Abstract The protozoans include many intracellular human pathogens. Accurate detection of these pathogens is necessary to treat the diseases. In clinical epidemiology, molecular identification of protozoan is considered a more reliable and rapid method for identification than microscopy. Among these protozoans, Cryptosporidium considered being one of the important water-borne zoonotic pathogens and a major cause of a diarrheal disease named cryptosporidiosis in humans, domestic animals, and wild animals. This study was aimed to identify Cryptosporidium in zoo felids (N= 56) belonging to different zoo of China, but accidentlly Colpodella was encountered in the zoo felids sample and phylogenetic data confirmed this unexpected amplification from fecal samples using two-step nested-PCR. Phylogenetic analysis revealed the fact about the specific primers used previously by many researchers and cross-genera amplification. We came to know that genetically sequenced amplicon gives more accurate identification of species. This study suggests more investigation on Colpodella which has been neglected previously but gains the attention of researchers after identified from humans and animals and has been known to correlate with neurological symptoms in patients.

Recordings of bat echolocation and social calls are used for many research purposes from ecological studies to taxonomy. Effective use of these relies on identification of species from the recordings, but comparative recordings or detailed call descriptions to support identification are often lacking for areas with high biodiversity. The ChiroVox website (www.chirovox.org) was created to facilitate the sharing of bat sound recordings together with their metadata, including biodiversity data and recording circumstances. To date, more than 30 researchers have contributed over 3,900 recordings of nearly 200 species, making ChiroVox the largest open-access bat call library currently available. Each recording has a unique identifier that can be cited in publications; hence the acoustic analyses are repeatable. Most of the recordings available through the website are from bats whose species identities are confirmed, so they can be used to determine species in recordings where the bats were not captured or could not be identified. We hope that with the help of the bat researcher community, the website will grow rapidly and will serve as a solid source for bat acoustic research and monitoring.

Modelling fire behaviour in forest fires is based on meteorological, topographical, and vegetation data, including species’ type. To accurately parameterise these models, an inventory of the area of analysis with the maximum spatial and temporal resolution is required. This study investigated the use of UAV-based digital aerial photogrammetry (UAV-DAP) point clouds to classify tree and shrub species in Mediterranean forests, and this information is key for the correct generation of wildfire models. In July 2020, two test sites located in the Natural Park of Sierra Calderona (eastern Spain) were analysed, registering 1036 vegetation individuals as reference data, corresponding to 11 shrub and one tree species. Meanwhile, photogrammetric flights were carried out over the test sites, using a UAV DJI Inspire 2 equipped with a Micasense RedEdge multispectral camera. Geometrical, spectral, and neighbour-based features were obtained from the resulting point cloud generated. Using these features, points belonging to tree and shrub species were classified using several machine learning methods, i.e., Decision Trees, Extra Trees, Gradient Boosting, Random Forest, and MultiLayer Perceptron. The best results were obtained using Gradient Boosting, with a mean cross-validation accuracy of 81.7% and 91.5% for test sites 1 and 2, respectively. Once the best classifier was selected, classified points were clustered based on their geometry and tested with evaluation data, and overall accuracies of 81.9% and 96.4% were obtained for test sites 1 and 2, respectively. Results showed that the use of UAV-DAP allows the classification of Mediterranean tree and shrub species. This technique opens a wide range of possibilities, including the identification of species as a first step for further extraction of structure and fuel variables as input for wildfire behaviour models.

In this paper different bioresources of the Loktak lake linked to the livelihood of the communities residing in the periphery of the lake is investigated using questionnaire survey and purposive sampling technique in 300 households from five selected villages. Identification of species and data analysis was taken up using appropriate methodologies. In all 38 type of fishes, 1 type of prawn, 2 type of mollusca, 1 type of mussel, 16 type of vegetables, 8 type of fodders, 6 type of fuelwoods, 3 type of thatch grasses, 12 type of plants with medicinal properties and 2 type of handicraft items were found used by the villagers from Loktak lake for consumption and household earning. People of the survey villages were found to be poor and depended on the lake’s resources for consumption and household financial earning. In some of these villages certain bioresources has been responded to be lost. The resources of the lake have been found degrading because of several anthropogenic activities. Suitable policies and conservation measures involving the villagers needed to be adopted urgently by the concerned authorities for the protection and long term management of the lake and its bioresources.

