The present study aimed to evaluate the filtration for separating seminal plasma of boars’ ejaculate by means of sperm viability and the occurrence of hyperactivation and lipid peroxidation in fresh semen and after cooling for up to 96 hours. The ejaculate of eight healthy boars of different breeds was collected and the gelatinous portion was separated and discarded. In the laboratory, the semen was fractioned into three aliquots (groups G1, G2 and G3) as follows: G1: semen with plasma diluted in BTS (TOTAL BTS); G2: semen centrifuged at 600G/10’ (BTS CEN); and G3: semen filtered with the Sperm-filter® following dilution of the retained cells with BTS (BTS FIL). The analyses were performed at three moments: with fresh samples (D0) and after 48 (D2) and 96 hours (D4) of cooling at 17ºC. The kinetic evaluation was performed using the CASA system, which provided data for the classification of sperm hyperactivity. For lipid stress analysis, the TBARS (thiobarbituric acid reactive substance) test was performed. The variance analysis test was conducted to compare the results between the groups and moments analyzed. The results showed better total motility values (%) for G1 at D0 (67.9, P= 0.001), D2 (36.6, P= 0.004) and D4 (26.1, P= 0.003). The occurrence of hyperactivity was observed in G2 and G3 at moments D2 and D4. In addition, TBARS showed higher peroxidation levels for G1 at D0 (8.1 mM MDA/ml, P= 0.01), D2 (7.4 mM MDA/ml, P= 0.02), and D4 (6.41mMol MDA/ml, P= 0.008) when compared to G2 and G3. Since the filtration method did not demonstrate any damage to the sperm viability, the study concluded that sperm filtration is an accessible and valid tool to replace centrifugation.

Resistance of erythrocytes from Brown trout (Salmo trutta m. trutta L.) affected by ulcerative dermal necrosis syndrome In the present work we evaluated the effect of ulcerative dermal necrosis (UDN) syndrome on resistance of erythrocytes to haemolytic agents and lipid peroxidation level in the blood from brown trout (Salmo trutta m. trutta L.). Results showed that lipid peroxidation increased in erythrocytes, as evidenced by high thiobarbituric acid reactive substance (TBARS) levels. Compared to control group, the resistance of erythrocytes to haemolytic agents was significantly lower in UDN-positive fish. Besides, UDN increased the percent of hemolysated erythrocytes subjected to the hydrochloric acid, urea and hydrogen peroxide. Results showed that UDN led to an oxidative stress in erythrocytes able to induce enhanced lipid peroxidation level, as suggested by TBARS level and decrease of erythrocytes resistance to haemolytic agents.

Background: Exogenous antioxidants are considered as a promising therapeutic approach to treat neurodegenerative diseases since they could prevent and/or minimize the neuronal damage by oxidation. Objective: Three series of lipophilic compounds structurally based on scutellarein (2), which is one metabolite of scutellarin (1) in vivo, have been designed and synthesized. Methods: Their antioxidant activity was evaluated by detecting the 2-thiobarbituric acid reactive substance (TBARS) produced in the ferrous salt/ascorbate-induced autoxidation of lipids, which were present in microsomal membranes of rat hepatocytes. The lipophilicity of these compounds indicated as partition coefficient between n-octanol and buffer was investigated by ultraviolet (UV) spectrophotometer. Results: This study indicated that compound 5e which had a benzyl group substituted at the C4'- OH position showed a potent antioxidant activity and good lipophilicity. Conclusion: 5e could be an effective candidate for preventing or reducing the oxidative status associated with the neurodegenerative processes.

