The last century has witnessed an increasing rate of new disease emergence across the world leading to permanent loss of biodiversity. Perkinsea is a microeukaryotic parasitic phylum composed of four main lineages of parasitic protists with broad host ranges. Some of them represent major ecological and economical threats because of their geographically invasive ability and pathogenicity (leading to mortality events). In marine environments, three lineages are currently described, the Parviluciferaceae, the Perkinsidae, and the Xcellidae, infecting, respectively, dinoflagellates, mollusks, and fish. In contrast, only one lineage is officially described in freshwater environments: the severe Perkinsea infectious agent infecting frog tadpoles. The advent of high-throughput sequencing methods, mainly based on 18S rRNA assays, showed that Perkinsea is far more diverse than the previously four described lineages especially in freshwater environments. Indeed, some lineages could be parasites of green microalgae, but a formal nature of the interaction needs to be explored. Hence, to date, most of the newly described aquatic clusters are only defined by their environmental sequences and are still not (yet) associated with any host. The unveiling of this microbial black box presents a multitude of research challenges to understand their ecological roles and ultimately to prevent their most negative impacts. This review summarizes the biological and ecological traits of Perkinsea—their diversity, life cycle, host preferences, pathogenicity, and highlights their diversity and ubiquity in association with a wide range of hosts.

Background. Surgical treatment of patients with talus posttraumatic aseptic necrosis and its consequences usually includes tibiotalocalcaneal arthrodesis with various foot joints according to additional indications. This type of surgical treatment has number of significant disadvantages: traumatic surgical technique, permanent loss of movement in functionally significant joints, high risk of non-union, high frequency of residual deformities, the need for long periods of limb immobilization. The question arises: how to overcome the existing disadvantages and improve the results of talus posttraumatic aseptic necrosis treatment? A potential solution to this problem is the total talus endoprosthetics. Clinical case. A 64-year-old patient came to the clinic complaining of pain and deformity of the right foot and ankle area. After the examination, talus posttraumatic aseptic necrosis was diagnosed. The patient underwent ankle joint arthroplasty using total talus ceramic endoprosthesis in combination with the tibial component of the ankle joint endoprosthesis, a course of rehabilitation treatment was performed. Results. The VAS and AOFAS scales indicators showed a significant improvement both in the pain decrease (from 75 mm before surgery to 10 mm after), and in the functional state according to AOFAS by 2.2 times (from 36 to 80 points 20 months after surgery). By the last follow-up the patient could take more than 8000 steps a day. Conclusion. Considering the good clinical result achieved, the ankle joint arthroplasty using total talus ceramic endoprosthesis in combination with the tibial component of the ankle joint endoprosthesis can be considered a promising method of treatment of this severe pathology.

Background: Low-income earners are particularly vulnerable to mental health, consequence of the coronavirus disease 2019 (COVID-19) lockdown restrictions, due to a temporary or permanent loss of income and livelihood, coupled with government-enforced measures of social distancing. This study evaluates the mental health status among low-income earners in southwestern Uganda during the first total COVID-19 lockdown in Uganda.Methods: A cross-sectional descriptive study was undertaken amongst earners whose income falls below the poverty threshold. Two hundred and fifty-three (n = 253) male and female low-income earners between the ages of 18 and 60 years of age were recruited to the study. Modified generalized anxiety disorder (GAD-7), Spielberger's State-Trait Anger Expression Inventory-2 (STAXI-2), and Beck Depression Inventory (BDI) tools as appropriate were used to assess anxiety, anger, and depression respectively among our respondents.Results: Severe anxiety (68.8%) followed by moderate depression (60.5%) and moderate anger (56.9%) were the most common mental health challenges experienced by low-income earners in Bushenyi district. Awareness of mental healthcare increased with the age of respondents in both males and females. A linear relationship was observed with age and depression (r = 0.154, P = 0.014) while positive correlations were observed between anxiety and anger (r = 0.254, P < 0.001); anxiety and depression (r = 0.153, P = 0.015) and anger and depression (r = 0.153, P = 0.015).Conclusion: The study shows the importance of mental health awareness in low resource settings during the current COVID-19 pandemic. Females were identified as persons at risk to mental depression, while anger was highest amongst young males.

Groundwater wells are widely used in the energy sector, including for drinking water supplies and as water source wells in the oil and gas industry to increase production of natural gas and petroleum. Water well clogging, which can happen to any well for various reasons, is a serious problem that can lead to increased power costs due to a higher head to the pump, a reduction in the flow rate and various drawdown issues. If rehabilitation procedures do not take place in time, this can result in permanent loss of the well, and a new well must be drilled, which is not a sustainable approach. Rehabilitation methods for water wells usually include mechanical and chemical treatments, and even though these methods are well established and have been used for many years we can still observe many abandoned wells which could be rehabilitated. In this study, sets of cavitation generators are developed and used in combination with common conic hydrodynamic nozzles. This combination reduces the pressure in the system and makes the cleaning setup much lighter and more mobile. The designed nozzles were successfully used in hydrodynamic cleaning of four water wells.

