Reproductive toxicity of triazole fungicides cyproconazole and epoxiconazole when exposed to male and female wistar rats during gametogenesis

Aim. Studying the effect of generic pesticides cyproconazole (98 %) and two samples of epoxiconazole (epoxiconazole 1 — 95,75 % and epoxiconazole 2 — 98,7 %) on the reproductive system of male and female Wistar Han rats at the level of the organism when exposed during gametogenesis, identification and characterization of their hazard, as well as assessment of the risk of reproductive toxicity of these compounds. Materials and Methods. The test samples were administered daily (5 days a week) by oral gavage at doses of 0.2 and 2.0 mg/kg for cyproconazole and 0.5 and 2.0 mg/kg for epoxiconazoles during 11 weeks for males, and 10 weeks for females. Also, there were kept intact males and females, intended for crossover mating with experimental animals. After the end of the exposure, functional indicators of the state of the gonads and the ability of animals to reproduce offspring were studied. The duration and the frequency of each stage of the estrous cycle in female rats and the number of motile sperm, the total amount of sperm and the number of abnormal forms of germ cells of the male rats were studied. The reproductive function state in females was evaluated on day 20th of pregnancy. Thereby the number of corpora lutea in the ovaries, number of alive, dead and resorbed foetuses and embryos, the foetus weight, total weight of litters were registered. The studies were carried out in accordance with the recommendations of the Bioethics Commission and the Centre’s standard operating procedures, developed in accordance with the recommendations and requirements of Good Laboratory Practice (GLP). Conclusions. Test substances at a maximum dose of 2.0 mg/kg of body weight have reproductive toxicity and endocrine-disruptive effect, exerting a significant antiandrogenic effect on males and antiestrogenic effect on female rats. No-observed-adverse-effect-level (NOАEL) for gonadal and reproductive toxicity for male and female Wistar Han rats were established. They are 0.2 mg/kg body weight for cyproconazole and 0.5 mg/kg body weight for epoxiconazole. Правозастосовча практика регулювання свободи наукових медико-біологічних досліджень в європейському союзі на прикладі Німеччини

Мета. На прикладі Німеччини, яка має передову економіку і дієву систему із забезпечення біологічної безпеки та біологічного захисту, дослідити практику правового регулювання в ЄС свобод наукових досліджень, зокрема в медико-біологічній царині. Матеріали та методи. На підставі аналізу законодавства цієї країни проаналізовано підходи ЄС до створення системи нормативно-правового забезпечення даного напрямку на рівні розробки та вдосконалення загальної стратегії з біобезпеки та біозахисту, яка обов'язкова для реалізації в національних програмах держав-учасниць. Створення і набуття чинності конкретних нормативно-правових актів, які детально регламентують порядок проведення медико-біологічних науково-дослідних робіт. У кожній державі-учасниці вони здійснюються індивідуально із урахуванням національних особливостей. Висновки. Запропоновано використання в Україні позитивного досвіду країн-учасниць ЄС із розробки національних програм біологічної безпеки і біологічного захисту та нормативно-правового забезпечення їхньої реалізації при створенні відповідної вітчизняної системи з протидії біотероризму, біологічним диверсіям та іншим загрозам біологічного характеру. Продукти природного опію в сучасній структурі наркотичних отруєнь (огляд літератури та результати скринінгу на наркотики у 1990-2020 рр.)

