Quantitative evaluation of the antioxidant capacity of foods is of great significance for estimating food’s nutritional value and preventing oxidative changes in food. Herein, we demonstrated an easy and selective colorimetric method for the total antioxidant capacity (TAC) assay based on 3,3’,5,5’-tetramethyl-benzidine (TMB), hydrogen peroxide (H2O2) and synthetic Lycium barbarum polysaccharide-iron (III) chelate (LBPIC) with high peroxidase (POD)-like activity. The results of steady-state kinetics study showed that the Km values of LBPIC toward H2O2 and TMB were 5.54 mM and 0.16 mM, respectively. The detection parameters were optimized, and the linear interval and limit of detection (LOD) were determined to be 2–100 μM and 1.51 μM, respectively. Additionally, a subsequent study of the determination of TAC in six commercial fruit and vegetable beverages using the established method was successfully carried out. The results implied an expanded application of polysaccharide-iron (III) chelates with enzymatic activity in food antioxidant analysis and other biosensing fields.

For measuring vitamins A and E in fortified palm oil, trichloroacetic acid (TCA) through visible spectrophotometry and total antioxidant capacity (TAC) through phenanthroline method were developed. Using TCA determination method, vitamin A was directly measured at λ = 620 nm, with 92.84 % ± 0.74 accuracy, 0.79 % RSD, 2.15 IU/g limit of detection (LOD) and 7.17 IU/g limit of quantitation (LOQ). Using the mole difference calculation method of Fe2+ after Fe3+ reduction by total antioxidants and vitamin A, vitamin E content was indirectly measured. Total antioxidant capacity required for reducing Fe3+ was determined using phenanthroline method at λ = 510 nm. The temperature and reaction time for the saponification process were also evaluated. Optimum saponification was obtained at 70 ºC in 20 min with a recovery percentage of 93.81 %. Determined Vitamin E had 93.94 % ± 0.63 accuracy, 0.68 % RSD, 0.11 IU/g LOD and 0.76 IU/g LOQ.

Curculigo orchioides Gaertn. is used in traditional medicine in Vietnam. Its antioxidant potential was evaluated through DPPH radical scavenging and the total antioxidant capacity method. The data resulted from DPPH radical scavenging activity indicate that Curculigo orchioides display high activity with a low IC50 value (22.78μg/mL), approximately 1.5 times less than that of curcumin (34.34 μg/mL). The total antioxidant capacity of the extract is equivalent to 132.48 ± 1.48 mg GA/g or 264.45 ± 2.34 μmol AS/g. The composition of Curculigo orchioides, including the total phenolic, total flavonoid, polysaccharides, and triterpenoid saponins, was examined by using the colorimetric method with reagents, and their quantity is equivalent to 196.24 ± 1.45 mg GAE/g, 78.49 ± 1.78 mg QE/g, 4.34 ± 0.08 %, 47.60 ± 0.24 mg Rb1/g (Rb1: Gypenoside III), respectively. Specifically, the total triterpenoid saponins content of Curculigo orchioides has been reported for the first time.

<p><em>Curculigo orchioides</em> Gaertn. is used in traditional medicine in Vietnam. Specifically, the total saponin - triterpenoid content of <em>Curculigo orchioides</em> has been reported for the first time.</p>

Compared with other sources, potato can bring multiple nutritional benefits because it’s naturally low energy food (0.7 kcal), having high water, fiber and starch content. Even if the consummation of potatoes is in decline, it is still considered a source of valuable nutrition. Depending on the method of preparation, potatoes contain significant level of proteins and antioxidants and can offer considerable protection against cardiovascular diseases and cancer. Natural antioxidants are present under different forms in all plants, being the base source of these compounds for humans. The objective of this study was to determine the moisture content, crude protein, total antioxidant capacity and phenolic content of three assortments of potatoes (Solanum tuberosum) available on the Romanian local market (Timis County). The study was carried out on raw, unpeeled, boiled and roasted potatoes. The moisture content was determinate thermogravimetrically using Sartorius thermo balance, crude protein quantified by using a rapid colorimetric method, total antioxidant capacity determinate using CUPRAC method and total polyphenols content using Folin-Ciocalteu assay. The experimental results show that blue roasted potatoes present the highest content of crude protein, total antioxidant capacity and total polyphenols content and the lowest water content. The obtained data are used to create a graphical fingerprint of raw and processed potatoes in order to identify the best options to mix different potatoes assortments and to create innovative nutritious food products

