Frustration is a negative emotional state implicated in a range of canine behaviour problems. The Canine Frustration Questionnaire (CFQ) is the first psychometric tool developed to assess frustration tendencies in dogs based on owner report. However, to date, no published studies have assessed behavioural and physiological correlates of this trait. A novel behaviour test battery was developed to induce frustration in dogs, mapping onto the CFQ. Forty-four dogs were recruited and filmed whilst undertaking the test battery, and a CFQ was completed by each owner. Targeted behavioural measures were assessed from this footage, based on hypotheses aimed at evaluating convergent and discriminant validity with facets of the CFQ. In addition, a saliva sample was collected pre- and post-testing for 39 dogs, and a cortisol assay performed using ELISA to provide a physiological measure of arousal. A range of predicted behavioural test measures (e.g., vocalising and lunging) positively correlated with CFQ scores. For 22 dogs with pre-test salivary cortisol levels of <4 ng/mL (indicative of normal arousal at baseline), cortisol change and post-test cortisol levels positively correlated with the CFQ PC5 ‘Frustration coping’ score. These results provide further evidence of the validity of frustration tendencies as measured by owner report through the CFQ.

<p>Background: MDMA preferentially releases serotonin (5HT) but following repeated exposure there is a decrease in this MDMA-produced effect. At the same time, some studies suggest an increase in MDMA-produced dopamine (DA) release following repeated exposure. The sensitised DA response is often accompanied by sensitisation of MDMA-produced locomotor activity. Because DAergic mechanisms have been implicated in the positively reinforcing properties of MDMA, these neuroadaptations might be relevant to MDMA self-administration.  Objectives: The main objective of this study was to determine whether MDMA self-administration and non-contingent MDMA exposure differentially affected the development of sensitisation to MDMA-produced hyperactivity. Additionally, the relationship between MDMA-produced hyperactivity and changes in c-fos expression in DA terminal regions was determined.  Methods: Triads of rats were designated ‘master’, ‘yoked MDMA’, or ‘yoked saline’. Lever press responding by the master rat resulted in an intravenous infusion of MDMA for both the master rat and the yoked MDMA rat, as well as an equal infusion of vehicle for the yoked control rat. Daily tests continued until a total of 350 mg/kg MDMA had been self-administered. Three days following the last self-administration session, forward and vertical locomotion produced by MDMA (5.0 mg/kg, i.p) were measured during a 2 hr test. Rats were sacrificed immediately following the behavioural test, and c-fos immunohistochemistry was measured.  Results: Repeated MDMA exposure resulted in sensitised forward and vertical locomotor activity. Sensitisation of the increase in forward locomotion was produced only in rats that self-administered MDMA; non-contingent MDMA administration failed to sensitise this behavioural response. In contrast, sensitisation to MDMA-produced vertical activity was produced following both contingent and non-contingent MDMA exposure. C-fos expression was reduced in ventrolateral, and ventromedial areas of the dorsal striatum, as well as the infralimbic cortex, after MDMA exposure, regardless of whether the exposure was via self-administration or yoked administration. A selective decrease in c-fos expression in the nucleus accumbens (NAc) core and the cingulate cortex was produced by MDMA self-administration. There was a negative correlation between MDMA-produced forward locomotor activity and MDMA-produced c-fos expression in the NAc core, cingulate cortex and infralimbic cortex. A negative correlation between rearing activity and MDMA-produced c-fos expression in the NAc core, NAc shell, cingulate cortex, and infralimbic cortex was also found.  Conclusions: These data provide evidence of behavioural sensitisation as a result of repeated MDMA exposure. Furthermore, MDMA-produced behavioural sensitisation was associated with a decrease in c-fos expression that was evident in the NAc and prefrontal cortex. Finally, region-specific changes in c-fos expression suggest an important role of neuroadaptations in the NAc core and the infralimbic cortex as a consequence of MDMA self-administration.</p>