ABSTRACT Background Wide variation in fertility rates is observed when using frozen bull semen, even when the bulls have met quality standards for semen production. Therefore, a simple and reliable test to assess the freezing potential of bull semen based on the analysis of fresh semen or blood would be of great value. Attention is now turning to assessment of seminal plasma components such as proteins and elements. In the present study, the concentrations of macro- and microelements in fresh bull semen plasma and in serum and their correlation with quality characteristics of fresh semen and with semen quality after freezing and thawing were determined. Ejaculates were collected from 30 mature bulls, and semen volume, concentration, sperm motility, morphology, tail membrane integrity, plasma membrane permeability and DNA fragmentation were determined on the day of collection and after freezing and thawing. The concentrations of macroelements (Na, Mg, K and Ca) and microelements (Cu, Fe, Zn and Se) were determined in the seminal plasma and serum. The semen samples were classified into satisfactory and unsatisfactory groups according to the fresh semen quality. Results Zinc and Se levels measured in serum were associated with almost all fresh and frozen-thawed semen quality characteristics, while Fe levels were associated only with acrosomal defects in fresh semen. Zinc and Fe levels in fresh seminal plasma were associated with various quality characteristics of fresh and frozen-thawed semen, while Se level in fresh seminal plasma was not associated with any of the semen quality characteristics. Conclusions Microelements were shown to be useful as biomarkers involved in the analysis of bull sperm quality and could be used as an additional tool to predict bull semen quality after freezing and thawing. Our results confirm that the analysis of Zn and Se levels in serum and Zn, Cu and Fe levels in fresh seminal plasma can provide information to discriminate between bull semen samples with spermatozoa with high or low cryotolerance.

This experiment aims to evaluate the correlation between lipid peroxidation levels in serum and seminal plasma in equines. Also, it investigates the lipid peroxidation in extended semen samples and its effects and sperm motility during a 72 hr refrigeration period. Blood and semen were collected from fertile Crioulo stallions. Serum and seminal plasma lipid peroxidation levels were analyzed by thiobarbituric acid reactive substances (TBARS) immediately after semen collection. After addition of extender (hour = 0), diluted semen was refrigerated and stored at 5 °C. Semen analyses, TBARS and catalase activity were performed in extended semen at 0, 24, 48, and 72 hours. We noted that levels of plasma lipid peroxidation can be used as an indicative of seminal oxidative stress. Also, lipid peroxidation does not increase substantially during semen storage. Lipid peroxidation and the antioxidant enzyme catalase do not seem to be the major cause of loss and motility and consequently reduction in fertility in stallion semen during storage for 72 h at 5 °C.

This study investigated the effects of searing process before sous-vide (SV) treatment on quality traits, visual attributes, palatability, and storage stability of SV cooked pork patties. Patties were seared on each side by pan-frying for 0 (control), 30 (S30), 60 (S60), 90 (S90), or 120 (S120) s in a stainless-steel pan, and all patties were then vacuum-packed and cooked under thermally controlled conditions at 75 °C for 2 h. Marked differences were observed in quality properties between the control and searing groups, and the S120 group exhibited greater brown surface color and cooking loss compared to the other groups (p < 0.001) due to the additional heating process. Patties from the S60 group showed greater appearance and tenderness acceptability scores compared to patties from the S30 and S120 groups (p < 0.001). On another note, the effects of searing on storage stability were somewhat limited, as they were measured by 2-thiobarbituric acid reactive substance, volatile basic nitrogen, total aerobic bacterial count, and coliforms during 49 d of cold storage. Therefore, searing process before SV treatment can improve the visual attributes and palatability of cooked pork patties, and the optimum searing condition was for 60 s, without impairing the storage stability.

