Many species are threatened with extinction, and captive breeding programs are becoming more common to avoid this outcome. These programs serve to prevent extinction and produce individuals for eventual reintroduction to natural populations in historical habitat. Captive animals experience different energetic demands than those in the wild, however, and as a result may have different levels of glucocorticoid hormones. Glucocorticoids help with responses to energetically expensive and potentially stressful situations. Elevated glucocorticoid levels can also potentially alter reproduction and other key behaviors, thus complicating successful captive breeding. The Puerto Rican parrot (Amazona vittata) is a critically endangered parrot that currently exists in only two wild and two captive populations. Its recovery program provides a good platform to better understand how glucocorticoid levels may relate to reproductive success under captive conditions. We validated a corticosterone assay in this species and used non-invasive techniques of measuring fecal glucocorticoid metabolites of males and females from two captive populations (Rio Abajo and El Yunque) of Puerto Rican parrots over two consecutive breeding seasons, 2017 and 2018, and the pre-breeding season of 2018, which occurred just after Hurricane Maria struck Puerto Rico. Our results show that levels of fecal glucocorticoid metabolites of males measured during the breeding season of 2018 negatively correlated to the number of total eggs and fertile eggs laid by pairs. In contrast, there was a positive relationship of female fecal glucocorticoid metabolite levels during the pre-breeding season of 2018 with total eggs laid. In males from the Rio Abajo population, we found seasonal differences in fecal glucocorticoid metabolite levels, with higher levels during the pre-breeding season of 2018 compared to both 2017 and 2018 breeding seasons. There was no difference in the mean value of male fecal glucocorticoid metabolites between the 2017 breeding season and 2018 breeding season which started four months after Hurricane Maria struck Puerto Rico. We did find sex differences during the pre-breeding season of 2018 in birds from the Rio Abajo population. Adjustments in the care routine of both populations that could reduce circulating baseline glucocorticoids and avoid frequent, sudden elevations of glucocorticoids should be considered. These results provide a baseline for future comparison with reintroduced populations of this endangered species and other species with captive breeding programs.

Abstract Host’s fitness can be affected by its genotype and gut microbiota, defined as the microbes living in the host’s intestinal tract. This study explored how the genetic diversity of the host influences its bacterial communities in the context of captive breeding programs, for the critically endangered European mink (Mustela lutreola). As stated by the ecosystem on a leash model, loss of host genetic diversity may lead to changes in immunomodulation and will therefore induce modifications of the gut microbiota. We investigated variation in the gut bacteria through 16S rRNA metabarcoding, related to the genetic diversity of European mink held in captivity in two breeding centers representing separate breeding stocks originating from the western and eastern populations. The genetic diversity of the host was assessed through diversity analysis of the adaptive MHC class I and II genes as well as neutral microsatellite markers. Results indicate lower diversity in neutral and MHC class I genes for the western population, and the opposite for MHC class II. A lower MHC class II gene variability led to an increase in microbial phylogenetic diversity and in abundance depending on the presence of specific MHC-II motifs. Those results seem to be linked to management practices that differs between the two programs, especially the number of generations in captivity. Long term Ex situ conservation practices can thus modulate gut microbial communities, that might potentially have consequences on the survival of reintroduced animals. We suggest strategies to foster genetic diversity in captive breeding program to mitigate the effects of genetic drift on those small, isolated populations.

Captive breeding programs are crucial to ensure the survival of endangered species and ultimately to reintroduce individuals into the wild. However, captive-bred populations can also deteriorate due to inbreeding depression and reduction of genetic variability. We genotyped a captive population of 82 individuals of the endangered Hume’s pheasant (Syrmaticus humiae, Hume 1881) at the Doi Tung Wildlife Breeding Center to assess the genetic consequences associated with captive breeding. Analysis of microsatellite loci and mitochondrial D-loop sequences reveal significantly reduced genetic differentiation and a shallow population structure. Despite the low genetic variability, no bottleneck was observed but 12 microsatellite loci were informative in reflecting probable inbreeding. These findings provide a valuable source of knowledge to maximize genetic variability and enhance the success of future conservation plans for captive and wild populations of Hume’s pheasant.

Despite a profound knowledge on reproduction biology in greater one-horned (GOH) rhinoceros, many individuals cope with sub or infertility or an-ovulatory estrous. At the same time, early and regular captive breeding is of high importance in female GOH rhinoceros due to their high prevalence to develop genital tract tumors with consequent cessation of reproduction. Thus, mature, an-ovulatory GOH rhinoceros represent a challenge for captive breeding programs and warrant for means of reliable ovulation induction. Here, we used hCG and GnRH analog histrelin to induce ovulation in an-ovulatory GOH rhinoceros. Upon ultrasound diagnosis of a preovulatory follicle hCG or GnRH were injected to induce ovulation (n = 11). As a result, 75% of the hCG (n = 6/8) and 33% of GnRH (n = 1/3) inductions resulted in ovulation. Ovulation occurred when fecal estrogen concentration increased before and pregnane concentration after induction. Thirty-six percent of all treatments (n = 4/11) failed to induce ovulation. When ovulation induction by hCG/GnRH injection failed, estrogen and pregnane concentrations were significantly lower compared to ovulatory estrous (P < 0.001). Our results suggest that hCG and GnRH analog facilitate an easily applicable treatment to induce ovulation in females with behavioral but at times an-ovulatory estrous. Frequent use of hCG as an ovulation inducer might help to achieve pregnancies in genetically important but an-ovulatory GOH rhinoceroses.

