We describe three new parastenocaridid: Cottarellicaris sanctiangeli Bruno & Cottarelli sp. nov., Stammericaris vincentimariae Bruno & Cottarelli sp. nov. and Proserpinicars specincola Bruno & Cottarelli sp. nov., collected in the pools of five different caves located in Calabria (Southern Italy). We conducted a phylogenetic analysis based on the mitochondrial COI and ribosomal 18S sequences of C. sanctiangeli sp. nov. and S. vincentimariae sp. nov., and of four more species of Stammericaris and one Proserpinicaris available from literature. Based on the molecular study, the specimens of C. sanctiangeli sp. nov. are clearly separated from the species belonging to the closely-related genus Stammericaris. The morphological and molecular data indicate that the genera belonging to the two subfamilies Parastenocaridinae and Fontinalicaridinae form two monophyletic and distinct clades, thus supporting their status. We also provide insights on the genus Proserpinicaris based on morphological data only; in particular, the most important synapomorphic character of the genus, i.e., the taxonomic value of the hyaline structure inserted on the anterior surface of the male leg 4 basis, is discussed based on the development of the P4 endopod, as observed in the last copepodid stage of some species of this genus. Finally, we widen the biogeographic and ecological knowledge of the three genera.

The genus Helicotylenchus contains cosmopolitan, ubiquitous plant-parasitic nematodes with some species capable of causing significant economic damage to agricultural crops. Accurate species identification in this genus is essential in recognizing the damaging species and establishing effective management options. In a study on cocoa plantations in Nigeria, two species of spiral nematodes were found in high numbers from soil samples obtained from a six-decade old cocoa plantation at the Cocoa Research Institute of Nigeria. An integrated approach involving a combination of morphology, morphometrics, and molecular tools was used to identify the nematode species. Morphological data indicate the presence of both H. multicinctus and H. dihystera. There is congruence in the morphological and molecular data obtained for H. multicinctus. However, phylogenetic analysis of the 28S rDNA expansion segment revealed a high variability in the sequences of the Nigerian population of H. dihystera, suggesting the need for a careful appraisal and more comparative studies.

A total of 38 eggplant genotypes, of which 32 were heirloom accessions collected from different regions of Burdur province five were different local genotypes from other provinces, and one was a cultivar, were used as reference in this study. The phylogenetic relationships among these heirlooms were evaluated using 40 morphologic descriptors and five randomly amplified polymorphic RAPD markers. The horizontal dendrograms were created by using UPGMA with both morphologic and molecular data. Burdur heirloom accessions showed high genetic diversity based on morphological and molecular data. The genetic similarity rates ranged from 0.29 to 0.91 according to the morphological data, and ranged from 0.84 to 0.98 according to the molecular data. Molecular data generated by RAPD method, compared to morphological data, were insufficient to reveal genetic diversity. Therefore, in order to confirm genetic variations, studies based on other molecular methods are necessary. The regional genetic populations include a wide eggplant genetic diversity which can be good source for the breeding studies performed in the future.

AbstractFour strains of entomopathogenic nematodes were isolated with a live trap method in southern Poland. The isolates were identified asSteinernema silvaticumbased on morphological, morphometric and molecular data. Infective juveniles of PolishS. silvaticumisolates differ in body length fromS. weiseri(951 vs 740 μm, respectively), and in the hyaline tail portion fromS. kraussei(48 vs 38%, respectively). First-generation males ofS. silvaticumare longer than those ofS. kraussei,S. weiseriandS. ichnusae(1829 vs 1400, 1180 and 1341 μm, respectively). Males ofS. silvaticumand a sister speciesS. krausseican be distinguished by the distance from the anterior end to the nerve ring (142 vs 105 μm), spicule (66 vs 49 μm) and gubernaculum length (45 vs 33 μm), and the presence of a mucron. The analysis of internal transcribed spacer (ITS), D2-D3 andcox1sequences of the tested nematodes revealed differences of 3–5%, 3% and 12–13%, respectively, fromS. krausseistrains. The phylogeny of both nuclear and mitochondrial genes indicated close relationships of the PolishS. silvaticumisolates withS. kraussei,S. oregonenseandS. cholashanense. The reproductive isolation of the studied isolates was confirmed by hybridization tests with other Europeanfeltiae–krausseigroup representatives. This study has supplemented the original description ofS. silvaticumwith morphological and morphometric characterization of the first-generation males and females. This is also the first molecular study of this species based on a multi-gene approach.

Abstract. Jannual N, Nipitwattanaphon M, Hasin S, Kaewgrajang T. 2020. Morphological and molecular characterization of Termitomyces (Lyophyllaceae, Agaricales) in Thailand. Biodiversitas 21: 2481-2491. Termitomyces is considered to be a highly prized delicacy collected both for home consumption and for sale in local markets. Although the taxonomic information about this genus is well known in Africa, the identification of Termitomyces species in Thailand is unclear. Therefore, this study presented an assessment of phylogenetic relationships in the genus Termitomyces by means of sequencing of the ITS1-5.8S-ITS2 region and the species characterization by combination of morphological data and molecular data. Total of 61 Termitomyces specimens was collected from a variety of geographical localities of Thailand. They were classified into six species, including T. clypeatus, T. cylindricus, T. fuliginosus, T. heimii, T. microcarpus, and T. striatus based on morphological characteristics combining with molecular characteristics. This is the first diagnostic key of Thai Termitomyces.

