Streptococcus pneumoniae carriage in school children

Purpose. To investigate the serotype distribution, clonal structure and antimicrobial resistance of pneumococci isolated from schoolchildren.Materials and methods. During the period from 2012 to 2018 we examined 498 healthy school children aged 6 to 17 years. Oropharyngeal swab was taken from each child for culture, after that all S. pneumoniae strains were genotyped for serotype and ST-type deduction (PCR and sequencing, respectively). Antimicrobial resistance was also determined.Results. Pneumococcal culture was positive in 10.6 % of children. S. pneumoniae isolates belonged to seven serogroups and seven serotypes. Serogroup 6 and serotype 19F strains (15.1% each), and serogroup 9 strains (13.2%) were the most prevalent. S. pneumoniae33FA/37 and 3 (9.4 and 5.7%), serogroups 15 and 18 (7.6 and 5.7%), and 10A serotype (3.8%) were determined at a lower frequency. 20 detected ST-types belonged to 14 clonal complexes (CCs); CC156, CC447, and CC320 were predominant. 1.9% of isolates were penicillin-resistant; 13.2% – macrolide-, clindamycin-, and tetracycline-resistant. S. pneumoniae antibiotic resistant strains belonged to multidrug-resistant CCs 320, 315, and 156.Conclusion. S. pneumoniae prevalence in school children is not high. Pneumococcal population is characterized by serotype and clonal diversity including ‘invasive’ serotypes and genotypes. Most of strains are susceptible to antimicrobials.

Determination of nasal carriage and skin colonization, antimicrobial susceptibility and genetic relatedness of Staphylococcus aureus isolated from patients with atopic dermatitis in Szczecin, Poland

Background Atopic dermatitis (AD) is one of the most frequent chronic and inflammatory skin condition. AD is characterized by damaged epidermal barrier, xerosis and pruritus of eczematous skin lesions which tend to flare. The duration and frequency of exacerbation of AD symptoms markedly affects the quality of patient life. AD results from the interplay between host genetics, immunity, and environmental factors, however the detailed pathogenesis of this disease is still not entirely cleared. Furthermore, disturbances of the skin microbiota and skin functional impairment predispose to secondary skin infections. Staphylococcus aureus colonizes skin and mucous membranes of 20 to 80% of healthy individuals and of 90% of patients with AD in whom this bacterium is accounted as an important AD exacerbating factor. It is also proven, that S. aureus nasal carriage significantly increases the risk for self-transmission and endogenous infection. In the current study the presence of S. aureus either in nasal vestibule and on lesioned skin of 64 patients with AD enrolled in 10-year autovaccination program was determined. The genetic relatedness of 86 S. aureus isolated from patients nose and skin using Pulsed Field Gel Electrophoresis (PFGE) and antimicrobial susceptibility of all strains to methicillin, erythromycin, clindamycin, mupirocin, gentamicin, amikacin, tetracycline, chloramphenicol and cotrimoxazole was also evaluated. Results In total 23 PFGE genotypes and 24 unique patterns were distinguished. 34 patients were S. aureus nasal carriers. Simultaneous presence of S. aureus in nose and on affected skin was found in 16 carriers colonized by indistinguishable or potentially related S. aureus vs 2 carriers colonized with non-related S. aureus in nasal vestibule and on skin. 4 isolates were methicillin resistant (MRSA) among which 3 showed constitutive MLSB resistance phenotype and remaining one was resistant to tetracycline and chloramphenicol. In 4 isolates inducible MLSB resistance phenotype was found, one of them was additionally resistant to tetracycline. 7 S. aureus were mupirocin resistant among them 3 - isolated from one patient, were resistant simultaneously to tetracyclines and chloramphenicol. 7 strains demonstrated resistance to chloramphenicol and susceptibility to all tested antimicrobial agents. The susceptibility to gentamicin, amikacin and cotrimoxazole among all examined S. aureus was confirmed. Conclusion The obtained results indicated non-clonal structure of S. aureus circulating in AD patients. PFGE results showed the clonal-structure of vast majority of S. aureus isolated from nose and skin from nasal carriers what may prove the autoinfection in these patients. All examined patients the moderate or strong severity of AD was reported. Susceptibility to most antibiotics among isolated strains was also observed.

The molecular make up of smoldering myeloma highlights the evolutionary pathways leading to multiple myeloma

Smoldering myeloma (SMM) is associated with a high-risk of progression to myeloma (MM). We report the results of a study of 82 patients with both targeted sequencing that included a capture of the immunoglobulin and MYC regions. By comparing these results to newly diagnosed myeloma (MM) we show fewer NRAS and FAM46C mutations together with fewer adverse translocations, del(1p), del(14q), del(16q), and del(17p) in SMM consistent with their role as drivers of the transition to MM. KRAS mutations are associated with a shorter time to progression (HR 3.5 (1.5–8.1), p = 0.001). In an analysis of change in clonal structure over time we studied 53 samples from nine patients at multiple time points. Branching evolutionary patterns, novel mutations, biallelic hits in crucial tumour suppressor genes, and segmental copy number changes are key mechanisms underlying the transition to MM, which can precede progression and be used to guide early intervention strategies.