The superorder Batoidea (rays, skates, and relatives), constitutes one of the most threatened group of vertebrates. Strengthening ex situ conservation programs developed in research centers and public aquaria could be a way of addressing this situation. However, captive breeding programs must be improved to prevent the capture of wild animals and to develop proper in situ reintroduction strategies. Sperm extraction and artificial insemination are two techniques commonly used in other threatened species, which could also be used in rays and the like. However, the different reproductive morphologies present within this group of animals may hamper both processes. Here, we present a comparison of the reproductive anatomies of 11 distinct batoid species, emphasizing the important differences between the species when performing sperm extraction or artificial insemination. Both male and female animals, belonging to the Rajidae, Dasyatidae, Torpedinidae and Myliobatidae families, from the Mediterranean Sea were studied. In addition, we describe the procedure to extract sperm using both cannulation and abdominal massage, either from live or dead batoids Finally, the obtention of motile sperm recovered from the oviducal gland of females is described. These techniques generate a new range of possibilities for the conservation of these threatened species.

Modern zoos are increasingly taking a leading role in emergency management and wildlife recovery. In the face of climate change and the predicted increase in frequency and magnitude of catastrophic events, zoos provide specialised expertise to assist wildlife welfare and endangered species recovery. In the 2019–2020 Australian bushfire season, now called Australia’s Black Summer, a state government-directed response was developed, assembling specialised individuals and organisations from government, non-government organisations, research institutions, and others. Here, we detail the role of Zoos Victoria staff in wildlife triage and welfare, threatened species evacuation and recovery, media and communications, and fundraising during and after the fires. We share strategies for future resilience, readiness, and the ability to mobilise quickly in catastrophic events. The development of triage protocols, emergency response kits, emergency enclosures, and expanded and new captive breeding programs is underway, as are programs for care of staff mental health and nature-based community healing for people directly affected by the fires. We hope this account of our response to one of the greatest recent threats to Australia’s biodiversity, and steps to prepare for the future will assist other zoos and wildlife organisations around the world in preparations to help wildlife before, during, and after catastrophic events.

Captive breeding programs are widely applied by conservation practitioners as a means of conserving, reintroducing, and supplementing populations of imperilled freshwater fishes and mussels. We conducted a systematic map to provide an overview of the existing literature base on the effectiveness of captive breeding and release programs. A key finding is that there is limited evaluation of the effectiveness of such programs at all three stages (i.e., broodstock collection, rearing/release methods, and post-release monitoring). We identified clusters of evidence for supplementation evaluations related to rearing/release methods for fish growth and survival metrics, and the monitoring stage for fish genetic diversity, growth, and survival metrics, primarily focused on salmonids. However, many studies had inadequate experimental designs (i.e., lacked a comparator). Overall, there was a paucity of studies on the effectiveness of captive breeding programs for imperilled freshwater mussels, highlighting the need to make such information broadly available when studies are undertaken. Outputs from this systematic map (i.e., the map database and heatmaps) suggest that the effectiveness of captive breeding and release programs requires further systematic evaluation.

Background: Study of behavioral conformations play a significant role in ex-situ conservation of ungulates particularly of deer to propagate deer farming. Due to advancement in animal hunting techniques, captive breeding is the best solution to propagate animals for sports or other traditional & medicinal uses. Due to the lack of management methods, captive breeding programs have been used on trial and error basis for rearing of wild animals like Chinkara. Objectives: The present study was planned to investigate behavioral patterns in captivity to explore factors over reproductive success of the Chinkara and potential of this wild animal (Chinkara) for deer farming practices in Pakistan. Methodology: Present study was conducted on the behavior of Chinkara (Gazella bennettii) [wild-caught (WC)=20, captive-bred (CB)=10] for a period of one year from April 2013 to March 2014 at Captive breeding facilities for ungulates, Ravi campus Pattoki, University of Veterinary and Animal Sciences (UVAS), Pakistan. Results: The behavior of captive-born and wild-caught animals was observed by focal sampling pattern to frame conservation strategies for successful management practices for promotion of deer farming in Pakistan. Similar behavioral patterns were observed in both WC and CB animals but WC male Chinkara displayed a higher degree of agonistic interaction than of CB males. From these results it is predicted that there is no obvious immediate effect of captivity on behavioral configurations up to 10 generations in Chinkara. Conclusion: It is suggested that Chinkara is not suitable for domestication like Goat until further studies on ethology of Chinkara.
