AbstractIn spite of tremendous advances made in the comprehension of mechanotransduction, implementation of mechanobiology assays remains challenging for the broad community of cell biologists. Hydrogel substrates with tunable stiffness are essential tool in mechanobiology, allowing to investigate the effects of mechanical signals on cell behavior. A bottleneck that slows down the popularization of hydrogel formulations for mechanobiology is the assessment of their stiffness, typically requiring expensive and sophisticated methodologies in the domain of material science. Here we overcome such barriers offering the reader protocols to set-up and interpret two straightforward, low cost and high-throughput tools to measure hydrogel stiffness: static macroindentation and micropipette aspiration. We advanced on how to build up these tools and on the underlying theoretical modeling. Specifically, we validated our tools by comparing them with leading techniques used for measuring hydrogel stiffness (atomic force microscopy, uniaxial compression and rheometric analysis) with consistent results on PAA hydrogels or their modification. In so doing, we also took advantage of YAP/TAZ nuclear localization as biologically validated and sensitive readers of mechanosensing, all in all presenting a suite of biologically and theoretically proven protocols to be implemented in most biological laboratories to approach mechanobiology.

Everyday experiences are the experiences available to shape developmental change. Remarkable advances in devices used to record infants' and toddlers' everyday experiences, as well as in repositories to aggregate and share such recordings across teams of theorists, have yielded a potential gold mine of insights to spur next-generation theories of experience-dependent change. Making full use of these advances, however, currently requires manual annotation. Manually annotating many hours of everyday life is a dedicated pursuit requiring significant time and resources, and in many domains is an endeavor currently lacking foundational facts to guide potentially consequential implementation decisions. These realities make manual annotation a frequent barrier to discoveries, as theorists instead opt for narrower scoped activities. Here, we provide theorists with a framework for manually annotating many hours of everyday life designed to reduce both theoretical and practical overwhelm. We share insights based on our team's recent adventures in the previously uncharted territory of everyday music. We identify principles, and share implementation examples and tools, to help theorists achieve scalable solutions to challenges that are especially fierce when annotating extended timescales. These principles for quantifying everyday ecologies will help theorists collectively maximize return on investment in databases of everyday recordings and will enable a broad community of scholars—across institutions, skillsets, experiences, and working environments—to make discoveries about the experiences upon which development may depend.

Abstract Background It is believed that natural selection acts on the phenotypical changes caused by mutations. Phenotypically, from fishes to amphibians to reptiles, the emergence of limbs greatly facilitates the landing of ancient vertebrates, but the causal mutations and evolutionary trajectory of this process remain unclear. Results We serendipitously obtained a pig of limbless phenotype. Mutations specific to this handicapped pig were identified using genome re-sequencing and comparative genomic analysis. We narrowed down the causal mutations to particular chromosomes and even several candidate genes and sites, such like a mutation-containing codon in gene BMP7 (bone morphogenetic protein) which was conserved in mammals but variable in lower vertebrates. Conclusions We parsed the limbless-related mutations in the light of evolution. The limbless pig shows phenocopy of the clades before legs were evolved. Our findings might help deduce the emergence of limbs during vertebrate evolution and should be appealing to the broad community of human genetics and evolutionary biology.

In Memory and Utopia, Manus O’Dwyer offers a new insight into Valente’s poetics. Contrary to the view that Valente detached his verse from any kind of social or political commitment, O’Dwyer claims that the notions of void and self-negation are key to understand his desire to make his lines reach a broad community and recover the memory of the dead. The author delineates Valente’s poetic career on the basis on the identification between desolation and the Francoist dictatorship. Valente’s verse points at a new nothingness, but not with the selfish aim to enjoy an isolation from reality. Quite on the contrary, in his poems and essays, the non-place or desert, together with other poetic motifs that have been previously analysed from an erotic perspective, allows the poet to portray an impossible community that accepts all those who have been denied participation in the discourse of History. It is only by means of a language that has not been corrupted by the institutional discourse that the poet can draw the map of that utopian, literary space.

