The glucotoxicity caused by long-term exposure of β-cells to high glucose (HG) conditions may lead to the generation of more reactive oxygen species (ROS), reduce the activity of antioxidant enzymes, cause cell damage and apoptosis, and induce insulin secretion dysfunction. Siegesbeckia orientalis linne is a traditional folk herbal medicine used to treat snake bites, rheumatoid arthritis, allergies, and immune deficiencies. In this study, we evaluated the protective effect of S. orientalis ethanol extract (SOE) on cell death and oxidative stress in RIN-m5f pancreatic β-cells stimulated by two HG concentrations (50–100 mM). In the cell viability assay, SOE could significantly increase the survival rate of pancreatic β-cells under HG-induced conditions. For the oxidative stress induced by HG condition, the treatment of SOE effectively reduced the ROS formation, increased the content of intracellular glutathione, and up-regulated the expression of antioxidant enzymes, catalase, superoxide dismutase, and glutathione peroxidase. As a result, the SOE treatment could decrease the glucotoxicity-mediated oxidative damage on RIN-m5F β-cells. Moreover, SOE had the function of regulating insulin secretion in pancreatic β-cells under different HG-mediated conditions. It could decrease the increasing intracellular insulin secretion under the low glucose concentration to normal level; while increase the decreasing intracellular insulin secretion under the relatively high glucose concentration to normal level. Taken together, this study suggests that SOE has a protective effect on pancreatic β-cells under the HG-stimulated glucotoxic environment.

Four new acyclic diterpenes, siegetalises A-D (1-4), were isolated from the aerial parts of Siegesbeckia orientalis. Their chemical structures were elucidated by extensive analysis of high-resolution electrospray ionization mass spectrometry and nuclear magnetic resonance spectral data. The effects of the isolated compounds on the activity of xanthine oxidase were evaluated by the oxidative reaction with xanthine as a substrate. At a concentration of 50 µM, compounds 1-4 exhibited xanthine oxidase inhibitory activity at levels of 13.59% ± 0.51%, 19.64% ± 1.54%, 17.45% ± 1.26%, and 21.36% ± 1.40%, respectively.

Siegesbeckia orientalis L. (SO) is a commonly used Chinese medicinal herb. It has long been used as a remedy in traditional Chinese medicine (TCM) for symptoms that resemble inflammatory joint disorders. However, it is slightly toxic. According to the TCM theory, processing can reduce the toxicity of the herbs. Here, we performed metabolomics to determine whether processing with rice wine reduces the toxicity of raw SO, and to explore the mechanisms underlying the raw SO–induced toxicity and the toxicity-reducing effect of processing. Our results showed that raw SO has long-term toxicity in rats. It significantly elevated the serum level of LDH and caused histopathological damages in the lung tissues. It is worth noting that the LDH level in the PSO group was lower than that in the raw SO group, and the damages in lung tissues were relatively mild in PSO-treated rats, suggesting that processing reduces the pulmonary toxicity of the raw. Moreover, a total of 32 significantly changed metabolites were identified. Based on the MetaboAnalyst pathway analysis, we found that two characteristic metabolic pathways including alanine, aspartate and glutamate metabolism and glycerophospholipid metabolism were only changed in the raw SO group, while histidine metabolism was only changed in the PSO group, which suggests that induction of oxidative stress contributes to raw SO–induced pulmonary toxicity, and free radical scavenging might be responsible for the toxicity-reducing effect of processing. Our data shed new light on how raw SO induces pulmonary toxicity and how the toxicity can be reduced by processing. This study not only provides scientific justifications for the traditional processing theory of SO, but also helps to optimize the processing protocol and the clinical drug combination of SO.

Weed commonly called ‘khar- kabad’ in Uttarakhand or ‘kharpatvar’ in India, and is one of the major biological constraints that limits crop productivity. The present communication pertains to survey and inventory of weed flora in Agastyamuni block of district Rudraprayag, Uttarakhand. The study was based on extensive and intensive field survey made during July 2018 to October 2019. During the study period the authors have reported a total 312 species belonging to 188 genera and 54 families from dicots, monocots and pteridophyta. Asteraceae was found to be the most dominant family followed by Poaceae, Lamiaceae and Fabaceae respectively. Survey results also revealed that most of the recorded species were annuals followed by perennials and biennials. Most abundant species were Bidense pilosa, Chenopodium album, Erigeron canadensis, Cynodon dactylon, Gallinsogo parviflora, Eupatorium adenophorum, Oxalis conrniculata, Parthenium hysterophorus, Lantana camara, Soncchus arvensis, Ageratum conozoides, Plantago major, Ganaphallium lutealbum, Siegesbeckia orientalis, Youngia japonica, Amaranthus virids, Stellaria media and Phalaris minor. Many weeds are ethnobotanically important and utilized by the local community.
