Mycoplasma gallisepticum induced poultry diseases are associated with a huge economic crisis and have a considerable impact on the poultry industry worldwide. The aim of the current study was to isolate and perform molecular detection of MG circulating pathogenic strain in the commercial layer farms in the Sylhet district of Bangladesh. The entire study was conducted from January 2018 to January 2019 at three Upazilas of Sylhet district in Bangladesh. A total of 50 dead layer chickens (indicating signs of respiratory distress before death) were collected randomly from 15 different layer farms. The tissue samples, such as air sacs, trachea, and lungs, were taken from suspected dead chickens. Both cultural and PCR-based techniques were applied to identify Mycoplasma from tissue samples. The conventional PCR technique was implemented to amplify 185 bp DNA fragments for the MG. Out of 50 samples, 36% (18/50) and 70% (35/50) of MG were identified by cultural method and PCR, respectively. Based on the results of the study, it can be concluded that PCR is an easier, more sensitive, and less time-consuming method for the early diagnosis of MG in chickens, compared to cultural isolation and hence can lower the economic burden to poultry farmers caused by this disease.

AbstractThis study reports the occurrence of pneumosteum (osteohistological structure related to an avian-like air sac system) in a nanoid (5.7-m-long) saltasaurid titanosaur from Upper Cretaceous Brazil. We corroborate the hypothesis of the presence of an air sac system in titanosaurians based upon vertebral features identified through external observation and computed tomography. This is the fifth non-avian dinosaur taxon in which histological traces of air sacs have been found. We provided a detailed description of pneumatic structures from external osteology and CT scan data as a parameter for comparison with other taxa. The camellate pattern found in the vertebral centrum (ce) of this taxon and other titanosaurs shows distinct architectures. This might indicate whether cervical or lung diverticula pneumatized different elements. A cotylar internal plate of bone tissue sustains radial camellae (rad) in a condition similar to Alamosaurus and Saltasaurus. Moreover, circumferential chambers (cc) near the cotyle might be an example of convergence between diplodocoids and titanosaurs. Finally, we also register for the first time pneumatic foramina (fo) and fossae connecting camellate structures inside the neural canal in Titanosauria and the second published case in non-avian dinosaurs. The extreme pneumaticity observed in this nanoid titanosaur contrasts with previous assumptions that this feature correlates with the evolution of gigantic sizes in sauropodomorphs. This study reinforces that even small-bodied sauropod clades could present a hyperpneumatized postcranial skeleton, a character inherited from their large-bodied ancestors.

Forty five cases, 7-14 day-old chicks were studied, they showed respiratory and nervous signs with yellowish granulomatous necrotic foci on the air sacs, liver, kidneys, cerebellum and cerebrum. Swabs were taken 7 days post inoculation, culture was made on Sabouraud's dextros agar,. Aspergillus fumigatus was isolated. Diagnosis was confirmed by experimentally exposed one day-old chicks to aerosols of A. fumigatus conidia and also by microscopic demonstration of typical fruiting bodies and spores of the fungus in fresh preparations made from Sabouraud's agar. tissues from fungal-infected Examination of histological sections of cerebrum and cerebellum chicks stained with hematoxylin and eosin and PAS stains revealed granulomas lesion with a central area of necrosis containing heterophils surrounded by macrophages, gaint cells, lymphocytes and layer of fibrous tissue. It was concluded that the cases deal within this study had been infected with A. fumigataus.

Abstract The occurrence of lateral openings and pleurocoels (lateral fossae) in the corpus of the thoracic vertebrae of extant and fossil neornithine birds is reviewed, with both features having been identified as osteological correlates of the avian pulmonary system. Openings mainly occur in larger species with a high overall bone pneumatization but do not seem to serve for the passage of lung or air sac diverticula. Pleurocoels, on the other hand, are not directly related to pneumatic features and constitute a plesiomorphic trait that was widespread in Mesozoic non-neornithine birds. It is noted that an inverse correlation exists between the occurrence of pleurocoels and the pneumatization of the humerus, with pleurocoels being mainly found in extant and fossil taxa, in which the humerus is not pneumatized by diverticula of the clavicular air sac. Here it is hypothesized that pleurocoels primarily serve to increase the structural resistance of the vertebral body and were reduced multiple times in neornithine birds. In some taxa, their reduction may be related to the development of the furcula, which assists ventilation of the clavicular and cervical air sacs and may thereby contribute to the pneumatization of both, the humerus and the thoracic vertebrae. If so, Mesozoic non-neornithine birds, which had a rigid furcula with massive shafts as well as non-pneumatic humeri and pronounced pleurocoels, are likely to have differed in functional aspects of their air sac system from extant birds.

