Abstract L-3,4-dihydroxy-phenylalanine (L-dopa) is the most widely used drug in Parkinson's disease treatment. However, development of cost-effective and high-throughput sensors to accurate enantioselective discrimination of L-dopa and D-dopa remains challenging to date. Herein, on the basis of the peroxidase-mimic activity of chiral FexCuySe nanoparticles, we demonstrated a novel colorimetric sensor for determination of chiral dopa. The surface chiral ligand, L/D-histidine (L/D-His), endowed the nanozymes with enantioselectivity in catalyzing the oxidation of dopa enantiomers. According to the values of kcat/Km, the efficiency of L-His modified nanoparticle (L-FexCuySe NPs) towards L-dopa was 1.56 times higher than that of D-dopa. While, D-His can facilely reverse the preference of the nanozyme to D-dopa. On the basis of high catalytic activity and enantioselectivity of L-FexCuySe NPs in oxidation of L-dopa, the L-FexCuySe NPs based system can be utilized for detection of L-dopa. The linear ranges for L-dopa determination were 5 µM to 0.125 mM and 0.125 mM to 1 mM with a detection limit of 1.02 µM. Critically, the developed sensor has been successfully applied in the quality control of clinical used L-dopa tablets. Our work sheds light on developing simple and sensitive chiral nanomaterials-based sensors for drug analysis.

Quantitative evaluation of the antioxidant capacity of foods is of great significance for estimating food’s nutritional value and preventing oxidative changes in food. Herein, we demonstrated an easy and selective colorimetric method for the total antioxidant capacity (TAC) assay based on 3,3’,5,5’-tetramethyl-benzidine (TMB), hydrogen peroxide (H2O2) and synthetic Lycium barbarum polysaccharide-iron (III) chelate (LBPIC) with high peroxidase (POD)-like activity. The results of steady-state kinetics study showed that the Km values of LBPIC toward H2O2 and TMB were 5.54 mM and 0.16 mM, respectively. The detection parameters were optimized, and the linear interval and limit of detection (LOD) were determined to be 2–100 μM and 1.51 μM, respectively. Additionally, a subsequent study of the determination of TAC in six commercial fruit and vegetable beverages using the established method was successfully carried out. The results implied an expanded application of polysaccharide-iron (III) chelates with enzymatic activity in food antioxidant analysis and other biosensing fields.

Contemporary immunoassays commonly used in clinical diagnostics mostly utilize enzymes, such as horseradish peroxidase, for signal generation. Numerous research is dedicated to the development of artificial peroxidase-mimicking catalysts with lower cost, high activity, better operational stability, and tunable properties. Herein we synthesized hemin-loaded bovine serum albumin (BSA) nanoparticles and applied them as catalytic labels (nanozymes) in colorimetric immunoassay of anti-tetanus antibodies. Hemin is a key part of the peroxidase catalytic center, possessing peroxidase like-activity. Albumin nanoparticles were loaded with multiple hemin molecules and decorated with Streptococcal protein G. Resulting nanozymes possessed good colloidal stability and allowed for antibody detection in blood serum. The sensitivity of antibody detection was sufficient for the assessment of post-vaccination immunity.

The assessment of glutathione (GSH) levels is associated with early diagnostics and pathological analysis for various disorders. Among all kinds of techniques for detecting GSH, the colorimetric assay relying on the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) catalyzed by many nanomaterials with peroxidase-like activity attracts increasing attention owing to its outstanding merits, such as high sensitivity and high selectivity. However, the aggregation between the nanomaterials severely hinders the entrance of TMB into the “active site” of these peroxidase mimics. To address this problem, the D-amino acid incorporated nanoflowers possessing peroxidase-like activity with a diameter of 10–15 μm, TMB and H2O2 were employed to establish the detection system for determining the level of glutathione. The larger diameter size of the hybrid nanoflowers substantially averts the aggregation between them. The results confirm that the hybrid nanoflowers detection system presents a low limit of detection, wide linear range, perfect selectivity, good storage stability and desired operational stability for the detection of GSH relying on the intrinsic peroxidase-like activity and favorable mechanical stability of the hybrid nanoflowers, indicating that the hybrid nanoflowers detection system has tremendous application potential in clinical diagnosis and treatment.
