacteria isolated from Mulkimchi. Among lactic acid bacteria derived from Mulkimchi, 41 strains were selected based on the culture suitability such as growth rate and antibiotic resistance. DPPH assay, ABTS assay, nitrite radical scavenging activity assay, and total phenolic contents were measured for 41 strains. Based on Lactobacillus plantarum LP299v, 1 strain was selected through DPPH assay, 1 strain through ABTS assay, 1 strain through nitrite radical scavenging activity assay, and 1 strain through total phenolic contents measurement. Among these, we isolated Lactobacillus sp. P100, which showed excellent antioxidant activity in all experimental methods, and the result of identification through 16s rRNA showed Lactobacillus plantarum. The strain showed 3-10% better antioxidant activity than Lactobacillus plantarum LP299v. And we measured the cell toxicity and anti-inflammatory activity. Lactobacillus sp. P100 showed the lowest cell toxicity and the highest NO production inhibitory rate. In other words, we could isolate lactic acid bacteria that can be used as an antioxidant ingredient in cosmetics from Mulkimchi.

Jaruk tigarun is a traditional fermented food from South Borneo made from tigarun flower that was fermented in water without salt addition. The objectives of the research were to isolate and identify the Lactic Acid Bacteria (LAB) from jaruk tigarun then to determine the DPPH radical scavenging activities of jaruk tigarun extracts. LAB was isolated from jaruk tigarun which was fermented for 7 days at room temperature. Media of MRS+CaCO3 were used to isolate the LAB while API 50 CHL kit was utilised to identify them. Fresh flowers and jaruk tigarun were also freeze-dryed, crushed and extracted using methanol, ethanol and ethyl acetate. The total phenolic and antioxidant activity of each extract were determine with Folin-Ciocalteu method and DPPH. Three isolates of LAB were isolated and identified as Lactobacillus plantarum. The fermentation was able to increase total phenolic of jaruk tigarun which was extracted with methanol, ethanol, and ethyl acetate (53.24 ± 0.73, 44.86 ± 0.90 and 23.95 ± 0.13 mg GAE/g extract, respectively). Similarly, the antioxidant activity of jaruk tigarun that were extracted with methanol, ethanol and ethyl acetate increased to 92.68 ± 0.02 %, 92.43 ± 0.11 %, and 42.94 ± 0.02 %. Methanolic extract of jaruk tigarun has the highest IC50 that was equal to 1.511 μg/mL. UV-Vis spectrum analysis and FT-IR were used to identify the compounds isolated from methanolic extract of jaruk tigarun resulting flavonoid as tentative identified compounds. ABSTRAKJaruk tigarun adalah salah satu makanan fermentasi tradisional dari Kalimantan Selatan yang dibuat dengan cara merendam bunga tigarun dalam air matang selama beberapa hari. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri asam laktat yang terlibat selama fermentasi jaruk serta mengetahui aktivitas penghambatan radikal DPPH pada jaruk tigarun. BAL diisolasi dari bunga tigarun yang difermentasi selama 7 hari pada suhu kamar sampai menjadi jaruk. Isolasi BAL menggunakan media MRS+CaCO3 dan diidentifikasi dengan API 50 CHL Kit. Bunga segar dan jaruk tigarun juga masing-masing dikeringbekukan, dihaluskan dan diekstraksi dengan metanol, etanol dan etil asetat. Total fenolik dan aktivitas antioksidan dari masing masing ekstrak ditentukan dengan metoda Folin Ciocalteu dan DPPH. Tiga isolat BAL berhasil diisolasi dan diidentifikasi sebagai Lactobacillus plantarum. Fermentasi berhasil meningkatkan total fenolik dari jaruk tigarun yang diekstrak dengan metanol, etanol dan etil asetat berturut-turut sebesar 53,24 ± 0,73, 44,86 ± 0,90 dan 23,95 ± 0,13 mg GAE/g ekstrak. Demikian pula halnya dengan aktivitas antioksidan dari jaruk yang diekstrak dengan metanol, etanol dan etil asetat  meningkat menjadi 92,68 ± 0,02 %, 92,43 ± 0,11 %, dan 42,94 ± 0,02 %. Ekstrak metanol dari jaruk tigarun  memiliki IC50  yang paling baik yaitu sebesar 1.511 μg/mL. Hasil isolasi dan identifikasi senyawa yang dimurnikan dari ekstrak metanol jaruk menggunakan analisis spektrum UV-Vis dan FT-IR diperoleh dugaan senyawa adalah golongan flavonoid.Kata kunci: DPPH; flavonoid; jaruk tigarun; L. The present study examined the effect of Lactobacillus plantarum ATCC14917 fermentation on the chemical composition and antioxidant activity of apple juice. Apple juice was fermented and examined of its antioxidant activity using chemical models and cellular antioxidant assay. Furthermore, the chemical composition of fermented apple juice was characterized by LC-MS/MS. Lactobacillus plantarum ATCC14917 fermentation showed an increase in DPPH and ABTS radical scavenging activity as well as cellular antioxidant activity of apple juice. However, fermentation decreased the total phenolic and flavonoid content. Subsequent LC-MS/MS analysis of the phenolic profile indicated that the content of 5-O-caffeoylquinic acid (5-CQA), quercetin, and phloretin with strong antioxidant activity was increased significantly after fermentation. The modified phenolic composition may contribute to the increased antioxidant activity of fermented apple juice. Our findings showed that Lactobacillus plantarum ATCC14917 fermentation may be an efficient way to enhance the bioavailability of phenolic compounds and to protect cells from oxidative stress.