The recent catalogues of the family Dolichopodidae considered Syntormon pallipes (Fabricius, 1794) and S. pseudospicatus Strobl, 1899 as separate species. In this study, we used three approaches to estimate the significance of differences between the two species: molecular analysis (COI and 12S rRNA sequences), analysis of leg colour characters and geometric morphometric analysis of wing shape. The morphological data confirmed the absence of significant differences between S. pallipes and S. pseudospicatus found in the DNA analysis. Significant differences in the wing shape of two species have not been revealed. Hence, according to our data, there is no reason to consider S. pseudospicatus as a distinct species.

AbstractCtenophthalmus is considered the largest genus within the Order Siphonaptera. From a morphological point of view, only males of this genus can be identified at species and subspecies levels using morphological keys, whereas there are no morphological criteria in order to classify females at these taxonomical levels. Furthermore, the amount of available molecular and phylogenetic data for this genus is quite scarce so far. The main objective of this work was to assess the utility of the combination of nuclear and mitochondrial markers with respect to their ability to differentiate among different subspecies within the Ctenophthalmus genus. With this purpose, we carried out a comparative morphological and molecular study of three different subspecies (Ctenophthalmus baeticus arvernus, Ctenophthalmus nobilis dobyi, and Ctenophthalmus andorrensis catalaniensis) in order to clarify and discuss its taxonomic status. In addition, our study complemented the molecular data previously provided for Ctenophthalmus baeticus boisseauorum and Ctenophthalmus apertus allani subspecies. We sequenced five different molecular markers: EF1-α, ITS1, ITS2, cox1, and cytb. Our results confirmed that morphological data by themselves are not able to discriminate among Ctenophthalmus female taxa; however, the combination of the nuclear marker EF1-α together with mtDNA markers cytb and cox1 constituted a useful taxonomical and phylogenetic tool to solve this issue. Based on these results, we consider that the use of this molecular approach should be gradually used within Ctenophthalmus genus in order to complement its classical taxonomy and clarifying the complex taxonomy of other congeneric species of fleas.

Girardia Ball, 1974 is the most diverse and widely distributed genus of the family Dugesiidae (Platyhelminthes: Continenticola) in the Neotropical region. Seven out of the 52 species of the genus are known for Argentina. The Somuncurá Plateau is a region in northern Patagonia with several endemic flora and fauna, but little is known about the free-living Platyhelminthes. We describe two new species of Girardia partially inhabiting in sympatry in the Somuncurá Plateau: Girardia somuncura sp. nov. and Girardia tomasi sp. nov. The identification criteria that we followed was an integrative taxonomic approach based on morphological and molecular data. Thus, we used anatomical features focused on the reproductive system, together with a phylogenetic analysis, using a mitochondrial (COI barcode region) genetic marker. This study is the first phylogenetic analysis of the genus Girardia in which we include the southernmost representatives of America here described, thus making it possible to incorporate them in global phylogenies.

Autolytinae is revised based on available types, and newly collected specimens. Out of 170 nominal species, 18 are considered as incertae sedis, 43 are regarded as junior synonyms, and 25 are referred to as nomina dubia. The relationships of Autolytinae is assessed from 51 morphological characters and 211 states for 76 ingroup-taxa, and 460 molecular characters from mitochondrial 16S rDNA and nuclear 18S rDNA for 31 ingroup-taxa; outgroups include 12 non-autolytine syllid polychaetes. Two analyses are provided, one including morphological data only, and one with combined morphological and molecular data sets. The resulting strict consensus tree from the combined data is chosen for a reclassification. Three main clades are identified: Procerini trib. n., Autolytini Grube, 1850, and Epigamia gen. n. Proceraea Ehlers, 1864 and Myrianida Milne Edwards, 1845 are referred to as nomen protectum, while Scolopendra Slabber, 1781, Podonereis Blainville, 1818, Amytis Savigny, 1822, Polynice Savigny, 1822, and Nereisyllis Blainville, 1828 are considered

The Gracilariaceae comprises 242 mainly tropical species, including some with significant economic value for the food and pharmaceutical industry. Accurate taxonomy is crucial for the selection of species for cultivation. However, species identification is still challenging taxonomists. Nineteen species of Gracilariaceae have so far been referred to the Brazilian coast. We describe this diversity across approximately 32 degrees of latitude, using mitochondrial (COI-5P) and plastid (UPA and rbcL) markers in species delimitation analyses (ABGD and SPN). We provide phylogenetic (based on a concatenated three-genes tree and a taxa rich rbcL tree) and barcoding analyses from a broad and partially original collection combined with morphoanatomical data, including information resulting from analyses of type specimens and topotype material. We identified twenty-five specific entities occurring on the Brazilian coast, including four new species yet to be described. Spermatangial arrangements and aspects of thallus morphology were the most useful characters for species and genera delimitation, while characters based on the anatomy of the cystocarp overlap. Our morphological analyses demonstrate the lack of exclusive characters that justify the recently proposed Gracilariaceae infrafamilial categories, and therefore we use in this work the Gracilaria sensu lato genus concept. We provide descriptions, illustrations and a dichotomous key for the identification of species occurring in Brazil.

Torrent catfishes of the subfamily Glyptosterninae from Bhutan are examined based on morphological and molecular data. Five new species are described: Creteuchiloglanis bumdelingensis sp. nov., Exostoma mangdechhuensis sp. nov., Parachiloglanis benjii sp. nov., P. dangmechhuensis sp. nov., and P. drukyulensis sp. nov. Molecular data derived from the mitochondrial gene Cyt b and the nuclear gene RAG2 recovered relationships within Parachiloglanis and the Glyptosterninae. A dichotomous key to the Glyptosterninae of Bhutan is provided.