Незважаючи на стрімке збільшення обсягів споживання синтетичних наркотиків у США, країнах Європейського Союзу та багатьох інших регіонах світу, наркотичні та психоактивні речовини природного походження залишаються актуальним об’єктом дослідження токсикологів. Мета. Аналіз результатів скринінгу на наркотики групи опію за 1990 – 2020 рр. та їхній вплив на формування гострих ненавмисних отруєнь серед дорослого населення м. Києва. Матеріали та методи. Проведено ретроспективний аналіз звітів токсикологічної лабораторії Київської міської клінічної лікарні швидкої медичної допомоги за 1990 – 2020 рр. про результати хіміко-токсикологічного скринінгу на вміст наркотичних речовин у осіб з діагнозом: «Гостре наркотичне отруєння» (МКХ-10:Т40.0-Т40.3). Використано методи: імунохроматографічний аналіз (ІХА), хроматографія у тонкому шарі сорбенту (ТШХ) та газорідинна хроматографія з мас-спектральним детектуванням (ГХ/МС). Результати. За офіційними даними, в Україні у 2019 р. речовини, що стали причиною звернення осіб за медичною допомогою, розподілені наступним чином: опіоїди – 68,53%; канабіноїди – 6,84%; кокаїн – 0,08%; галюциногени – 0,04%; інші наркотики – 24,51%. Серед осіб, померлих внаслідок отруєнь наркотиками, частка опіатів Т40.2 (кодеїн, морфін) становила 16%; опій Т40.0 – 4%; метадон Т40.3 – 23%; інших наркотиків Т40.4 (петидин) – 2%, що разом становить 45%. За результатами хіміко-токсикологічного скринінгу на вміст опіатів встановлено, що першу сходинку за темпом середньорічного зростання посідають: бупренорфін (+7,95%), морфін (+7,6%) і героїн (+6,04%). Зменшення кількості позитивних тестів відбулося у групі опіатів – «ширки» (–9,38%) і алкалоїдів опію (–2,55%). Останні 10 років відбувається прогресивне зростання щорічної кількості позитивних тестів на вміст метадону (+39,3%) при R 2 =8904, що свідчить про стабільність темпу зростання показника. Висновки. Опіати продовжують займати вагомий сегмент у структурі наркотичних речовин, де свої позиції за період 1990 – 2020 рр. посилили бупренорфін, морфін і героїн на фоні послаблення позиції опію («ширки») та алкалоїдів опію; одночасно відбувається швидке збільшення частки напівсинтетичних і синтетичних опіоїдів. Зазначені зміни обумовлюють необхідність постійного удосконалення методів хіміко-токсикологічних досліджень продуктів опію, клінічної діагностики та профілактики пов’язаних з ними отруєнь. Socio-ecological and medical problems of iodine deficiency among the population of Ukraine

Aim. Study of iodine supply in different segments of the population throughout Ukraine, establishment of the morbidity level caused by iodine deficiency, analysis of preventive measures and their effectiveness and development of adequate ways of mass, group and individual prophylaxis of iodine deficiency in the body, which will reduce morbidity in areas of endemic iodine deficiency. The article considers the most important results of long-term epidemiological studies of iodine supply in different categories of the population of almost all regions of Ukraine. Significant volume of work was performed with the support of the Ministry of Healthcare of Ukraine, UNICEF Ukrainian office, and the CDC in Atlanta (USA), which for more than 19 years has provided external quality control of the study of iodine status of the population of Ukraine. The data on iodine content in food and biological fluids of the population affected by the Chernobyl accident are given and the dependence of the irradiation dose of the thyroid on the body supply with stable iodine and on the optimization of the latter after the adoption of the State Programme for Iodine Deficiency Prevention for 2002–2005 is shown. The effectiveness of mass iodine prophylaxis by using universally iodized table salt is emphasized. The article presents data on suggested and implemented ways of mass, group and individual iodine prophylaxis, studies of their effectiveness and shows the positive dynamics of changes in providing the population with dietary iodine. The need for constant consumption of food products with sufficient iodine content, including iodized salt, is proven. Significant changes in the prevalence and incidence of thyroid pathology, socio-ecological and medical benefits of prevention of iodine-dependent disorders are described. The article was prepared as a scientific information material for family doctors, endocrinologists, obstetricians and gynaecologists, hygienists, the public, the Ministry of Healthcare of Ukraine in connection with the need for a legislative solution to the prevention of iodine-dependent disorders in Ukraine. Materials and Methods. To study the problem and assess the state of iodine deficiency in Ukraine the criteria recommended by the WHO and the United Nations Children’s Fund (UNICEF) were applied for the first time, a method for determining the iodine content in the body was developed and implemented, which allowed to join the international “Equip” system of iodine supply control in the world and in Ukraine. This made it possible to move from indirect to direct methods of determining the intake of iodine in the body. Conclusions. A detailed study of iodine supply in different segments of the population of Ukraine proved the negative effects of iodine deficiency on health; in particular, it was found that the overall decrease in productivity of the generation of children in iodine deficiency populations is 5%. Measures to eliminate iodine-dependent disorder among the population of Ukraine have been developed and implemented. The development and production of iodized food salt, adequate to the needs of the population, has been carried out. The result of these measures, developed and implemented to eliminate iodine-dependent disorders and to improve the nation’s intelligence, is, according to the official data, the improvement of iodine supply of the population, significant reduction in the number of thyroid disorders, especially among those affected with 131I, which, even just because of this indicator, has significant social-economic effect. Natural opium products in the modern structure of drug poisoning (literature review and results of drug screening for 1990-2020)