Purpose This paper aims to study the effects of Zygophyllum album (Za) (Z. album) in hypercholesterolemic-diabetic rats. Design/methodology/approach Male Wistar rats (n = 36) weighing 200 ± 10 g, consumed an experimental diet containing 20 per cent casein were divided into three groups (n = 12). The first group consumed the diet enriched with 1 per cent cholesterol (CH), the second group is rendered diabetic by intraperitoneal injection of streptozotocin (STZ) (35 mg/kg body weight) (D) (DM). The third group is hypercholesterolemic and STZ-induced diabetic (CH-DM); each group was subdivided into two groups (n = 6), non-treated groups (CH, DM and CH-DM) and treated groups supplemented with 1 per cent Z. album lyophilized aqueous extract (CH-Z, DM-Z and CH-DM-Z). Findings In CH-DM group, Z. album decreases glycemia (−15 per cent) and inversely increases insulinemia (+28 per cent) and Homeostasis Model Assessment of Insulin Resistance (+19 per cent). In liver, total cholesterol (TC) and triacyglycerols (TAG) levels were reduced by −57 per cent and −29 per cent, respectively. In plasma, TC concentration was increased by +20 per cent, whereas those of TAG level were lowered by −56 per cent. Lecithin cholesterol acyl transferase and paraoxonase 1 activities were raised by +45 and +59 per cent, respectively. Inversely, thiobarbituric acid reactive substance levels were lowered significantly in liver, heart, kidney and adipose tissue (p < 0.05). Superoxide dismutase activity was enhanced in liver (+54 per cent), heart (+36 per cent), kidney (+45 per cent) and adipose tissue (59 per cent). Liver glutathione peroxidase (GSH-Px) activity was enhanced by +38 per cent and heart activities of GSH-Px, and glutathione reductase (GSSH-Red) were increased by +25 and +18 per cent. In kidney, GSH-Px activity was reduced by −26 per cent; in reverse, GSSH-Red activity was increased by +14 per cent. In adipose tissue, GSH-Px and GSSH-Red activities were augmented by +35 and +15 per cent. Originality/value These results suggest that Z. album aqueous extract has antihyperglycemic and antihyperlipemic actions. Also, Z. album protects against tissue oxidative damage; therefore, it can help to prevent cardiovascular complications of diabetes combined with hypercholesterolemia.

Oxidant and antioxidant activity in seminal plasma was compared with that in spermatozoa of teleost (common carp Cyprinus carpio and brook trout Salvelinus fontinalis) and chondrostean (Russian sturgeon Acipenser gueldenstaedtii, Siberian sturgeon Acipenser baerii, and sterlet Acipenser ruthenus) fishes. No differences were found between seminal plasma and spermatozoa in the level of thiobarbituric-acid-reactive substance (0.24 &plusmn; 0.08 to 0.33 &plusmn; 0.04 nmol/mg proteins) in Russian sturgeon, Siberian sturgeon, and sterlet. Carbonyl protein concentration was significantly higher in spermatozoa than in seminal plasma of all studied species. Analyzed antioxidants included superoxide dismutase, glutathione reductase, and glutathione peroxidase activity. Significant differences (P &lt; 0.05) were detected between seminal plasma and spermatozoa in total superoxide dismutase (SOD) and glutathione reductase (GR). Total glutathione peroxidase (GPx) activity was significantly higher in brook trout (12.56 &plusmn; 3.23 mU/mg proteins) and Russian sturgeon (11.56 &plusmn; 3.12 mU/mg proteins) spermatozoa compared to seminal plasma (6.81 &plusmn; 1.56 mU/mg proteins in brook trout and 9.56 &plusmn; 3.12 mU/mg proteins in Russian sturgeon). This study provides new data on oxidant and antioxidant balance between spermatozoa and seminal plasma that may be of value in the development of methods for artificial reproduction of teleost and chondrostean species. &nbsp;

Sudarshanapowder (SP) is one of the most effective Ayurveda powder preparations for paediatric febrile conditions. The objective of the present study was to evaluate thein vitroandin vivoantioxidant potentials of SP. Thein vitroantioxidant effects were evaluated using ABTS radical cation decolourization assay where the TROLOX equivalent antioxidant capacity (TEAC) was determined. Thein vivoantioxidant activity of SP was determined in Wistar rats using the Lipid Peroxidation (LPO) assay in serum. Thein vitroassay was referred to as the TROLOX equivalent antioxidant capacity (TEAC) assay. For thein vivoassay, animals were dosed for 21 consecutive days and blood was drawn to evaluate the MDA level. Thein vitroantioxidant activity of 0.5 μg of SP was equivalent to 14.45 μg of standard TROLOX. The percentage inhibition against the radical formation was50.93±0.53%. The SP showed a statistically significant (p<0.01) decrease in the serum level of thiobarbituric acid-reactive substance in the test rats when compared with the control group. These findings suggest that the SP possesses potent antioxidant activity which may be responsible for some of its reported bioactivities.