Spinal cord injury (SCI) is an incurable condition in which the brain is disconnected partially or completely from the periphery. Mainly, SCIs are traumatic and are due to traffic, domestic or sport accidents. To date, SCIs are incurable and, most of the time, leave the patients with a permanent loss of sensitive and motor functions. Therefore, for several decades, researchers have tried to develop treatments to cure SCI. Among them, recently, our lab has demonstrated that, in mice, repetitive trans-spinal magnetic stimulation (rTSMS) can, after SCI, modulate the lesion scar and can induce functional locomotor recovery non-invasively. These results are promising; however, before we can translate them to humans, it is important to reproduce them in a more clinically relevant model. Indeed, SCIs do not lead to the same cellular events in mice and humans. In particular, SCIs in humans induce the formation of cystic cavities. That is why we propose here to validate the effects of rTSMS in a rat animal model in which SCI leads to the formation of cystic cavities after penetrating and contusive SCI. To do so, several techniques, including immunohistochemical, behavioral and MRI, were performed. Our results demonstrate that rTSMS, in both SCI models, modulates the lesion scar by decreasing the formation of cystic cavities and by improving axonal survival. Moreover, rTSMS, in both models, enhances functional locomotor recovery. Altogether, our study describes that rTSMS exerts positive effects after SCI in rats. This study is a further step towards the use of this treatment in humans.

Reptile euthanasia techniques are poorly described. This non-blinded randomized study compared pentobarbital, lidocaine, and potassium chloride (KCl) for leopard geckos (Eublepharis macularius) euthanasia at 20° Celsius and evaluated for post-mortem artifacts. Fifty-three non-clinical adult leopard gecko were scheduled for euthanasia due to Cryptosporidium varanii exposure. Based on pilot study results, geckos randomly received one of six treatments: intracoelomic (ICo) pentobarbital 400mg/kg (ICo-P4, n = 9) or 800mg/kg (ICo-P8, n = 9), intramuscular (IM) alfaxalone 20mg/kg followed by intracardiac (ICa) pentobarbital 800mg/kg (ICa-P, n = 9), KCl 10mEq/kg (ICa-K, n = 9), or lidocaine 100mg/kg (ICa-L, n = 9) or IM alfaxalone 20mg/kg (IM-A, n = 8, control). Serial Doppler heart rate assessments were performed until cardiac arrest, 30 minutes (ICa groups), or 60 minutes (ICo groups). Cardiac arrest occurred in 7/9, 9/9, 9/9, 8/9, and 8/9 geckos in ICo-P4, ICo-P8, ICa-P, ICa-K, and ICa-L, respectively, with median (range) times of 35 (25 - 45), 30 (15-60), 0 (0 - 6.33), 0 (0 - 0.05), and 0 (0 - 0.03) minutes. Side effects were noted in a subset of ICo-P (arched posture) and ICa-K (muscle spasms) geckos. Six geckos in ICa-L had Doppler sound resumption several hours following cessation, with three displaying spontaneous movement. All geckos in IM-A recovered. Intracardiac pentobarbital following IM alfaxalone caused rapid, permanent loss of heartbeat in all tested geckos. Intracoelomic pentobarbital was also effective but had a prolonged time to cardiac arrest. Intracardiac lidocaine is not recommended for leopard gecko euthanasia.

Peripheral anterior synechiae (PAS) after corneal transplantation leads to refractory glaucoma and permanent loss of vision. However, the exact mechanism remains elusive. This study aimed to evaluate the association between cytokine levels in the aqueous humor (AqH) and the progression of PAS after penetrating keratoplasty (PKP). We measured 20 cytokine levels in AqH and assessed the correlation with PAS progression after PKP in 85 consecutive patients who underwent PKP. We also evaluated age-dependent alterations in PAS and cytokine levels in DBA2J mice. PAS developed in 38 (44.7%) of 85 eyes after PKP. The incidence of intraocular pressure increase after PKP was significantly greater in eyes with PAS (26.3%) than in those without PAS (2%, p = 0.0009). The PAS area at 12 months after PKP was significantly positively correlated with the preoperative levels of interleukin (IL)-6, interferon (IFN)-γ and monocyte chemotactic protein (MCP)-1 (p ≤ 0.049). In the DBA2J mice, an experimental glaucoma model that developed PAS at 50 weeks, the AqH levels of IL-2, IL-6, IL-10, IFN-γ, tumor necrosis factor-α, MCP-1 and granulocyte-macrophage colony-stimulating factor (GM-CSF) significantly increased at 50 weeks compared to 8 weeks (p ≤ 0.021). In conclusion, inflammatory alterations in the AqH microenvironment, such as high preoperative specific cytokine levels, can lead to PAS formation and glaucoma.

Mining of coal (fossil fuel) resources in Enugu resulted in groundwater pollution/depletion and left the mine site with the potentials of air pollution, loss of landscape/aesthetic degradation and soil contamination. Other Environmental impacts were extensive soil damage, alteration in microbial communities and affecting vegetation leading to destruction of vast amounts of land and displacement of dwellers. Reclamation is the process to restore the ecological integrity of these disturbed mine land areas. It includes the management of all types of physical, chemical and biological disturbances of soils such as soil pH, fertility, microbial community and various soil nutrient cycles that makes the degraded land soil productive. Mining does not mean permanent loss of land for other use. On the other hand it holds potential for altered and improved use apart from restoring for agriculture, forestry and irrigation. This paper attempts to view the best practices for reclaiming the abandoned Enugu coal mine site which ceased production since 2002.

Abstract: Diabetic retinopathy is the leading cause of blindness in diabetic patients. Screening of diabetic retinopathy using fundus image is the most effective way. As the time increases this DR leads to permanent loss of vision. At present, Diabetic retinopathy is still being treated by hand by an ophthalmologist which is a time-consuming process. Computer aided and fully automatic diagnosis of DR plays an important role in now a day. Data-set containing a collection of fundus images of different severity scale is used to analyze the fundus image of DR patients. Here the deep neural network model is trained by using this fundus image and five-degree classification task is performed. We were able to produce an sensitivity of 90%.