Despite the rapid increase in synthetic drug use in the United States, the European Union, and many other parts of the world, narcotic and psychoactive substances of natural origin remain relevant to toxicologists. Aim. To analyze the results of drug screening of opium drugs in 1990-2020 and their impact on the formation of acute unintentional poisoning among the adult population of Kyiv. Materials and Мethods. A retrospective analysis of the reports of the toxicological laboratory of the Kyiv City Clinical Emergency Hospital for 1990-2020 on the results of chemical-toxicological screening for drug content in persons diagnosed with "Acute drug poisoning" (ICD-10: T40.0-T40.3). Methods used: Immuno-chromatographic analysis (ICA), thin layer chromatography (TLC) and gas-liquid chromatography with mass spectral detection (GC/MS). Results. According to official data in Ukraine in 2019, the substances that caused people to seek medical help are distributed as follows: opioids – 68.53%; cannabinoids – 6.84%; cocaine – 0.08%; hallucinogens – 0.04%; other drugs – 24.51%. Among those who died as a result of drug poisoning, the proportion of opiates T40.2 (codeine, morphine) was 16%; opium T40.0 – 4%; methadone T40.3 – 23%; other drugs T40.4 (pethidine) – 2%, which together is 45%. According to the results of chemical-toxicological screening for opiate content, it was found that the first step in the average annual growth rate is occupied by: buprenorphine (+7.95%), morphine (+7.6%) and heroin (+6.04%). The number of positive tests decreased in the group of opiates –"shirka" (–9.38%) and opium alkaloids (–2.55%). Over the last 10 years, there has been a progressive increase in the annual number of positive tests for methadone content (+39.3%) at R 2 =8904, which indicates a high stability of the growth rate. Conclusions. Opiates continue to occupy an important segment in the structure of drugs, where their positions for the period 1990-2020 strengthened buprenorphine, morphine and heroin against the weakening of the position of opium ("shirka") and opium alkaloids; at the same time there is a rapid increase in the proportion of semisynthetic and synthetic opioids. These changes necessitate continuous improvement of methods of chemical and toxicological studies of opium products, clinical diagnosis and prevention of related poisonings.

