The raspberry tree, a species from the Rosaceae family and the Rubus genus, is included in the group of small fruit that has been drawing the attention of the global consuming market for carrying antioxidant properties and substances capable of fighting free radicals, bringing health benefits. The objective of this work was to evaluate the effect of collection periods on the in vitro establishment of raspberry trees (Rubus idaeus L.). Explants were collected in different seasons of the year (winter, spring, summer and autumn), and were inoculated in a MS culture medium. The experimental design was totally randomized, and arranged in a factorial scheme. At 28 days, the following variables were evaluated: oxidation, fungal and bacterial contamination survival and establishment. There was less phenolic oxidation in raspberry tree explants collected in winter. Explants contamination by endogenic bacteria occurred in higher percentages in winter. Although summer was the season of the year with higher percentage of explants’ phenolic oxidation for both cultivars, the highest in vitro establishment percentages for the Heritage cultivar occurred in summer and fall. However, for the Fallgold cultivar, spring and winter were the seasons with greater in vitroestablishment.

Microshoot and callus cultures of Cornus florida (flowering dogwood), which were grown on woody plant medium amended with BA, were inoculated with Microsphaera pulchra (an obligate plant parasite) by gently shaking infected leaves bearing numerous conidia over the tissue. Culture dishes were sealed with parafilm and incubated at 24 °C with 25 mol·m–2·s–1 provided by cool fluorescent bulbs for 15 h. Cultures were examined with a dissecting scope every 24 h and cultures transferred when contaminating fungi were present. Specimens were prepared light microscopy and SEM. The fungus infected individual callus cells, but did not sporulate. In contrast, powdery mildew was well-established (both primary and secondary hyphae) in 70% of the microshoot cultures after 6 days and sporulated on 20% by 7 to 8 days. The cellular relationship between host and pathogen in vitro was similar to that found in greenhouse-grown plants. This technique has possible applications in maintaining fungal culture collections and studying host–pathogen relationships under more stringently controlled conditions.

Telangiectatic osteosarcoma (TOS) is an aggressive variant of osteosarcoma (OS) with distinctive radiographic, gross, microscopic features, and prognostic implications. Despite several studies on OS, we are still far from understanding the molecular mechanisms of TOS. In recent years, many studies have demonstrated not only that microRNAs (miRNAs) are involved in OS tumorigenesis, development, and metastasis, but also that the presence in high-grade types of OS of cancer stem cells (CSCs) plays an important role in tumor progression. Despite these findings, nothing has been described previously about the expression of miRNAs and the presence of CSCs in human TOS. Therefore, we have isolated/characterized a putative CSC cell line from human TOS (TOS-CSCs) and evaluated the expression levels of several miRNAs in TOS-CSCs using real-time quantitative assays. We show, for the first time, the existence of CSCs in human TOS, highlighting the in vitro establishment of this unique stabilized cell line and an identification of a preliminary expression of the miRNA profile, characteristic of TOS-CSCs. These findings represent an important step in the study of the biology of one of the most aggressive variants of OS and the role of miRNAs in TOS-CSC behavior.

ABSTRACT: The objective of this study was to evaluate the in vitro establishment of 'Xavante' blackberry under different conditions of plant luminosity and concentrations of salts of MS medium. The maintenance of 'Xavante' blackberry plants in the absence of light, with the use of MS medium with salt concentrations of 75% and 100% reduce bacterial contamination and the percentage of explants oxidized and provide the highest percentage of surviving and established explants. The 50% salt dilution in MS medium is not recommended when plants are subjected to the absence of light due to the high percentage of oxidized explants and consequently lower survival and establishment.

ABSTRACT: The goal of the present study was to evaluate the germination, initial growth, and in vitro co-cultivation of Comanthera curralensis Moldenke, a "sempre viva" native of the Chapada Diamantina state of Bahia. Full strength (MS) and half-strength MS (MS1/2) growth media supplemented with two different sucrose concentrations (15 and 30g L-1) were tested for germination and initial plant growth. Three different plant densities were tested by in vitro culture (8, 10 and 12 plants per container). MS1/2 medium with 15g L-1 sucrose resulted in a higher percentage of germination and plant growth for the in vitro establishment of C. curralensis. The use of 12 plants per container is indicated for cost reduction in C. curralensis in vitro production.