Infectious laryngotracheitis (ILT) is a severe respiratory disease, which causes high morbidity and mortality in affected birds. In our study, ILT were reported in 42 farms from nine governates over two years (2018–2020) that showed clinical signs of ILT including dyspnea, blood expectoration of, excessive lacrimation, rattling, conjunctivitis. The disease affected different chicken breeds and age groups despite vaccination with licensed and commonly used vaccines. Samples of larynx, trachea, lungs and air sacs were examined and collected for histopathological, ultrastructural, immunohistochemical examination and molecular detection. Gross examination of laryngeal and tracheal lumen revealed different types of exudate varied from catarrhal to fibrinonecotric, also pneumonia and airsacculitis were detected. Histopathological examination showed different alternation in larynx, trachea, lung and air sac as characteristic syncytial cells containing intranuclear inclusion body hanged in fibrinoheterphilic exudate that precent in laryngeal, tracheal, bronchial and parabronchial lumen and air sacs. Tracheal lesion scoring system was used to categorize the severity of lesion in different governates. Tracheal lesion score showed that 6.02%, 26.5%, 43.3% of the birds exhibited mild, moderate, and severe changes, respectively, while 24.18% of the birds exhibited very severe changes. Furthermore, severe cases were related to the Qalyubia , Fayoum then Sharkia Governorate. Moreover, immunohistochemistry was used to detect viral particles in syncytial cells, inflammatory cells beside epithelium of trachea and lung. Transmission electron microscopy enabled the detection of virus particles and demonstrated that heterophils could be infected. PCR targeting a region in the thymidine kinase gene and glycoprotein gJ gene confirmed the presence of infectious laryngotracheitis ILT virus-specific DNA. In conclusion, anatomopathological, immunohistochemical, molecular and ultrastructural findings showed increased of ILTV severity in Egypt. Larynx, trachea, lungs and air sac should be collected and examined that aid in diagnosis. Importance of good biosecurity level to be considered.

AbstractHere, the ultrastructure and development of the white patches on thorax and head of Bactrocera oleae are analysed using scanning electron microscopy, transmission electron microscopy, and fluorescence microscopy. Based on these analyses and measurements of patch reflectance spectra, we infer that white patches are due to modified air sacs under transparent cuticle. These air sacs show internal arborisations with beads in an empty space, constituting a three-dimensional photonic solid responsible for light scattering. The white patches also show UV-induced blue autofluorescence due to the air sac resilin content. To the best of our knowledge, this research describes a specialized function for air sacs and the first observation of structural color produced by tracheal structures located under transparent cuticles in insects. Sexual dimorphism in the spectral emission also lays a structural basis for further investigations on the biological role of white patches in B. oleae.

Abstract Stylops advarians Pierce (Strepsiptera: Stylopidae) is a prevalent parasite of adult Andrena milwaukeensis Graenicher (Hymenoptera: Andrenidae) in Saskatoon, Saskatchewan, Canada. By dissecting adult bees and examining histological sections, we sought to determine how neotenic females of S. advarians impact female hosts of A. milwaukeensis anatomically. Adult bees with 1–3 females of S. advarians within their gasters were compared to nonstylopised bees (control). The presence of a single female parasite inhibited development of the host’s ovaries. The bee’s foregut shifted laterally when one parasite occupied the gaster and ventrally when two or three were present, thereby reducing the crop’s expandable capacity and the amount of nectar and pollen that stylopised bees can ingest. The midgut and hindgut were less significantly affected by stylopisation. Female parasites typically occupied the host’s gaster dorsolaterally, where each was supported by one of the host’s air sacs. If a third female parasite was present, she resided dorsally along the midline of the host’s gaster, mostly supported by the two female parasites to either side. Asynchronous development within neotenic female parasites was demonstrated, wherein mature first-instar larvae occupied the cephalothorax and abdomen at the same time that the adult female was still supporting multiple embryos.

Aspergillosis is important in poultry production and human health but difficulty in its ante mortem diagnosis in addition to the  unsuccessful outcome of most management options has made the disease a lingering nightmare to farmers. This study presents the clinicopathological features and management of aspergillosis in some poultry farms in Jos metropolis, Nigeria. Thirteen cases of  aspergillosis from 12 poultry farms were confirmed and managed from April, 2019 to March, 2020 with 7.6% cases in a mixed turkey-broiler farm, 15.4% in broilers and 76.9% in layers. The ages affected ranged from 3 weeks to 21 weeks with 69.2% cases in pullet grower birds of 10 – 21 weeks compared to young chicks, 3 – 9 weeks with 30.8%. The rainy season accounted for 61.8% cases while mortality ranged from 1.0% to 17.4%. The clinical features were consistent and included dyspnoea; inappetence; retarded growth and mortality. While necropsy showed consolidated lungs with caseous nodules; liver with raised grayish or yellowish irregular nodules; peritonitis with the peritoneum and air sacs laced with caseous nodules; diphtheritic membrane and caseous nodules on intestinal mucosa; haemorrhagic bursitis with granulomata; caseous nodules on osseous tissues as well as nodules on the skin and skeletal muscles. Confirmatory diagnoses of  aspergillosis were made by culturing nodules on Sabouraud’s Dextrose Agar. Antifungal susceptibility testing showed CuSO4 to be more susceptible which was used at the dosage of 3g/10L of drinking water for at least 7 days in all the cases. Concurrent bacterial infections were seen in some of the cases and were treated concurrently with antibiotics based on susceptibility testing while farmers using moldy feeds were asked to withdraw such feeds. Conclusively, aspergillosis is prevalent in Jos metropolis with severe impact on production and  may be a silent killer disease in poultry due to constraints in diagnosis and its management.