Modeling of content of biologically active hepatoprotective substances in the composition of natural honey

Aim. The search for ways to create hepatoprotective food additives based on biologically active substances of plant origin and bee honey is an important area of research in the field of biology and medicine. Materials and Methods. The material for the research was bees of Ukrainian breed (Apis mellifera sossimai). The process of enriching the natural properties of natural honey was carried out in a natural way through the bee family by feeding colonies of bees with a special composition of natural origin components based on Avéna satíva and Silibum marianum. Sieve honey with monofloral honey from white acacia (Robinia pseudoacacia) 55.0–55.0% was used for feeding of the bee families. The first research group was exclusively fed honey water (control). For the second and third experimental groups, Avena Sativa and Milk Thistle Dry Extract were introduced into the main feed, respectively. Preparation of honey water was carried out on distilled water. Each group of bee families was evaluated according to the complex of biological and economic-useful features during the whole study period. The concentrations of active substances in the obtained batches of honey were determined. Ferulic acid was chosen as the active substance for oat, sum of Silibinin A and Silibinin B for milk thistle. The mass concentration of ferulic acid was measured according to European Standards EN 15662:2008 using Gas Chromatography-Tandem Mass Spectrometry following acetonitrile extraction/partitioning and clean-up by dispersive solid phase extraction (Quechers) method. The limits of quantitation of ferulic acid and sum of Silibinin A and Silibinin B were 0.02 mg/kg. The high content (83.3±0.4 mg/kg) of Silibinin flavonoids (A and B), Isosilibin (A and B), Silichristin A, etc. in honey persists. Instead, ferulic acid precipitated in fairly low concentrations of honey (1.13±0.02 mg/kg). Conclusions. The obtained results indicate that the extract of milk thistle is promising for the creation of enriched natural honey with hepatoprotective properties, as the biologically active components of this plant have characteristics that allow to obtain the final product with a high content of active substances. Testing methods for the assessment of chemical neurotoxic effects on the developing organisms in pre- and postnatal period

Aim of the research. Analysis of approaches to the assessment of neurotoxic effects of chemicals during ontogenesis. The dangerous tendency of the increase in the incidence of pathology of the nervous system in the child and the mother, who during pregnancy was exposed to neurotoxicants, necessitates the protection of the child’s body from such a negative effect of chemicals. One of the possible preventive ways to solve this problem is screening of xenobiotics before their widespread application, as well as identification of their ability to cause neurotoxic effects on the body during its development, and banning substances, neurotoxic effects of which on the development of progeny are proven. Materials and Methods. Detection of adverse effects at the stage of foetal development and then at the beginning of the functioning of a nervous system in the postnatal period requires complex experimental studies in laboratory animals, which are described in the recommendations of OECD guideline 426 (OECD Guideline for Testing of Chemicals; Guideline 426: Developmental Neurotoxicity Study, 2007). The OECD guideline 426 protocol provides for a wide range of methods for assessing sensory, motor, behavioural and cognitive functions. Conclusions. Analysis of data given in publications and methodological approaches to research and evaluation of neurotoxic effects of chemicals on the developing organism showed that to obtain representative results it is important to take into account all factors that may affect the result, select adequate informative tests and comply with all requirements. Law enforcement practice of regulating freedom of scientific biomedical research in the european union on the example of Germany

Aim. Using the example of Germany, which has an advanced economy and an effective system for ensuring biological safety and biological protection, to study the practice of legal regulation of freedom of scientific research in the EU, in particular in the medical and biological field. Materials and Methods. Based on the analysis of the legislation of this country, the EU approaches to the creation of a system of regulatory support for this area at the level of development and improvement of the general strategy on biosafety and bioprotection, which is mandatory for implementation in the national programs of the member states, have been analysed. The creation and entry into force of specific regulatory legal acts which in detail administer the procedure for conducting biomedical research work is carried out individually in each participating state, considering national characteristics. Conclusions. It is proposed for Ukraine when creating an appropriate domestic system to counter bioterrorism, biological sabotage, and other biological threats to use both the positive experience of the EU member states in the development of national programs of biological safety and biological protection as well as regulatory support for their implementation. Соціально-екологічна та медична проблеми нестачі йоду серед населення України

