In spite of the rapid progress in the field of medicine, humanity still striving for cure of diabetes, a disorder of metabolism. From literature study it is inferred that no one therapeutic agent is isolated and/ or synthesized to treat diabetes, but to manage the disease. Therefore, it is of much importance to address this medical issue with phytochemical synergism for boosting immunity and limiting the severe side effects of synthetic hypoglycemic agents. The antioxidant and hypoglycemic effects of black seeds and chickpea have been determined previously, however the combined hypoglycemic effect of these seeds has not been studied until now. In present study the combined methanolic extract (CME) of Nigella sativa (black cumen) and Cicer arietinum (chickpea) seeds has shown the significant antioxidant potential as revealed by its significantly reduced IC50 value for radical scavenging activity in DPPH assay (-36.39%, p<0.001) compared to the standard compound butylated hydroxyanisole (BHA), and also due to the presence of significant (p<0.001) amount of total phenolic contents (TPC) in the mixture (254.2+2.75 mg GAE/g of dry weight). The CME was also fractionated through column chromatography and based on the phytochemical analysis, the fractions enriched with phenolic compounds were isolated and characterized by FT-IR spectroscopy. The IR-spectra have shown the presence of respective functional groups, indicating the isolation of specific phenol enriched compounds from CME which may be further used for drug development. During the evaluation of antidiabetic potential of CME in alloxan induced diabetic rats, the animals showed non-significant (p>0.05) increase (2.12%) in observed body weights, however, showed the significant decrease in blood glucose levels (-42.24%, p<0.001) compared to the diabetic control rats. Our findings suggest that CME may be used as a potential hypoglycemic herbal medication or might be used in conjunction with the usual synthetic drugs to boost activity and limit the side effects.

Objective: To investigate the presence of phytochemical components and to evaluate the in vitro antioxidant activity of pet. ether, chloroform, methanol and aqueous extracts of Grewia heterotricha mast leaves.Methods: The leaves of Grewia heterotricha mast were dried and extracts were prepared using a pet. ether, chloroform, methanol by soxhlet extraction method. The aqueous extract was prepared using distilled water by cold extraction method. The preliminary phytochemical analysis was carried out on aqueous, methanol, chloroform and pet. either leaf extracts of the plant using standard qualitative procedures. The total phenolic content (TPC) was estimated using modified Folin-Ciocalteau method, tannin content by Folin-Denis method and total flavonoids by aluminum chloride method. In vitro, antioxidant activities were evaluated by 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, H2O2 scavenging activity and FRAP assay.Results: The preliminary phytochemical analysis revealed the presence of complex bioactive constituents like phenols, tannins, alkaloids, terpenoids, flavonoids, saponins, steroids, glycosides, coumarins, proteins and carbohydrates. Methanolic extract showed highest total phenolic content (87.58±2.52 mg CE/g) than aqueous extract (78.46±5.36 mg CE/g). Higher tannin content was found in the aqueous extract (148.0±8.96 mg TAE/g). Total flavonoids were highest in chloroform extract (314.9±25.06 mg QE/g) followed by aqueous (242.98±32.42 mg QE/g) and methanolic extract (217.0±18.32 mg QE/g) and lowest in a pet. ether extract (188.86±23.35 mg QE/g). The methanolic extract had shown very significant DPPH radical scavenging activity (IC50 98.95 µg/ml) and H2O2 scavenging activity (IC50110.1µg/ml) compared to the standard ascorbic acid. Higher reducing ability was observed in methanol extract (131.8±11.67 mg AE/g).Conclusion: The results obtained reveal that the leaves of Grewia heterotricha mast have potent antioxidant property. The observed activity may be associated with bioactive components like phenolics, flavonoids present in the leaf extracts and could have greater importance as therapeutic agents in oxidative stress-related degenerative diseases. Further studies are needed in order to purify bioactive compounds responsible for the antioxidant property.