In our present study, we performed a quick test of the antioxidant capacity of domestic fruit and vegetable cultivars with a PHOTOCHEM® instrument (Analytik Jena AG, Germany). The instrument is special because it can measure both the total lipid-soluble antioxidant capacity (ACL method) and the total water-soluble antioxidant capacity. Another advantage of the system is its quick measurement. With similar instruments, such measurement can take several hours, while PHOTOCHEM® can perform it within a few minutes.In our model studies, we examined the changes in the antioxidant capacity of sour cherry, green pepper, carrot, pumpkin, red beet and Brassica sp. cultivars. We aimed to determine how suitable the instrument is for quick, routine measurements in the case of the different horticultural products and which species have high antioxidant capacity values.On the basis of our examinations, we can state that the PHOTOCHEM® system is suitable for routine, semi-automated measurements of the total antioxidant capacity at several vegetables and fruits. Besides this, the system can be used for testing the quality of food products, for monitoring storability in post-harvest technologies, and for special breeding programmes and for physiological and biotechnological studies.

Background: Total antioxidant capacity (TAC) is one of the biomarkers to determine a person's health condition through antioxidant and oxidant activity or oxidative stress so that it can help in determining the therapy needed. However, currently, TAC levels in patients with chronic kidney disease (CKD) undergoing hemodialysis (HD) and non-hemodialysis (non-HD) are still controversial, so further research is needed. Objective: To analyze the comparison of TAC in HD and non-HD CKD patient. Material and Method: This was an analytical cross-sectional study with clinical observation in CKD patient. A total of 71 CKD patients consisting of 28 CKD HD patients and 43 non-HD CKD patients were enrolled in this study. Data about the characteristics of the subjects were taken using the anamnesis method, while data about TAC were taken using the colorimetric method with COBAS C-501 0837-19 series and 0835-13 series to measure TAC levels in the serum of CKD patients. The data that has been obtained were analyzed using independent sample t-tests. Result: TAC levels were significantly higher in CKD patients undergoing HD compared to non-HD CKD patients (p <0.05). There are data on albumin as an antioxidant and MDA as an oxidative stress biomarker that affects TAC levels. Conclusion: TAC levels found to be higher in hemodialysis CKD patient compared to non-hemodialysis CKD patient.

Abstract. Physical exercise increases free radicals production; antioxidant supplementation may improve the muscle fiber’s ability to scavenge ROS and protect muscles against exercise-induced oxidative damage. This study was designed to examine the effects of all-trans resveratrol supplementation as an antioxidant to mediate anti-oxidation and lipid per-oxidation responses to exercise in male Wistar rats. Sixty-four male Wistar rats were randomly divided into four equal number (n = 16) including training + supplement (TS), training (T), supplement (S) and control (C) group. The rats in TS and S groups received a dose of 10 mg/kg resveratrol per day via gavage. The training groups ran on a rodent treadmill 5 times per week at the speed of 10 m/min for 10 min; the speed gradually increased to 30 m/min for 60 minutes at the end of 12th week. The acute phase of exercise protocol included a speed of 25 m/min set to an inclination of 10° to the exhaustion point. Superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) activity, non-enzymatic antioxidants bilirubin, uric acid, lipid peroxidation levels (MDA) and the total antioxidant capacity (TAC) were measured after the exercise termination. The data were analyzed by using one-way ANOVA. The result showed that endurance training caused a significant increase in MDA level [4.5 ± 0.75 (C group) vs. 5.9 ± 0.41 nmol/l (T group)] whereas it decreased the total antioxidant capacity [8.5 ± 1.35 (C group) vs. 7.1 ± 0.55 mmol/l (T group)] (p = 0.001). In addition, GPx and CAT decreased but not significantly (p > 0.05). The training and t-resveratrol supplementation had no significant effect on the acute response of all variables except MDA [4.3 ± 1.4 (C group) vs. 4.0 ± 0.90 nmol/l (TS group)] (p = 0.001) and TAC [8.5 ± 0.90 (C group) vs. 6.6 ± 0.80 mmol/l (TS group)] (p = 0.004). It was concluded that resveratrol supplementation may prevent exercise-induced oxidative stress by preventing lipid peroxidation.
