Plants with improved nutrient use efficiency are needed to maintain and enhance future crop plant production. The aim of this study was to explore candidate traits for pre-breeding to improve nutrient accumulation and early vigor of spring wheat grown at high latitudes. We quantified shoot and root traits together with nutrient accumulation in nine contrasting spring wheat genotypes grown in rhizoboxes for 20 days in a greenhouse. Whole-plant relative growth rate was here correlated with leaf area productivity and plant nitrogen productivity, but not leaf area ratio. Furthermore, the total leaf area was correlated with the accumulation of six macronutrients, and could be suggested as a candidate trait for the pre-breeding towards improved nutrient accumulation and early vigor in wheat to be grown in high-latitude environments. Depending on the nutrient of interest, different root system traits were identified as relevant for their accumulation. Accumulation of nitrogen, potassium, sulfur and calcium was correlated with lateral root length, whilst accumulation of phosphorus and magnesium was correlated with main root length. Therefore, special attention needs to be paid to specific root system traits in the breeding of wheat towards improved nutrient accumulation to counteract the suboptimal uptake of some nutrient elements.

Cereals grains are the prime component of the human diet worldwide. To promote food security and sustainability, new approaches to non-chemical weed control are needed. Early vigor cultivars with enhanced weed-competitiveness ability are a potential tool, nonetheless, the introduction of such trait in breeding may be a long and labor-intensive process. Here, two image-driven plant phenotyping methods were evaluated to facilitate effective and accurate selection for early vigor in cereals. For that purpose, two triticale genotypes differentiating in vigor and growth rate early in the season were selected as model plants: X-1010 (high) and Triticale1 (low). Two modeling approaches, 2-D and 3-D, were applied on the plants offering an evaluation of various morphological growth parameters for the triticale canopy development, under controlled and field conditions. The morphological advantage of X-1010 was observed only at the initial growth stages, which was reflected by significantly higher growth parameter values compared to the Triticale1 genotype. Both modeling approaches were sensitive enough to detect phenotypic differences in growth as early as 21 days after sowing. All growth parameters indicated a faster early growth of X-1010. However, the 2-D related parameter [projected shoot area (PSA)] is the most available one that can be extracted via end user-friendly imaging equipment. PSA provided adequate indication for the triticale early growth under weed-competition conditions and for the improved weed-competition ability. The adequate phenotyping ability for early growth and competition was robust under controlled and field conditions. PSA can be extracted from close and remote sensing platforms, thus, facilitate high throughput screening. Overall, the results of this study may improve cereal breeding for early vigor and weed-competitiveness.

Direct-seeded rice is one of the solutions against the issues of limited labor and time in the rice cropping system. Improved useful traits, such as fertilizer uptake and anaerobic germination, are needed to increase yield and efficiency in the direct seeding system in rice. Pup1 (Phosphorous uptake1) containing PSTOL1 is useful in improving the phosphate uptake under rainfed/upland conditions. OsTPP7 is the major gene of AG1 (Anaerobic Germination), which shows anaerobic germination. IR64-Pup1-AG1 (I-PA) was developed by pyramiding Pup1 and AG1. Around 20% of the chromosomal segments from the donor remained in I-PA. Phenotypic analysis revealed that I-PA showed better phenotypic performance under low and normal P conditions by enhancing the root system and tiller numbers during the early stage. Significantly better P uptake capacity of I-PA was observed upon a P-supplied soil condition. The coleoptile length and germination rate of I-PA showed tolerance under anaerobic-germinated conditions. PSTOL1 and OsTPP7 were independently expressed under different P conditions of soils, as well as anaerobic conditions. The newly developed breeding lines, I-PA, showed early vigor capacity through a high number of tillers, better P uptake, and germination in low-oxygen conditions. It will be a useful and improved breeding line for direct seeding rice breeding programs.

AbstractChemical weed-control is the most effective practice for wheat, however, rapid evolution of herbicide-resistant weeds threat food-security and calls for integration of non-chemical practices. We hypothesis that integration of GAR dwarfing-genes into elite wheat cultivars can promote early vigor and weed-competitiveness under Mediterranean climate. We develop near-isogenic lines of bread wheat cultivars with GAR dwarfing genes and evaluate them for early vigor and weed-competitiveness under various environmental and management conditions to identify promising NIL for weed-competitiveness and grain yield. While all three NILs, OC1 (Rht8,12), ZC4 (Rht12,13), and BNMF12 (Rht12), responded to gibberellic acid, they exhibited differences in early vigor. Greenhouse and field evaluation highlighted OC1 as a promising line, with significant advantage in early vigor over its parental. To facilitate accurate and continuous early vigor data collection, we applied non-destructive image-based phenotyping approaches which offers non-expensive and end-user friendly solution for selection. NIL OC1 was tested under different weed density level, infestation waves, and temperatures and highlight the complex genotypic × environmental × management interactions. Our findings demonstrate the potential of genetic modification of dwarfing genes as promising approach to improve weed-competitiveness, and serve as basis for future breeding efforts to support sustainable wheat production under semi-arid Mediterranean climate.

Abstract Huanghuazhan (HHZ) and 9,311 are two elite rice cultivars in China. They have achieved high yield through quite different mechanisms. One of the major features that gives high yield capacity to 9,311 is its strong early vigor, i.e., faster establishment of its seedling as well as its better growth in its early stages. To understand the mechanistic basis of early vigor in 9,311, as compared to HHZ the cultivar, we have examined, under controlled environmental conditions, different morphological and physiological traits that may contribute to its early vigor. Our results show that the fresh weight of the seeds, at germination, not only determined the seedling biomass at 10 days after germination (DAG), but was also responsible for ~ 80% of variations in plant biomass between the two cultivars even up to 30 DAG. Furthermore, the 9,311 cultivar had a larger root system, which led to its higher nitrogen uptake capacity. Other noteworthy observations about 9,311 being a better cultivar than HHZ are: (i) Ten out of 15 genes involved in nitrogen metabolism were much more highly expressed in its roots; (ii) it had a higher water uptake rate, promoting better root-to-shoot nitrogen transfer; and (iii) consistent with the above, it had higher leaf photosynthetic rate and stomatal conductance. All of the above identified features explain, to a large extent, why the 9,311, as compared to HHZ, exhibits much more vigorous early growth.

A worldwide food shortage has been projected as a result of the current increase in global population and climate change. In order to provide sufficient food to feed more people, we must develop crops that can produce higher yields. Plant early vigor traits, early growth rate (EGR), early plant height (EPH), inter-node length, and node count are important traits that are related to crop yield. Glycine soja, the wild counterpart to cultivated soybean, Glycine max, harbors much higher genetic diversity and can grow in diverse environments. It can also cross easily with cultivated soybean. Thus, it holds a great potential in developing soybean cultivars with beneficial agronomic traits. In this study, we used 225 wild soybean accessions originally from diverse environments across its geographic distribution in East Asia. We quantified the natural variation of several early vigor traits, investigated the relationships among them, and dissected the genetic basis of these traits by applying a Genome-Wide Association Study (GWAS) with genome-wide single nucleotide polymorphism (SNP) data. Our results showed positive correlation between all early vigor traits studied. A total of 12 SNPs significantly associated with EPH were identified with 4 shared with EGR. We also identified two candidate genes, Glyma.07G055800.1 and Glyma.07G055900.1, playing important roles in influencing trait variation in both EGR and EPH in G. soja.