<p><strong>Objective: </strong>To evaluate the <em>in vitro </em>antioxidant activity of liquorice (<em>Glycyrrhiza glabra) </em>against H<sub>2</sub>O<sub>2</sub> induced oxidative stress in HepG2 cell line.</p><p><strong>Methods: </strong>Antioxidant activity of methanolic extracts of <em>Glycyrrhiza glabra</em> was investigated by measuring total phenolic content using folin-ciocalteu reagent (FCR), free radical scavenging activity by DPPH and ferric reducing antioxidant power (FRAP). The presence of phenolic compounds and flavonoids in the extract was confirmed by Liquid Chromatography-Mass Spectrometry (LC-MS) analysis. Furthermore, the protective effect of methanolic extract of <em>Glycyrrhiza glabra</em> against oxidative stress induced by H<sub>2</sub>O<sub>2 </sub>in HepG2 cells was investigated by MTT assay. HepG2 cells were exposed with five different treatments viz. liquorice, H<sub>2</sub>O<sub>2</sub>, ascorbic acid, H<sub>2</sub>O<sub>2</sub>+liquorice and H<sub>2</sub>O<sub>2</sub>+ascorbic acid, to explore the effect of the extract on malondialdehyde (MDA) production, catalase activity, and glutathione reductase levels.<strong></strong></p><p><strong>Results: </strong>The total phenolic content estimated in <em>Glycyrrhiza glabra </em>extract was found to be 241.47 µg per 1000 µg/ml of methanolic extract. It was found that as the concentration of the extract was increased both the free radical scavenging activity and ferric ion reducing power was also found to increase. LC-MS analysis confirmed the presence of eight different phenolic compounds in the methanolic extract which are possibly contributing to the antioxidant activity exhibited by the extract. It was also observed that liquorice treated HepG2 cells showed lower MDA and higher glutathione and catalase levels as compared to only H<sub>2</sub>O<sub>2 </sub>treated HepG2 cells where increased MDA production, decreased glutathione reductase and catalase production was observed.</p><p><strong>Conclusion: </strong>Our results thus conclude that, the methanolic extract of <em>Glycyrrhiza glabra </em>can be used as natural supplements in various disease conditions where oxidative stress has been reported. <strong></strong></p><p> </p>

The present study investigates the total phenol amount, radical scavenging and anticancer activity of methanolic extract of Phlomis brevibracteata Turrill (PBT) leaves; an endemic plant of North Cyprus. The antiradical activity and cytotoxicity of 70% methanolic extract of PBT leaves were analyzed by the implementation of DPPH, ABTS radical scavenging activity tests and MTT assay, respectively. Our results showed significant antiradical scavenging activity in the total phenolic content of the PBT leaves. MTT assay proved potent cytotoxic activity against SK-HEP-1 cell line not only for 70% methanolic extract of PBT, but also for gallic acid and caffeic acid constituents of the total phenolic extract.

Objective: To evaluate the preliminary phytochemical content and antioxidant potential of the hydroalcoholic leaf extracts of Hemionitis arifolia. Methods: Total phenolic, flavonoid and alkaloid contents were evaluated using spectrophotometric methods. The free radical scavenging activity of the leaf hydroalcoholic extract were evaluated against DPPH+, ABTS+, Reducing power assay and nitric oxide assay were determined. Results: The hydroalcoholic concentrate of H. arifolia uncovered the most elevated polyphenol content when contrasted and the other phytoconstituents. Absolute phenol content of the hydroalcoholic separate was observed to be 31.78%, flavonoid content is 1.02% and Alkaloid content is 30.40% individually. The Solvent concentrates showed huge cell reinforcement movement, with hydroalcoholic extract. ABTS Assay, DPPH assay, Reducing power assay and Nitric oxide assay where the Inhibition concentration were 667.75µg/ml, 734.25 µg/ml, 791.58 µg/ml and 899.67 µg/ml. Conclusion: This study suggests that hydroalcoholic leaf extracts of H. arifolia could be a potential source of natural antioxidant and justifies its use in ethno-medicine.