Instagram is a free, online social media application that facilitates social networking. Since Instagram is image dependent, educators create visuals accompanied by captions of up to 2,200 characters. By adding specific hashtags to captions, educator posts are curated by the algorithm into a broad community of practice, colloquially known as “Teachergram.” As a technology-facilitated PD tool, Teachergram lends itself to many of the characteristics of high quality and sustainable professional development (PD). In this paper, we focus on how Instagram can be used as a PD tool that supports collaboration, reflection, and feedback loops of educators. We focus on how the unique characteristics of Teachergram lead to the creation of communities and affinity spaces, aligned with existing PD frameworks, built to support the retention of special education teachers and related service providers.

<p>Four instruments mounted at the Telescopio Nazionale Galileo (Roque de Los Muchachos Observatory, La Palma) are currently used to study exoplanets.  They are HARPS-N and GIANO-B, also combined in the new GIARPS configuration, Dolores and Nics. They exploit both the visible and near-infrared domains. I present the scientific highlights obtained in the last years by different teams, since  the access to the TNG is open to the international community through distinct calls. </p>

Phage lytic proteins are a clinically advanced class of novel enzyme-based antibiotics, so-called enzybiotics. A growing community of researchers develops phage lytic proteins with the perspective of their use as enzybiotics. A successful translation of enzybiotics to the market requires well-considered selections of phage lytic proteins in early research stages. Here, we introduce PhaLP, a database of phage lytic proteins, which serves as an open portal to facilitate the development of phage lytic proteins. PhaLP is a comprehensive, easily accessible and automatically updated database (currently 16,095 entries). Capitalizing on the rich content of PhaLP, we have mapped the high diversity of natural phage lytic proteins and conducted analyses at three levels to gain insight in their host-specific evolution. First, we provide an overview of the modular diversity. Secondly, datamining and interpretable machine learning approaches were adopted to reveal host-specific design rules for domain architectures in endolysins. Lastly, the evolution of phage lytic proteins on the protein sequence level was explored, revealing host-specific clusters. In sum, PhaLP can act as a starting point for the broad community of enzybiotic researchers, while the steadily improving evolutionary insights will serve as a natural inspiration for protein engineers.

While tracking-data analytics can be a goldmine for institutions and companies, the inherent privacy concerns also form a legal, ethical and social minefield. We present a study that seeks to understand the extent and circumstances under which tracking-data analytics is undertaken with social licence—that is, with broad community acceptance beyond formal compliance with legal requirements. Taking a University campus environment as a case, we enquire about the social licence for Wi-Fi-based tracking-data analytics. Staff and student participants answered a questionnaire presenting hypothetical scenarios involving Wi-Fi tracking for university research and services. Our results present a Bayesian logistic mixed-effects regression of acceptability judgements as a function of participant ratings on 11 privacy dimensions. Results show widespread acceptance of tracking-data analytics on campus and suggest that trust, individual benefit, data sensitivity, risk of harm and institutional respect for privacy are the most predictive factors determining this acceptance judgement.

Background Broad community acceptance of a COVID-19 vaccination will be critical for effectively halting the spread of the virus. In this study, we focus on factors that differentiate those who are undecided from those who are either willing or unwilling to accept a prospective COVID-19 vaccine. Methods An online survey in May 2020 assessed Australian adults’ willingness to receive a COVID-19 vaccine (yes, maybe, no). A multinomial logistical regression of responses (N = 1,313) was used to identify correlates of vaccine willingness between the three groups. Results 65% were willing to vaccinate, with 27% being in the ‘maybe’ category. Respondents were more likely to be in the ‘maybe’ than the ‘yes’ group when they perceived COVID-19 to be less severe, had less trust in science, were less willing to vaccinate for influenza, and were female. They were more likely to be in the ‘maybe’ than ‘no’ group when they perceived COVID-19 as severe, and less likely to be a hoax, had more trust in science, and greater willingness to vaccinate for influenza. A repeat of the survey in November 2020 with a subset of participants found fewer of them saying yes to the vaccine (56%) and more saying maybe (31%). Conclusions The effectiveness of any COVID-19 vaccine rollout will be reliant on maximizing uptake. The significant number of people who remain undecided about whether or not to get a COVID-19 vaccine, despite the ongoing devastating consequences of the virus for individuals, communities, and economies, is concerning. Our findings aid current research seeking to inform policy regarding how to convince the undecided to vaccinate.
