Clerodendrum viscosum (CV) has been used traditionally to treat medical problems like asthma, ulcer, inflammation, pyrexia, diabetes, malaria, skin diseases, snakebite and tumor by folk practitioners. The present study evaluated the analgesic, antiinflammatory, neuropharmacological and cytotoxic properties C. viscosum (leaves) in rodents. Swiss albino mice of either sex weighing 25-30 gm and SD rats (150-180 mg) were divided into control (DW), standard (model specific) as well as test groups (n=6). Analgesic potential was evaluated using acetic acid-induced writhing and formalin induced pawlicking test. Anti-inflammatory properties were evaluated by xylene and croton oil induced ear edema test. Glucose tolerance was evaluated by OGTT in normal rats. Pentobarbital induced sleeping time test was applied to assess neuropharmacological activity. Also, Brine shrimp lethality bioassay method was employed for cytotoxicity evaluation. The alcoholic extracts showed significant antinociceptive activity in acetic acid test (p<0.01) and formalin test (p<0.05) at the dose of 1000mg/kg bw. The crude extract reduced inflammation significantly (p<0.01) in both xylene and croton oil induced ear edema test. At the dose of 1000mg/kg it increased glucose tolerance significantly (p<0.05) in normal rats. CV extract significantly (p<0.01) increased sleeping time indicating CNS depressant effect. The extract exhibited a potent cytotoxicity against brine shrimp (LC50=316.23μg/ml). C. viscosum leaves showed analgesic, antiinflammatory, hypoglycemic and CNS depressant effect against experimentally induced model mice. It also possessed cytotoxic properties and further studies are required to evaluate these effects and the potential of the plant.

Background: Clerodendrum viscosum is one of the most well-known plants in traditional practices. The study aims to explore the antidiabetic, antidiarrheal and sedative effects of methanolic extract of Clerodendrum viscosum roots (MECV) in mice. Methods: For each experiment, four groups of animals were used consisting of three mice per group. Each group was treated individually: group I treated as negative control, Group II received reference drug of each experiment, Group III and IV received MECV at 200 and 400 mg/kg body weight, respectively. Antidiabetic effect of MECV was evaluated by oral glucose tolerance test. Antidiarrheal activity was examined by the method of castor oil induced diarrhea and reduction in amount of diarrheal feces was determined. To assess the sedative activity of MECV, total sleeping time was determined in phenobarbitone induced mice. Results: MECV showed statistically significant (p<0.05) blood glucose lowering activity which was comparable to the standard drug, glibenclamide and the effect was found to be dose dependent. The extract reduced the diarrheal feces in mice by 51.85% and 65.56% at 200 and 400 mg/kg doses, respectively. The observed activity was found significant (p<0.05) in comparison to the positive control. Also, the test extract showed significant (p<0.05) sedative activity with respect to the control suggesting that the extract potentiated the phenobarbitone induced sleeping time. Conclusion: This is the first report on the therapeutic importance of methanolic extract of Clerodendrum viscosum roots in diabetes, diarrhea and sedation and thus supporting the uses of the plant in traditional medicine.

Clerodendrum viscosum is a traditionally used medicinal plant. The present study aimed to analyze a detailed cytotoxic effect of the nonpolar petroleum ether fraction (AQPEF) of leaf aqueous extract of C. viscosum Vent. (LAECV) in Allium cepa root tip cells. The LAECV was fractionated with petroleum ether and tested for A. cepa toxicity at early hours (2 and 4 h treatment) at concentrations 0, 0.050, 0.100, 0.150 and 0.200 mg mL-1. The highest aberrant cell percentage (10.45%) was scored from 0.150 mg mL-1 followed by0.100 mg mL-1 (8.75%) concentration at 4 h treated samples. The AQPEF treatment induced a significant (p<0.0001) increase in micronuclei frequency; 4.31, 5.08, 5.05 and 3.05% respectively at concentrations 0.50, 0.100, 0.150, and 0.200 mg mL-1. The highest polyploid frequency (20.14%) induced with 0.100 mg mL-1 of AQPEF at 16 h recovery. 0.150 mg mL-1 is the most effective concentration of AQPEF to decipher its activity similar to colchicine (0.150 mg mL-1). In summary, the present study indicates petroleum ether is suitable for extraction of the active phytochemicals of LAECV having cytotoxic effects on A. cepa root tip cells. The AQPEF has colchicine like micronuclei, polyploidy, and mitotic abnormality inducing potentials in A. cepa root apical meristem cells.

AbstractClerodendrum viscosum is a traditionally used medicinal plant and the earlier reports indicate its leaf aqueous extract (LAECV) contains metaphase arresting, cell cycle delay, and mitotic abnormality inducing active principles. The present study aimed to isolate pro-metaphase arresting, polyploidy, micronuclei, and mitotic abnormality inducing active principles of LAECV. The LAECV was successively fractionated as petroleum ether (PEF), chloroform (CHF), and ethyl acetate (EAF) fractions. All the extract fractions were tested for Allium cepa and Triticum aestivum root swelling and root growth inhibition analyses. The petroleum ether fraction was selected for further cytotoxicity analysis on A. cepa root tip cells and was processed for detection of the active principles through HPLC, LC-MS, GC-MS, and IR analyses. The comparative seedlings’ root growth and swelling patterns indicate the bioactive principles are effectively fractionated in PEF and GC-MS analysis revealed the presence of Clerodin (m/z 434.3), 15-hydroxy-14, 15-dihydroclerodin (m/z 452), 15-methoxy-14, 15-dihydroclerodin (m/z 466), and 14, 15-dihydroclerodin (m/z 436) with a retention time of 14.038, 14.103, 14.480 and 14.655 respectively. Thus the present study explores clerodane diterpenoids of LAECV as pro-metaphase arresting, polyploidy, micronuclei, and mitotic abnormality inducing active principles.

it is observed that the anxiolytic activity of Clerodendrum viscosum may be due to Scutellarin, Xylitol and 3-deoxy d-mannoic lactone either alone or synergistic effect of two or more compound interactions. So, for the molecular interaction analysis such as Scutellarin and Xylitol. Molecular docking with GABA glide score was presented as a method in this research.
