The role of MT1-MMP in the progression and metastasis of osteosarcoma

The dysregulation of Membrane - type 1 matrix metalloproteinase (MT1-MMP) has been extensively studied in numerous cancer types, and plays key roles in angiogenesis, cancer progression, and metastasis. MT1-MMP is a predictor of poor prognosis in osteosarcoma (OS), yet the molecular mechanisms of disease progression are unclear. This review provides a summary of the literature relating to the gene and protein expression of MT1-MMP (MMP-14) in OS clinical samples, evaluates the expression in cell lines and experimental models, and analyses its potential role in the progression and metastasis of OS. In addition, the therapeutic potential of MT1-MMP as a drug target has been assessed. Due to the biological complexity of MMPs, inhibition has proven to be challenging. However, exploiting the expression and proteolytic capacity of MT1-MMP could open new avenues in the search for novel, safer and selective drugs for use in OS.

Creation of Sealed Strong Structures of Rocket and Space Equipment FDM Printing Methods by Ultem ™ 9085 Pei Plastic

The work shows the possibility of manufacturing products for rocket and space technology using the additive FDM-printing technology. The object of research is the nozzle plugs of the "membrane" type. Considering the specific-strength properties of the product during its operation during operation, as well as the features of the FDM-process, the design was optimized, the regularities of the formation of its properties were established. An impregnation technology has been developed to seal the product. The equipment was designed, and pneumatic tests were carried out. The properties of materials were investigated considering their guaranteed shelf life for 12 years of operation under accelerated climatic tests. It is shown that the production of products by FDM-printing methods is promising and expedient, since the properties being formed are predictable, achievable, and stable.

Application of Waste Glycerol as a Draw Solution for Forward Osmosis

Waste glycerol generated during biofuel production accounts for ~10% of total biodiesel volume. Increasing the use of renewable energy sources, including so-called biodiesel, will significantly increase the amount of waste glycerol for disposal. One possible route for waste glycerol reuse is to use it as a draw solution in forward osmosis (FO). Glycerol solutions are particularly suited as FO draw solutions due to their high osmotic pressures. In this work, the effects of waste glycerol composition on FO draw solution osmotic pressures, as well as the effects of membrane type and linear flow velocities on FO water and reverse flux, were investigated. Those results indicated the feasibility of using waste glycerol as a draw solution in FO, allowing the reuse of significant amounts of this by-product.

Increased Risk of Aortic Dissection with Perlecan Deficiency

Perlecan (HSPG2), a basement membrane-type heparan sulfate proteoglycan, has been implicated in the development of aortic tissue. However, its role in the development and maintenance of the aortic wall remains unknown. Perlecan-deficient mice (Hspg2−/−-Tg: Perl KO) have been found to show a high frequency (15–35%) of aortic dissection (AD). Herein, an analysis of the aortic wall of Perl KO mice revealed that perlecan deficiency caused thinner and partially torn elastic lamina. Compared to the control aortic tissue, perlecan-deficient aortic tissue showed a significant decrease in desmosine content and an increase in soluble tropoelastin levels, implying the presence of immature elastic fibers in Perl KO mice. Furthermore, the reduced expression of the smooth muscle cell contractile proteins actin and myosin in perlecan-deficient aortic tissue may explain the risk of AD. This study showed that a deficiency in perlecan, which is localized along the elastic lamina and at the interface between elastin and fibrillin-1, increased the risk of AD, largely due to the immaturity of extracellular matrix in the aortic tissue. Overall, we proposed a new model of AD that considers the deficiency of extracellular molecule perlecan as a risk factor.

Interference-free SERS nanoprobes for labeling and imaging of MT1-MMP in breast cancer cells

The expression of membrane type-1 matrix metalloproteinase (MT1-MMP) in cancer cells is critical for understanding the development, invasion and metastasis of cancers. In this study, we devised an interference-free surface-enhanced Raman scattering (SERS) nanoprobe with high selectivity and specificity for MT1-MMP. The nanoprobe was comprised of silver core-silica shell nanoparticle with a Raman reporter tag (4-mercaptobenzonitrile) embedded in the interface. Moreover, the nitrile group in 4-mercaptobenzonitrile shows a unique characteristic peak in the Raman-silent region (1800-2800 cm-1), which eliminates spectral overlapping or background interference in the Raman fingerprint region (500-1800 cm-1). After surface modification with a targeting peptide, the nanoprobe allowed visualization and evaluation of MT1-MMP in breast cancer cells via SERS spectrometry. This interference-free, peptide functionalized SERS nanoprobe is supposed to be conducive to early diagnosis and invasive assessment of cancer in clinical settings.