Investigation of Clonal Structure and Self-incompatibility in Japanese Endemic Snow Camellia (Camellia rusticana)

We investigated the clonal structure, self-incompatibility, and number of seeds per fruit in Camellia rusticana, a Japanese endemic species, in comparison to the closely related C. japonica. Clonal propagation was more vigorous in C. rusticana than in C. japonica and the clonal structure of C. rusticana varied among populations. C. rusticana can maintain genets for extended periods, even in harsh environments such as high-altitude areas with considerable snow accumulation, because even a single surviving genet can propagate clonally. However, sexual reproduction (i.e., reproduction by seed) is advantageous for dispersal to distant suitable habitats. An artificial crossing test revealed high self-incompatibility in C. rusticana, as observed in C. japonica. In addition, the number of seeds per fruit was lower in C. rusticana than in C. japonica. Self-incompatibility in C. rusticana may play a role in production of high-quality seed, despite low seed production.

Genetic Differentiation of Ukrainian Populations of Eusomus ovulum (Coleoptera, Curculionidae): Evidence of Multiple Hybrid Speciation

Th e clonal structure of populations of the weevil Eusomus ovulum Germar, 1824 (Coleoptera, Curculionidae) from several regions of Ukraine was analyzed. Th e signifi cant diff erentiation between populations from dif-ferent region was demonstrated. Th e hypothesis of multiple origins of the hybrid form E. ovulum from several parental species is proposed.

Genomic surveillance of Neisseria meningitidis serogroup B invasive strains: Diversity of vaccine antigen types, Brazil, 2016-2018

Background Neisseria meningitidis serogroup B remains a prominent cause of invasive meningococcal disease (IMD) in Brazil. Because two novel protein-based vaccines against serogroup B are available, the main purpose of this study was to provide data on the diversity and distribution of meningococcal vaccine antigen types circulating in Brazil. Methodology Genetic lineages, vaccine antigen types, and allele types of antimicrobial-associated resistance genes based on whole-genome sequencing of a collection of 145 Neisseria meningitidis serogroup B invasive strains recovered in Brazil from 2016 to 2018 were collected. Results A total of 11 clonal complexes (ccs) were identified among the 145 isolates, four of which were predominant, namely, cc461, cc35, cc32, and cc213, accounting for 72.0% of isolates. The most prevalent fHbp peptides were 24 (subfamily A/variant 2), 47 (subfamily A/variant 3), 1 (subfamily B/variant 1) and 45 (subfamily A/variant 3), which were predominantly associated with cc35, cc461, cc32, and cc213, respectively. The NadA peptide was detected in only 26.2% of the isolates. The most frequent NadA peptide 1 was found almost exclusively in cc32. We found seven NHBA peptides that accounted for 74.5% of isolates, and the newly described peptide 1390 was the most prevalent peptide exclusively associated with cc461. Mutated penA alleles were detected in 56.5% of the isolates, whereas no rpoB and gyrA mutant alleles were found. Conclusion During the study period, changes in the clonal structure of circulating strains were observed, without a predominance of a single hyperinvasive lineage, indicating that an epidemiologic shift has occurred that led to a diversity of vaccine antigen types in recent years in Brazil.

Effects of a spatially heterogeneous nutrient distribution on the growth of clonal wetland plants

Background Clonal plants are important in maintaining wetland ecosystems. The main growth types of clonal plants are the guerrilla and phalanx types. However, little is known about the effects of these different clonal growth types on plant plasticity in response to heterogeneous resource distribution. We compared the growth performance of clonal wetland plants exhibiting the two growth forms (guerrilla growth form: Scirpus yagara, Typha orientalis, Phragmites australis and Sparganium stoloniferum; phalanx growth form: Acorus calamus, Schoenoplectus tabernaemontani and Butomus umbellatus) grown in soil substrates that were either homogeneous or heterogeneous but had the same total amount of nutrients. Results We found that the morphological traits (plant height, ramet number, spacer diameter and length) and biomass accumulation of the guerrilla clonal plants (T. orientalis) were significantly enhanced by heterogeneity, but those of the phalanx clonal plants (A. calamus, S. tabernaemontani and B. umbellatus) were not. The results showed that the benefits of environmental heterogeneity to clonal plants may be correlated with the type of clonal structure. Conclusions Guerrilla clonal plants, which have a dispersed, flexible linear structure, are better suited to habitats with heterogeneous resources. Phalanx clonal plants, which form compact structures, are better suited to habitats with homogeneous resources. Thus, wetland clonal species with the guerrilla clonal structure benefit more from soil nutrient heterogeneity.
