Colorectal cancer (CRC) is one of the leading causes of cancer-related mortality worldwide. The current TNM (Tumor, Node, and Metastasis) classification approach is suboptimal in determining the prognosis of CRC patients. The prognosis for CRC is affected by a variety of features that are present at the initial diagnosis. Herein, we performed a systematic exploration and established a novel five-panel gene signature as a prognostic and early diagnosis biomarker after performing differential gene expression analyses in five independent in silico CRCs cohort and independently validating it in one clinical cohort, using immunohistochemistry. Four genes (BDNF, PTGS2, GSK3B, and CTNNB1) were significantly upregulated and one gene (HPGD) was significantly downregulated in primary tumor tissues compared with adjacent normal tissues throughout all the five in silico datasets. The univariate CoxPH analysis yielded a five-gene signature that accurately predicted overall survival (OS) and recurrence-free survival (RFS) in the in silico training (AUC = 0.73 and 0.69, respectively) and one independent in silico validation cohort (AUC = 0.69 and 0.74, respectively). This five-gene signature demonstrated significant associations with poor OS in independent clinical validation cohorts of colon cancer (CC) patients (AUC = 0.82). Intriguingly, a risk stratification model comprising of the five-gene signature together with TNM stage and gender status achieved an even superior AUC of 0.89 in the clinical cohorts. On the other hand, the circulating mRNA expression of the upregulated four-gene signature achieved a robust AUC = 0.83 with high sensitivity and specificity as a diagnosis marker in plasma from CRC patients. We have identified a novel, five-gene signature as an independent predictor of OS, which in combination with TNM stage and gender offers an easy-to-translate and facile assay for the personalized risk-assessment in CRC patients.

We present a survey on the French research community and climate change carried out in 2020. It is one of the largest surveys ever conducted on this issue: it is based on a sample of more than 6,000 respondents representative of the French public sector research community, regardless of their status and discipline. On the one hand, it measures practices that emit large amounts of greenhouse gases, such as air travel, and addresses the differences between disciplines and within them according to different individual characteristics (gender, status, location, etc.). On the other hand, it questions the representations of research actors concerning the climate emergency, and what they are willing to do to reduce their emissions. The survey highlights three results: first, an acute awareness of environmental and climate issues widely shared by members of the scientific community; second, a willingness to implement changes; and third, a clear gap between these attitudes and practices that still emit large amounts of greenhouse gases. This raises the question of the role of research institutions, whose support is required to implement profound reforms in the organization of research activities.

Abstract: Postgenderism is a social movement about understanding the balance between men, women and nature. The development of the times and technological advances has brought people to various conditions with various attitudes to life, including a new way of looking at humans regarding gender status. This is intended as a space for human potential by eliminating gender status biologically and psychologically because it is considered an arbitrary limitation space. Postgenderism faces the limitations of social constructionist views on gender and sexuality, as well as the possibility of gender transcendence through social and political means that can be resolved by means of technology. Maqashid Syariah is here to provide a bridge of thought in viewing discourses and phenomena that move with the times. This is as a role in providing a legal footing that is oriented to the benefit of the people. In order to provide a sharp analysis, this study uses a qualitative library research with the theory of Maqashid Syariah. The method used in this research is descriptive analytical by describing a problem, and qualitative analysis with reference to the literature and applicable legal provisions. So that through the research method used, the use of Sex Reassigment Surgery technology as a re-determination of gender status can be studied using the Maqashid Syariah theory which legally has benefits or brings new problems in its implementation in social life. Keywords: Maqashid Syariah Postgender, Sex Reassigment Surgery, Technology Abstrak: Postgenderisme adalah gerakan sosial tentang memahami keseimbangan antara laki-laki, perempuan dan alam. Inequality in all its forms is common to human societies, and thus its understanding is part of the central mission of anthropology. Social structural elements such as race, class, gender, status, and caste have a decisive influence on human well-being and have been a persistent focus of inquiry over the course of more than a century of anthropological investigations. It is also fair to say that anthropologists over time have adopted as core values equal protection and equal opportunity for all peoples. These principles have been applied by extension most recently to sexual minorities, stateless refugees, and migrants.

As a white, Western-educated man, undertaking research in Kakuma refugee camp, Kenya, I encountered ethical dilemmas related to my privileged racial and gender status. These include power imbalances between researchers and refugees and conducting research in the face of human suffering. Through critical self-reflexivity, I analyze my own experiences to reveal the personal and professional vulnerabilities that researchers from high-income countries (HICs) may encounter when working in contexts where oppression and forced displacement are prominent. I conclude that researchers who work in contexts of forced migration must extend beyond the boundaries of procedural research ethics and include components of relational ethics. This involves close collaboration with refugee participants to develop more culturally relevant research ethics guidelines for refugee-specific populations.

Background: Arthroscopic drilling is a well-established treatment for stable intact OCDs of the knee in children when non-operative treatment fails. The decision to proceed to operative treatment requires a discussion regarding the risks and benefits of surgical intervention. While there is abundant data suggesting that OCD drilling is a highly effective treatment strategy, there is little data specifically evaluating the risks and complications of this procedure. Purpose: The goal of this study was to assess the complications of drilling of stable OCDs of the knee, and the risk factors for developing complications. Methods: In a retrospective chart review from 2009-2017, data from patients <18 years old who underwent arthroscopic drilling for stable intact OCD lesions of knee were collected. Lesions treated with other techniques (fixation, chondroplasty, OATS), and those with <3 months of follow-up were excluded. Characteristics of the lesions, treatment approach, and post-operative course were recorded. For bilateral surgeries, each knee was considered a separate record. Results: 139 knees in 131 patients were evaluated, of which 102 (73%) were male. The mean age was 12.7years. 108 knees (77%) had an open physis at the time of operation. Patients were managed conservatively for a mean of 6.76 months pre-operatively (SD=6.6). Average follow-up was 14.8 months after the initial surgery (SD=13.25). 53 patients (38%) were obese or overweight at the time of surgery. All patients regained full ROM within 5° of the contralateral knee at a mean 12.9 weeks post-op (SD=3.2), with all but 7 (5 %) returning to activities of daily living at the 3-month visit. No cases of infection, stiffness, arthrofibrosis, or other procedure-related complications were recorded. 6 knees (4.3%) underwent 7 additional surgeries during the follow-up period, all of which were for treatment failure in non-healing lesions (loose body removals, chondroplasty, or repeat drilling). In a multiple logistic regression model, age, gender, status of the physis at the time of surgery, BMI percentile, and OCD size were not predictive of the need for additional surgeries. Discussion: Our findings suggest that arthroscopic drilling for OCDs of the knee is a safe procedure with minimal risk of complications. The majority of patients return to their pre-operative daily activity level with full ROM by 3 months after surgery. Complications, including reoperation, were related to the progression of the OCD, rather than the surgical procedure.