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.

Melastoma malabathricum Linn. commonly known as ‘senduduk’ in Malaysia, belongs to Melastomataceae family. The study was carried out to screen the phytochemicals present in methanol extract of leaf, flower, fruit, and stem and to evaluate the antioxidant activity, total flavonoid and total phenolic contents of these different parts. Phytochemical screening showed that all parts of this plant contained tannins, steroids, phenols and flavonoids. The flower extract was found to have highest total phenolic whereas the leaf demonstrated the highest flavonoid content followed by flower. The results of antioxidant activity from the DPPH assay showed that the flower has the highest radical scavenging activity comparable to quercetin standard while stem showed the lowest activity. The higher content of total phenolics, flavonoids and antioxidant capacity of flower suggests the possibility of its incorporation and exclusion of stem in M. malabathricum preparations for development of newer effective drugs.

In this study, total phenolic, flavonoid contents, antioxidant capacity, and phenolic compositions of Castanopsis phuthoensis and Castanopsis grandicicatricata (Fagaceae family) were investigated. It was found that bark extracts were rich of phenolic contents, whereas leaf extracts were abundant of flavonoids. The total phenolics varied from 11.20 to 35.47 mg gallic acid equivalent g-1 dry weight (DW), and the total flavonoids were from 2.24 to 12.55 mg rutin equivalent g-1 DW. The results of antioxidant activity showed that the DPPH (1,1-diphenyl-2-picryl-hydrazyl) radical scavenging activity of the free phenolic extracts were higher than the bound phenolic extracts. Regarding the reducing power and β-carotene bleaching assays, the free phenolic extracts showed remarkably strong antioxidant capacity that were similar to the levels of the standard BHT (dibutyl hydroxytoluene) did. It could be concluded that free phenolic extracts were more effective in antioxidant activities than bound phenolic extracts. A highly significant correlation between phenolic contents and antioxidant activity in extracts were observed. By HPLC analysis, seven phenolic acids were detected including gallic, p-hydroxybenzoic, vanillic, sinapic, p-coumaric, ellagic, and vanillin. Of which, gallic, ellagic, and sinapic acids were the most abundant compounds in the two species. The results suggest C. phuthoensis and C. grandicicatricata contain rich sources of natural antioxidants and phenolic compounds which are probably considered in pharmaceutical use.

Through different extraction techniques, the potential of Centella asiatica as a natural source of antioxidant was investigated. The C. asiatica aqueous extracts were obtained via infusion, decoction and ultrasonic-assisted extraction (UAE) techniques. The effects of different extraction techniques were studied on the extraction yield, total phenolic contents and antioxidant activity. The total phenolic contents of the extracts and antioxidant activity were examined using the Folin-Ciocalteu’s reagent and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity assay, respectively. Results indicated that the UAE exhibited the highest extraction yield, highest total phenolic contents, as well as highest antioxidant activity. The yield of the extracts increased in the order of infusion < decoction < UAE which were 18.2 %, 23.6% and 25.4 %, respectively. All extraction techniques had a significant effect (p <0.05) on the total phenolic contents and antioxidant activity of C. asiatica extracts. The total phenolic contents was ranged from 3.42 ± 0.030 to 8.32 ± 0.105 mg GAE/g dry extract while the antioxidant activity was in the range of 75 to 86 %. This study confirms that C. asiatica has the potential to be a good resource for the future development of natural antioxidant. In addition, extraction via UAE can be an ideal technique to obtain phytochemical-rich extracts from medicinal plants.