Мета. Вивчення йодного забезпечення різних верств населення на всій території України, встановлення рівня захворюваності, зумовленого нестачею йоду, стан профілактичних заходів, їх ефективність та розробка адекватних шляхів масової, групової та індивідуальної профілактики йододефіциту в організмі сприятиме зниженню рівня захворюваності населення ендемічних на йод регіонів. У статті розглянуто найважливіші результати багаторічних епідеміологічних досліджень вивчення йодного забезпечення різних категорій населення практично всіх регіонів України. Значний об'єм цієї роботи виконано за підтримки МОЗ України, представництва Дитячого фонду ЮНІСЕФ в Україні, а також центру СDC в Атланті (США), який впродовж понад 19 років здійснював зовнішній контроль за якістю вивчення йодного статусу населення України. Наведено дані щодо вмісту йоду в харчуванні та біологічних рідинах населення регіонів, постраждалих внаслідок Чорнобильської катастрофи та показано залежність дози опромінення щитоподібної залози від забезпечення організму стабільним йодом і оптимізації вмісту останнього в організмі людей після прийняття Державної програми профілактики йодної недостатності населення на 2002–2005 роки. Підкреслено ефективність масової йодної профілактики шляхом вживання універсально йодованої харчової солі. У статті представлені дані про запропоновані та впроваджені шляхи масової, групової та індивідуальної йодопрофілактики, вивчена їх ефективність і показана позитивна динаміка змін при забезпеченні населення аліментарним йодом, а також доведено необхідність постійного вживання продуктів із достатнім вмістом аліментарного йоду, зокрема йодованої кухонної солі. Описано суттєві зміни щодо поширеності та захворюваності населення на тиреоїдну патологію, соціально-екологічні та медичні переваги профілактики йодозалежних захворювань. Стаття підготовлена як науковий інформаційний матеріал для сімейних лікарів, ендокринологів, акушеров-гінекологів, гігієністів, громадськості, МОЗ України у зв'язку з необхідністю законодавчого вирішення проблеми профілактики йодозалежних захворювань в України. Матеріали та методи. Для вивчення проблеми та оцінки стану нестачі йоду на території України вперше застосовано критерії, рекомендовані ВООЗ та дитячого фонду ООН (ЮНІСЕФ), розроблено та впроваджено методику визначення вмісту йоду в організмі, що дозволило приєднатися до міжнародної системи "Equip" контролю за станом йодного забезпечення у світі та в Україні. Це надало можливість перейти від опосередкованих до прямих методів визначення надходження йоду в організм. Висновки. Детальне вивчення йодного забезпечення різних верств населення України довело, що дефіцит йоду має негативні наслідки для здоров'я, зокрема встановлено, що загальне зниження продуктивності покоління дітей у йододефіцитних популяціях становить 5%. Розроблено та запроваджено заходи щодо ліквідації йодозалежних захворювань серед населення України. Здійснено розробку та започатковано випуск харчової йодованої солі, адекватної потребам населення. Наслідком розроблених та впроваджених заходів щодо ліквідації йодозалежних захворювань стане поліпшення інтелекту нації. Покращення йодної забезпеченості населення, за офіційними даними, суттєво знизить кількість захворювань щитоподібної залози, особливо серед осіб, які зазнали впливу 131 І і що має значний соціально-економічний ефект. Water-soluble vitamins: their physiological significance, role in life of humans and their content in the nutrition of the population suffered as a result of Chernobyl accident

Aim of the Research. Generalization of information about the properties and history of discovery of water-soluble vitamins, their up-to-date classification, sources of their intake and daily consumption by people of different ages and for people affected by the Chernobyl accident and living in environmentally hazardous areas. The article describes the results and data of long-term studies of the actual consumption of water-soluble vitamins by the population living in radioactively contaminated areas as a result of the Chernobyl accident. Materials and Methods. The presented data of the academic literature on the history of discovery of vitamins reveal their main characteristics and significance for the functioning of the human body. The information about physiological needs and the main sources of vitamins intake are presented. Conclusions. The article is relevant for family doctors, hygienists, nutritiologists, nutritionists and others as a scientific information material on the prevention and correction of vitamin deficiency.