Laboratory animals often develop abnormal repetitive (stereotypic) behaviour that can influence both physiology and behavioural test results. Such abnormal behaviours usually develop in suboptimal environment and increase over time. To explore the development of stereotypic forms of activity night home-cage behaviour of laboratory mice was analysed and collated with the behaviour in open field (OFT), hole-board (HBT) and tail suspension (TST) tests twice within 16 weeks. Mice expressed few stereotypies and their duration lessened over time from median 8.2 to 1.0 %. In contrast, grooming behaviour increased significantly from 29.5 to 49.6 %. Home-cage grooming correlated with the latency of locomotion start in OFT and with the immobility time in TST. Intensity and stability of stereotypic activity and grooming inf­luenced the duration of grooming in OFT: the mice with unstable stereotypies groomed more than others, and mice with the lowes home-cage grooming level also groomed in OFT the most. Intensity and stability of night grooming influenced the behaviour in TST: the mice with unstable level of grooming were the most mobile in this test. Abnormal home-cage activity may indicate impaired welfare, and that, in turn, may affect test activity, so researchers need to keep it in mind when planning animal behaviour experiments.

Blowflies are vectors of microorganisms and human pathogens, and their maggots cause myiasis in vertebrates and infest and spoil meat and fish products. Essential oils (EOs) from spices were proven to be a safer and more sustainable alternative to synthetic insecticides for the control of blowflies and are suitable for protecting food from such pests. However, some EOs are not acceptable for environmental or topical applications due to their strong, unpleasant odour. In this study, we measured the acute toxicity and the repellence of two EOs extracted from the Andean plants Aloysia citrodora Palau and Bursera graveolens (Kunth) Triana and Planch., both known for their pleasant odour, against the blue blowfly Calliphora vomitoria (L.) (Diptera: Calliphoridae). We also compared their bioactivity with that of the Allium sativum L. EO, which is very effective but bad-smelling. The A. citrodora EO was mainly rich in oxygenated monoterpenes, the most abundant of which were geranial (26.8%) and neral (21.0%). The B. graveolens EO was chiefly composed of monoterpene hydrocarbons, mostly represented by limonene (46.2%). According to the sensory description, the best odour profile was associated with the A. citrodora EO, while the olfactory expression of the EO from B. graveolens was negatively affected by a strong odour of “mouldy lemon”. The behavioural test showed that the A. citrodora EO was more repellent than that of A. sativum and, on the contrary, that the B. graveolens EO was attractive. The toxicity tests showed that the EOs of both Andean plants have a clear toxic effect on the C. vomitoria eggs and adults. In terms of ovicidal activity, there were no significant differences among the effects of the three tested EOs. On the adult flies, the toxicity of the two EOs of the Andean plants was much lower than that of A. sativum (LC50 fumigation = 1.86 μL EO L−1 air; LC50 ingestion = 8.10 μL EO mL−1 gel) both by fumigation (LC50 = 23.66 and 25.30 μL EO L−1 air for A. citrodora and B. graveolens, respectively) and ingestion (LC50 = 36.65 and 44.97 μL EO mL−1 gel for A. citrodora and B. graveolens, respectively), while, by contact, the A. citrodora EO (LD50 = 0.27 μL EO insect−1) was more toxic than the A. sativum EO (LD50 = 0.46 μL EO insect−1).

Horses in a herd develop and maintain a dominance hierarchy between all individuals. There are many situations in riding facilities and studs in which horses have to be separated out of a group. The aim of the study was to determine the rate of behaviours, level of locomotor activity and cardiac activity variables in a herd of horses during a short social separation of individuals differently ranked in the dominance hierarchy. Twelve adult Arabian mares were involved. A behavioural test had been performed before the main experiment to determine the rank order of the mares in this social herd. Three tests were performed when a dominant, mixed and submissive three-member group of mares was separated for 10 min. The response of the remaining herd was determined by a rate of behaviours, time of locomotor activity and cardiac parameters. The results of the experiment reveal evident changes towards emotional arousal in the social herd elicited by a short separation of some conspecifics. The herd created by humans preserves the sensitivity to a temporary loss of its members. The response of the remaining herd does not depend strictly on the composition of the separated mares regarding their rank in the dominance hierarchy.