The antioxidant activity (AA), total phenolic content (TPC), and total flavonoid content (TFC) of selected Indonesian Zingiberaceae herbs were determined. An optimization extraction procedure was conducted by using Taguchi L16 orthogonal array. Four chemical assays were applied, including 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity assay, H2O2 scavenging activity assay, Folin–Ciocalteau (F–C) assay, and NaNO2-AlCl3-NaOH assay, which revealed remarkable differences in AA, TPC, and TFC. The result indicated the diversity of AA composition among the herbs, and C. longa exhibited the highest AA. HPLC-PAD analysis revealed that curcumin was present in five high antioxidant herbs, and the highest amount was in C. longa. Pearson correlation analysis indicated that the identified TPC and TFC were significant contributors to AA, and curcumin was likely the main contributing antioxidant compound. Our approach concluded that C. longa is the greatest source of natural antioxidants among 12 Indonesian indigenous Zingiberaceae herbs. The use of a mixed-method approach to augment the findings of solitary methods might facilitate future researchers to uncover deeper and hidden meanings.

Objectives.Salvia fruticosaMill. (S. fruticosa) is widely used in folk medicine. Accordingly, the present study was designed to evaluate the antioxidant and anti-inflammatory activities ofS. fruticosa, and to determine the phenolic constituents of its extracts.Methods. The antioxidant activity was determined using 2,2-diphenylpicrylhydrazyl assay. Total phenolic contents were estimated using Folin-Ciocalteu reagent, and high-performance liquid chromatography was performed to identify phenolic constituents. To evaluate the anti-inflammatory activity, carrageenan-induced mouse paw edema was determined plethysmographically.Key Findings. Different plant extracts demonstrated strong radical scavenging activity, where the ethyl acetate extract had the highest value in the roots and the lowest in the aerial parts. This antioxidant activity was correlated to the total phenolic content of different extracts, where rutin and luteolin were the most abundant constituents. Interestingly, both the roots and aerial parts revealed a significant anti-inflammatory activity comparable to diclofenac.Conclusions. This study is the first to demonstrate pharmacologic evidence of the potential anti-inflammatory activity ofS. fruticosa. This activity may partly be due to the radical scavenging effects of its polyphenolic contents. These findings warrant the popular use of the East Mediterranean sage and highlight the potential of its active constituents in the development of new anti-inflammatory drugs.

ABSTRACTA pool of selected lactic acid bacteria was used for the sourdough fermentation of various cereal flours with the aim of synthesizing antioxidant peptides. The radical-scavenging activity of water/salt-soluble extracts (WSE) from sourdoughs was significantly (P< 0.05) higher than that of chemically acidified doughs. The highest activity was found for whole wheat, spelt, rye, and kamut sourdoughs. Almost the same results were found for the inhibition of linoleic acid autoxidation. WSE were subjected to reverse-phase fast protein liquid chromatography. Thirty-seven fractions were collected and assayedin vitro. The most active fractions were resistant to further hydrolysis by digestive enzymes. Twenty-five peptides of 8 to 57 amino acid residues were identified by nano-liquid chromatography-electrospray ionization-tandem mass spectrometry. Almost all of the sequences shared compositional features which are typical of antioxidant peptides. All of the purified fractions showedex vivoantioxidant activity on mouse fibroblasts artificially subjected to oxidative stress. This study demonstrates the capacity of sourdough lactic acid bacteria to release peptides with antioxidant activity through the proteolysis of native cereal proteins.

Abstract Objectives Immature Canavalia Gladiata Hull (ICGH) has been used traditionally as food but the research data on its functionality is insufficient. Antioxidant and anti-inflammation activity of ICGH and fermentation was performed using lactic acid bacteria (LAB) with glycosidase activity to confirm whether the bioconversion affect the functionality. Methods LAB was screened from Kimchi for the glycosidase activity. Antioxidant activity was evaluated by ABTS, DPPH, cellular antioxidant activity (CAA), and nitrite oxide (NO) production was evaluated with RAW264.7 cell treated with lipopolysaccharide (LPS). Active component in ICGH was analyzed using HPLC-PDA, MS/MS, and NMR. Results LABs could grow in medium with ICGH as a sole nutrient source. After fermentation the contents of lactic acid was increased as sugar contents declined and asparagine and γ-aminobutyrate(GABA) were detected as main metabolite, and some flavonoids were identified. After fermentation, higher free radical scavenging activity was confirmed in ABTS and DPPH assay and the tendency to decrease in reactive oxygen species and nitrite production was found in a dose dependent manner. Conclusions The fermentation of ICGH with two selected strains enhanced the antioxidant activity and decreased NO production by increase in the bioavailability of active compounds in ICGH.