The goal of this study was to investigate in vitro multiplication protocols for use with red currant cultivars grown in the Czech Republic. Cultivars Detvan, Vitan and Rotte Höllandische were successfully established in vitro using mercuric chloride in a concentration of 0.15% as a sterilization solution. The overall rate of contamination was 25.7%. Two proliferation media Murashige and Skoog medium (MS) and McCown woody plant medium (WPM) containing 1 or 2 mg/l of 6-benzylaminopurine (BAP) were tested. Initial explants produced new plants in the form of rosettes. Rosettes arose from the base of the initial explants in the form of adventitious bud formation. The shoot number was relatively low and varied between 1.0 and 2.1. Generally, the highest number was obtained for cultivar Rotte Holländische that produced 2.1 ± 0.1 new rosettes on MS medium containing lower concentration 1 mg/l BAP. In contrary, Vitan cv. had significantly lower shoot number ranging from 1.0 to 1.3. WPM medium with a lower concentration of mineral salts proved to be unsuitable for the multiplication of tested cultivars.

Bamboo species are an alternative for the composition of forest plantations. However, their potential has not been explored due to the hard time in producing large-scale clonal plants. Thus, the aim this work was to evaluate the in vitro establishment, bud multiplication and ex vitro rooting of Bambusa vulgaris. The first experiment tested different systemic and contact fungicide solutions, based on exposure time, during the establishment phase. Established explants were subjected to evaluation of residual fungicide effect on subcultures during the multiplication and elongation phases. The second experiment evaluated the influence of activated carbon on ex vitro survival and on adventitious rooting. Explant immersion in liquid culture medium added with 1.0 mL of fungicide for 120 hours has favored the in vitro establishment and reduced fungal contamination. On the other hand, it favored the shoot emission of shoots per explant during the multiplication phase. Both rooting induction culture medium and mini-incubator system use were effective in enabling adventitious root formation. The presence of activated carbon in the rooting induction culture medium resulted in a higher clonal plant survival rate.

The barueiro (Dipteryx alata Vog.) is a native fruit species of the Cerrado ecoregion that has multiple uses. It is a wild species, and its cultivation is difficult. Furthermore, it is threatened with extinction. Plant tissue culture is a major tool for the conservation of germplasm, as well as a means of propagating high-quality seedlings on a large scale. However, this technique has not been used with barueiro, although it might provide valuable contributions to the process of barueiro domestication. The most popular method of cultivation is the use of the Murashige and Skoog medium (MS), which is considered one of the most nutritionally complete media. Woody plant medium (WPM) is indicated for the propagation of woody species, but there are no reports of its use for barueiro cultivation. Woody plants tend to have problems with rust in vitro during the establishment phase. Activated charcoal acts as an adjuvant for the adsorption of phenolic compounds, mitigating its effects in the medium. Thus, the objective of this study was to test four activated charcoal doses (0, 2, 4 and 6 g L-1) and three culture media: MS, WPM, and AA (over water agar) in the in vitro establishment of barueiro. The experimental design was a completely randomised (DIC), 4 × 3 factorial design with three replications. At 60 days after inoculation, the explants were evaluated for dry matter, fresh weight, stem diameter, shoot length, number of leaves, longest root length, germination rate, and chlorophyll contents. The MS medium supplemented with 3,0 g L-1 activated charcoal appeared to be the best for in vitro establishment of barueiro.

Red Araçá’s (Psidium cattleianum) micropropagation processes have shown enormous potential both in terms of research and as a sustainable native resource to be used in the areas of food production, ecology, and pharmacology. Currently, however, despite that potential, research efforts involving this myrtaceae, native to the Brazilian Atlantic Forest, have been scarce. With that in mind, this study set out to establish micropropagation techniques that would allow the development of a feasible protocol to be used with Red Araçá, achieving its mircropropagation from in vitro germinated seeds. Different types of explants were tested for in vitro establishment. For the multiplication of nodal segments, different concentrations of BAP and IAA combinations were tested in an MS medium. Using the same medium, different concentrations of ampicillin were applied in order to determine its influence on the decontamination of the apical segments. The BAP and IAA combinations were also used to test their effects on the in vitro explants’ development and rooting. During pre-acclimatization, survival of in vitro rooted plants was tested in a nebulizer chamber, using a commercial substrate and that same substrate mixed with washed sand (1:1). In essence, it was indeed possible to develop a complete protocol for the micropropagation of the Red Araçá from seedlings obtained by in vitro germination. The in vitro introduction of the Red Araçá was rather efficient, independently of the type of explants used. As the BAP and IAA concentrations increased, so did the in vitro seedlings’ development (7 leaves explant-1) and rooting (67%). Additionally, the in vitro rooted plants exhibited a high rate of survival (80%) in the pre-acclimatization phase, independently of the substrate used.