Regulation EC 1/2005 has stricter rules for transportation of unbroken (untamed) vs. broken (tamed) horses, but does not provide adequate tools for their identification. This study aimed to develop and validate such a tool. A behavioural test (Broken/Unbroken Test (BUT)) based on approaching, haltering, and leading was applied to 100 horses. Physiological and additional behavioural data were also collected, and the horses’ status (broken/unbroken) was assessed by the expert who administered the BUT. Each horse’s behaviour during the BUT was scored by four trained observers blinded to the horse’s history. The BUT score showed excellent inter-observer, intra-observer, and test–retest reliability (all intraclass correlation coefficients (ICCs) > 0.75). It was also negatively associated with respiratory rate, avoidance distance, and time needed to approach, halter, and lead the horse (p < 0.05 for all). The optimal BUT score cut-off for discrimination between broken and unbroken horses (gold standard: expert judgment) showed 97.8% sensitivity and 97.3% specificity. There was almost perfect agreement between BUT-based and expert classification of horses (ICC = 0.940). These findings confirm the BUT’s construct and criterion validity. The BUT could provide officials with a feasible, reliable, and valid tool to identify a horse’s broken/unbroken status and, consequently, direct stakeholders towards correct transport procedures.

Drosophila melanogaster is a candidate species to replace rodents in some neurobiological studies, encouraging attempts to develop behavioural tests for these flies. This study aimed to develop a behavioural test to simultaneously evaluate ethological (categorical) aspects of the motor and fluid intake activities, which may be used to assess sucrose preference in flies. For that, a lane-maze was 3D-printed to accommodate up to 14 individual flies in a single trial. Each lane had a capillary filled with 5% sucrose solution attached to one of the extremities. To validate a 5-min lane-maze test, male and female flies (adults, 5-6 days of age) underwent 0, 2, 8 or 20 h of food deprivation (FD, n=9-11/group) before testing. Duration of locomotion, immobility and grooming in the lane or capillary were scored from the video-recorded trials using EthoWatcher software. Minor effects of sex or FD were observed in the behaviours of flies. Independent of sex or FD, flies spent proportionally longer on the capillary than on the lane. Flies exhibited a significantly higher preference than expected for the capillary zone when food-deprived for 2h (males) or 20 h (females). Data suggest that short lane-maze test is a feasibly high throughput assessment of sucrose preference in flies, which may be sexually dimorphic as in other species studied so far.

AbstractPrimate colour vision depends on a matrix of photoreceptors, a neuronal post receptoral structure and a combination of genes that culminate in different sensitivity through the visual spectrum. Along with a common cone opsin gene for short wavelengths (sws1), Neotropical primates (Platyrrhini) have only one cone opsin gene for medium-long wavelengths (mws/lws) per X chromosome while Paleotropical primates (Catarrhini), including humans, have two active genes. Therefore, while female platyrrhines may be trichromats, males are always dichromats. The genus Alouatta is inferred to be an exception to this rule, as electrophysiological, behavioural and molecular analyses indicated a potential for male trichromacy in this genus. However, it is very important to ascertain by a combination of genetic and behavioural analyses whether this potential translates in terms of colour discrimination capability. We evaluated two howler monkeys (Alouatta spp.), one male A. caraya and one female A. seniculus, using a combination of genetic analysis of the opsin gene sequences and a behavioral colour discrimination test not previously used in this genus. Both individuals completed the behavioural test with performances typical of trichromatic colour vision and the genetic analysis of the sws1, mws, and lws opsin genes revealed three different opsin sequences in both subjects. These results are consistent with uniform trichromacy in both male and female, with presumed spectral sensitivity peaks similar to Catarrhini, at ~ 430 nm, 532 nm, and 563 nm for S-, M- and L-cones, respectively.