The petroleum ether, chloroform and ethyl acetate soluble fractions of methanolic extract of Litsea lancifolia Roxb. leaves were subjected to different pharmacological screenings to explore its potential as anti-oxidant, antimicrobial, peripheral analgesic, hypoglycemic and CNS depressing agent. The ethyl acetate soluble fraction showed highest total phenolic content and free radical scavenging activity compared to the standard, acetyl salicylic acid. Potential antimicrobial activity was shown against P. aeruginosa (23.50 mm), E. coli (22.33 mm), B. cereus (18 mm) and S. paratyphi (18 mm). The crude extract demonstrated significant peripheral analgesic (p < 0.01) activity with 69.45 and 77.96% inhibition of acetic acid-induced writhing at 100 and 200 mg/kg b.w., respectively. The crude methanolic extract also showed significant hypoglycemic activity (p < 0.01) at a dose of 500 mg/kg/day on the 7th day of treatment. All the organic soluble fractions exhibited noteworthy (p < 0.001) CNS depressant activity. Taken together, the plant can be considered as a good material for further chemical investigation to isolate the bioactive constituents.

Abutilon indicum is consider to be used in the traditional system of medicine. It is found in tropical and subtropical regions of the world. It is used to treat various diseases. This plant does not cause any side effects to humans. As the plant has wide variety of medicinal properties, the present study aimed to comparative between plant and callus extract of Abutilon indicum (L.) sweet for antioxidant, antibacterial, antidiabetic and anti- proliferative activity. The highest percentage of callus induction (89.50%) and callus weight (1.26 g) was observed in T5 (MS + 2, 4-D (2.5 mg/l) + BAP (2 mg/l) and T8 [IBA (4 mg/l)] respectively. Phytochemical analysis of aqueous and ethyl acetate extracts of A. indicum in vivo plant and in vitro grown callus showed the presence of alkaloids, flavonoids, phenols, carbohydrates, glycosides, protein, terpenoids, saponins, tannins and coumarin. The total phenolic content was high in aqueous extract of callus (30.68 mg TAE/g). Maximum DPPH radical scavenging activity was found in aqueous extract of callus (86%) with IC50 value of 68.49 µg/ml. FT-IR analysis of aqueous extract of A. indicum plant and callus showed the presence of characteristic stretching at 2930.28 and 2927.75 indicating the presence of C-H stretching respectively. GC-MS analysis revealed the presence of 17 compounds in ethyl acetate plant extract, whereas 7 compounds in ethyl acetate callus extract such as tetradecane, 1-chloro, Sulfurous acid 2-prophytridecyl ester and 1- ethyl-3-[2-(octadecylthio) ethyl] thiourea. The ethyl acetate extracts of callus and plant and was found to be effective against Bacillus subtilis (3.1 mm) and Staphylococcus aureus (2.9 mm). Maximum α-amylase inhibitory activity was observed in aqueous callus extract (32.65%) with IC50 value of 833.61 µg/ml. HeLa cell viability was found to be 26.8% and 21.8% in plant and callus extract respectively.

Stephania glabra is a wild medicinal plant possessing multiple uses as anti-cancer, antimicrobial and antioxidant activities. Different extracts and fractions were prepared from S. glabra tubers and were analyzed for different activities. Evaluation of the total phenolic and flavonoid content revealed that chloroform extract of S. glabra contain maximum phenolic content and maximum flavonoid content was found in methanolic extract. Dry powder analysis of the tubers revealed the presence of saponins, alkaloids and proteins. Ethyl acetate extract of S. glabra tubers showed highest antioxidant potential as revealed by ABTS radical scavenging activity. Methanolic extract has the highest anti-cancer activity while chloroform extracts and fractions have highest antimicrobial activity. These activities can be attributed to the high amount of the phytoconstituents present in these extracts

