Utilization of Porphyridium sp. began to be developed as an anticancer, anti-inflammatory, antibacterial, antifungal, and production of biogas and biodiesel so that necessary to carry out culture activities for ensure continuity of Porphyridium sp. This study aims to determine the type of culture container that showed the best growth response of Porphyridium sp. with the highest biomass. The stages in this research were preparation of containers and media sterilization, making diatom fertilizers and agar media, Culture of Porphyridium sp. on agar media, Culture of Porphyridium sp. in the test tube, and Culture of Porphyridium sp. in glass and plastic containers. The results showed that the growth response of Porphyridium sp. that cultured in glass containers (18,9 ± 0,21 x 105 cells/mL) was higher than plastic (15.57 ± 0,03 x 105 cells/ml).

The purpose of this study was to determine the risk management of oyster mushroom agribusiness in Jember District. Oyster mushroom farming as an alternative to post harvest and can be used for meat and fish substitution foods. In fact there is a factors that are difficult to predict, easy to change and cannot be controlled. Constraints in agricultural excellence such as environmental risk, production risk, supply risk and demand risk. Implementation management needs to be done to minimize the aspects to be disputed. one of the risk management methods is the FMEA method (Failure Mode and Effect Analysis), this method is useful to identify and allow as much as possible. The research method used in literature studies. In mushroom cultivation of white oyster mushroom (P. ostreatus), problems in the contamination during the production are typically found. This contamination is due to the issue in controlling temperature during the media sterilization for growing the fungus (bag-log). This suboptimal sterilization causes unwanted bacteria and spores to grow. To reduce the amount of contamination, the purpose of this study was to demonstrate and evaluate the temperature distribution using one convection pipe with diameter of 6 and 8 cm. The fuel used for this sterilization process is rice husk. Sterilization itself aims to kill other unwanted bacteria and spores on the bag-log. Sterilization was done by traditional steaming bag-log drums arranged in four rows upwards for 6 hours. Convection pipes were built in drums. The steamer was done by performing two retrievals of data. From the experiment results, the use of convection pipe of 8 cm was better than the convection pipe of 6 cm. This is shown from the amount of fungal contamination in convection pipes were less in 8. The result of temperature measurement using dual laser infrared thermometer was also completed and interpreted using Matrix Laboratory with interpolation method to get heat distribution result.

Clinoptilolite has been proven useful in cancer therapy in the last decade. The use of natural zeolite such as clinoptilolite requires some purification of the raw material before it can be applied in biomedical applications, yet microorganism contamination is still viewed as a possible issue for most researchers. Therefore, the clinoptilolite-treated media must undergo filter sterilization process prior to its use in any in vitro research of human cell studies. The aim of this study was to determine whether clinoptilolite cytostatic effect on cervical cancer (HeLa) cells will differ when a different media-sterilization technique is used on the clinoptilolite-treated medium. Comparison of the effect of clinoptilolite-treated medium on cell cytostatic activity was made between three sterilization methods; heat sterilization using an autoclave, UV radiation and filter sterilization. Results showed that there were inhibition of HeLa cell growth after clinoptilolite treatment in all three media-sterilization techniques. Significant differences were shown in all except between sterilizing using UV radiation and filter sterilization. This proved that in vitro cytostatic activity of clinoptilolite on human cervical cancer cells may be affected if different sterilization methods are used to sterilize the pre-treated media. Nonetheless, filter sterilization is still the preferred choice for sterilizing treated-media in cell culture. Keywords: Zeolite, clinoptilolite, cytotoxicity, cancer cell lines, sterilization.
