The vertebrate endocytic receptor CUBAM, consisting of three cubilin monomers complexed with a single amnionless molecule, plays a major role in protein reabsorption in the renal proximal tubule. Here, we show that Drosophila CUBAM is a tripartite complex composed of dAmnionless and two cubilin paralogues Cubilin and Cubilin-2, and that it is required for nephrocyte slit diaphragm (SD) dynamics. Loss of CUBAM-mediated endocytosis induces dramatic morphological changes in nephrocytes and promotes enlarged ingressions of the external membrane and SD mislocalisation. These phenotypes result in part from an imbalance between endocytosis, strongly impaired in CUBAM mutants, and exocytosis in these highly active cells. Noteworthy, rescuing receptor-mediated endocytosis by Megalin/LRP2 or Rab5 expression only partially restores SD-positioning in CUBAM mutants, suggesting a specific requirement of CUBAM in SD degradation and/or recycling. This finding and the reported expression of CUBAM in podocytes argue for a possible unexpected conserved role of this endocytic receptor in vertebrate SD remodelling.

The aim of this paper is to demonstrate the evaluation process to select the Aerosol Suction Machine (ASM) design to be implemented in a dental treatment practice to prevent the spread of COVID-19. There are two designs evaluated, the movable ASM and the fixed mounting ASM. The evaluation process involved four evaluation criteria: flexibility, safety level, manufacturability, and maintenance. The evaluation is conducted based on a specific requirement of a small dental clinic located in Surakarta, Indonesia. The evaluation results show that the fixed mounting is more suitable for implementing in a small dental clinic with many natural ventilation openings.

Ram Air Turbines (RAT) are used for emergency on-board power generation on aircraft and associated systems. Many studies on usage of RATs have shown promising results in terms of using RATs as a source of emergency on-board power generation. Many external podded systems on aircraft utilise RATs for self-sufficient adaptation. These pods generate their own power using RATs for their power requirements instead of depending on the mother aircraft power. Commercial cargo planes use RATs for generating emergency hydraulic power. A RAT was suggested to be used for emergency power, during failure of main alternator on a prototype aircraft. A specific requirement of the RAT was also to produce high drag for aerodynamic braking when deployed and concurrently generate electrical energy. Three models with different solidity were studied in wind tunnel at different wind speeds for suitability of this drag-energy combination. This paper presents the results of the study. Based on the results, a suitable RAT was selected for further analysis and ground trials.

ABSTRACT Yin Yang 1 (YY1) is a ubiquitous transcription factor and mammalian Polycomb Group protein (PcG) with important functions for regulating lymphocyte development and stem cell self-renewal. YY1 mediates stable PcG-dependent transcriptional repression via recruitment of PcG proteins that result in histone modifications. Many questions remain unanswered regarding how cell- and tissue-specificity is achieved by PcG proteins. Here, we demonstrate that a conditional knockout of Yy1 in the hematopoietic system results in an early T cell developmental blockage at the double negative (DN) 1 stage with reduced Notch1 signaling. There is a lineage-specific requirement for YY1 PcG function. YY1 PcG domain is required for T and B cell development but not necessary for myeloid cells. YY1 functions in early T cell development are multicomponent and involve both PcG-dependent and -independent regulations. Although YY1 promotes early T cell survival through its PcG function, its function to promote the DN1-to-DN2 transition and Notch1 expression and signaling is independent of its PcG function. Our results reveal how a ubiquitously expressed PcG protein mediates lineage-specific and context-specific functions to control early T cell development.

AbstractHuman SAGA is an essential co-activator complex that regulates gene expression by interacting with enhancer-bound activators, recruiting transcriptional machinery, and modifying chromatin near promoters. Subunit variations and the metazoan-specific requirement of SAGA in development hinted at unique structural features of the human complex. Our 2.9 Å structure of human SAGA reveals intertwined functional modules flexibly connected to a core that distinctively integrates mammalian paralogs, incorporates U2 splicing subunits, and features a unique interface between the core and the activator-binding TRRAP. Our structure sheds light on unique roles and regulation of human coactivators with implications for transcription and splicing that have relevance in genetic diseases and cancer.

AbstractNucleic acids are powerful triggers of innate immunity and can adopt the unusual Z-conformation. The p150 isoform of adenosine deaminase acting on RNA 1 (ADAR1) prevents aberrant interferon (IFN) induction and contains a Z-nucleic acid binding (Zα) domain. We report that knock-in mice bearing two point mutations in the Zα domain of ADAR1, which abolish binding to Z-form nucleic acids, spontaneously induced type I IFNs and IFN-stimulated genes (ISGs) in multiple organs. This included the lung where both stromal and haematopoietic cells displayed ISG induction in Adar1mZα/mZα mice. Concomitantly, Adar1mZα/mZα mice showed improved control of influenza A virus. The spontaneous IFN response in Adar1mZα/mZα mice required MAVS, implicating cytosolic RNA sensing. Finally, analysis of A-to-I changes revealed a specific requirement of ADAR1’s Zα domain in editing of a subset of RNAs. In summary, our results reveal that endogenous RNAs in Z-conformation have immunostimulatory potential that is curtailed by ADAR1.

Abstrak. Sistem informasi yang mengotomatiskan proses bisnis, terutama dengan persyaratan khusus masih relevan saat ini. Clarias Makmur, sebuah usaha mikro di Indonesia yang membiakkan dan menjual ikan lele menggunakan sistem informasi ini untuk menjalankan penjualan, pengeluaran, modal, dan pelaporan mereka. Penjualan ikan lele sebagai makhluk hidup memiliki ciri khas tersendiri yang menunjukkan pola musiman yang unik. Sebuah model bernama SARIMA (Seasonal Autoregressive Integrated Moving Average) kemudian diusulkan untuk memprediksi penjualan. Lebih lanjut, sistem yang disebut SITRAN dibuat online supaya perusahaan mikro ini beroperasi secara fleksibel. Ada 400 data penjualan yang digunakan dalam metode ini untuk membuat model dan prediksi, sedangkan 100 data lainnya digunakan untuk menguji akurasi metode. Hasil penelitian menunjukkan bahwa SARIMAX (21,2,0) (1,0,0,12) adalah model terbaik yang ditemukan dalam percobaan dan yang memberikan RMSE terkecil.  Kata kunci: prediksi; penjualan ikan lele; pola musiman; SARIMA Abstract. An information system that automates a business process, especially with specific requirement is still relevant these days. Clarias Makmur, a micro cooperation in Indonesia that breeds and sells catfish uses such an information system to carry out their sales, expenses, capital and reporting. The sales of catfish as a living creature have their own characteristics showing the unique seasonal pattern. A model named SARIMA (Seasonal Autoregressive Integrated Moving Average) is then proposed to predict the sales. Furthermore the system called SITRAN is made to be online for the cooperation to operate flexibly. There are 400 sales data used for the method to model and predict, while another 100 are used to test the method accuracy. The result shows that SARIMAX(21,2,0)(1,0,0,12) is the best model found in the experiment giving the smallest RMSE.  Keywords: prediction; catfish sales; seasonal pattern; SARIMA