Carissa edulis is a plant used in the management of oxidative stress and inflammatory related disorders such as malaria, rheumatism inflammation, and cardiovascular diseases. The present study evaluates the total phenolic content, antioxidant capacity (DPPH, ABTS, and FRAP), and the bioactive compounds present in the various extracts of C. edulis (HEC, MEC, AEC, and PC). An HPLC analysis determined the different compounds present in the extracts. High concentration of total phenolic content was observed in aqueous and methanolic extracts more than in the hydroethanolic extract though not significantly different. Flavonoids were higher in the hydroethanolic and methanolic extracts, respectively, with 14.84 mg RE/g extract and 12.02 mg RE/g extract. Tannins were also found in large amounts in the same two extracts with 26.76 mg TAE/g extract and 34.67 mg TEE/g extract. The percentage radical scavenging activity DPPH ranged between 58.63% and 94.67% for aqueous extract and for ABTS between 51.39% and 94.12% for the methanolic extract. The highest FRAP was obtained in the methanolic extract (6.73 g AAE/100 g extract). HPLC analysis revealed the presence of quercetin, rutin, and gallic acid in the different extracts. C. edulis represents a potential source of bioactive components with antioxidant capacity.

Objective: To standardise the protocol for rapid callogenesis in Mussaenda frondosa L. using leaf explants. Qualitative and quantitative phytochemical analysis of leaf, stem and callus cultures.Methods: The leaf explants were inoculated onto MS medium supplemented with varying concentrations of growth regulators such as 2, 4 - D, NAA, BAP, Kn for the induction of callus. Qualitative and quantitative analysis of total phenol, flavonoids and alkaloids contents of leaf, stem and callus were tested by standard methods.  The antioxidant activities were investigated using DPPH radical scavenging method and reducing power assay. The anti - inflammatory activity was evaluated by membrane stabilizing activity.Results: Pale green, healthy, friable and fast growing callus was obtained on the medium enriched with NAA (2mg/l) + Kn (4mg/l). Quantitative determination showed the highest concentration of total phenolics in the methanolic extract of in vitro grown callus (10 ± 1.1 mg of GA/g of extract), flavonoids in methanolic stem extract (137±1.6 mg of Quercitin/g of extract) and alkaloids in methanolic extract of leaf (118.3±1.5 mg/10g of extract). The methanolic leaf extract exhibited highest free radical scavenging activity with IC50 value of 40.6±10.06 μg/ml. The highest membrane stabilizing activity was shown by chloroform extract of the leaf (66.02%).Conclusion: The present preliminary phytochemical and pharmacological analysis may form the basis for drug development in future using callus cultures of M. frondosa.

Objective: The present study was carried out to evaluate the in vitro preliminary phytochemical analysis and antioxidant activity of methanolic leaf extracts of Corchorus olitorius L. The in vitro antioxidant activity was evaluated by DPPH radical scavenging activity method.Methods: Fresh jute leaves (immature, mature and senescence) were collected, air dried and the crude powder was prepared for phytochemical and antioxidant analysis. The powder was mixed with 70% methanol and the supernatant was separated. The antioxidant activity of this methanolic extract was measured on the basis of the scavenging activity of the stable 1, 1-diphenyl 2-picrylhydrazyl (DPPH) free radical with slight modifications. Phytochemical analysis was performed according to standard laboratory protocol.Results: The results indicated the presence of different phytochemicals viz. glycosides, steroids, cholesterol, alkaloids, phenols, flavonoids, riboflavin, saponins and terpenoids. The sample also showed antioxidant activity by inhibiting DPPH radical. The significant antioxidant activity of methanolic leaf extract of might be due to the presence of saponins, phenols, flavonoids and alkaloids found in the preliminary phytochemical analysis.Conclusion: Present study reveals that the jute leaf possesses different phytonutrients and exhibited DPPH radical scavenging activity, and therefore, may be used for therapeutic purposes.