The identification of species based on anatomical characters is valuable to investigate their taxonomic status, phylogeny and understand their autoecology. The current study analysed the stem and leaf properties of 20 species of the subgenus Cyanus (Mill.) Hayek in Turkey. The Cyanus species exhibited xeromorphic anatomical features, such as tight palisade parenchyma and induced spongy parenchyma, which indicated adaptations to the arid soil and a forest ecosystem in which they occurred, as well as to high light intensity and precipitation. Anatomical features such as the midrib shape and number of vascularization patterns differed among the species. Similar anatomical characters were analysed for the investigated species. These properties can provide an important database for future studies including the phylogeny within the subgenus Cyanus. Bangladesh J. Plant Taxon. 28(2): 295-309, 2021 (December)

The correct identification of species is an essential step before any study on biodiversity, ecology or genetics. Keratella is a genus with a predominantly temperate distribution and with several species being endemics or restricted geographically. Its diversity may be underestimated considering the confusing taxonomy of species complexes such as K. cochlearis. In this study, we examined genetic diversity and morphology among some Keratella populations from Mexico in order to determine if these populations represent different species. We analyzed a dataset of previously published and newly generated sequences of the mitochondrial COI gene and the nuclear ITS1 marker. We conducted phylogenetic analyses and applied three methods of species delimitation (ABGD, PTP and GMYC) to identify evolutionary significant units (ESUs) equivalent to species. Morphological analyses were conducted through scanning electron microscope (SEM) and morphometry under a compound microscope. In the present study, three new species Keratella cuitzeiensis sp. nov., Keratella huapanguensis sp. nov., and Keratella albertae sp. nov., are formally described. These species were collected in high-altitude water bodies located in the Central Plateau of Mexico. Combining DNA results through COI and ITS1 molecular markers and morphology it was possible to confirm the identity of the new species.

The biodiversity of the Mediterranean Sea is currently threatened by the introduction of Non-Indigenous Species (NIS). Therefore, monitoring the distribution of NIS is of utmost importance to preserve the ecosystems. A promising approach for the identification of species and the assessment of biodiversity is the use of DNA barcoding, as well as DNA and eDNA metabarcoding. Currently, the main limitation in the use of genomic data for species identification is the incompleteness of the DNA barcode databases. In this research, we assessed the availability of DNA barcodes in the main reference libraries for the most updated inventory of 665 confirmed NIS in the Mediterranean Sea, with a special focus on the cytochrome oxidase I (COI) barcode and primers. The results of this study show that there are no barcodes for 33.18% of the species in question, and that 45.30% of the 382 species with COI barcode, have no primers publicly available. This highlights the importance of directing scientific efforts to fill the barcode gap of specific taxonomic groups in order to help in the effective application of the eDNA technique for investigating the occurrence and the distribution of NIS in the Mediterranean Sea.

The Elaeagnus L. species are trees and bushes that mainly grow in temperate zones of Western Europe; Minor, Central, and Southeast Asia; the Far East; and North America. Some species are used as fruit or ornamental plants and have economic value. Problems with the identification of species in the Elaeagnus genus by molecular genetical methods arise in the study of populations, systematics, breeding, and other areas of plant science and practice. Recently, the polymorphism of 5S ribosomal DNA non-transcribed spacers (5S rDNA NTSs) in Elaeagnaceae Adans. has been described. The results were used in our study as a basis for development of new species-specific molecular markers for some members of the Elaeagnus genus. The author’s method was applied for finding regions that were potentially applicable for species-specific primer design. As a result, some species-specific molecular markers were developed for Elaeagnus angustifolia L., E. commutata Bernh., E. pungens Thunb., and E. multiflora Thunb. These markers were tested in a range of samples and showed the presence of amplified fragments in lanes of the marked species only. Samples of other species showed no amplifications. Thus, the developed markers may be useful for the species identification of the studied Elaeagnus plants in botanical, dendrological, and genetic research (especially in a leafless period of year), as well as in breeding and hybridization experiments.

A new leptodirine leiodid beetle species belonging to the genus Leonhardia Reitter, 1901, L. solakisp. nov., from a pit in Bosnia and Herzegovina (western Balkan Peninsula) is described and diagnosed. Important morphological features of the new species are listed and photographed. The new species is endemic to the Dinarides of Bosnia and Herzegovina. A key for identification of species and subspecies of the genus Leonhardia is also provided.
