Background: Bacteria coordinate their behavior as a group via communication with their peers, known as ‘ quorum sensing’. Enterococcus faecalis employs quorum sensing via RNPP-peptides which were not yet reported to be present in mammalian biofluids. Results: Solid phase extraction of murine feces was performed, followed by ultra high performance liquid chromatography (UHPLC–MS/MS) in multiple reaction monitoring (MRM) mode (in total <90 min/sample) for the nine known RNPP peptides. Limits of detection ranged between 0.045 and 52 nM. Adequate identification criteria allowed detection of RNPP quorum sensing peptides in 2/20 wild-type murine feces samples (i.e., cAM373 and cOB1). Conclusion: A fit-for-purpose UHPLC–MS/MS method detected these RNPP peptides in wild-type murine feces samples.

The sound propagation speed measurement us is used for monitoring triglyceride ethanolysis in a broad range of reaction conditions (mainly, temperature: 23–50 °C; ethanol/oil: from 6 to 24 mol/mol). Experimentally, us slightly increased with the reaction time in all cases as a result of the contribution of its dynamic mixture components. Nomoto’s expression for homogeneous mixtures offered suitable us estimation but with values notably higher than the experimental ones due to the resistance to sound propagation offered by the ethanol/oil interphase (non-homogeneous medium). Our strategy was based on both the comparison of the experimental us values and the theoretical ones correlated by means of triglyceride conversion and on the estimation of the sound speed of oil/ethanol that could emulate the resistance offered by the interphase. The evolution of the reactions was predicted quite well for all the experiments carried out with very different reaction rates. Nevertheless, at the beginning of the reaction, the estimated conversion (outside of industrial interests) showed important deviations. The presence of the intermediate reaction products, diglycerides, and monoglycerides could be responsible for those deviations.

Despite the prevalence of ortho- and pyrophosphate in biochemistry, operationally simple and versatile high-throughput methodologies for their quantification are lacking. We herein introduce PUB, a module for phosphate detection by continuous UV-spectroscopic monitoring of 5-bromouridine phosphorolysis. The PUB module employs cheaply available, bench-stable reagents and can be employed for continuous and discontinuous reaction monitoring in biochemical assays to detect (pyro-)phosphate concentrations spanning almost four orders of magnitude, as demonstrated with representative use-cases.
